Abstract: The NOvA collaboration reports cross-section measurements for $\nu_{\mu}$ charged-current interactions with low hadronic energy (maximum kinetic energy of 250 MeV for protons and 175 MeV for pions) in the NOvA Near Detector. The results are presented as a double-differential cross section as a function of the direct observables of the final-state muon kinematics. Results are also presented as a single-differential cross section as a function of the derived square of the four-momentum transfer, $Q^{2}$, and as a function of the derived neutrino energy. The data correspond to an accumulated 8.09$\times10^{20}$ protons-on-target (POT) in the neutrino mode of the NuMI beam, with a narrow band of neutrino energies peaked at 1.8 GeV. The analysis provides a sample of neutrino-nucleus interactions with an enhanced fraction of quasi-elastic and two-particle-two-hole (2p2h) interactions. This enhancement allows quantitative comparisons with various nuclear models. We find strong disagreement between data and theory-based models in various regions of the muon kinematic phase space, especially in the forward muon direction., Comment: 20 pages, 12 figures. Abstract: Double- and single-differential cross sections for inclusive charged-current neutrino-nucleus scattering are reported for the kinematic domain 0 to 2 GeV/c in three-momentum transfer and 0 to 2 GeV in available energy, at a mean muon-neutrino energy of 1.86 GeV. The measurements are based on an estimated 995,760 muon-neutrino CC interactions in the scintillator medium of the NOvA Near Detector. The subdomain populated by 2-particle-2-hole reactions is identified by the cross-section excess relative to predictions for neutrino-nucleus scattering that are constrained by a data control sample. Models for 2-particle-2- hole processes are rated by chi-square comparisons of the predicted-versus-measured muon-neutrino CC inclusive cross section over the full phase space and in the restricted subdomain. Shortfalls are observed in neutrino generator predictions obtained using the theory-based Val`encia and SuSAv2 2p2h models., Comment: 20 pages, 14 figures

Abstract: This Letter reports a search for charge-parity (CP) symmetry violating non-standard interactions (NSI) of neutrinos with matter using the NOvA Experiment, and examines their effects on the determination of the standard oscillation parameters. Data from $\nu_{\mu}(\bar{\nu}_{\mu})\rightarrow\nu_{\mu}(\bar{\nu}_{\mu})$ and $\nu_{\mu}(\bar{\nu}_{\mu})\rightarrow\nu_{e}(\bar{\nu}_{e})$ oscillation channels are used to measure the effect of the NSI parameters $\varepsilon_{e\mu}$ and $\varepsilon_{e\tau}$. With 90% C.L. the magnitudes of the NSI couplings are constrained to be $|\varepsilon_{e\mu}| \, \lesssim 0.3$ and $|\varepsilon_{e\tau}| \, \lesssim 0.4$. A degeneracy at $|\varepsilon_{e\tau}| \, \approx 1.8$ is reported, and we observe that the presence of NSI limits sensitivity to the standard CP phase $\delta_{\tiny\text{CP}}$.

Abstract: NOvA is a long-baseline neutrino oscillation experiment that measures oscillations in charged-current $\nu_{\mu} \rightarrow \nu_{\mu}$ (disappearance) and $\nu_{\mu} \rightarrow \nu_{e}$ (appearance) channels, and their antineutrino counterparts, using neutrinos of energies around 2 GeV over a distance of 810 km. In this work we reanalyze the dataset first examined in our previous paper [Phys. Rev. D 106, 032004 (2022)] using an alternative statistical approach based on Bayesian Markov Chain Monte Carlo. We measure oscillation parameters consistent with the previous results. We also extend our inferences to include the first NOvA measurements of the reactor mixing angle $\theta_{13}$ and the Jarlskog invariant. We use these results to quantify the strength of our inferences about CP violation, as well as to examine the effects of constraints from short-baseline measurements of $\theta_{13}$ using antineutrinos from nuclear reactors when making NOvA measurements of $\theta_{23}$. Our long-baseline measurement of $\theta_{13}$ is also shown to be consistent with the reactor measurements, supporting the general applicability and robustness of the PMNS framework for neutrino oscillations., Comment: 20 pages, 17 figures; version accepted by Phys. Abstract: Measuring observables to constrain models using maximum-likelihood estimation is fundamental to many physics experiments. Wilks' theorem provides a simple way to construct confidence intervals on model parameters, but it only applies under certain conditions. These conditions, such as nested hypotheses and unbounded parameters, are often violated in neutrino oscillation measurements and other experimental scenarios. Monte Carlo methods can address these issues, albeit at increased computational cost. In the presence of nuisance parameters, however, the best way to implement a Monte Carlo method is ambiguous. Here, we present the method used in the NOvA experiment, which we call `Profiled Feldman--Cousins.' We show that it achieves more accurate frequentist coverage in toy experiments approximating a neutrino oscillation measurement than other methods commonly in use. Abstract: The inclusive electron neutrino charged-current cross section is measured in the NOvA near detector using $8.02\times10^{20}$ protons-on-target (POT) in the NuMI beam. The sample of GeV electron neutrino interactions is the largest analyzed to date and is limited by $\simeq$ 17\% systematic rather than the $\simeq$ 7.4\% statistical uncertainties. The double-differential cross section in final-state electron energy and angle is presented for the first time, together with the single-differential dependence on $Q^{2}$ (squared four-momentum transfer) and energy, in the range 1 GeV $ \leq E_{\nu} < $6 GeV. Detailed comparisons are made to the predictions of the GENIE, GiBUU, NEUT, and NuWro neutrino event generators. The data do not strongly favor a model over the others consistently across all three cross sections measured, though some models have especially good or poor agreement in the single differential cross section vs. $Q^{2}$.

Abstract: We report cross-section measurements of the final-state muon kinematics for \numu charged-current interactions in the NOvA near detector using an accumulated 8.09$\times10^{20}$ protons-on-target (POT) in the NuMI beam. We present the results as a double-differential cross section in the observed outgoing muon energy and angle, as well as single-differential cross sections in the derived neutrino energy, $E_\nu$, and square of the four-momentum transfer, $Q^2$. We compare the results to inclusive cross-section predictions from various neutrino event generators via $\chi^2$ calculations using a covariance matrix that accounts for bin-to-bin correlations of systematic uncertainties. These comparisons show a clear discrepancy between the data and each of the tested predictions at forward muon angle and low $Q^2$, indicating a missing suppression of the cross section in current neutrino-nucleus scattering models.

Abstract: We present new $\nu_\mu\rightarrow\nu_e$, $\nu_\mu\rightarrow\nu_\mu$, $\overline{\nu}_\mu\rightarrow\overline{\nu}_e$, and $\overline{\nu}_\mu\rightarrow\overline{\nu}_\mu$ oscillation measurements by the NOvA experiment, with a 50% increase in neutrino-mode beam exposure over the previously reported results. The additional data, combined with previously published neutrino and antineutrino data, are all analyzed using improved techniques and simulations. A joint fit to the $\nu_e$, $\nu_\mu$, $\overline{\nu}_e$, and $\overline{\nu}_\mu$ candidate samples within the 3-flavor neutrino oscillation framework continues to yield a best-fit point in the normal mass ordering and the upper octant of the $\theta_{23}$ mixing angle, with $\Delta m^{2}_{32} = (2.41\pm0.07)\times 10^{-3}$ eV$^2$ and $\sin^2\theta_{23} = 0.57^{+0.03}_{-0.04}$. The data disfavor combinations of oscillation parameters that give rise to a large asymmetry in the rates of $\nu_e$ and $\overline{\nu}_e$ appearance. This includes values of the CP-violating phase in the vicinity of $\delta_\text{CP} = \pi/2$ which are excluded by $>3\sigma$ for the inverted mass ordering, and values around $\delta_\text{CP} = 3\pi/2$ in the normal ordering which are disfavored at 2$\sigma$ confidence., Comment: 11 pages, 6 figures. Abstract: A search is performed for supernova-like neutrino interactions coincident with 76 gravitational wave events detected by the LIGO/Virgo Collaboration. For 40 of these events, full readout of the time around the gravitational wave is available from the NOvA Far Detector. For these events, we set limits on the fluence of the sum of all neutrino flavors of $F < 7(4)\times 10^{10}\mathrm{cm}^{-2}$ at 90% C.L. assuming energy and time distributions corresponding to the Garching supernova models with masses 9.6(27)$\mathrm{M}_\odot$. Under the hypothesis that any given gravitational wave event was caused by a supernova, this corresponds to a distance of $r > 29(50)$kpc at 90% C.L. Abstract: This Letter reports results from the first long-baseline search for sterile antineutrinos mixing in an accelerator-based antineutrino-dominated beam. The rate of neutral-current interactions in the two NOvA detectors, at distances of 1 km and 810 km from the beam source, is analyzed using an exposure of $12.51\times10^{20}$ protons-on-target from the NuMI beam at Fermilab running in antineutrino mode. A total of $121$ of neutral-current candidates are observed at the Far Detector, compared to a prediction of $122\pm11$(stat.)$\pm15$(syst.) assuming mixing between three active flavors. No evidence for $\bar{\nu}_{\mu}\rightarrow\bar{\nu}_{s}$ oscillation is observed. Interpreting this result within a 3+1 model, constraints are placed on the mixing angles ${\theta}_{24} < 25^{\circ}$ and ${\theta}_{34} < 32^{\circ}$ at the 90% C.L. for $0.05$eV$^{2} \leq \Delta m^{2}_{41} \leq 0.5$eV$^{2}$, the range of mass splittings that produces no significant oscillations at the Near Detector. Abstract: We report the rate of cosmic ray air showers with multiplicities exceeding 15 muon tracks recorded in the NOvA Far Detector between May 2016 and May 2018. The detector is located on the surface under an overburden of 3.6 meters water equivalent. We observe a seasonal dependence in the rate of multiple-muon showers, which varies in magnitude with multiplicity and zenith angle. During this period, the effective atmospheric temperature and surface pressure ranged between 210 K to 230 K and 940mbar to 990mbar, respectively; the shower rates are anti-correlated with the variation in the effective temperature. The variations are about 30% larger for the highest multiplicities than the lowest multiplicities and 20% larger for showers near the horizon than vertical showers.

Abstract: The continuing improvement in quantum efficiency (above 90% for single visible photons), reduction in noise (below 1 electron per pixel), and shrink in pixel pitch (less than 1 micron) motivate billion-pixel X-ray cameras (BiPC-X) based on commercial CMOS imaging sensors. We describe BiPC-X designs and prototype construction based on flexible tiling of commercial CMOS imaging sensors with millions of pixels. Device models are given for direct detection of low energy X-rays ($<$ 10 keV) and indirect detection of higher energies using scintillators. Modified Birks's law is proposed for light-yield nonproportionality in scintillators as a function of X-ray energy. Single X-ray sensitivity and spatial resolution have been validated experimentally using laboratory X-ray source and the Argonne Advanced Photon Source. Possible applications include wide field-of-view (FOV) or large X-ray aperture measurements in high-temperature plasmas, the state-of-the-art synchrotron, X-ray Free Electron Laser (XFEL), and pulsed power facilities., Comment: 6 pages, 8 figures

Abstract: We report a search for a magnetic monopole component of the cosmic-ray flux in a 95-day exposure of the NOvA experiment's Far Detector, a 14 kt segmented liquid scintillator detector designed primarily to observe GeV-scale electron neutrinos. No events consistent with monopoles were observed, setting an upper limit on the flux of $2\times 10^{-14} \mathrm{cm^{-2}s^{-1}sr^{-1}}$ at 90% C.L. for monopole speed $6\times 10^{-4} < \beta < 5\times 10^{-3}$ and mass greater than $5\times 10^{8}$ GeV. Abstract: The two-detector design of the NOvA neutrino oscillation experiment, in which two functionally identical detectors are exposed to an intense neutrino beam, aids in canceling leading order effects of cross-section uncertainties. However, limited knowledge of neutrino interaction cross sections still gives rise to some of the largest systematic uncertainties in current oscillation measurements. We show contemporary models of neutrino interactions to be discrepant with data from NOvA, consistent with discrepancies seen in other experiments. Adjustments to neutrino interaction models in GENIE that improve agreement with our data are presented. Abstract: The NOvA long-baseline neutrino experiment uses a pair of large, segmented, liquid-scintillator calorimeters to study neutrino oscillations, using GeV-scale neutrinos from the Fermilab NuMI beam. These detectors are also sensitive to the flux of neutrinos which are emitted during a core-collapse supernova through inverse beta decay interactions on carbon at energies of $\mathcal{O}(10~\text{MeV})$. This signature provides a means to study the dominant mode of energy release for a core-collapse supernova occurring in our galaxy. We describe the data-driven software trigger system developed and employed by the NOvA experiment to identify and record neutrino data from nearby galactic supernovae. This technique has been used by NOvA to self-trigger on potential core-collapse supernovae in our galaxy, with an estimated sensitivity reaching out to 10~kpc distance while achieving a detection efficiency of 23\% to 49\% for supernovae from progenitor stars with masses of 9.6\~M$_\odot$ to 27\~M$_\odot$, respectively., Comment: 30 pages, 17 figures

18. What is the lived experience for doctors when they are involved in a serious incident? : a descriptive phenomenological analysis

Abstract: Within the National Health Service (NHS), when a serious incident (SI) occurs that harms a patient, understandably, the patient and their families are the primary focus. However, Wu (2000) recognised that the medical doctors involved in the SI, whom he termed "second victims", can also experience psychological distress. This research aimed to explore the lived experiences of second victims, to better understand the second victim phenomenon, as given. The rationale for this research is highlighted through a critical review of the relevant existing literature, which revealed this to be predominately empirically driven, or otherwise interpreted or thematicised. Seven medical doctors were interviewed utilizing a semi-structured interview questionnaire. A Descriptive Phenomenological Analysis was undertaken to conduct an in-depth analysis of the data. The data were sectioned into meaning units and further scrutinised, to identify any psychologically sensitive elements. These were then transformed into nine constituents, which were synthesised to construct the general psychological second victim phenomenon. The findings indicate that doctors experience profound psychological distress at the moment they realise a SI has occurred, from which they never fully recover, concealing and suppressing the emotions the SI has provoked to their own psychological detriment. They feel changed, subsequently altering their work practices, becoming hypervigilant and avoidant. Their self-identities are damaged along with their sense of expertise as doctors, with symptoms indicative of post-traumatic stress disorder manifesting. Nonetheless, they do not consider seeking psychological support, as it is not a part of the clinical culture they have been socialised into since medical school. These findings provide a unique insight into the lived experience of doctors who have been involved in SIs. The clinical implications are discussed in the context of the current literature, leading to recommendations for training and psychological support for clinicians as crucial interventions to help alleviate the psychological distress of second victims.

Abstract: BackgroundSurvival from metastatic cutaneous melanoma is substantially lower than for localized disease. Treatments for metastatic melanoma have been limited, but remarkable clinical improvements have been reported in clinical trials in the last decade. We described the characteristics of US patients diagnosed with cutaneous melanoma during 2001-2013 and assessed trends in short-term survival for distant-stage disease.MethodsTrends in 1-year net survival were estimated using the Pohar Perme estimator, controlling for background mortality with life tables of all-cause mortality rates by county of residence, single year of age, sex, and race for each year 2001-2013. We fitted a flexible parametric survival model on the log-hazard scale to estimate the effect of race on the hazard of death because of melanoma and estimated 1-year net survival by race.ResultsOnly 4.4% of the 425 915 melanomas were diagnosed at a distant stage, cases diagnosed at a distant stage are more commonly men, older patients, and African Americans. Age-standardized, 1-year net survival for distant-stage disease was stable at approximately 43% during 2001-2010. From 2010 onward, survival improved rapidly, reaching 58.9% (95% confidence interval = 56.6% to 61.2%) for patients diagnosed in 2013. Younger patients experienced the largest improvement. Survival for distant-stage disease increased in both Blacks and Whites but was consistently lower in Blacks.ConclusionsOne-year survival for distant-stage melanoma improved during 2001-2013, particularly in younger patients and those diagnosed since 2010. This improvement may be a consequence of the introduction of immune-checkpoint-inhibitors and other targeted treatments for metastatic and unresectable disease. Persistent survival inequalities exist between Blacks and Whites, suggesting differential access to treatment.

22. Priority research directions for in situ data management: Enabling scientific discovery from diverse data sources

Abstract: In January 2019, the US Department of Energy, Office of Science program in Advanced Scientific Computing Research, convened a workshop to identify priority research directions (PRDs) for in situ data management (ISDM). A fundamental finding of this workshop is that the methodologies used to manage data among a variety of tasks in situ can be used to facilitate scientific discovery from many different data sources—simulation, experiment, and sensors, for example—and that being able to do so at numerous computing scales will benefit real-time decision-making, design optimization, and data-driven scientific discovery. This article describes six PRDs identified by the workshop, which highlight the components and capabilities needed for ISDM to be successful for a wide variety of applications—making ISDM capabilities more pervasive, controllable, composable, and transparent, with a focus on greater coordination with the software stack and a diversity of fundamentally new data algorithms.

Abstract: BACKGROUND: Systemic therapy options for salivary cancers are limited. MyPathway (NCT02091141), a phase IIa study, evaluates targeted therapies in non-indicated tumor types with actionable molecular alterations. Here, we present the efficacy and safety results for a subgroup of MyPathway patients with advanced salivary gland cancer (SGC) matched to targeted therapies based on tumor molecular characteristics. PATIENTS AND METHODS: MyPathway is an ongoing, multiple basket, open-label, non-randomized, multi-center study. Patients with advanced SGC received pertuzumab + trastuzumab (HER2 alteration), vismodegib (PTCH-1/SMO mutation), vemurafenib (BRAF V600 mutation), or atezolizumab [high tumor mutational burden (TMB)]. The primary endpoint is the objective response rate (ORR). RESULTS: As of January 15, 2018, 19 patients with SGC were enrolled and treated in MyPathway (15 with HER2 amplification and/or overexpression and one each with a HER2 mutation without amplification or overexpression, PTCH-1 mutation, BRAF mutation, and high TMB). In the 15 patients with HER2 amplification/overexpression (with or without mutations) who were treated with pertuzumab + trastuzumab, 9 had an objective response (1 complete response, 8 partial responses) for an ORR of 60% (9.2 months median response duration). The clinical benefit rate (defined by patients with objective responses or stable disease >4 months) was 67% (10/15), median progression-free survival (PFS) was 8.6 months, and median overall survival was 20.4 months. Stable disease was observed in the patient with a HER2 mutation (pertuzumab + trastuzumab, n = 1/1, PFS 11.0 months), and partial responses in patients with the PTCH-1 mutation (vismodegib, n = 1/1, PFS 14.3 months), BRAF mutation (vemurafenib, n = 1/1, PFS 18.5 months), and high TMB (atezolizumab, n = 1/1, PFS 5.5+ months). No unexpected toxicity occurred. CONCLUSIONS: Overall, 12 of 19 patients (63%) with advanced SGC, treated with chemotherapy-free regimens matched to specific molecular alterations, experienced an objective response. Data from MyPathway suggest that matched targeted therapy for SGC has promising efficacy, supporting molecular profiling in treatment determination.

25. Long-Term Measurements Show Little Evidence for Large Increases in Total U.S. Methane Emissions Over the Past Decade

Abstract: Recent studies show conflicting estimates of trends in methane (CH4) emissions from oil and natural gas (ONG) operations in the United States. We analyze atmospheric CH4 measurements from 20 North American sites in the National Oceanic and Atmospheric Administration Global Greenhouse Gas Reference Network and determined trends for 2006–2015. Using CH4 vertical gradients as an indicator of regional surface emissions, we find no significant increase in emissions at most sites and modest increases at three sites heavily influenced by ONG activities. Our estimated increases in North American ONG CH4 emissions (on average approximately 3.4 ± 1.4 %/year for 2006–2015, ±σ) are much smaller than estimates from some previous studies and below our detection threshold for total emissions increases at the east coast sites that are sensitive to U.S. outflows. We also find an increasing trend in ethane/methane emission ratios, which has resulted in major overestimation of oil and gas emissions trends in some previous studies.

Abstract: Mucin-4 (Muc4) is a large cell surface glycoprotein implicated in the protection and lubrication of epithelial structures. Previous studies suggest that aberrantly expressed Muc4 can influence the adhesiveness, proliferation, viability and invasiveness of cultured tumor cells, as well as the growth rate and metastatic efficiency of xenografted tumors. Although it has been suggested that one of the major mechanisms by which Muc4 potentiates tumor progression is via its engagement of the ErbB2/HER2 receptor tyrosine kinase, other mechanisms exist and remain to be delineated. Moreover, the requirement for endogenous Muc4 for tumor growth progression has not been previously explored in the context of gene ablation. To assess the contribution of endogenous Muc4 to mammary tumor growth properties, we first created a genetically engineered mouse line lacking functional Muc4 (Muc4ko), and then crossed these animals with the NDL (Neu DeLetion mutant) model of ErbB2-induced mammary tumorigenesis. We observed that Muc4ko animals are fertile and develop normally, and adult mice exhibit no overt tissue abnormalities. In tumor studies, we observed that although some markers of tumor growth such as vascularity and cyclin D1 expression are suppressed, primary mammary tumors from Muc4ko/NDL female mice exhibit similar latencies and growth rates as Muc4wt/NDL animals. However, the presence of lung metastases is markedly suppressed in Muc4ko/NDL mice. Interestingly, histological analysis of lung lesions from Muc4ko/NDL mice revealed a reduced association of disseminated cells with platelets and white blood cells. Moreover, isolated cells derived from Muc4ko/NDL tumors interact with fewer blood cells when injected directly into the vasculature or diluted into blood from wild type mice. We further observed that blood cells more efficiently promote the viability of non-adherent Muc4wt/NDL cells than Muc4ko/NDL cells. Together, our observations suggest that Muc4 may facilitate metastasis by promoting the association of circulating tumor cells with blood cells to augment tumor cell survival in circulation.

Abstract: The lethality of the aggressive brain tumor glioblastoma multiforme (GBM) results in part from its strong propensity to invade surrounding normal brain tissue. Although oncogenic drivers such as epidermal growth factor receptor activation and Phosphatase and Tensin homolog inactivation are thought to promote the motility and invasiveness of GBM cells via phosphatidylinostitol 3-kinase activation, other unexplored mechanisms may also contribute to malignancy. Here we demonstrate that several components of the planar cell polarity (PCP) arm of non-canonical Wnt signaling including VANGL1, VANGL2 and FZD7 are transcriptionally upregulated in glioma and correlate with poorer patient outcome. Knockdown of the core PCP pathway component VANGL1 suppresses the motility of GBM cell lines, pointing to an important mechanistic role for this pathway in glioblastoma malignancy. We further observe that restoration of Nrdp1, a RING finger type E3 ubiquitin ligase whose suppression in GBM also correlates with poor prognosis, reduces GBM cell migration and invasiveness by suppressing PCP signaling. Our observations indicate that Nrdp1 physically interacts with the Vangl1 and Vangl2 proteins to mediate the K63-linked polyubiquitination of the Dishevelled, Egl-10 and Pleckstrin (DEP) domain of the Wnt pathway protein Dishevelled (Dvl). Ubiquitination hinders Dvl binding to phosphatidic acid, an interaction necessary for efficient Dvl recruitment to the plasma membrane upon Wnt stimulation of Fzd receptor and for the propagation of downstream signals. We conclude that the PCP pathway contributes significantly to the motility and hence the invasiveness of GBM cells, and that Nrdp1 acts as a negative regulator of PCP signaling by inhibiting Dvl through a novel polyubiquitination mechanism. We propose that the upregulation of core PCP components, together with the loss of the key negative regulator Nrdp1, act coordinately to promote GBM invasiveness and malignancy.

28. Considerable contribution of the Montreal Protocol to declining greenhouse gas emissions from the United States

Abstract: Ozone depleting substances (ODSs) controlled by the Montreal Protocol are potent greenhouse gases (GHGs), as are their substitutes, the hydrofluorocarbons (HFCs). Here we provide for the first time a comprehensive estimate of U.S. emissions of ODSs and HFCs based on precise measurements in discrete air samples from across North America and in the remote atmosphere. Derived emissions show spatial and seasonal variations qualitatively consistent with known uses and largely confirm U.S. Environmental Protection Agency (EPA) national emissions inventories for most gases. The measurement-based results further indicate a substantial decline of ODS emissions from 2008 to 2014, equivalent to ~50% of the CO2-equivalent decline in combined emissions of CO2 and all other long-lived GHGs inventoried by the EPA for the same period. Total estimated CO2-equivalent emissions of HFCs were comparable to the sum of ODS emissions in 2014, but can be expected to decline in the future in response to recent policy measures.

29. Lower-tropospheric CO2 from near-infrared ACOS-GOSAT observations

Abstract: We present two new products from near-infrared Greenhouse Gases Observing Satellite (GOSAT) observations: Lowermost tropospheric (LMT, from 0 to 2.5km) and upper tropospheric-stratospheric (U, above 2.5km) carbon dioxide partial column mixing ratios. We compare these new products to aircraft profiles and remote surface flask measurements and find that the seasonal and year-to-year variations in the new partial column mixing ratios significantly improve upon the Atmospheric CO2 Observations from Space (ACOS) and GOSAT (ACOS-GOSAT) initial guess and/or a priori, with distinct patterns in the LMT and U seasonal cycles that match validation data. For land monthly averages, we find errors of 1.9, 0.7, and 0.8ppm for retrieved GOSAT LMT, U, and XCO2; for ocean monthly averages, we find errors of 0.7, 0.5, and 0.5ppm for retrieved GOSAT LMT, U, and XCO2. In the southern hemispheric biomass burning season, the new partial columns show similar patterns to MODIS fire maps and MOPITT multispectral CO for both vertical levels, despite a flat ACOS-GOSAT prior, and a CO-CO2 emission factor comparable to published values. The difference of LMT and U, useful for evaluation of model transport error, has also been validated with a monthly average error of 0.8 (1.4)ppm for ocean (land). LMT is more locally influenced than U, meaning that local fluxes can now be better separated from CO2 transported from far away.

31. The influence of daily meteorology on boreal fire emissions and regional trace gas variability

Abstract: Relationships between boreal wildfire emissions and day-to-day variations in meteorological variables are complex and have important implications for the sensitivity of high-latitude ecosystems to climate change. We examined the influence of environmental conditions on boreal fire emissions and fire contributions to regional trace gas variability in interior Alaska during the summer of 2013 using two types of analysis. First, we quantified the degree to which meteorological and fire weather indices explained regional variability in fire activity using four different products, including active fires, fire radiative power, burned area, and carbon emissions. Second, we combined daily emissions from the Alaskan Fire Emissions Database (AKFED) with the coupled Polar Weather Research and Forecasting/Stochastic Time-Inverted Lagrangian Transport model to estimate fire contributions to trace gas concentration measurements at the Carbon in Arctic Reservoirs Vulnerability Experiment-NOAA Global Monitoring Division (CRV) tower in interior Alaska. Tower observations during two high fire periods were used to estimate CO and CH4 emission factors. We found that vapor pressure deficit and temperature had a level of performance similar to more complex fire weather indices. Emission factors derived from CRV tower measurements were 134 ± 25 g CO per kg of combusted biomass and 7.74 ± 1.06 g CH4 per kg of combusted biomass. Predicted daily CO mole fractions from AKFED emissions were moderately correlated with CRV observations (r = 0.68) and had a high bias. The modeling system developed here allows for attribution of emission factors to individual fires and has the potential to improve our understanding of regional CO, CH4, and CO2 budgets.

32. A multi-scale comparison of modeled and observed seasonal methane emissions in northern wetlands

Abstract: Wetlands are the largest global natural methane (CH4/ source, and emissions between 50 and 70° N latitude contribute 10-30% to this source. Predictive capability of land models for northern wetland CH4 emissions is still low due to limited site measurements, strong spatial and temporal variability in emissions, and complex hydrological and biogeochemical dynamics. To explore this issue, we compare wetland CH4 emission predictions from the Community Land Model 4.5 (CLM4.5-BGC) with siteto regional-scale observations. A comparison of the CH4 fluxes with eddy flux data highlighted needed changes to the model's estimate of aerenchyma area, which we implemented and tested. The model modification substantially reduced biases in CH4 emissions when compared with CarbonTracker CH4 predictions. CLM4.5 CH4 emission predictions agree well with growing season (May-September) CarbonTracker Alaskan regional-level CH4 predictions and sitelevel observations. However, CLM4.5 underestimated CH4 emissions in the cold season (October-April). The monthly atmospheric CH4 mole fraction enhancements due to wetland emissions are also assessed using the Weather Research and Forecasting-Stochastic Time-Inverted Lagrangian Transport (WRF-STILT) model coupled with daily emissions from CLM4.5 and compared with aircraft CH4 mole fraction measurements from the Carbon in Arctic Reservoirs Vulnerability Experiment (CARVE) campaign. Both the tower and aircraft analyses confirm the underestimate of cold-season CH4 emissions by CLM4.5. The greatest uncertainties in predicting the seasonal CH4 cycle are from the wetland extent, coldseason CH4 production and CH4 transport processes. We recommend more cold-season experimental studies in highlatitude systems, which could improve the understanding and parameterization of ecosystem structure and function during this period. Predicted CH4 emissions remain uncertain, but we show here that benchmarking against observations across spatial scales can inform model structural and parameter improvements.

Abstract: We describe a method for removing systematic biases of column-averaged dry air mole fractions of CO2 (XCO2) and CH4 (XCH4) derived from short-wavelength infrared (SWIR) spectra of the Greenhouse gases Observing SATellite (GOSAT). We conduct correlation analyses between the GOSAT biases and simultaneously retrieved auxiliary parameters. We use these correlations to bias correct the GOSAT data, removing these spurious correlations. Data from the Total Carbon Column Observing Network (TCCON) were used as reference values for this regression analysis. To evaluate the effectiveness of this correction method, the tnzuncorrected/corrected GOSAT data were compared to independent XCO2 and XCH4 data derived from aircraft measurements taken for the Comprehensive Observation Network for TRace gases by AIrLiner (CONTRAIL) project, the National Oceanic and Atmospheric Administration (NOAA), the US Department of Energy (DOE), the National Institute for Environmental Studies (NIES), the Japan Meteorological Agency (JMA), the HIAPER Pole-to-Pole observations (HIPPO) program, and the GOSAT validation aircraft observation campaign over Japan. These comparisons demonstrate that the empirically derived bias correction improves the agreement between GOSAT XCO2/XCH4 and the aircraft data. Finally, we present spatial distributions and temporal variations of the derived GOSAT biases.

Abstract: LRIG1 (leucine-rich repeat and immunoglobulin-like domain containing), a member of the LRIG family of transmembrane leucine-rich repeat-containing proteins, is a negative regulator of receptor tyrosine kinase signaling and a tumor suppressor. LRIG1 expression is broadly decreased in human cancer and in breast cancer and low expression of LRIG1 has been linked to decreased relapse-free survival. Recently, low expression of LRIG1 was revealed to be an independent risk factor for breast cancer metastasis and death. These findings suggest that LRIG1 may oppose breast cancer cell motility and invasion, cellular processes that are fundamental to metastasis. However, very little is known of LRIG1 function in this regard. In this study, we demonstrate that LRIG1 is downregulated during epithelial-to-mesenchymal transition (EMT) of human mammary epithelial cells, suggesting that LRIG1 expression may represent a barrier to EMT. Indeed, depletion of endogenous LRIG1 in human mammary epithelial cells expands the stem cell population, augments mammosphere formation and accelerates EMT. Conversely, expression of LRIG1 in highly invasive Basal B breast cancer cells provokes a mesenchymal-to-epithelial transition accompanied by a dramatic suppression of tumorsphere formation and a striking loss of invasive growth in three-dimensional culture. LRIG1 expression perturbs multiple signaling pathways and represses markers and effectors of the mesenchymal state. Furthermore, LRIG1 expression in MDA-MB-231 breast cancer cells significantly slows their growth as tumors, providing the first in vivo evidence that LRIG1 functions as a growth suppressor in breast cancer.

Abstract: ErbB3, a member of the ErbB family of receptor tyrosine kinases, is a potent activator of phosphatidyl inositol-3 kinase (PI3K) and mammalian target of rapamycin (mTOR) signaling, driving tumor cell survival and therapeutic resistance in breast cancers. In luminal breast cancers, ErbB3 upregulation following treatment with the antiestrogen fulvestrant enhances PI3K/mTOR-mediated cell survival. However, the mechanism by which ErbB3 is upregulated in fulvestrant-treated cells is unknown. We found that ErbB3 protein levels and cell surface presentation were increased following fulvestrant treatment, focusing our attention on proteins that regulate ErbB3 at the cell surface, including Nrdp1, NEDD4 and LRIG1. Among these, only LRIG1 correlated positively with ERα, but inversely with ErbB3 in clinical breast cancer data sets. LRIG1, an estrogen-inducible ErbB downregulator, was decreased in a panel of fulvestrant-treated luminal breast cancer cells. Ectopic LRIG1 expression from an estrogen-independent promoter uncoupled LRIG1 from estrogen regulation, thus sustaining LRIG1 and maintaining low ErbB3 levels in fulvestrant-treated cells. An LRIG1 mutant lacking the ErbB3 interaction motif was insufficient to downregulate ErbB3. Importantly, LRIG1 overexpression improved fulvestrant-mediated growth inhibition, whereas cells expressing the LRIG1 mutant were poorly sensitive to fulvestrant, despite effective ERα downregulation. Consistent with these results, LRIG1 expression correlated positively with increased disease-free survival in antiestrogen-treated breast cancer patients. These data suggest that ERα-dependent expression of LRIG1 dampens ErbB3 signaling in luminal breast cancer cells, and by blocking ERα activity with fulvestrant, LRIG1 is decreased thus permitting ErbB3 accumulation, enhanced ErbB3 signaling to cell survival pathways and blunting therapeutic response to fulvestrant.

36. U.S. emissions of HFC-134a derived for 2008–2012 from an extensive flask-air sampling network

Abstract: U.S. national and regional emissions of HFC-134a are derived for 20082012 based on atmospheric observations from ground and aircraft sites across the U.S. and a newly developed regional inverse model. Synthetic data experiments were first conducted to optimize the model assimilation design and to assess model-data mismatch errors and prior flux error covariances computed using a maximumlikelihood estimation technique. The synthetic data experiments also tested the sensitivity of derived national and regional emissions to a range of assumed prior emissions, with the goal of designing a system that was minimally reliant on the prior. We then explored the influence of additional sources of error in inversions with actual observations, such as those associated with background mole fractions and transport uncertainties. Estimated emissions of HFC-134a range from 52 to 61 Gg yr-1 for the contiguous U.S. during 20082012 for inversions using air transport from Hybrid Single-Particle Lagrangian Integrated Trajectory (HYSPLIT) model driven by the 12km resolution meteorogical data from North American Mesoscale Forecast System (NAM12) and all tested combinations of prior emissions and background mole fractions. Estimated emissions for 20082010 were 20% lower when specifying alternative transport from Stochastic Time-Inverted Lagrangian Transport (STILT) model driven by the Weather Research and Forecasting (WRF) meteorology. Our estimates (for HYSPLIT-NAM12) are consistent with annual emissions reported by U.S. Environmental Protection Agency for the full study interval. The results suggest a 1020% drop in U.S. national HFC-134a emission in 2009 coincident with a reduction in transportation-related fossil fuel CO2 emissions, perhaps related to the economic recession. All inversions show seasonal variation in national HFC-134a emissions in all years, with summer emissions greater than winter emissions by 2050%.

37. Drake passage oceanic pCO2: Evaluating CMIP5 coupled carbon-climate models using in situ observations

Abstract: Surface water partial pressure of CO2 (pCO2) variations in Drake Passage are examined using decade-long underway shipboard measurements. North of the Polar Front (PF), the observed pCO2 shows a seasonal cycle that peaks annually in August and dissolved inorganic carbon (DIC)-forced variations are significant. Just south of the PF, pCO2 shows a small seasonal cycle that peaks annually in February, reflecting the opposing effects of changes in SST and DIC in the surface waters. At the PF, the wintertime pCO2 is nearly in equilibrium with the atmosphere, leading to a small sea-to-air CO2 flux. These observations are used to evaluate eight available Coupled Model Intercomparison Project, phase 5 (CMIP5), Earth system models (ESMs). Six ESMs reproduce the observed annual-mean pCO2 values averaged over the Drake Passage region. However, the model amplitude of the pCO2 seasonal cycle exceeds the observed amplitude of the pCO2 seasonal cycle because of the model biases in SST and surface DIC. North of the PF, deep winter mixed layers play a larger role in pCO2 variations in the models than they do in observations. Four ESMs show elevated wintertime pCO2 near the PF, causing a significant sea-to-air CO2 flux. Wintertime winds in these models are generally stronger than the satellite-derived winds. This not only magnifies the sea-to-air CO2 flux but also upwells DIC-rich water to the surface and drives strong equatorward Ekman currents. These strong model currents likely advect the upwelled DIC farther equatorward, as strong stratification in the models precludes subduction below the mixed layer. © 2014 American Meteorological Society.

38. A low-cost system for measuring horizontal winds from single-engine aircraft

Abstract: The implementation and accuracy of a low-rate (~1 Hz) horizontal wind measurement system is described for a fixed-wing aircraft without modification to the airframe. The system is based on a global positioning system (GPS) compass that provides aircraft heading and a ground-referenced velocity, which, when subtracted from the standard true airspeed, provides estimates of the horizontal wind velocity. A series of tests was performed flying "L"-shaped patterns above the boundary layer, where the winds were assumed to be horizontally homogeneous over the area bounded by the flight (approximately 25km2). Four headings were flown at each altitude at a constant airspeed. Scaling corrections for both heading and airspeed were found by minimizing the variance in the 1-s wind measurements; an upper limit to the error was then computed by calculating the variance of the corrected wind measurements on each of the four headings. A typical uncertainty found in this manner tends to be less than 0.2ms-1. The measurement system described herein is inexpensive and relatively easy to implement on single-engine aircraft. © 2014 American Meteorological Society.

Abstract: Climate models incorporate photosynthesis-climate feedbacks, yet we lack robust tools for large-scale assessments of these processes. Recent work suggests that carbonyl sulfide (COS), a trace gas consumed by plants, could provide a valuable constraint on photosynthesis. Here we analyze airborne observations of COS and carbon dioxide concentrations during the growing season over North America with a three-dimensional atmospheric transport model. We successfully modeled the persistent vertical drawdown of atmospheric COS using the quantitative relation between COS and photosynthesis that has been measured in plant chamber experiments. Furthermore, this drawdown is driven by plant uptake rather than other continental and oceanic fluxes in the model. These results provide quantitative evidence that COS gradients in the continental growing season may have broad use as a measurement-based photosynthesis tracer.
