46. The Effects of Phenylpropanolamine on Zucker Rats Selected for Fat Food Preference.

Abstract

Treatments of human and rodent obesity frequently involve administration of amphetamine derivatives, much like phenylpropanolamine, which suppress food intake. The Zucker rat is a commonly employed model of youth-onset obesity in which the homozygous genotype manifests hyperphagia as well as other characteristics that parallel human obesity. Using a macronutrient selection procedure, we examined phenylpropanolamine's differential actions in controlling dietary intake, spontaneous open-field activity, and regional hypothalamic neurotransmitter levels in obese female Zucker rats of varying fat food preference. We hypothesized that phenylpropanolamine would alter hypothalamic monoamine levels differently in low-fat preferring and high-fat preferring Zucker rats, and hence affect feeding behavior and activity differently in these two groups. It was found that in high-fat preferring animals, phenylpropanolamine significantly decreased spontaneous open-field activity, decreased only carbohydrate caloric intake, and increased serotonin and 5-HIAA levels in the paraventricular nucleus (PVN). In low-fat preferring animals, phenylpropanolamine decreased carbohydrate, protein, and total caloric intake, had no significant effect of spontaneous activity, and increased serotonin and 5-hydroxyindole acetic acid levels in the PVN. Inherent and induced physiological differences of low-fat and high-fat preferring animals are discussed as well as phenylpropanolamine's potential in combination drug therapy for the treatment of human hyperphagic obesity. 49. Chiral Recognition:  Design and Preparation of Chiral Stationary Phases Using Selectors Derived from Ugi Multicomponent Condensation Reactions and a Combinatorial Approach

Abstract

Combinatorial approaches together with high-throughput screening have been used to develop highly selective stationary phases for chiral recognition. Libraries of potential chiral selectors have been prepared by the Ugi multicomponent condensation reactions and screened for their enantioselectivity using the reciprocal approach involving a chiral stationary phase with immobilized model target compound N-(3,5-dinitrobenzoyl)-α-l-leucine. The best candidates were identified from the library of phenyl amides of 2-oxo-azetidineacetic acid derivatives. This screening also enabled specification of the functionalities of the selector desired to achieve the highest level of chiral recognition. The substituents of the phenyl ring adjacent to the chiral center of the selector candidates exhibited the most profound effect on the chiral recognition. The best candidate was then synthesized on a larger scale, resolved into single enantiomers using preparative enantioselective HPLC, and attached to porous poly(2-hydroxyethyl methacrylate-co-ethylene dimethacrylate) beads via an ester linkage to afford the desired stationary phase. Selectivities α as high as 3.2 were found for the separation of a variety of amino acid derivatives.

50. Photografting and the Control of Surface Chemistry in Three-Dimensional Porous Polymer Monoliths

Abstract

The photografting of porous three-dimensional materials has been achieved using a benzophenone-initiated surface photopolymerization within the pores of a macroporous polymer monolith contained in a fused silica capillary. Despite the relatively high thickness (100 μm or more) of the layer of material involved, the photografting process occurs efficiently throughout its cross section as confirmed by electron probe microanalysis. In addition, the use of photomasks during grafting enables the precise placement of specific functionalities in selected and predetermined areas of a single monolith for use in a variety of applications ranging from supported catalysis to microfluidics. For example, we have demonstrated the fast and selective incorporation of chains of poly(2-acrylamido-2-methyl-1-propanesulfonic acid) into the irradiated areas of pores of a 100 μm thick monolith and monitored the extent of grafting through measurements of the electroosmotic flow afforded by the newly introduced ionized functionalities. Grafting of the porous polymer with 4,4-dimethyl-2-vinylazlactone was also successful and could be monitored visually by fluorescence measurements following fluorescent labeling of the grafted chains with Rhodamine 6G.
