High-biomass blooms of the toxic dinoflagellate Pyrodinium bahamense occur most summers in Tampa Bay, Florida, USA, posing a recurring threat to ecosystem health. Like many dinoflagellates, P. bahamense forms immobile resting cysts that can be deposited on the seafloor-creating a seed bank that can retain the organism within the ecosystem and initiate future blooms when cysts germinate. In this study, we examined changes in the dormancy status of cysts collected from Tampa Bay and applied lessons from plant ecology to explore dormancy controls. Pyrodinium bahamense cysts incubated immediately after field collection displayed a seasonal pattern in dormancy and germination that matched the pattern of cell abundance in the water column. Newly deposited (surface) cysts and older (buried) cysts exhibited similar germination patterns, suggesting that a common mechanism regulates dormancy expression in new and mature cysts. Extended cool- and warm-temperature conditioning of field-collected cysts altered the cycle of dormancy compared with that of cysts in nature, with the duration of cool temperature exposure being the best predictor of when cysts emerged from dormancy. Extended warm conditioning, on the other hand, elicited a return to dormancy, or secondary dormancy, in nondormant cysts. These results directly demonstrate environmental induction of secondary dormancy in dinoflagellates-a mechanism common and thoroughly documented in higher plants with seasonal growth cycles. Our findings support the hypothesis that a seasonal cycle in cyst germination drives P. bahamense bloom periodicity in Tampa Bay and point to environmentally induced secondary dormancy as an important regulatory factor of that cycle.

Recent increases in global urea usage, including its incorporation in slow-release fertilizers commonly used in lawn care in Florida, have the potential to alter the form and amount of nitrogen inputs to coastal waters. This shift may, in turn, impact phytoplankton community diversity and nutrient cycling processes. An autonomous water quality monitoring and sampling platform containing meteorological and water quality instrumentation, including urea and phycocyanin sensors, was deployed between June and November of 2009 in Sarasota Bay, Florida. This shallow, lagoonal bay is characterized by extensive and growing urban and suburban development and limited tidal exchange and freshwater inputs. During the monitoring period, three high-biomass (up to 40 &micro, g chlorophyll-a&middot, L&minus, 1) phytoplankton blooms dominated by picocyanobacteria or picoeukaryotes were observed. Each bloom was preceded by elevated (up to 20 &mu, M) urea concentrations. The geolocation of these three parameters suggests that &ldquo, finger canals&rdquo, lining the shore of Sarasota Bay were the source of urea pulses and there is a direct link between localized urea inputs and downstream picoplankton blooms. Furthermore, high frequency sampling is required to detect the response of plankton communities to pulsed events.

The ongoing restoration of the Florida Everglades has changed the hydrology in south Florida and increasing freshwater discharge has contributed to a shift in nutrients and phytoplankton in northern Florida Bay. To understand the effect of the changing nitrogen (N) forms and nitrogen:phosphorus (N:P) ratios on phytoplankton biomass and assemblage composition, five mesocosm experiments were conducted. Nutrient additions included a matrix of different forms of N (NO 3 − , NH 4 + , and DON) and P (as sodium β-glycerophosphate pentahydrate) in differing N:P molar ratios (4, 16, 32), a P alone addition, and a control. In general, chlorophyll a increased three-fold in the + P treatments and ten-fold in the + N + P treatments, while it did not increase in the + N treatments. Typically the + N + P treatments, particularly the + NO 3 − +P at the N:P molar ratio of 32, promoted high concentrations of fucoxanthin (generally indicative of diatoms) relative to chlorophyll a . While chlorophyll a did not increase significantly in the + N alone treatments, there was a change in the phytoplankton assemblage. The + N treatments, when N was in the form of + NH 4 + yielded proportionally higher zeaxanthin (generally indicative of picocyanobacteria). When the + N:P ratio increased, the relative concentrations of fucoxanthin and alloxanthin (generally indicative of cryptophytes) to chlorophyll a increased, whereas the relative concentrations of zeaxanthin and dinoflagellates (generally indicative of photosynthetic dinoflagellates) declined. This study highlights the importance of dual P and N control, particularly N in the forms of NH 4 + and DON if picocyanobacterial blooms are to be controlled.

The largest estuarine flow restoration project is the Comprehensive Everglades Restoration Plan (CERP). In 2012, one of its first phases, the C-111 Project, was initiated and designed to increase freshwater flow into northern Florida Bay. Effects of changes in flow and associated nutrients on phytoplankton abundance and composition were studied in two sets of interconnected, quasi-enclosed saline lakes at the southern end of the Everglades that discharge into northern Florida Bay. After C-111 implementation, total dissolved nitrogen (TDN) decreased, driven by especially large declines in NOx−. In spite of N reduction, those lakes that has been oligotrophic prior to the project experienced an approximate doubling of phytoplankton biomass as nutrient ratios converged on Redfield proportions, and a shift to smaller sized cells and a loss of diatoms as NOx− availability declined. The other set of these lakes, previously highly eutrophic, sustained a significant, nearly 50%, decline in overall phytoplankton biomass, particularly larger sized cells, which, in turn, increased downstream. A chemostat model was invoked to explain these changes based on changes in flow and nutrients. In both systems the proportions of picocyanobacteria increased in the upper lakes as the relative availability of chemically reduced forms of N increased. Both lake systems may export algal biomass, changing the phytoplankton assemblage in northern Florida Bay. These lakes may also serve as buffers, damping the effects in Florida Bay itself, and they illustrate broad implications for estuaries that are impacted by natural or anthropogenic changes in flow.

In the past four decades, consistent and coordinated management actions led to the recovery of Tampa Bay, FL (USA) – an estuary that was declared dead in the 1970s. An exception to this success story is Old Tampa Bay, the northernmost subestuary of the system. Compared to the other bay segments, Old Tampa Bay is characterized by poorer water quality and spring and summer blooms of cyanobacteria, picoplankton, diatoms, and the saxitoxin-producing dinoflagellate Pyrodinium bahamense. Together, these blooms contribute to light attenuation and lagging recovery of seagrass beds. Yet, studies of phytoplankton dynamics within Old Tampa Bay have been limited – both in number and in their spatiotemporal resolution. In this study, we used field sampling and continuous monitoring to (1) characterize temporal and spatial variability in phytoplankton biomass and community composition and (2) identify key drivers of the different phytoplankton blooms in Old Tampa Bay. Overall, temporal variability in phytoplankton biomass (using chlorophyll a as a proxy) and community composition surpassed spatial variability of these parameters. We found a base community of small diatoms and flagellates, as well as certain dinoflagellates, that persisted year round in the system. Seasonally, freshwater runoff stimulated phytoplankton growth, specifically that of chlorophytes, cyanobacteria and other dinoflagellates -- consistent with predictions based on ecological theory. On shorter time scales, salinity, visibility, and freshwater inflows were important predictors of phytoplankton biomass. With respect to P. bahamense, environmental drivers including salinity, temperature and dissolved nutrient concentrations explained ∼24% of the variability in cell abundance, indicating missing explanatory parameters in our study for this taxon, such as cyst density and location of cyst beds. Spatially, we found differences in community trajectories across north–south and west–east gradients, with the northernmost sampling station being the most unique in the region. This work contributes to the knowledge of phytoplankton biomass and community composition in Tampa Bay by generating spatially and temporally rich phytoplankton community and environmental data for the Old Tampa Bay subestuary. Moreover, it enhances our understanding of bloom drivers and provides recommendations for ecosystem management. Specifically, our findings support continued nutrient reduction measures as a way to mitigate seasonal blooms of diatoms, cyanobacteria and chlorophytes, but not necessarily blooms of P. bahamense. Prediction and mitigation of P. bahamnese blooms should incorporate first order drivers such as cyst location and abundance.

Identifying nutrient sources, primarily nitrogen (N) and phosphorus (P), sufficient to support high biomass blooms of the red tide dinoflagellate, Karenia brevis, has remained problematic. The West Florida Shelf is oligotrophic, yet populations >106 cells L−1 frequently occur and blooms can persist for months. Here we examine the magnitude and variety of sources for N and P that are available to support blooms. Annual average in situ or background concentrations of inorganic N in the region where blooms occur range 0.02–0.2 μM while inorganic P ranges 0.025–0.24 μM. Such concentrations would be sufficient to support the growth of populations up to ∼3×104 cells L−1 with at least a 1 d turnover rate. Organic N concentrations average 1–2 orders of magnitude greater than inorganic N, 8–14 μM while organic P concentrations average 0.2–0.5 μM. Concentrations of organic N are sufficient to support blooms >105 cells L−1 but the extent to which this complex mixture of N species is utilizable is unknown. Other sources of nutrients included in our analysis are aerial deposition, estuarine flux, benthic flux, zooplankton excretion, N2-fixation, and subsequent release of organic and inorganic N by Trichodesmium spp., and release of N and P from dead and decaying fish killed by the blooms. Inputs based on atmospheric deposition, benthic flux, and N2-fixation, were minor contributors to the flux required to support growth of populations >2.6×104 cells L−1. N and P from decaying fish could theoretically maintain populations at moderate concentrations but insufficient data on the flux and subsequent mixing rates does not allow us to calculate average values. Zooplankton excretion rates, based on measured zooplankton population estimates and excretion rates could also supply all of the N and P required to support populations of 105 and 106 cells L−1, respectively, but excretion is considered as “regenerated” nutrient input and can only maintain biomass rather than contribute to “new” biomass. The combined estuarine flux from Tampa Bay, Charlotte Harbor, and the Caloosahatchee River can supply a varying, but at times significant level of N and P to meet growth and photosynthesis requirements for populations of approximately 105 cells L−1 or below. Estimates of remineralization of dead fish could supply a significant proportion of bloom maintenance requirements but the rate of supply must still be determined. Overall, a combination of sources is required to maintain populations >106 cells L−1.

Blooms of the toxic dinoflagellate Karenia brevis are common in the Gulf of Mexico, yet no in situ studies of zooplankton and K. brevis have been conducted there. Zooplankton abundance and taxonomic composition at non-bloom and K. brevis bloom stations within the Ecology of Harmful Algal Blooms (ECOHAB) study area were compared. At non-bloom stations, the most abundant species of zooplankton were Parvocalanus crassirostris, Oithona colcarva, and Paracalanus quasimodo at the 5-m isobath and P. quasimodo, O. colcarva, and Oikopleura dioica at the 25-m isobath. There was considerable overlap in dominance of zooplankton species between the 5 and 25-m isobaths, with nine species contributing to 90% of abundance at both isobaths. At stations within K. brevis blooms however, Acartia tonsa, Centropages velificatus, Temora turbinata, Evadne tergestina, O. colcarva, O. dioica, and P. crassirostris were dominant. Variations in abundance between non-bloom and bloom assemblages were evident, including the reduction in abundance of three key species within K. brevis blooms.

Florida Bay, a shallow, seagrass-dominated bay in southern Florida, USA, receives significant nutrient inputs and has experienced seagrass losses and microalgal blooms within the last several decades. Inorganic nutrient inputs have been well characterized, but the role of organic nutri- ents, specifically of dissolved organic nitrogen (DON) and organic phosphorus (DOP), in supporting microbial processes in the bay is unknown. In this study various techniques were used to assess the importance of these nutrients along a transect in Florida Bay when a cyanobacterial bloom occurred in the central region in November 2002. These techniques included measurements of ambient particu- late and dissolved nutrients, enzyme (urease and alkaline phosphatase) activities, and experiments to determine rates of 15 N uptake (nitrate, ammonium, urea, and amino acids over a period of 0.5 h) and long-term (48 h) changes in microbial biomass and 15 N natural abundance in enrichment bioassays. The cyanobacterial bloom in central Florida Bay was associated with the highest concentrations of DON and DOP, whereas the microflagellate- and diatom-dominated eastern bay region was associ- ated with the highest concentrations of inorganic nutrients. The zeaxanthin:chlorophyll a ratio (an indicator of the relative contribution of cyanobacteria to phytoplankton biomass) was positively corre- lated with the rate of uptake of urea, and negatively correlated with the rate of uptake of inorganic nitrogen. The opposite pattern was observed for the fucoxanthin:chlorophyll a ratio (indicative of relative diatom biomass) and the peridinin:chlorophyll a ratio (indicative of relative photosynthetic dinoflagellate biomass), suggesting that different algal groups were using different N substrates. Bio- mass responses in the bioassay experiments showed that phytoplankton (as chlorophyll a) responded to DON additions in the western region and to DOP additions in the eastern region, but heterotrophic bacteria, in contrast, responded to DOP additions in the west and DON additions in the east. These findings thus demonstrate the potential for different sources of N, including DON, to stimulate differ- ent components of the algal community, and for the phytoplankton and bacteria to respond differently to N and P.

The effect of equivalent additions of nitrogen (N, 30-40 μM-N) in different forms (ammonium, NH4+, and nitrate, NO3-) under conditions of different light exposure on phytoplankton community composition was studied in a series of four, 5-day enclosure experiments on water collected from the nutrient-rich San Francisco Bay Delta over two years. Overall, proportionately more chlorophyll a and fucoxanthin (generally indicative of diatoms) was produced per unit N taken up in enclosures enriched with NO3- and incubated at reduced (~15% of ambient) light intensity than in treatments with NO3- with high (~60% of ambient) light exposure or with NH4+ under either light condition. In contrast, proportionately more chlorophyll b (generally indicative of chlorophytes) and zeaxanthin (generally indicative of cyanobacteria) was produced in enclosures enriched with NH4+ and incubated under high light intensity than in treatments with low light or with added NO3- at either light level. Rates of maximal velocities (Vmax) of uptake of N substrates, measured using 15N tracer techniques, in all enclosures enriched with NO3- were higher than those enriched with NH4+. Directionality of trends in enclosures were similar to phytoplankton community shifts observed in transects of the Sacramento River to Suisun Bay, a region in which large changes in total N quantity and form occur. These data substantiate the growing body of experimental evidence that dichotomous microbial communities develop when enriched with the same absolute concentration of oxidized vs. reduced N forms, even when sufficient N nutrient was available to the community prior to the N inoculations.
