Abstract

Research using alfalfa seedlings indicates that plants may not be especially vulnerable to the DNA-damaging effects of increasing ultraviolet radiation due to diminishing stratospheric ozone. The action spectrum or wavelength-dependent sensitivity at which alfalfa DNA-injury occurs in the form of cyclobutyl pyrimidine dimer induction was found to be at the unexpectedly long wavelength of 365 nm rather than 280 to 313 nm as had been postulated in previous forecasts of how ozone depletion will affect plants.

10. Advanced regression methods for combustion modelling using principal components

Abstract

Modelling the physics of combustion remains a challenge due to a large range of temporal and physical scales which are important in these systems. Detailed chemical kinetic mechanisms are used to describe the chemistry involved in the combustion process yielding highly coupled partial differential equations for each of the chemical species used in the mechanism. Recently, Principal Components Analysis (PCA) has shown promise in its ability to identify a low-dimensional manifold describing the reacting system. Several PCA-based models have been developed which may be well-suited for combustion problems; however, several challenging aspects of the model must be addressed. In this paper, the parameterization of state-space variables and PC-transport equation source terms are investigated. The ability to achieve highly accurate mapping through various nonlinear regression methods is shown. In addition, the effect of PCA-scaling on the ability to regress the surface is investigated. Abstract

The action spectrum for photoreactivation by enzymes from human leukocytes and fibroblasts extends from 300 to approximately 600 nm with a maximum near 400 nm. The ability of the human enzymes to utilize light of wavelengths greater than 500 nm suggested that yellow or gold lights conventionally used as safelights for photoreactivation might serve as sources of photoreactivating light for these enzymes. Experiments using lights with a range of spectral outputs confirm that the standard yellow “safe” lights do produce photoreactivation by the human but not the Escherichia coli enzyme.

32. Probes of DNA Structure and Interactions Effects of Copper II on Ultraviolet-Induced Pyrimidine Dimer Formation

Abstract

Cu(II) affects the yield of cyclobutyl dimers induced in DNA by 254 nm radiation. The effects are a function of r, the ratio of Cu(II) to DNA phosphate, and of the ultraviolet (UV) fluence; they seem to reflect two types of copper complexes with DNA. The first probably involves “exterior” binding to the bases of native DNA and increases [unk]TT formation (without affecting [unk]UT yield) by raising the energy levels of bases other than thymine. The second seems to occur only at high ratios (rs) and only after the structure has been opened locally by UV radiation; it involves “interior” binding of Cu(II) to the bases. This complex tends to decrease dimer yield by holding the bases apart and/or by lowering the energy levels of bases other than thymine. These results illustrate the potential use of DNA photoproducts and ligands to probe the structure and interactions of DNA in vitro and perhaps also in vivo.

Abstract

In addition to the acridine dyes, acridine orange and proflavine, we find that three other cationic molecules which bind to DNA—ethidium bromide, chloroquine, and methyl green—inhibit the production of cyclobutyl pyrimidine dimers by ultraviolet radiation. Intercalation is not necessary for dimer inhibition. The long range nature of the inhibition implies that energy transfer is responsible. The transfer is between the lowest excited singlet state of DNA and the acceptor singlet, and seems to involve the Förster mechanism.
