This study sought to determine direct and indirect annual costs stemming from treatment-related travel, appointments, waiting times and medication costs, before exploring likely CO2 emission-related cost-savings for 20,330 patients receiving SCIT or SLIT-tablets in Sweden.Methods: A model was developed in Python to capture each category of costs in the target patient population. Absenteeism costs arising from treatment-related travel were determined by obtaining average hourly pay data from Swedish Government sources. Absenteeism costs were also calculated for 30-minute post-dose observation times, which occurred during one clinical appointment for SLIT patients, and all clinical appointments for SCIT patients. Clinical appointment costs were obtained from healthcare price lists for Sweden. Medication costs were retrieved from the Pharmaceutical Specialities in Sweden (Fass) website, and treatment doses required for SCIT and SLIT-tablets were determined based on product labels and previously-calculated dosage regimes. High-cost protection and reimbursement scheme payment caps were applied when determining patient appointment and medication costs, respectively, and when identifying financial burdens for individual payers.Results: Mean total annual costs for SCIT were Swedish Krona (SEK) 604.1 million (m), with clinical appointments contributing the largest share of these costs (52.7%), followed by medication (34.4%), travel-related absenteeism (8.9%), waiting time-related absenteeism (2.7%) and private transportation (1.3%). Mean total annual costs for SLIT-tablets were SEK 336.2m. Medication contributed the most to these costs (72.3%), followed by clinical appointments (22.7%), travel-related absenteeism (3.8%), waiting time-related absenteeism (0.6%) and private transportation (0.6%).Conclusion: For patients with moderate-severe AR receiving AIT in Sweden, SLIT-tablets displayed large potential cost savings to patients, the healthcare system, and the government, whilst possessing reduced societal costs of carbon emissions relative to SCIT.Keywords: allergic rhinitis, costs, subcutaneous immunotherapy, sublingual immunotherapy

In this paper we design high-order accurate and stable finite difference schemes for the initial–boundary–value problem associated with the magnetic induction equation with resistivity. We use summation-by-parts finite difference operators to approximate spatial derivatives and a simultaneous approximation term technique for implementing boundary conditions. The resulting schemes are shown to be energy stable. Various numerical experiments demonstrating both the stability and the high order of accuracy of the schemes are presented. Griseofulvin represents a rare case of a close-packed organic apohost that can clathrate selected volatile guests in a solid-gas fashion. Inclusion mechanisms and solvent exchange were investigated by a combination of single crystal and powder X-ray diffraction, coupled to optical microscopy and thermal analyses. The nickel metal hydride (NiMH) battery technology has been designed for use in electric vehicles, solar-powered applications and power tools. These batteries contain the critical and strategic raw materials cobalt, nickel and several rare earth elements (REE). When designing a battery recycling process, there are several choices to be made regarding end-products and process chemicals. The aim of this study is to investigate and compare the environmental and economic sustainability of different recycling options for NiMH batteries by taking projected market developments into consideration and by applying life cycle assessment and life cycle costing methods. The comparative study is limited to recovery of the REEs. Two hydrometallurgical processes for recovery of the REEs from the anode material are compared with extraction of REEs from primary sources in China. The processes compared are a high-temperature sulfation roasting process and a process based on hydrochloric acid leaching followed by precipitation of REE oxalates. By comparing the different recycling approaches, the hydrochloric acid process performs best. However, the use of oxalic acid has a large impact on the overall sustainability footprint. For the sulfation roasting process, the energy, sodium hydroxide and sulphuric acid consumption contribute most to the total environmental footprint. In this study L-(+)-tartaric acid was used to extract metals from either pure cathode material (NMC111) or black mass from spent lithium-ion batteries. The leaching efficiencies of Li, Co, Ni, and Mn from NMC111 are >87 % at 70 °C, with an initial solid to liquid ratio of 17, and >72.4±1.0 % from black mass under corresponding conditions. The metals tend to form mixed phases in antisolvent crystallization and seeding has a minimal effect on the final solid composition. Impurities influence both crystal nucleation and growth. By controlling the antisolvent addition rate crystal growth can be promoted. The theoretical dielectric constant of the solution is shown to correlate excellently to the recovery efficiency across different antisolvents, where a value <52 results in over 95 % total transition metal recovery efficiency. The correlation can be a powerful tool for quantitative prediction of optimal solvent composition for effective antisolvent crystallization., (© 2024 The Author(s). Deep eutectic solvents (DESs) have garnered attention in Li-ion battery (LIB) recycling due to their declared eco-friendly attributes and adjustable metal dissolution selectivity, offering a promising avenue for recycling processes. However, DESs currently lack competitiveness compared to mineral acids, commonly used in industrial-scale LIB recycling. Current research primarily focuses on optimizing DES formulation and experimental conditions to maximize metal dissolution yields in standalone leaching experiments. While achieving yields comparable to traditional leaching systems is important, extensive DES reuse is vital for overall recycling feasibility. To achieve this, evaluating the metal dissolution mechanism can assist in estimating DES consumption rates and assessing process makeup stream costs. The selection of appropriate metal recovery and DES regeneration strategies is essential to enable subsequent reuse over multiple cycles. Finally, decomposition of DES components should be avoided throughout the designed recycling process, as by-products can impact leaching efficiency and compromise the safety and environmental friendliness of DES. The early stages of the molecular self-assembly pathway leading to crystal nucleation have a significant influence on the properties and purity of organic materials. This mini review collates the work on organic mesoscale clusters and discusses their importance in nucleation processes, with a particular focus on their critical properties and susceptibility to sample treatment parameters. This is accomplished by a review of detection methods, including dynamic light scattering, nanoparticle tracking analysis, small angle X-ray scattering, and transmission electron microscopy. Considering the challenges associated with crystallisation of flexible and large-molecule active pharmaceutical ingredients, the dynamic nature of mesoscale clusters has the potential to expand the discovery of novel crystal forms. By collating literature on mesoscale clusters for organic molecules, a more comprehensive understanding of their role in nucleation will evolve and can guide further research efforts., (© 2023 The Authors. In Sweden, allergy immunotherapy (AIT) is available as either subcutaneous immunotherapy (SCIT) injections or sublingual immunotherapy (SLIT) tablets and is used to treat moderate-severe allergic rhinitis (AR). This study sought to determine treatment-related CO 2 emissions and travel times in Swedish patients receiving either SCIT or SLIT-tablets. A list of specialized Swedish AR clinics that administer AIT was determined, and respective co-ordinates retrieved. Swedish municipality population data were obtained from a national database. The mean distance from each Swedish municipality to the nearest AR clinic was calculated, adjusted using a detour index, and weighted by estimated patient population size. Transport modality data were obtained from a Swedish urban transport study and CO 2 emissions were obtained from Government sources. The mean number of annual SLIT-tablets and SCIT doses required were calculated based on product labels and clinical expert input. The annual number of healthcare professional interactions were layered into the model to estimate changes in mean patient travel time, distance, and travel-related CO 2 emissions associated with using SCIT versus SLIT-tablets. Mean annual travel-related CO 2 emissions were 410 tonnes (to two significant figures [s.f.]; standard deviation [SD] 90) with SLIT-tablets, versus 1700 tonnes (SD 380) for SCIT, resulting in mean annual savings of approximately 1300 tonnes (SD 290) of CO 2 if all AIT patients were to receive SLIT-tablets instead of SCIT, over 380 times greater than 2021 average Swedish CO 2 emissions per capita. Approximate mean annual travel times for patients taking SLIT-tablets were 66,500 h (three s.f.; SD 14,400), and 278,000 h (SD 60,200) for SCIT, resulting in mean annual savings of 211,000 h (SD 45,800) if all AIT patients were to receive SLIT-tablets instead of SCIT. This work presents two new solid forms, a polymorph and a solvate, of the antifungal active pharmaceutical ingredient griseofulvin (GSF). The novel forms were characterized by powder X-ray diffraction, differential scanning calorimetry, and thermogravimetric analysis, and their crystal structures were determined by single-crystal X-ray diffraction. The new polymorphic form (GSF Form VI) was obtained upon drying at room temperature the GSF-acetonitrile solvate. GSF Form VI is a relict structure related to reported solvates of GSF. Thermal stability studies show that Form VI is metastable and monotropically related to the stable GSF Form I. The new GSF- n -butyl acetate solvate was obtained by crystallization from an n -butyl acetate solution. The stoichiometry of the n -butyl acetate solvate is 1:0.5. The recovery of scandium from waste streams of other mining and metallurgical processing industries is gaining research interest due to the scarcity of scandium-containing ores. Hydrometallurgical techniques such as leaching, solvent extraction and crystallization amongst others have been successfully applied to recover scandium salts from such waste streams. Scandium can be recovered as (NH 4 ) 3 ScF 6 by antisolvent crystallization from NH 4 F strip liquors obtained after solvent extraction. The coextraction of metal impurities such as Fe, Al, Zr and Ti causes contamination of the final solid product. The extent of coprecipitation of ammonium metal fluorides depends on their initial concentration in the strip liquor and their solubility in the NH 4 F-antisolvent mixtures. Here, the solubility of ammonium metal fluorides of Sc, Zr, Fe, Al and Ti is reported separately in 3 mol L -1 NH 4 F-ethanol mixtures at 25 °C as well as in a system containing all five solid phases. The solubility of (NH 4 ) 3 ZrF 7 is slightly higher than that of (NH 4 ) 3 ScF 6 , while the solubilities of (NH 4 ) 3 FeF 6 and (NH 4 ) 3 AlF 6 are significantly lower in comparison to (NH 4 ) 3 ScF 6 . The solubility of (NH 4 ) 2 TiF 6 is 1-2 orders of magnitude higher than those of other ammonium metal fluorides. When a mixture of ammonium metal fluoride salts is dissolved in the same 3 mol L -1 NH 4 F-ethanol mixture as for the individual salts, the resultant solubility of the ammonium metal fluorides of Sc, Zr and Fe decreases significantly, while the resultant solubility of ammonium aluminum hexafluoride increases. The flexible and reversible preparation of columns for use in high-performance solid phase extraction chromatography by physisorption of organophosphorus acid extractants has been investigated in detail. Two extractants have been evaluated, bis (2-ethyl-1-hexyl) phosphoric acid (HDEHP) and 2-ethyl-1-hexyl (2-ethyl-1-hexyl) phosphonic acid (HEHEHP), but the developed procedure should be broadly applicable to other extractants. The liquid-liquid solubility of the extractants in feed solvents consisting of aqueous ethanol solutions of varying composition has been determined. The total amount of adsorbed extractant has been quantified by complete desorption and elution with ethanol followed by acid-base titrimetry. Column impregnation with feed solutions of varying concentration in the undersaturated region has been systematically evaluated, and the influence of a subsequent water wash step has been explored. It is shown that to achieve a robust and reproducible physisorption, the adsorbed amount of extractant should be determined after the wash step, and care must be taken when using indirect methods of measurement. Equilibrium Langmuir-type adsorption isotherms as a function of the extractant concentration in the feed solution have been determined. Adsorption of HEHEHP is higher than HDEHP for equal feed compositions, but the solubility of HEHEHP is lower, resulting in approximately identical maximum coverage levels. A new polymorph of the drug active pharmaceutical ingredient piracetam (Form VI) has been discovered and characterized by X-ray powder diffraction (PXRD), solid-state Raman, attenuated total reflectance infrared spectroscopy, and differential scanning calorimetry. The PXRD diffractogram of Form VI shows a distinct peak at 24.2° (2θ) that distinguishes it from the previously known polymorphs and solvates. Form VI is metastable with respect to the previously known polymorphs Form II and Form III; in ethanol solution at 288 K, Form VI transforms into Form II within 15 min, while in isopropanol solution Form VI is kinetically stable for at least 6 h. A total of 1200 crystal nucleation induction time experiments of piracetam in ethanol and isopropanol solutions have been conducted, in sets of 40-80 repeat experiments carried out at different temperatures and solute concentrations. Each solution nucleated as a single polymorph, and each set of repeat experiments resulted in different proportions of Form II, Form III, and Form VI, with Form VI dominating at low nucleation temperatures and Form II at higher nucleation temperatures. The induction time data for Form VI at 288 K have been evaluated within the framework of the classical nucleation theory. At equal driving force, nucleation of Form VI is less obstructed in ethanol than in isopropanol, as captured by a lower interfacial energy and higher pre-exponential factor in ethanol. When faced with an imminent collision threat, human vehicle drivers respond with braking in a manner which is stereotypical, yet modulated in complex ways by many factors, including the specific traffic situation and past driver eye movements. A computational model capturing these phenomena would have high applied value, for example in virtual vehicle safety testing methods, but existing models are either simplistic or not sufficiently validated. This paper extends an existing quantitative driver model for initiation and modulation of pre-crash brake response, to handle off-road glance behavior. The resulting models are fitted to time-series data from real-world naturalistic rear-end crashes and near-crashes. A stringent parameterization and model selection procedure is presented, based on particle swarm optimization and maximum likelihood estimation. A major contribution of this paper is the resulting first-ever fit of a computational model of human braking to real near-crash and crash behavior data. The model selection results also permit novel conclusions regarding behavior and accident causation: Firstly, the results indicate that drivers have partial visual looming perception during off-road glances; that is, evidence for braking is collected, albeit at a slower pace, while the driver is looking away from the forward roadway. Secondly, the results suggest that an important causation factor in crashes without off-road glances may be a reduced responsiveness to visual looming, possibly associated with cognitive driver state (e.g., drowsiness or erroneous driver expectations). It is also demonstrated that a model parameterized on less-critical data, such as near-crashes, may also accurately reproduce driver behavior in highly critical situations, such as crashes., (Copyright © 2021 The Authors. The present study investigates the solubility of famotidine polymorphs forms A and B between 298.15 K and 348.15 K in a range of pure solvents: water, methanol, ethanol, isopropanol and acetonitrile. Empirical and semi-empirical models have been fitted to solubility data determined experimentally by a gravimetric method. The solid phases have been characterized by FTIR and Raman spectroscopy, SEM and PXRD. In addition, heat capacities and melting data determined by DSC have been used to estimate the fusion thermodynamics and the activity of the solid phases as a function of temperature. The relationship between the famotidine polymorphs is monotropic, with form A being the stable polymorph. For both polymorphs, in terms of mass ratio, the solubility in the studied solvents decreases in the order methanol > water > ethanol > acetonitrile > isopropanol. The activity coefficient at saturation in all the solutions exceeds unity, showing a positive deviation with respect to ideality, which translates into solubilities significantly lower than the ideal values. Among the alcohols, a consistent correlation is observed between the polarity and the order of solubility.The Hildebrand solubility parameter is also well correlated with the order of solubilities in the studied solvents, with a higher solubility in more polar solvents, revealing the importance of the hydrogen bonding of the sulfamoyl group oxygens., (Copyright © 2021 Elsevier B.V. Studies show high correlations between drivers' off-road glance duration or pattern and the frequency of crashes. Understanding drivers' use of peripheral vision to detect and react to threats is essential to modelling driver behavior and, eventually, preventing crashes caused by visual distraction. A between-group experiment with 83 participants was conducted in a high-fidelity driving simulator. Each driver in the experiment was exposed to an unexpected, critical, lead vehicle deceleration, when performing a self-paced, visual-manual, tracking task at different horizontal visual eccentricity angles (12°, 40° and 60°). The effect of visual eccentricity on threat detection, glance and brake response times was analyzed. Contrary to expectations, the driver glance response time was found to be independent of the eccentricity angle of the secondary task. However, the brake response time increased with increasing task eccentricity, when measured from the driver's gaze redirection to the forward roadway. High secondary task eccentricity was also associated with a low threat detection rate and drivers were predisposed to perform frequent on-road check glances while executing the task. These observations indicate that drivers use peripheral vision to collect evidence for braking during off-road glances. The thermodynamic relationship between FI and FII of ethyl 4-aminobenzoate (benzocaine) has been investigated. Slurry conversion experiments show that the transition temperature below which FI is stable is located between 302 K-303 K (29 °C-30 °C). The polymorphs FI and FII have been characterised by infrared spectroscopy (IR), Raman spectroscopy, transmission powder X-ray diffraction (XRPD) and differential scanning calorimetry (DSC). The isobaric solid state heat capacities have been measured by DSC. The quantitative thermodynamic stability relationship has been determined in a comprehensive thermodynamic analysis of the calorimetric data. The solubility of both polymorphs has been determined in eight pure organic solvents over the temperature range 278 K-323 K by a gravimetric method. The mole fraction solubility of benzocaine decreases in the order: 1,4-dioxane, acetone, ethyl acetate, chloroform, acetonitrile, methanol, n-butanol and toluene. Comparison with the determined activity of solid benzocaine forms shows that negative deviation from Raoult's law ideality is found in dioxane, acetone and ethyl acetate solutions, and positive deviation in solutions of the other investigated solvents., (Copyright © 2020 The Authors. The solubility of the racemic solid phase of ketoprofen (KTP) in methanol, ethanol, isopropanol, butanol, acetonitrile, ethyl acetate, 1,4-dioxane and toluene has been determined between 273 and 303 K by a gravimetric method. FTIR and Raman spectroscopy, SEM and PXRD, have been used to characterise the solid phase. The melting data and heat capacity of solid and melt have been determined by DSC, and used to estimate fusion thermodynamics and the activity of the solid phase as functions of temperature. Empirical and semi-empirical models have been fitted to experimental solubility data. The solution activity coefficients reveal positive deviation from ideality in all solvents except for in dioxane, and very close to ideality in methanol. The solubility is fairly high in the alcohols but decrease with increasing hydrocarbon chain. Generally and due to the presence of the carboxylic acid group, KTP is more readily dissolved in polar protic solvents, followed in order by polar aprotic and non-polar solvents. However, the highest solubility is found in dioxane, classified as a non-polar solvent, but notably though the molecule having two strong hydrogen bond accepting functionalities, and no hydrogen bond donation capability., (Copyright © 2020 Elsevier B.V. The solid-liquid solubility of two polymorphs of the title compound has been measured in n-propanol over the temperature range (278 K-303 K) by an isothermal, gravimetric method and a low heating rate polythermal method. Due to marked differences in the settling behavior of crystals of the two polymorphs in the investigated solvent, it is found that the low heating rate polythermal method gives the overall best performance for this particular system. Systematic slurry conversion experiments show that FII is the stable polymorph over the investigated temperature range (268 K-308 K). Solubility data for both polymorphs is well correlated, and has been extrapolated to the melting point, by a previously proposed semi-empirical regression model based on solid-phase calorimetric data. The system exhibits a marked positive deviation from Raoult's law, with solute activity coefficients at equilibrium decreasing with increasing temperature., (Copyright © 2020 The Authors. The solubility of butamben has been measured gravimetrically in pure methanol, 1-propanol, 2-propanol, 1-butanol, and toluene over the temperature range 268-298 K. Polymorph transition and melting temperatures, associated enthalpy changes, and the heat capacity of the solid forms and the supercooled melt have been measured by differential scanning calorimetry. Based on extrapolated calorimetric data, the Gibbs energy, enthalpy and entropy of fusion, and the activity of solid butamben (the ideal solubility) have been calculated from below ambient temperature up to the melting point. Activity coefficients of butamben at equilibrium in the different solvents have been estimated from solubility data and the activity of the solid, revealing that all investigated systems exhibit positive deviation from Raoult's law. Solubility data are well correlated by a semiempirical regression model. On a mass basis, the solubility is clearly higher in methanol than in the other solvents, but mole fraction solubilities are very similar across all 5 solvents. The 2 known polymorphs are enantiotropically related, and the transition point is located at 283 K. Polymorph interconversions occur within 0.3 K of the transition point even in the solid state, and the 2 forms exhibit strong similarities in investigated properties., (Copyright © 2019 American Pharmacists Association®. Molecular clustering and solvent-solute interactions in isopropanol solutions of fenoxycarb have been thoroughly and systematically investigated by dynamic light scattering, small-angle X-ray scattering, and nanoparticle tracking, supported by infrared spectroscopy and molecular dynamics simulations. The existence of molecular aggregates, clusters, ranging in size up to almost a micrometre is clearly recorded at undersaturated as well as supersaturated conditions by all three analysis techniques. The results systematically reveal that the cluster size increases with solute concentration and time at stagnant conditions. For most concentrations the time scale of cluster growth is of the order of days. In undersaturated solutions the size appears to eventually reach a maximum value, higher the higher the concentration. Below a certain concentration threshold clusters are significantly smaller. Clusters are found to be smaller in solutions pre-heated at a higher temperature, which offers a possible explanation for the so-called "history of solution" effect. The cluster distribution is influenced by filtration through membranes with a pore size of 0.1 μm, offering an alternative explanation for the "foreign particle-catalysed nucleation" effect. At moderate concentrations larger clusters appear to be sheared into smaller ones, but the original size distribution is rapidly re-established. At higher concentrations, although still well below solubility, the cluster size as well as solute concentration are strongly affected, suggesting that larger clusters contain at least a core of more organized molecules not able to pass through the filter.

Background and Objectives: In Sweden, approximately 6% of children and 10% of adults suffer from house dust mite (HDM) allergy with symptoms of allergic rhinitis and allergic asthma. Treatment is aimed at reducing HDM exposure and to control the symptoms of allergic rhinitis and allergic asthma by symptom-relieving pharmacotherapy. This pharmacotherapy is often effective, but some patients remain inadequately controlled. For these patients, allergy immunotherapy (AIT, subcutaneous or sublingual) with repeated administration of HDM allergen should be considered. The objective of this study was to compare the costs for sublingual AIT (SLIT; SQ ® SLIT-tablet) to the costs for subcutaneous AIT (SCIT; SQ ® SCIT) for the treatment of HDM allergy in a cost-minimisation analysis (CMA)., Methods: The CMA included resources (and costs) for treatment, healthcare visits, travelling and lost productivity. Resource use based on Swedish clinical treatment practice and costs were obtained from medical price lists. Analyses were conducted from the societal, as well as healthcare perspective, by use of a time horizon of 3 years., Results: The results show that SQ ® SLIT-tablet is a cost-saving treatment as compared to SQ ® SCIT for the treatment of HDM allergy (€6800 over 3 years). The results are mainly driven by the cost of healthcare visits and the frequency of SCIT administrations., Conclusion: In conclusion, cost-savings of €6800 over 3 years are expected from treating HDM allergy with SQ ® SLIT-tablet as compared to SQ ® SCIT, including costs for treatment, healthcare visits, travelling and lost productivity. The reduced number of healthcare visits compensates for higher medication costs.

Melting temperatures and enthalpies of fusion have been determined by differential scanning calorimetry (DSC) for 2 polymorphs of the drug tolbutamide: FI(H) and FV. Heat capacities have been determined by temperature-modulated DSC for 4 polymorphs: FI(L), FI(H), FII, FV, and for the supercooled melt. The enthalpy of fusion of FII at its melting point has been estimated from the enthalpy of transition of FII into FI(H) through a thermodynamic cycle. Calorimetric data have been used to derive a quantitative polymorphic stability relationship between these 4 polymorphs, showing that FII is the stable polymorph below approximately 333 K, above which temperature FI(H) is the stable form up to its melting point. The relative stability of FV is well below the other polymorphs. The previously reported kinetic reversibility of the transformation between FI(L) and FI(H) has been verified using in situ Raman spectroscopy. The solid-liquid solubility of FII has been gravimetrically determined in 5 pure organic solvents (methanol, 1-propanol, ethyl acetate, acetonitrile, and toluene) over the temperature range 278 to 323 K. The ideal solubility has been estimated from calorimetric data, and solution activity coefficients at saturation in the 5 solvents determined. All solutions show positive deviation from Raoult's law, and all van't Hoff plots of solubility data are nonlinear. The solubility in toluene is well below that observed in the other investigated solvents. Two crystal polymorphs of 1,7-bis-(4-hydroxy-3-methoxyphenyl)-1,6-heptadiene-3,5-dione (curcumin) have been obtained by crystallization from ethanol (EtOH) solution. The polymorphs have been characterized by differential scanning calorimetry, infrared spectroscopy, and X-ray powder diffraction and shown to be the previously described forms I and III. The solubility of both polymorphs in EtOH and of one polymorph in ethyl acetate (EA) has been measured between 10°C and 50°C with a gravimetric method. Primary nucleation of curcumin from EtOH solution has been investigated in 520 constant temperature crystallization experiments in sealed, magnetically stirred vials under different conditions of supersaturation, temperature, and agitation rate. By a thermodynamic analysis of the melting data and solubility of form I, the solid-state activity is estimated from 10°C up to the melting point. The solubility is lower in EtOH than in EA, and in both solvents, a positive deviation from Raoult's law is observed. Form I has lower solubility than form III and is accordingly thermodynamically more stable over the investigated temperature interval. Extrapolation of solubility regression models indicates that there should be a low-temperature enantiotropic transition point, below which form I will be metastable. By slurry conversion experiments, it is established that this temperature is below -30°C. All nucleation experiments resulted in the stable form I. The induction time is observed to decrease with increasing agitation rate up to a certain point, and then increase with further increasing agitation rate; a trend previously observed for other compounds. By correlating the induction time data obtained at different supersaturation and temperature, the interfacial energy of form I in EtOH is estimated to be 3.0 mJ/m(2) ., (© 2015 Wiley Periodicals, Inc. and the American Pharmacists Association.)

In this paper, we have explored the relationship between surface structure and crystal growth and morphology of fenoxycarb (FC). Experimental vs. predicted morphologies/face indices of fenoxycarb crystals are presented. Atomic-scale surface structures of the crystalline particles, derived from experimentally indexed single crystals, are also modelled. Single crystals of fenoxycarb exhibit a platelet-like morphology which closely matches predicted morphologies. The solvent choice does not significantly influence either morphology or crystal habit. The crystal morphology is dominated by the {001} faces, featuring weakly interacting aliphatic or aromatic groups at their surfaces. Two distinct modes of interaction of a FC molecule in the crystal can be observed, which appear to be principal factors governing the microscopic shape of the crystal: the relatively strong collateral and the much weaker perpendicular bonding. Both forcefield-based and quantum-chemical calculations predict that the aromatic and aliphatic terminated {001} faces have comparably high stability as a consequence of weak intermolecular bonding. Thus we predict that the most developed {001} surfaces of fenoxycarb crystals should be terminated randomly, favouring neither aliphatic nor aromatic termination., (Copyright © 2014 Elsevier Inc. Erv41p is a conserved integral membrane protein that is known to play a role in transport between the endoplasmic reticulum and Golgi apparatus, part of the early secretory pathway of eukaryotes. However, the exact function of the protein is not known, and it shares very low sequence identity with proteins of known structure or function. Here we present the structure of the full lumenal domain of Erv41p from Saccharomyces cerevisiae, determined by X-ray crystallography to a resolution of 2.0Å. The structure reveals the protein to be composed predominantly of two large β-sheets that form a twisted β-sandwich. Comparison to structures in the Protein Data Bank shows that the Erv41p lumenal domain displays only limited similarity to β-sandwich domains of other proteins. Analysis of the surface properties of the protein identifies an extensive patch of negative electrostatic potential on the exposed surface of one of the β-sheets, which likely forms a binding site for a ligand or interaction partner. A predominantly hydrophobic region close to the membrane interface is identified as a likely site for protein-protein interaction. This structure, the first of Erv41p or any of its homologues, provides a new starting point for studies of the roles of Erv41p and its interaction partners in the eukaryotic secretory pathway., (Copyright © 2013 Elsevier Ltd. The membrane protein Erv41p is a major component of COPII-coated vesicles and is thought to play a role in the early secretory pathway in eukaryotic cells. In this study, the full lumenal domain of Erv41p from Saccharomyces cerevisiae (ScErv41p&#95;LD) was recombinantly expressed in Sf9 insect cells and purified by nickel-affinity, ion-exchange and size-exclusion chromatography. ScErv41p&#95;LD crystals were obtained using the sitting-drop vapour-diffusion method and native X-ray diffraction data were collected to 2.0 Å resolution. The crystals belonged to space group P21, with unit-cell parameters a = 49.8, b = 76.9, c = 65.1 Å, α = γ = 90.0, β = 104.8°.

P58(IPK) is one of the endoplasmic reticulum- (ER-) localised DnaJ (ERdj) proteins which interact with the chaperone BiP, the mammalian ER ortholog of Hsp70, and are thought to contribute to the specificity and regulation of its diverse functions. P58(IPK), expression of which is upregulated in response to ER stress, has been suggested to act as a co-chaperone, binding un- or misfolded proteins and delivering them to BiP. In order to give further insights into the functions of P58(IPK), and the regulation of BiP by ERdj proteins, we have determined the crystal structure of human P58(IPK) to 3.0 Å resolution using a combination of molecular replacement and single wavelength anomalous diffraction. The structure shows the human P58(IPK) monomer to have a very elongated overall shape. In addition to the conserved J domain, P58(IPK) contains nine N-terminal tetratricopeptide repeat motifs, divided into three subdomains of three motifs each. The J domain is attached to the C-terminal end via a flexible linker, and the structure shows the conserved Hsp70-binding histidine-proline-aspartate (HPD) motif to be situated on the very edge of the elongated protein, 100 Å from the putative binding site for unfolded protein substrates. The residues that comprise the surface surrounding the HPD motif are highly conserved in P58(IPK) from other organisms but more varied between the human ERdj proteins, supporting the view that their regulation of different BiP functions is facilitated by differences in BiP-binding.

Human MCFD2 (multiple coagulation factor deficiency 2) is a 16-kDa protein known to participate in transport of the glycosylated human coagulation factors V and VIII along the secretory pathway. Mutations in MCFD2 or in its binding partner, the membrane-bound transporter ERGIC (endoplasmic reticulum-Golgi intermediate compartment)-53, cause a mild form of inherited hemophilia known as combined deficiency of factors V and VIII (F5F8D). While ERGIC-53 is known to be a lectin-type mannose binding protein, the role of MCFD2 in the secretory pathway is comparatively unclear. MCFD2 has been shown to bind both ERGIC-53 and the blood coagulation factors, but little is known about the binding sites or the true function of the protein. In order to facilitate understanding of the function of MCFD2 and the mechanism by which mutations in the protein cause F5F8D, we have determined the structure of human MCFD2 in solution by NMR. Our results show the folding of MCFD2 to be dependent on availability of calcium ions. The protein, which is disordered in the apo state, folds upon binding of Ca(2+) to the two EF-hand motifs of its C-terminus, while retaining some localized disorder in the N-terminus. NMR studies on two disease-causing mutant variants of MCFD2 show both to be predominantly disordered, even in the presence of calcium ions. These results provide an explanation for the previously observed calcium dependence of the MCFD2-ERGIC-53 interaction and, furthermore, clarify the means by which mutations in this protein result in inefficient secretion of blood coagulation factors V and VIII.

Vanillin crystals in a saturated aqueous solution disappear and a second liquid phase emerges when the temperature is raised above 51 degrees C. The phenomenon has been investigated with crystallization and equilibration experiments, using DSC, TGA, XRD and hot-stage microscopy for analysis. The new liquid solidifies on cooling, appears to melt at 51 degrees C, and has a composition corresponding to a dihydrate. However, no solid hydrate can be detected by XRD, and it is shown that the true explanation is that a liquid-liquid phase separation occurs above 51 degrees C where the vanillin-rich phase has a composition close to a dihydrate. To our knowledge, liquid-liquid phase separation has not previously been reported for the system vanillin-water, even though thousands of tonnes of vanillin are produced globally every year., ((c) 2007 Wiley-Liss, Inc. and the American Pharmacists Association.)

The BOEL test was originally devised for the early discovery of communication disorders in infants. The sound stimuli employed for hearing testing have been calibrated and standardized and any deviation from normal hearing responses to these stimuli indicate a hearing impairment. Since 1971 the BOEL screening program has been applied experimentally in a number of child health centers in Stockholm. Up to 1975, more than 30 000 infants have been examined and approximately 5% have not responded fully regarding visual or tactile attention, auditory, motor or mental functions or social contact. The results of the audiological follow-up are reported. Application of the BOEL hearing subtest proved very effective. It is pointed out that the BOEL test covers not only hearing defects but also other communication malfunctions and as such it is a more useful technique than simple hearing screening methods.

Calcium binding to bone gamma-carboxyglutamic acid protein (BGB) from calf has been studied using 43Ca NMR. The temperature dependence of the 43Ca NMR signal has been used to calculate the calcium ion exchange rate, koff. The dependence of the 43Ca NMR band shape on the [Ca2+]/[BGP] ratio fits well to a chemical equilibrium model having a single Ca2+-binding site with an association constant in the range of 5 X 10(3)-1 X 10(5) M-1. The pH dependence of the 43Ca NMR line-width shows a single apparent pKa value of 5.1.

Cation binding to three apoparvalbumins was studied by means of 113Cd NMR. The 3 parvalbumins that were investigated were carp pI 4.25, rabbit pI 5.5 and pike pI 5.0. The results showed that Cd2+ ions bind to the EF and CD sites of carp apoparvalbumin pI 4.25 with about the same affinity. For rabbit (pI 5.5) apoparvalbumin, Cd2+ binds preferentially to the EF site, while for pike (pI 5.0) apoparvalbumin, it was the CD site that exhibited somewhat higher affinity for Cd2+. The effect of Mn2+ on the 113Cd signals of rabbit parvalbumin was used to assign the 113Cd NMR signals to the EF and CD sites. The Mn2+ paramagnetic effect on rabbit and pike parvalbumins differed from that obtained for carp parvalbumin. This is in agreement with the assumption that the beta-lineage parvalbumins possess a third external site of higher affinity than the alpha-lineage parvalbumins. Furthermore, 23Na NMR was used to study Na+-Mg2+ competition in the native carp (pI 4.25) parvalbumin. The results showed that Na+ and Mg2+ compete for the same site, the third external site.
