15. Conformational preference of 2-(4-methoxyphenyl)ethanol studied by supersonic jet spectroscopy: Intramolecular OH/π interaction.

Abstract

A π-type hydrogen bonding between the OH group and the π electron is a crucial factor for the conformational preference of the molecular structure with a flexible group. However, the information on the effect of the substituent on the OH/π interaction is insufficient. The laser-induced fluorescence (LIF) excitation, the dispersed fluorescence (DF), the IR–UV hole-burning, and the IR dip spectra of jet-cooled 2-(4-methoxyphenyl)ethanol were measured for the first time. Almost all bands observed in the spectral region of 35 550–36 500 cm−1 in the LIF excitation spectrum were successfully assigned with the DF and the IR–UV hole-burning spectra coupled with the quantum chemical calculation at M06-2x/6-311G and MP2/6-311G levels. Five conformers were found in the LIF excitation spectrum. The most stable conformer was Ggπ, and the second most stable conformer was Ggπ′ (the trans rotamer of the methoxy group for Ggπ). Ggπ and Ggπ′ had the OH group directed toward the π electron system of the benzene ring. The OH stretching frequency of Ggπ/Ggπ′ of MPE in the IR dip spectra was red-shifted against that of Ggπ of phenylethanol, indicating that the introduction of the methoxy group would enhance the intramolecular OH/π interaction. In addition, the torsional vibration between the benzene ring and the side chain (−CH2CH2OH) (mode 63) was observed in the DF spectra of the Ggπ–00 and Ggπ′–00 band excitation, but their intensities were rather different, resulting from the different orientation of the OH group for each conformer toward the π electron system. The methoxy group would increase the negative charge on the benzene ring and would enhance the intramolecular OH/π interaction through the electrostatic interaction. [ABSTRACT FROM AUTHOR]

Abstract

The cytosolic peptide:N‐glycanase (PNGase) is involved in the quality control of N‐glycoproteins via the endoplasmic reticulum‐associated degradation (ERAD) pathway. Mutations in the gene encoding cytosolic PNGase (NGLY1 in humans) cause NGLY1 deficiency. Recent findings indicate that the F‐box protein FBS2 of the SCFFBS2 ubiquitin ligase complex can be a promising drug target for NGLY1 deficiency. Here, we determined the crystal structure of bovine FBS2 complexed with the adaptor protein SKP1 and a sugar ligand, Man3GlcNAc2, which corresponds to the core pentasaccharide of N‐glycan. Our crystallographic data together with NMR data revealed the structural basis of disparate sugar‐binding specificities in homologous FBS proteins and identified a potential druggable pocket for in silico docking studies. Abstract

Early detection and treatment of diseases are crucial in preventing and controlling their progression. Developing methods for a super-early screening is highly desirable. We have continuously conducted health survey sessions for elderly adults, and have collected information on their health, such as their living environment and physical functions. This paper presents the design of the measurement session. At the latest measurement session, we focus on DLB (Dementia with Lewy Bodies). We then try to construct a screening method for Parkinsonism, which is one of the prodromes of DLB. In the survey, we collected not only health information but also gait measurements. It attempts to realize a highly accurate screening tool with modality fusion; each modality is based on the gait analysis, in addition to measurements, questionnaires, and interviews. Next, we show the difference between the active and inactive groups in the last five sessions of information. The results show each of the two groups made contact with different types of communities. The results also show that the active group excelled in activities of daily living and working memory and had few concerns about dementia. [ABSTRACT FROM AUTHOR]

44. Correlations of sarcopenia with phase angle and Timed Up and Go test in patients on maintenance hemodialysis

Abstract

Objective: This study aimed to investigate the correlations of sarcopenia with phase angle（PhA）and timed up and go test（TUG）in patients on maintenance hemodialysis（MHD）. Methods: A total of 133 patients（median age = 67.3［59.3-72.7］years, 89 males）who underwent hemodialysis 2-3 times per week between July and September 2019 at Oyokyo Hirosaki Hospital were included. Subject demographic and clinical data were collected. Handgrip strength（HGS）and TUG were measured. Body composition was measured using bioelectrical impedance analysis to determine appendicular skeletal muscle mass（ASM）and PhA. Diagnostic criteria from the Asian Working Group for Sarcopenia 2019（AWGS2019）were used for confirming sarcopenia. Logistic regression analysis was used to examine the factors associated with sarcopenia. Single correlation analysis and multiple regression analysis were used to examine the factors associated with PhA and TUG. Results: The prevalence of sarcopenia was 37.6%. Predictors for sarcopenia were age, body mass index（BMI）, and PhA. Factors associated with PhA were HGS, creatinine, the geriatric nutritional risk index（GNRI）, and HD vintage, while factors associated with TUG were age, BMI, and PhA. Conclusion: PhA is a predictor of both sarcopenia and TUG, and the predictors of TUG are the same as those of sarcopenia.
