Abstract

Copper (Cu) is a critical element for cancer cell proliferation and considerably accumulates in the nucleus. 64Cu2+ is an anticancer radiopharmaceutical that targets the copper requirement of cancer cells. However, intravenously injected 64Cu2+ ions primarily accumulate in the liver. Ligand complexation of 64Cu2+ may be a promising method for increasing tumor delivery by reducing liver uptake. In this study, we used three tripodal amine ligands [tris(2-aminoethyl)amine (Tren), diethylenetriamine (Dien), and tris(2-pyridylmethyl)amine (TPMA)] to enclose 64Cu2+ ions and compared their in vivo tumor and liver uptakes using a tumor-bearing xenograft mouse model of the extrahepatic bile duct carcinoma cell line TFK-1. We examined intracellular Cu distribution using microparticle-induced X-ray emission (micro-PIXE) analysis of these compounds. 64Cu2+-Tren and 64Cu2+-Dien showed higher tumor uptake than 64Cu2+-TPMA and 64Cu2+ ions in TFK-1 tumors. Among the three 64Cu2+ complexes and 64Cu2+ ions, liver uptake was inversely correlated with tumor uptake. Micro-PIXE analysis showed that in vitro cellular uptake was similar to in vivo tumor uptake, and nuclear delivery was the highest for 64Cu2+-Tren. Conclusively, an inverse correlation between tumor and liver uptake was observed using three 64Cu2+ complexes of tripodal amine ligands and 64Cu2+ ions. Abstract

[64Cu]Cu-diacetyl-bis(N4-methylthiosemicarbazone) ([64Cu]Cu-ATSM) is a radioactive hypoxia-targeting therapeutic agent being investigated in clinical trials for malignant brain tumors. For the quality management of [64Cu]Cu-ATSM, understanding trace metal impurities' effects on the chelate formation of 64Cu and ATSM is important. In this study, we conducted coordination chemistry studies on metal–ATSM complexes. First, the effects of nonradioactive metal ions (Cu2+, Ni2+, Zn2+, and Fe2+) on the formation of [64Cu]Cu-ATSM were evaluated. When the amount of Cu2+ or Ni2+ added was 1.2 mol or 288 mol, equivalent to ATSM, the labeling yield of [64Cu]Cu-ATSM fell below 90%. Little effect was observed even when excess amounts of Zn2+ or Fe2+ were added to the ATSM. Second, these metals were reacted with ATSM, and chelate formation was measured using ultraviolet–visible (UV-Vis) absorption spectra. UV-Vis spectra showed a rapid formation of Cu2+ and the ATSM complex upon mixing. The rate of chelate formation by Ni2+ and ATSM was lower than that by Cu-ATSM. Zn2+ and Fe2+ showed much slower reactions with the ATSM than Ni2+. Trace amounts of Ni2+, Zn2+, and Fe2+ showed little effect on [64Cu]Cu-ATSM' quality, while the concentration of impurity Cu2+ must be controlled. 30. Impact of Using Facebook as a Social Learning Platform to Connect High School Students with Working Adults

Abstract

This study examines "the Socla study program" to build a social learning community for high school students using Facebook and other Internet services. In the two-week program, the students worked on individual study projects that focused on their future plans. With the help of volunteer supporters and facilitators, the students found relevant information and received constructive feedback about their progress. We investigated how the program was accepted by the students and how it affected their views on the future using pre- and post-survey data. The results indicated that the students (a) viewed their own future more positively, (b) realized that learning about unknown subjects can be interesting, and (c) discovered that advice from their superiors was useful. Responses from the volunteer supporters showed that the program also worked as a reflective learning opportunity for them. Abstract

先進核医学基盤研究部では、臨床診断・治療や生体機能の計測に有用な放射性核種標識薬剤の開発、標識薬剤合成に必要な放射性核種の製造及び標識・分析技術の開発研究を行っている。また、新規放射性薬剤を開発し、多種多様な動物モデルを含めた動物実験による薬剤の有効性と前臨床評価、臨床共同研究を実施している。さらに、安全で高品位な標識核種及び診断・治療用の放射性核種を製造し、脳機能イメージング研究部、分子イメージング診断治療研究部のみならずQST病院や外部の大学・研究機関・企業の研究者に広く提供している。 当研究部によって製造される臨床研究用の放射性薬剤は、1) HIMACや量子メスを用いた腫瘍の治療効果の評価や転移の有無などの判定、2) がん患者への診断・治療研究、3) 認知症をはじめとする各種の脳疾患の診断、治療効果の評価及び病態発生メカニズムの解明研究などに利用されている。本報告書では、新規標識薬剤の開発状況及び放射性核種・薬剤の製造状況を報告する。

39. Harnessing PD-L1 interface peptide for positron emission tomography imaging of the PD-1 immune checkpoint

Abstract

Interface peptides that mediate protein-protein interactions (PPI) are a class of important lead compounds for designing PPI inhibitors. However, Their potential as precursors for radiotracers has never been exploited. Here we reported that the interface peptides from programmed death-ligand 1 (PD-L1) can be used in positron emission tomography (PET) imaging of the programmed cell death 1 (PD-1) with high accuracy and sensitivity. Moreover, the performance differentiation between murine PD-L1 derived interface peptide (mPep-1) and human PD-L1 derived interface peptide (hPep-1) as PET tracers for PD-1 unveiled an unprecedented role of a non-critical residue in target binding, highlighting of the significance of PET imaging as a companion diagnostics in drug development. Collectively, this study not only provided a first-of-its-kind peptide-based PET tracer for PD-1 but also conveyed a unique paradigm for developing imaging agents for highly challenging protein targets, which could be used to identify other protein biomarkers involved in the PPI networks.

40. Short-Term Magnesium Deficiency Triggers Nutrient Retranslocation in Arabidopsis thaliana

Abstract

Magnesium (Mg) is essential for many biological processes in plant cells, and its deficiency causes yield reduction in crop systems. Low Mg status reportedly affects photosynthesis, sucrose partitioning and biomass allocation. However, earlier physiological responses to Mg deficiency are scarcely described. Here, we report that Mg deficiency in Arabidopsis thaliana first modified the mineral profile in mature leaves within 1 or 2 days, then affected sucrose partitioning after 4 days, and net photosynthesis and biomass production after 6 days. The short-term Mg deficiency reduced the contents of phosphorus (P), potassium, manganese, zinc and molybdenum in mature but not in expanding (young) leaves. While P content decreased in mature leaves, P transport from roots to mature leaves was not affected, indicating that Mg deficiency triggered retranslocation of the mineral nutrients from mature leaves. A global transcriptome analysis revealed that Mg deficiency triggered the expression of genes involved in defence response in young leaves.
