Managed honey bee populations have fluctuated over the past several decades in the U.S. While a single factor has not been identified for these losses, the interaction between multiple biotic and abiotic stressors have been suggested to be responsible. Of major concern are several invasive parasite and pathogen species as well as colony management. A single honey bee colony often suffers from multiple harmful agents, that may act synergistically and cause greater declines in bee health. We conducted a survey to detect known and lesser-known honey bee parasites and pathogens. While previous research has primarily focused on commercially managed colonies, research is limited to pertaining parasite and pathogen prevalence in hobbyist managed colonies. Molecular diagnostics were used to screen 541 Arkansas (AR) honey bee colonies from 107 hobbyist beekeepers for eight A. mellifera parasite and pathogen species. Colony samples were obtained between 2015–2016 and represented forty-seven of the seventy-five AR counties. Vairimorpha ceranae (11.6% occurrence) and parasite Varroa destructor (49.4% occurrence) were relatively common in AR hobbyist colonies. Interestingly, the lesser-studied pathogenic trypanosome species, Lotmaria passim, was detected in 11.3% of the colonies and widespread in twenty of the forty-seven counties sampled. None of the honey bee pathogens Vairimorpha apis, Spiroplasma apis, S. melliferum, Crithidia mellificae, or the parasitic phorid fly, Apocephalus borealis, were detected in the colonies sampled. This study provides an extensive assessment of the parasite and pathogen species occurring at the AR state-level in hobbyist-managed honey bee colonies.

A study was conducted on the mitochondrial DNA genetic diversity of feral colonies and swarms of Apis mellifera from ten counties in Utah by sequencing the intergenic region of the cytochrome oxidase (COI-COII) gene region. A total of 20 haplotypes were found from 174 honey bee colony samples collected from 2008 to 2017. Samples belonged to the A (African) (48%); C (Eastern Europe) (43%); M (Western Europe) (4%); and O (Oriental) lineages (5%). Ten African A lineage haplotypes were observed with two unique to Utah among A lineage haplotypes recorded in the US. Haplotypes belonging to the A lineage were observed from six Utah counties located in the southern portion of the State, from elevations as high as 1357 m. All five C lineage haplotypes that were found have been observed from queen breeders in the US. Three haplotypes of the M lineage (n=7) and two of the O lineage (n=9) were also observed. This study provides evidence that honey bees of African descent are both common and diverse in wild populations of honey bees in southern Utah. The high levels of genetic diversity of A lineage honey bee colonies in Utah provide evidence that the lineage may have been established in Utah before the introduction of A lineage honey bees from Brazil to Texas in 1990.

Honey bee populations in the United States have been under stress for the past decade. Although no one cause has been able to explain this, recent studies have shown that multiple interactions among microorganisms may be behind honey bee decline. Several internal parasites may contribute to this, including the trypanosome Lotmaria passim and the microsporidian, Nosema ceranae. This study combined novel data with data from several previous molecular diagnostic surveys for L. passim and N. ceranae to determine co-infection rates of these honey bee parasites. A total of 782 honey bee samples from Arkansas, Oklahoma, Texas, Utah, Mississippi, and Hawaii were analyzed for co-occurrence of N. ceranae and L. passim. Of the 782 samples, 37 (4.7%) were co-infected with both N. ceranae and L. passim, while 110 (14.1%) were infected with N. ceranae alone, and 51 (6.5%) with L. passim alone. Co-infections occurred most frequently in honey bees from Hawaii (11.8%), followed by Utah (2.4%) and Arkansas (1.2%), and no co-infections were observed in samples from Mississippi, Oklahoma, or Texas. Statistical analysis revealed that N. ceranae and L. passim co-occur more frequently than expected in samples from Hawaii and Utah. Infection rates for N. ceranae, L. passim, and co-infections were similar between managed (n = 393) and swarms (n = 73) but were both greater than feral (n = 316) honey bee colonies. Abstract Background In the Eastern and Upper Midwestern regions of North America, Ixodes scapularis (L.) is the most abundant tick species encountered by humans and the primary vector of B. burgdorferi, whereas in the southeastern region Amblyomma americanum (Say) is the most abundant tick species encountered by humans but cannot transmit B. burgdorferi. Surveys of Borreliae in ticks have been conducted in the southeastern United States and often these surveys identify B. lonestari as the primary Borrelia species, surveys have not included Arkansas ticks, canines, or white-tailed deer and B. lonestari is not considered pathogenic. The objective of this study was to identify Borrelia species within Arkansas by screening ticks (n = 2123), canines (n = 173), and white-tailed deer (n = 228) to determine the identity and locations of Borreliae endemic to Arkansas using PCR amplification of the flagellin (flaB) gene. Methods Field collected ticks from canines and from hunter-killed white-tailed were identified to species and life stage. After which, ticks and their hosts were screened for the presence of Borrelia using PCR to amplify the flaB gene. A subset of the positive samples was confirmed with bidirectional sequencing. Results In total 53 (21.2%) white-tailed deer, ten (6%) canines, and 583 (27.5%) Ixodid ticks (252 Ixodes scapularis, 161 A. americanum, 88 Rhipicephalus sanguineus, 50 Amblyomma maculatum, 19 Dermacentor variabilis, and 13 unidentified Amblyomma species) produced a Borrelia flaB amplicon. Of the positive ticks, 324 (22.7%) were collected from canines (151 A. americanum, 78 R. sanguineus, 43 I. scapularis, 26 A. maculatum, 18 D. variabilis, and 8 Amblyomma species) and 259 (37.2%) were collected from white-tailed deer (209 I. scapularis, 24 A. maculatum, 10 A. americanum, 10 R. sanguineus, 1 D. variabilis, and 5 Amblyomma species). None of the larvae were PCR positive. A majority of the flaB amplicons were homologous with B. lonestari sequences: 281 of the 296 sequenced ticks, 3 canines, and 27 deer. Only 22 deer, 7 canines, and 15 tick flaB amplicons (12 I. scapularis, 2 A. maculatum, and 1 Amblyomma species) were homologous with B. burgdorferi sequences. Conclusions Data from this study identified multiple Borreliae genotypes in Arkansas ticks, canines and deer including B. burgdorferi and B. lonestari; however, B. lonestari was significantly more prevalent in the tick population than B. burgdorferi. Results from this study suggest that the majority of tick-borne diseases in Arkansas are not B. burgdorferi.
