Abstract

Healthy and nutritious food is a basic need of humans; therefore, the food industry is striving to develop healthy food products with fewer calories and improved nutritional quality. Nowadays, one of the major challenges to the food industry is developing novel wheat-free products, especially for celiac patients, as people are gluten intolerant to this disease. Celiac disease is spreading day by day, and it affects 1-2% of the population all over the world. Thus, the food industry needs to use new sustainable wheat alternatives for the development of bakery products. Therefore, the present study is designed to develop gluten-free muffins from plant-based sustainable raw materials such as maize, sorghum, and chickpea. Maize flour (50, 60, and 70%), sorghum flour (20, 25, 30, 40, and 50%), and chickpea flour (20, 25, 30, 40, and 50%) were utilized to prepare composite flour. The composite flour was then analyzed for proximate composition and rheological characteristics. The findings revealed that different flour blend compositions had significantly affected flour's nutritional profile and rheological properties. Afterward, gluten-free muffins were developed and examined for proximate composition, mineral, color, texture, and sensory assessment. The results showed a significant increase in protein (30.8%), fiber (5.31%), ash (2.93%), Cu (0.35 mg/L), Mg (1.23mg/L), Fe (1.30mg/L) content and decrease in moisture (9.20%), NFE (15.9%), and Cd (0.10mg/L) content of gluten-free muffins as compared to wheat muffins. Moreover, during the storage study, an increase in texture hardness (30.9 to 34.3%) value and a decrease in moisture (9.72 to 6.59%) value for developed muffins was observed from day 1 to day 21. The crust color (l*, a*, b*) values and sensory score decrease significantly as compared to control. Conclusively, maize (60%), sorghum (20%), and chickpea (20%) act as appropriate combinations of flour to prepare gluten-free muffins with excellent nutritional value, color attributes, and consumer acceptance. Abstract

Objective: Posaconazole(POS) is an antifungal drug widely used in treatment of fungal infections. However, the drug has very low oral bioavailability due to high first-pass metabolism, thereby limiting its therapeutic effectiveness. The present study is aimed to prepare nanoemulsion of posaconazole to increase its solubility and to develop posaconazole-loaded nanoemulsion based gel for single dose topical administration to sustain the release of the drug, improve patient compliance and avoid repeated administration. Method:-FTIR-study was performed to determine compatibility of drug with excipients. Pseudoternary-phase diagram was constructed to optimize the Smix ratio. Optimization of nanoemulsion was done by Box-Behnken Design. The optimized formulation was evaluated for particle-size (nm), polydispersity index (PDI), (%) drug content and in-vitro drug release. POS-nanoemulgel (FNG) was developed using carbopol-934P (1%), which was evaluated for homogeneity, pH, viscosity, spreadability, percentage-yield and in-vitro drug release. The anti-fungal activity of POS-nanoemulgel was evaluated against Candida Albicans. Results: The FTIR-study confirmed that there was no interaction between drug and excipients. The optimized selected nanoemulsion (FN1) exhibited particle size 79.19nm, PDI 0.357, drug content 95.90% and in-vitro release of 79.40% at 24 hour. The prepared nanoemulgel was homogenous, opaque with good spreadability 13.91±1.19, viscosity 3760±0.014, pH 5.6±0.06 and the zeta-potential was -29mV, indicating good stability. The drug release at 24h was 72.01%. Further, the developed nanoemulgel formulation exhibited better antifungal activity in comparison to the pure drug gel. Conclusion: Hence, it can be concluded that nanoemulgel of posaconazole can provide better antifungal activity and improve patient compliance. Abstract

Bovine serum albumin (BSA) nanoparticles (BSA-MTX-CUR-NPs) encapsulating methotrexate (MTX) and curcumin (CUR) was developed with an aim to co-deliver the drugs at the inflamed joint so as to maximize the therapeutic efficacy and alleviate toxic side effects associated with MTX. Nanoparticle albumin-bound technology was used to formulate nanoparticles, followed by characterization for its particle size, polydispersity index, encapsulation efficiency, zeta potential, surface morphology, in-vitro drug release and drug release kinetics. Further, we investigated the pharmacokinetics and pharmacodynamics of the developed nanoparticles in the adjuvant-induced arthritis model. BSA-MTX-CUR-NPs exhibited particle size of 163.05 ± 1.708 nm, polydispersity index of 0.195 ± 0.0024 and % encapsulation efficiency of 68.23 ± 0.640% for MTX and 75.71 ± 0.216% for CUR with controlled release pattern for both the drugs. The scanning electron microscopy revealed nanoparticles exhibited a spherical shape. DSC study confirmed the absence of incompatibility between the drugs and the excipients. Half-life and area under the curve were significantly higher for MTX in the nanoparticulate form in comparison to free MTX. Pharmacodynamic studies revealed that BSA-MTX-CUR-NPs possessed better disease-modifying effects in comparison to free MTX. Hence, it can be concluded that albumin nanoparticles constitute a viable method for delivering MTX and CUR to inflamed joints simultaneously, because of the strong affinity of albumin and enhanced permeability and retention effect at the inflamed joint. Abstract

Blow Molding is one of the most versatile and economical process available for molding hollow materials. When polyethylene is stretched, it exhibits strain-hardening properties, which are temperature, pressure, velocity and strain-rate dependent. In this paper, preform is made by extrusion and forced between two halves by pressurization. This process includes isothermal and transient flow of Newtonian fluid in complex geometries simultaneous with structuring and solidification. A time dependent problem is defined and setting material properties and boundaries condition for bottle blow molding. Numerical data available in POLYDATA for a time dependent problem using ANSYS POLYFLOW were applied. Results display in form contours associated with different variables at different time steps and good agreement with the bottle thickness profile is observed. In this paper, the analysis of the stretch-blow molding (SBM) process of polyethylene terephthalate (PET), parison plastic bottles is studied by the finite element method (FEM). A hyper elastic constitutive behavior was calibrated using material data available in literature in variant high temperatures and strain rates and was used in the numerical simulation. Hydrostatic pressure with convention heat transfer has been used instead of a blowing process. Comparisons of numerical results with experimental observations demonstrate that the model can predict an overall trend of thickness distribution. Through the study, it becomes clear that the proposed model is applicable for simulating the stretch-blow molding process of PET bottles, and is capable of offering helpful knowledge in the production of bottles and the design of an optimum preform.
