Abstract

This study uses anthropological and forensic medical analyses to determine the cause of fractures found in the remains of 15 individuals buried at a site associated with the Globular Amphora Culture (2875-2670 BC). The intent was to determine the mechanism underlying the injuries and to indicate the types of tools that might have inflicted the blows. The fractures were diversified in their forms, but the majority of the injuries appear to have been inflicted by a flint axe, which is frequently found in graves of the Globular Amphora Culture. Apart from the forearm being severed in one of the victims, all the remaining skeletons showed from 1 to 4 injuries involving solely the skulls. The grave might contain victims attacked by invaders who executed the captives, or else the feature is ritual in character and it reflects the beliefs of the Neolithic community.

Abstract

This is the dataset of imputed downsampled ancient genomes that we used to assess imputation accuracy of ancient human genomes (for more information, please read our manuscript "Imputation of ancient human genomes"). Abstract

In the area of the Małopolska Upland there are known eleven sites with graves of the Bell Beaker culture. A new single grave of this culture was discovered in 2019, at Site 3 in Kazimierza Wielka. The grave inventory consisted of three vessels. The remains of a male aged 40-50 years were discovered in this feature. Analysis of stable carbon and nitrogen isotopes in the bones of this individual indicate a terrestrial diet based on C3 plants supplemented with animal protein. Signatures of strontium isotopes suggest that the man may have spent his childhood in the area of the Pińczów Hummock. The date 3885±35 BP was obtained from the bones of this person (Poz-134513), which can be calibrated with a probability of 60.8% to the years 2456-2340 BC. The conducted modelling of the obtained AMS dates from Bell Beaker culture graves allows us to establish a range of the presence of this culture in the Małopolska Upland for the years 2400/2350-2200 BC. Abstract

Due to postmortem DNA degradation, most ancient genomes sequenced to date have low depth of coverage, preventing the true underlying genotypes from being recovered. Genotype imputation has been put forward to improve genotyping accuracy for low-coverage genomes. However, it is unknown to what extent imputation of ancient genomes produces accurate genotypes and whether imputation introduces bias to downstream analyses. To address these questions, we downsampled 43 ancient genomes, 42 of which are high-coverage (above 10x) and three constitute a trio (mother, father and son), from different times and continents to simulate data with coverage in the range of 0.1x-2.0x and imputed these using state-of-the-art methods and reference panels. We assessed imputation accuracy across ancestries and depths of coverage. We found that ancient and modern DNA imputation accuracies were comparable. We imputed most of the 42 high-coverage genomes downsampled to 1x with low error rates (below 5%) and estimated higher error rates for African genomes, which are underrepresented in the reference panel. We used the ancient trio data to validate imputation and phasing results using an orthogonal approach based on Mendel’s rules of inheritance. This resulted in imputation and switch error rates of 1.9% and 2.0%, respectively, for 1x genomes. We further compared the results of downstream analyses between imputed and high-coverage genomes, notably principal component analysis (PCA), genetic clustering, and runs of homozygosity (ROH). For these three approaches, we observed similar results between imputed and high-coverage genomes using depths of coverage of at least 0.5x, except for African genomes, for which the decreased imputation accuracy impacted ROH estimates. Altogether, these results suggest that, for most populations and depths of coverage as low as 0.5x, imputation is a reliable method with potential to expand and improve ancient DNA studies.

Abstract

This article presents an inhumation burial of a young woman from a biritual Early Roman Iron Age cemetery in Kazimierza Wielka, site 12, western Lesser Poland. The text draws attention to the furnishing of buried woman unusual of the Przeworsk Culture environment. She was furnished with ornaments and parts of clothing that have analogies in the Eastern Zone of the Przeworsk Culture and in the Wielbark Culture. Additionally, the funerary rites observed were not characteristic of the area, indicating the foreign origin of the buried woman. The article also draws attention to a further 17 inhumations from this site, as well as other newly discovered cemeteries of the Przeworsk Culture located a short distance from Kazimiera Wielka, where inhumations burials have also been discovered.
