Hydrangea spp. is renowned for its variety of color changes in its developmental stage and before and after aluminum treatment. We analyzed gene expression in hydrangeas sepals to study the causes of color change. The accuracy of quantitative RT-qPCR analysis depends on the reliability of reference genes. We selected reference genes for hydrangea of varying cultivars, at different developmental stages, and in aluminum treatment groups. We chose ‘Bailmer’ and ‘Duro’ as subject species. We selected eight candidate genes, all of which were ranked by geNorm, NormFinder, BestKeeper, and RefFinder. CCR, NHX1, and LODX were used to verify the exactitude of reference genes. According to the ranking result of RefFinder, the top-ranked reference genes in each group were different; the top four candidate reference genes in each group mostly included EF1-β, RPL34, GADPH, and RPL10. EF1-β and RPL34 ranked top in the ‘all materials’ group, and their expression trends, obtained from the analysis of CCR, NHX1, and LODX, were consistent. From the results, we gather that EF1-β and RPL34 can be used as reference genes to quantify target gene expression. In this study, we screened for reference genes in hydrangeas to provide a technical basis for hydrangea sepal formation and transformation for further experiments.

Lilium L. is a perennial ornamental bulbous species, belonging to Liliaceae family, which consists of about 100 species. One of the most important hybrids in Lilium L. is the Oriental hybrid lily. Different cross combinations have been done in the lily family such as AA (Asiatic × Asiatic), AL (Asiatic × Longiflorum), and OT (Oriental × Trumpet). The OO (Oriental × Oriental) combination is a new one. SSR and AFLP markers were used to overlap each other and the genetic linkage groups were created according to the haploid number of lily chromosomes (12 linkage groups). In this experiment, the final F(1) population, which creates a genetic linkage group, was 100 individuals. For map construction, JOINMAP 4.0 software by treating segregation data of markers as a CP (out breeder full-sib family) model was used. After evaluation of ornamental traits, MapQTL 4.0 software was also used to find possible QTLs on these linkage maps. A total of 940 primers were tested and the best ones, which were 172 primer pairs (96 AFLP and 76 SSR markers), were used for map construction and the total of 616 loci (465 loci for AFLP marker and 151 loci for SSR marker) were scored. The entire mapped length was 2144.2 cM. 8 QTLs were obtained for flower number which is an important trait in lily. Each QTL locus explained the phenotypic variation of 2.4–89.5%. The highest amount of LOD (35.21) was found in LG-F1P2 for FN4 QTL. For leaf number, one-QTL was mapped with LOD of 7.08 between 2 markers on the LG-M10 of maternal maps. The QTL for petal length was placed on the LG-F1P2 of the F1 hybrid maps on the E-CGC/M-CGC-4 primer combination. The petal width QTLs also were mapped on the E-CGC/M-CGC-4. Qualitative locus named LN was mapped on the LG-M10 of the maternal maps. PW2 QTL was also localized on the LG-F4 of the paternal maps. In this experiment, 5 QTLs also were mapped for spot number in all F(1) hybrids and paternal and maternal maps, and spot size. Moreover, one QTL with the length of 51 cM was measured on the LG-M8 of the maternal maps. Plant height QTL with the LOD of 12.54 was mapped on the primer combination of E-CGC/M-CGC-4 on the LG-F1P2 of the F(1) hybrid maps.

Photosynthesis is closely related to the growth of plants. A stable reference gene is fundamental for studies of the molecular mechanism of photosynthesis in Lilium regale. Therefore, it is very important to select a suitable reference gene for qRT-PCR analysis on genes of photosynthetic system, chlorophyll biosynthetic pathway and chloroplast development in Lilium regale. Three kinds of tissues, leaves and bulbs (abnormal leaves) of tissue culture plantlets and cotyledons of seedlings of the wild-type and mutant Lilium regale were selected as materials for qRT-PCR. Six housekeeping genes were selected as candidate genes from transcriptome sequencing data of the wild-type and yellow seedling lethal mutant of Lilium regale. Finally, the expression stability of six candidate reference genes was analyzed using geNorm, NormFinder, and BestKeeper software, the comparative ∆Ct method, and the RefFinder program. The results showed that LrActin2 was the best reference gene for qRT-PCR analysis of photosynthesis-related genes expression in leaves of tissue culture plantlets and seedlings of Lilium regale. This study provided useful data for further research on molecular mechanism of photosynthesis in the Lilium.

Gene function analysis is challenging for Lilium due to the lack of an efficient method for stable genetic transformation. Virus-induced gene silencing (VIGS) is an attractive tool for determining gene function in plants. This study reported that Tobacco rattle virus (TRV)-based VIGS of a PHYTOENE DESATURASE (PDS) ortholog (LhPDS) can be achieved in Lilium × formolongi seedlings, with a survival rate of 92%, using the inoculation method of rubbing plus injection. Compared with untreated and mock-treated seedlings, the photobleached leaf phenotype of silenced plants significantly correlates with down-regulation of endogenous LhPDS (P ≤ 0.05). In addition, the silencing phenomenon can be observed in the growing points of plants, indicating that systemic viral infection was achieved using the protocol. The results indicate that TRV-based VIGS can be used to characterize gene function in Lilium × formolongi. This work lays the foundation for gene function analysis and molecular breeding in Lilium spp. Keywords: Lilium, photobleaching, seedling, TRV, VIGS, PDS

Glucoraphanin is a major secondary metabolite found in Brassicaceae vegetables, especially broccoli, and its degradation product sulforaphane plays an essential role in anticancer. The fine mapping of sulforaphane metabolism quantitative trait loci (QTLs) in broccoli florets is necessary for future marker-assisted selection strategies. In this study, we utilized a doubled haploid population consisting of 176 lines derived from two inbred lines (86,101 and 90,196) with significant differences in sulforaphane content, coupled with extensive genotypic and phenotypic data from two independent environments. A linkage map consisting of 438 simple sequence repeats markers was constructed, covering a length of 1168.26 cM. A total of 18 QTLs for sulforaphane metabolism in broccoli florets were detected, 10 were detected in 2017, and the other 8 were detected in 2018. The LOD values of all QTLs ranged from 3.06 to 14.47, explaining 1.74–7.03% of the biochemical variation between two years. Finally, 6 QTLs (qSF-C3-1, qSF-C3-2, qSF-C3-3, qSF-C3-5, qSF-C3-6 and qSF-C7) were stably detected in more than one environment, each accounting for 4.54–7.03% of the phenotypic variation explained (PVE) and a total of 30.88–34.86% of PVE. Our study provides new insights into sulforaphane metabolism in broccoli florets and marker-assisted selection breeding in Brassica oleracea crops.

Lilium davidii var. unicolor Salisb is a cultivar of Lilium (Liliaceae) with important edible and ornamental characteristic. The application and production of Lilium davidii var. unicolor Salisb were still facing large problems because of its several disadvantages such as narrow range of adaptability, small annual growth increment, and low fertility. To achieve broader environmental adaptability and obtain a more nutritious germplasm, we used colchicine and oryzalin to induce chromosome doubling via the soaking method. Tissue culture bulbs were treated with colchicine at 0.03%, 0.05%, or 0.08% for 32, 40, or 48 hours or with oryzalin at 0.002%, 0.005%, 0.008%, or 0.01% for 3, 6, 9, 12, or 24 hours before being transferred to a differentiating medium. The results showed that colchicine treatment resulted in the highest induction rate when applied at 0.05% for 48 hours, whereas oryzalin treatment produced fewer tetraploid plants. The chromosome number of induced plants with small stoma density and longer guard cells is twice than that of the diploid. The plants were identified as tetraploid. In this study, a new germplasm of Lilium davidii var. unicolor Salisb was innovative and showed novel genetic characteristic.

Leaf color mutants are ideal materials for exploring plant photosynthesis mechanisms, chlorophyll biosynthetic pathways and chloroplast development. The yellow seedling lethal mutant lrysl1 was discovered from self-bred progenies of Lilium regale; however, the mechanism of leaf color mutation remains unclear. In this study, the ultrastructural and physiological features and de novo RNA-Seq data of a L. regale leaf color mutant and wild-type L. regale were investigated. Genetic analysis indicated that the characteristics of the lrysl1 mutant were controlled by a recessive nuclear gene. The chlorophyll a, chlorophyll b and carotenoid contents in the mutant leaves were lower than those in the wild-type leaves. Furthermore, the contents of the chlorophyll precursors aminolevulinic acid (ALA), porphobilinogen (PBG), protoporphyrin IX (ProtoIX), Mg-protoporphyrin IX (Mg-ProtoIX), and protochlorophyll (Pchl) decreased significantly in mutant leaves. Transcriptome data from the mutant and wild type showed that a total of 892 differentially expressed genes were obtained, of which 668 and 224 were upregulated genes and downregulated genes in the mutant, respectively. Almost all genes in the photosynthesis pathway and chlorophyll biosynthetic pathway were downregulated in the mutant, which corroborated the differences in the physiological features mentioned above. Further research indicated that the chloroplasts of the mutant leaves exhibited an abnormal morphology and distribution and that the expression of a gene related to chloroplast development was downregulated. It was concluded that abnormal chloroplast development was the main cause of leaf color mutation in the mutant lrysl1 and that LrGLK was a gene related to chloroplast development in L. regale. This research provides a foundation for further research on the mechanism by which LrGLK regulates chloroplast development in L. regale.

The planting density of broccoli can directly affect the yield and overall health of plants. Nonetheless, studies on the genetic base of planting density in broccoli are rather limited. In this study, the important planting density-dependent factors of broccoli, plant height (PH), maximum outer petiole length (PL) and leaf width (LW), were investigated during 2017 and 2018. In this study, mapping of QTL for PH, PL and LW based on a DH population in broccoli, and as well as the interaction between QTLs and the environment. The DH population was constructed with 176 genotypes obtained from F1 hybrid produced by crossing two different inbred lines 86101 (P1) and 90196 (P2). In our study, a linkage group including a total of 438 SSR markers were constructed covering a length of 1168.26 cM using QTL IciMapping 4.0 software. Finally, there were mainly four QTLs (phc1, phc2, phc4−2, phc4−3), one QTL (plc6−2), and two QTLs (lwc1, lwc3−1) corresponding for PH, PL and LW recurred during the two years. In three environments, inclusive composite interval mapping (ICIM) analysis showed that there was a major QTL for PH at 7.20 cM on chromosome 1 with a high explanatory contribution rate of 20.05 %. The QTL at the 11.10 cM position of chromosome 6 was located for the PL with a high explanatory contribution rate of 20.02 %. The QTL at the 147.00 cM position of chromosome 3 was located on LW with a high explanatory contribution rate of 19.97 %. Interestingly, we found an overlap region (pplc4) in the chromosome 4 containing three QLTs for traits PH, PL, and LW (phc4−2, plc4−3, and lwc4−2). According to this study, the possible positions of candidate genes were screened to provide a basis for further locating and cloning genes for plant height, maximum outer petiole and leaf width, which would be beneficial to breeding of broccoli and the Brassica crops, especially in high planting density.

Virus-induced gene silencing (VIGS) is an attractive tool for determining gene function in plants. The present study constitutes the first application of VIGS in S. pseudocapsicum, which has great ornamental and pharmaceutical value, using tobacco rattle virus (TRV) vectors. Two marker genes, PHYTOENE DESATURASE (PDS) and Mg-chelatase H subunit (ChlH), were used to test the VIGS system in S. pseudocapsicum. The photobleaching and yellow-leaf phenotypes of the silenced plants were shown to significantly correlate with the down-regulation of endogenous SpPDS and SpChlH, respectively (P ≤ 0.05). Moreover, the parameters potentially affecting the efficiency of VIGS in S. pseudocapsicum, including the Agrobacterium strain and the inoculation method (leaf syringe-infiltration, sprout vacuum-infiltration and seed vacuum-infiltration), were compared. The optimized VIGS parameters were the leaf syringe-infiltration method, the Agrobacterium strain GV3101 and the growth of agro-inoculated plants at 25°. With these parameters, the silencing efficiency of SpPDS and SpChlH could reach approximately 50% in S. pseudocapsicum. Additionally, the suitability of various reference genes was screened by RT-qPCR using three candidate genes, and the results demonstrated that glyceraldehyde 3-phosphate dehydrogenase (GAPDH) can serve as a suitable reference for assessing the gene expression levels of VIGS systems in S. pseudocapsicum. The proven application of VIGS in S. pseudocapsicum and the characterization of a suitable reference gene in the present work will expedite the functional characterization of novel genes in S. pseudocapsicum.

Pollen viability and stigma receptivity are prerequisites for successful pollination and seed set in flowering plants. In this study, the pollen viabilities and stigma receptivities of nine Lilium genotypes (six cultivars and three species native to China) were assayed by in vitro pollen germination and the benzidine-H2O2 method, respectively. Embryo sac development during anthesis was observed to further ensure the timing of controlled pollination. In addition, the relationship between stigma secretion and stigma receptivity was studied to estimate the pollination time based on phenotype. Anthers cracked on the day of flowering in all genotypes, but pollen germination during anthesis was not observed in Asiatic hybrids excepted for ‘Tiny pudhye’, which exhibited low pollen viability for a short period of time (from 0 to 1 day after anthesis). In the other genotypes, pollen germination rates were highest on anthesis (five of seven genotypes), 0–1 day after anthesis (L. sulphureum), or 0–2 days after anthesis (one Longiflorum hybrid), and then gradually decreased with days after anthesis. While, stigma receptivity first increased and then decreased during anthesis. For most genotypes, stigmas began to be receptive 1 day after anthesis, and all genotypes exhibited stigma receptivity at 2 days after anthesis. The durations of stigma receptivity and strongest stigma receptivity, were genotype dependent, and were 5–8 days and 1–4 days, respectively. Moreover, on the first flowering day, 6 of 7 genotypes had mature embryo sacs, and at the time at which stigmas began to be receptive, all tested genotypes had mature embryo sacs. Some Lilium genotypes showed stigma secretion, which can be a sign of stigma receptivity. Stigmas became receptive and reached highest receptivity within 1 day of the first appearance of secretion on the surface of the stigma and at peaking, respectively. The results of this study are valuable for the implementation of successful Lilium breeding programs.

The bicolor Asiatic hybrid lily cultivar "Tiny Padhye" is an attractive variety because of its unique color pattern. During its bicolor tepal development, the upper tepals undergo a rapid color change from green to white, while the tepal bases change from green to purple. However, the molecular mechanisms underlying these changes remain largely uncharacterized. To systematically investigate the dynamics of the lily bicolor tepal transcriptome during development, we generated 15 RNA-seq libraries from the upper tepals (S2-U) and basal tepals (S1-D, S2-D, S3-D, and S4-D) of Lilium "Tiny Padhye." Utilizing the Illumina platform, a total of 295,787 unigenes were obtained from 713.12 million high-quality paired-end reads. A total of 16,182 unigenes were identified as differentially expressed genes during tepal development. Using Kyoto Encyclopedia of Genes and Genomes pathway analysis, candidate genes involved in the anthocyanin biosynthetic pathway (61 unigenes), and chlorophyll metabolic pathway (106 unigenes) were identified. Further analyses showed that most anthocyanin biosynthesis genes were transcribed coordinately in the tepal bases, but not in the upper tepals, suggesting that the bicolor trait of "Tiny Padhye" tepals is caused by the transcriptional regulation of anthocyanin biosynthetic genes. Meanwhile, the high expression level of chlorophyll degradation genes and low expression level of chlorophyll biosynthetic genes resulted in the absence of chlorophylls from "Tiny Padhye" tepals after flowering. Transcription factors putatively involved in the anthocyanin biosynthetic pathway and chlorophyll metabolism in lilies were identified using a weighted gene co-expression network analysis and their possible roles in lily bicolor tepal development were discussed. In conclusion, these extensive transcriptome data provide a platform for elucidating the molecular mechanisms of bicolor tepals in lilies and provide a basis for similar research in other closely related species.

To investigate the transcriptional network that governs purple ovary coloration in Asiatic hybrid lilies, we obtained transcriptome data from green ovaries (S1) and purple ovaries (S2) of Asiatic “Tiny Padhye”. Comparative transcriptome analysis revealed 4228 differentially expressed genes. Differential expression analysis revealed that ten unigenes including four CHS genes, one CHI gene, one F3H gene, one F3′H gene, one DFR gene, one UFGT gene, and one 3RT gene were significantly up-regulated in purple ovaries. One MYB gene, LhMYB12-Lat, was identified as a key transcription factor determining the distribution of anthocyanins in Asiatic hybrid lily ovaries. Further qPCR results showed unigenes related to anthocyanin biosynthesis were highly expressed in purple ovaries of three purple-ovaried Asiatic hybrid lilies at stages 2 and 3, while they showed an extremely low level of expression in ovaries of three green-ovaried Asiatic hybrid lilies during all developmental stages. In addition, shading treatment significantly decreased pigment accumulation by suppressing the expression of several unigenes related to anthocyanin biosynthesis in ovaries of Asiatic “Tiny Padhye”. Lastly, a total of 15,048 Simple Sequence Repeats (SSRs) were identified in 13,710 sequences, and primer pairs for SSRs were designed. The results could further our understanding of the molecular mechanisms of anthocyanin biosynthesis in Asiatic hybrid lily ovaries.

Simple-sequence repeat (SSR) markers are a common choice for assaying genetic diversity and genetic mapping due to their highly polymorphic and co-dominant nature. In this paper, we developed SSR markers derived from expressed sequence tags (EST-SSRs) based on transcriptome sequences in Lilium regale. In total, 59,046 UniGene sequences (approximately 27.37 Mb) were evaluated, where 1,716 SSR sites in the 1,606 UniGene sequences were identified by data mining. Of these SSRs, the trinucleotide repeats were the most common repeat types (631, 36.8 %), followed by hexa- (23.7 %), di- (23.4 %), penta- (11.5 %), tetra- (3.3 %), hepta- (0.9 %), deca- (0.3 %), and octanucleotide repeats (0.1 %). TC/GA and CCG/CGG predominated in dimeric and trimeric repeat motifs, respectively. In total, 1,072 primer pairs were designed, of which 494 were amplified to yield the expected PCR products in L. regale. The polymorphism and transferability study was performed in six species endemic to China and ten cultivars derived from intraspecific and interspecific hybridization. Of the 494 primer pairs, 172 exhibited clear polymorphisms with stable cross-species amplifications in the 16 individuals, which contained 537 alleles for the 181 loci. The polymorphism information content values for the 172 primers ranged from 0.111 to 0.830, with an average value of 0.493. The phylogenetic dendrogram derived from the amplification profiles of the 172 polymorphic primers was congruent with the genetic background of the hybrids and the species. In addition, the putative functions of the 109 UniGenes containing the 118 polymorphic and transferable EST-SSRs were identified. Thus, the informative EST-SSR markers will be very useful in subsequent Lilium genetic improvement projects.

Pancreatic hydrolysis of bovine casein was carried out at pH 8.0 in a batch stirred tank reactor. The resulting peptides were identified by liquid chromatography–tandem mass spectrometry and their release kinetics were tracked. During 24 h of digestion, a total of 297 peptides consisting of 109, 48, 126, and 14 peptides, from α s1 -, α s2 -, β-, and κ-caseins (α s1 -, α s2 -, β-, and κ-CN), respectively, were identified by LC–MS–MS. We found that pancreatin attacked different caseins with varying hydrolysis rates (β-CN > α s1 -CN > α s2 -CN > κ-CN). Based on our analysis of the peptide fragments produced, we determined that pancreatin preferentially cleaved the N-terminal phosphorylated region, the C-terminal hydrophobic region and the middle region of β-casein, the terminal regions of α s1 -casein, and both the terminals and the middle region of α s2 -casein. The hydrolysis of κ-casein occurred at the slowest rate, releasing peptides only after 30 min of hydrolysis.

The objective of this study was to improve induction of embryogenesis in white cabbage (Brassica oleracea var. capitata) microspore cultures. The effect of NLN-13 liquid medium pH on isolated microspore embryogenesis was investigated in five white cabbage genotypes. Relatively high pH (6.2 or 6.4) was more effective on microspore embryogenesis in most of the white cabbage genotypes than the pH of 5.8, especially for inducing microspore-derived embryos in recalcitrant genotype ‘Zhonggan No. 8’. Based on this, 2—(N-Morpholino) ethanesulfonic acid (MES) and the arabinogalactan-protein from gum arabic were tested on four out of five genotypes to see if they could increase embryo yield in microspore cultures. Adding MES or gum arabic alone was effective for these four genotypes, but the frequency of embryos derived from microspores was still low. However, the combination of 10 mg l−1 gum arabic and 3 mM MES in NLN-13 at pH 6.4 significantly enhanced microspore embryogenesis efficiency (with embryo production of 4.57–222.97 embryos per bud), especially with recalcitrant genotype “Zhonggan No. 8” for which it was increased by about 35-fold.

A prediction model of the peptide retention time was developed on the basis of 358 non-phosphorylated peptides, which had various amino acid residues in the C-terminal from the casein pancreatic hydrolysates. This model was applied to predict the retention times (RT) of another 43 peptides on C4 reversed phase columns with trifluoroacetic acid (TFA) as the ion-pairing reagent, with a relatively high R2 value (0.969). Furthermore, the experimental RTs of 32 phosphopeptides were compared with the predictive RTs of their non-phosphorylated cognates. Mono- and poly-phosphopeptides seemed to elute after or before the non-phosphorylated predictive cognates, respectively. The positive charges of peptides were partly masked by the ion-pairing reagent TFA. Single and multiple phosphorylation might generally lead to an increase or reduction in the overall hydrophobicity of peptides, respectively.

The relationship between the ploidy level of microspore-derived plants and chloroplast number in stomatal guard cells was studied in cabbage, broccoli, and Chinese kale. In the experiment, distribution statistics analysis and t-test were used to perform statistical analysis on chloroplast number of different ploidy level in those stomatal guard cells mentioned above, and morphology identifying and chromosome counting were used to test accuracy of counting chloroplast number in stomatal guard cells. The chloroplast average number in stomatal guard cells was very similar among the different leaf positions on the same plant and among the different locations in the same leaf, while the chloroplast number varied significantly among the different ploidy stoma in the same variety. All the distributions of the chloroplast number in different ploidy stoma were normal distribution fitted. A correlation has been established between ploidy and chloroplast number in the stomatal guard cells. In every single stoma of microspore-derived plants, the chloroplast number for a haploid should not be more than 10, diploids 11 to 15, and polyploids more than 15. The accuracy of this method for identification of different ploidy plants was 93.93%. Furthermore, the accuracy of this method was reliable and did not vary with the plants growth conditions. Therefore, the chromosome ploidy of plants derived from microspore culture in cabbage, broccoli, and Chinese kale can be identified by simply counting the chloroplast number in stomatal guard cells.

Bovine casein was digested with pancreatin at pH 8.0 in a batched stirred tank reactor. In total, 52 casein phosphopeptides (CPPs) in the time-course samples were separated and identified using liquid chromatography-tandem mass spectrometry by means of neutral loss scanning and database searching. Of these peptides, 23, 7, 8, 10 and 4 are singly, doubly, triply, quadruply and quintuply phosphorylated, respectively. Furthermore, 14 peptides contained the cluster sequence, S(P)S(P)S(P)EE, providing the mineral binding sites; these peptides were all formed after 10 min of hydrolysis, but none of them survived after 6 h of pancreatic digestion. A study of the release kinetics of CPPs allowed determination of the degrees of hydrolysis for the preparation of target peptides with high yields. The discrimination of pancreatic attack on different phosphorylated regions of bovine casein was also analyzed in terms of the release of CPPs.

Chromosome doubling of microspore-derived plants is an important factor in the practical application of microspore culture technology because breeding programs require a large number of genetically stable, homozygous doubled haploid plants with a high level of fertility. In the present paper, 29 populations of microspore-derived plantlets from cabbage (Brassica oleracea var. capitata) and broccoli (Brassica oleracea var. italica) were used to study the ploidy level and spontaneous chromosome doubling of these populations, the artificial chromosome doubling induced by colchicine, and the influence of tissue culture duration on the chromosomal ploidy of the microspore-derived regenerants. Spontaneous chromosome doubling occurred randomly and was genotype dependent. In the plant populations derived from microspores, there were haploids, diploids, and even a low frequency of polyploids and mixed-ploidy plantlets. The total spontaneous doubling in the 14 cabbage populations ranged from 0 - 76.9%, compared with 52.2 - 100% in the 15 broccoli populations. To improve the rate of chromosome doubling, an efficient and reliable artificial chromosome doubling protocol (i.e., the immersion of haploid plantlet roots in a colchicine solution) was developed for cabbage and broccoli microspore-derived haploids. The optimal chromosome doubling of the haploids was obtained with a solution of 0.2% colchicine for 9 - 12 h or 0.4% colchicine for 3 - 9 h for cabbage and 0.05% colchicine for 6 - 12 h for broccoli. This protocol produced chromosome doubling in over 50% of the haploid genotypes for most of the populations derived from cabbage and broccoli. Notably, after 1 or more years in tissue culture, the chromosomes of the haploids were doubled, and most of the haploids turned into doubled haploid or mixed-ploidy plants. This is the first report indicating that tissue culture duration can change the chromosomal ploidy of microspore-derived regenerants.

BackgroundBrassica oleraceaencompass a family of vegetables and cabbage that are among the most widely cultivated crops. In 2009, theB. oleraceaGenome Sequencing Project was launched using next generation sequencing technology. None of the available maps were detailed enough to anchor the sequence scaffolds for the Genome Sequencing Project. This report describes the development of a large number of SSR and SNP markers from the whole genome shotgun sequence data ofB. oleracea, and the construction of a high-density genetic linkage map using a double haploid mapping population.ResultsTheB. oleraceahigh-density genetic linkage map that was constructed includes 1,227 markers in nine linkage groups spanning a total of 1197.9 cM with an average of 0.98 cM between adjacent loci. There were 602 SSR markers and 625 SNP markers on the map. The chromosome with the highest number of markers (186) was C03, and the chromosome with smallest number of markers (99) was C09.ConclusionsThis first high-density map allowed the assembled scaffolds to be anchored to pseudochromosomes. The map also provides useful information for positional cloning, molecular breeding, and integration of information of genes and traits inB. oleracea. All the markers on the map will be transferable and could be used for the construction of other genetic maps.
