Healthcare has seen a big boon in technological advancement with the last few years with next-generation genome sequencing, robotics, artificial intelligence, and bioinformatics. While the origins of these breakthroughs are deeply rooted in biology, life sciences, and medical science, the new age of technological breakthrough require an interdisciplinary approach. Materials science itself is an interdisciplinary field, with a huge contribution of physics and chemistry, and within the past few decades, advances in nanotechnology has allowed integration into healthcare giving materials science a new emerging role. Such is the case with a vibrational spectroscopy technique called Surface Enhanced Raman Spectroscopy (SERS).SERS has the ability to measure the molecular bonds of the target analyte at the single molecular level. Yet, SERS is not a recognized spectrometry tool in healthcare. This dissertation examines the fundamental challenges that require a technique like SERS to transition from the academic research laboratories to clinical applications. While there are broad potential clinical applications, this dissertation focuses on the SERS measurement of diseases originating from cellular morphologies. The two main focal topics are cancer and microbiology research, where the proteome identification can provide new diagnostic and therapeutic treatment options. The first major section is about testing the capabilities of our group’s unique graphene gold nanopyramid hybrid platform in controlled research laboratory environments. Varies cases of cancers are examined: lung, skin, and breast, through their various size, morphology, and proteomic differences. Afterwards, the field of microbiology is explored with bacteria and fungus species.The second major section involves the SERS’s platform performance with clinical patient data. The first case study looks at the diagnosing meningitis from patients in China. The second case study looks at multidrug resistance detection of tuberculosis in rural Pakistan.The last major section tries to consolidate the major lessons in this journey of bringing SERS to clinical applications. In particular, this section tackles the main question of why a technique like SERS has not been fully recognized in clinics yet and brings the case to why our SERS hybrid platform is very close to it.

Surface enhanced raman scattering (SERS) technique has been widely implemented for the detection/quantifi- cation of numerous compounds. The development of reusable SERS substrates through regeneration is a constant concern of scientists in the field, related to the sustainability of the method. Cold atmospheric plasma (CAP) is an advantageous green method well-known for its effectiveness towards the successful degradation of organic molecules and materials’activation/modification. In the present study, we explored for the first time nanosecond pulsed dielectric barrier discharge (NSP-DBD) plasma as a rapid, energy efficient method for SERS solid sub- strates regeneration, implemented either directly in the gas or in the liquid phase as well as through immersion of the substrates in plasma activated water (PAW). We investigated the critical cold plasma factors (e.g. feeding gas, plasma treatment and retention time) in order to propose the most cost-effective alternative and shed light on the underlying regeneration mechanisms. The different SERS analysis case scenarios (analyte’s class , concentration andcross check) were considered, in order to simulate real SERS measurements conditions/requirements. In practical terms, such an approach will contribute to a significant reduction of the detection costs, revealing the NSP-DBD process as a flexible, fast, green, effective and low-cost solution towards the SERS substrates regeneration.

Nonlinear nanophotonics is a rapidly developing field of research that aims at detecting and disentangling weak congested optical signatures on the nanoscale. Sub-wavelength field confinement of the local electromagnetic fields and the resulting field enhancement is achieved by utilizing plasmonic near-field antennas. This allows for probing nanoscopic volumes, a property unattainable by conventional far-field microscopy techniques. Combination of plasmonics and nonlinear optical microscopy provides a path to visualizing a small chemical and spatial subset of target molecules within an ensemble. This is achieved while maintaining rapid signal acquisition, which is necessary for capturing biological processes in living systems. Herein, a novel technique, wide-field surface enhanced coherent anti-Stokes Raman scattering (\textit{wf}SE-CARS) is presented. This technique allows for isolating weak vibrational signals in nanoscopic proximity to the surface by using chemical sensitivity of coherent Raman microspectroscopy (CRM) and field confinement from surface plasmons supported on a thin gold film. Uniform field enhancement over a large field of view, achieved with surface plasmon polaritons (SPP) in \textit{wf}SE-CARS, allows for biomolecular imaging demonstrated on extended structures like phospholipid droplets and live cells. Surface selectivity and chemical contrast are achieved at $\sim$ 70 fJ/$\mu$m$^2$ incident energy densities, which is over five orders of magnitude lower than used in conventional point scanning CRM. Next, a novel surface sensing imaging technique, local field induced metal emission (LFIME), is introduced. Presence of a sample material at the surface influences the local fields of a thin flat gold film, such that nonlinear fluorescence signal of the metal can be detected in the far-field. Nanoscale nonmetallic, nonfluorescent objects can be imaged with high signal-to-background ratio and diffraction limited lateral resolution using LFIME. Additionally, structure of the extended samples' surfaces can be visualized with a nanoscale axial resolution providing topographic information. Finally, a platform for coherently interrogating single molecules is presented. Single-molecule limit SE-CARS on non-resonant molecules is achieved by means of 3D local field confinement in the nanojunctions between two spherical gold nanoparticles. Localized plasmon resonance of the dimer nanostructure confines the probe volume down to 1 nm$^3$ and provides the local field enhancement necessary to reach single-molecule detection limit. Nonlinear excitation of Raman vibrations in SE-CARS microspectroscopy allows for higher image acquisition rates than in conventionally used single-molecule surface enhanced Raman spectroscopy (SERS). Therefore, data throughput is significantly improved while preserving spectral information despite the presence of the metal. Data simultaneously acquired from hundreds of nanoantennas allows to establish the peak enhancement factor from the observed count rates and define the maximum allowed local-field that preserves the integrity of the antenna. These results are paramount for the future design of time resolved single-molecule studies with multiple pulsed laser excitations, required for single-molecule coherence manipulation and quantum computing.

The development of single molecule-based techniques in the last decades has enabled directly selecting, tracking, and measuring an individual molecule. In this thesis, the structural dynamics of a single quantum emitter, served by hypericin, is characterized. By using confocal scanning microscopy combined with radially/azimuthally polarized laser modes, three-dimensional reorientation of the transition dipole moment of a single molecule is observed. To quantify the temporal properties of the tautomerism, photon autocorrelation function is used to extract the intensity fluctuations. The results show the distinct influence of the local environment, such as PVA matrix and deuteration effect. The local photonic environment of a molecule is modified by the microcavity/nanocavity. A significant change of the radiative emission rate and of the fluorescence spectra is discussed. It allows us to measure the absolute quantum yield by using a tunable microcavity. The results show the possibility of controlling tautomerization by changing the photonic environment. Subsequently, molecular dissociation is discussed by single molecule surface-enhanced Raman spectra profiting from near field enhancement of nanocavity. A fast experimental optimization strategy towards optimal fluorescence enhancement is outlined.; Le développement de techniques basées sur une seule molécule au cours des dernières décennies a permis de sélectionner, de suivre et de mesurer directement une molécule individuelle. Dans cette thèse, la dynamique structurelle d'un seul émetteur quantique, servi par l'hypéricine, est caractérisée. En utilisant la microscopie à balayage confocale combinée à des modes laser polarisés radialement / azimutalement, une réorientation tridimensionnelle du moment dipolaire de transition d'une seule molécule est observée. Pour quantifier les propriétés temporelles de la tautomérie, la fonction d'autocorrélation des photons est utilisée pour extraire les fluctuations d'intensité. Les résultats montrent l'influence distincte de l'environnement local, comme la matrice PVA et l'effet de deutération. L'environnement photonique local d'une molécule est modifié par la microcavité / nanocavité. Un changement significatif du taux d'émission radiatif et des spectres de fluorescence est discuté. Il nous permet de mesurer le rendement quantique absolu en utilisant une microcavité accordable. Les résultats montrent la possibilité de contrôler la tautomérisation en modifiant l'environnement photonique. Par la suite, la dissociation moléculaire est discutée par des spectres Raman améliorés en surface à molécule unique bénéficiant de l'amélioration en champ proche de la nanocavité. We investigate the performance of vertical metal-insulator-metal (MIM) structures as refractive index (RI) sensor and SERS (surface enhanced Raman scattering) substrate. The disc-on-film structures with different geometrical parameters and different spacer configurations were studied systematically by experiments and simulations. The circular FP (Fabry-Pérot) cavity model was applied to describe the cavity plasmon modes which were divided into edge cavity modes and surface cavity modes, showing different optical response to wet etching, RI sensing and SERS. The dependencies of cavity plasmon modes as a function of periodicity, cavity radius and cavity height were studied. The reasons for different line shapes of far-field optical spectra were revealed. The influences of varying spacer on the locations of 'hot spots' and resonance wavelengths were systematically studied. The 'hot spots' of cavity plasmon modes were exposed by wet etching. The RI sensitivities of MIM structures were thoroughly studied including different types plasmonic modes, sensing based on spectral shifts and intensity changes, and bulk/molecular RI sensing configurations. Notable SERS EFs (enhancement factors) were achieved by underetching. EF peaks attributing to different orders of cavity plasmon modes were observed in both experiments and simulations. Good qualitative agreement was found between the experimental and simulated results. Similar depnedency of effective SERS EF as a function of cavity radius was found for vertical disc dimers compared to disc-on-film substrate., Nous avons étudié les performances des structures verticales métal-isolant-métal (MIM) en tant que capteurs d'indice de réfraction (RI) et substrats SERS (Raman augmenté en surface). Les structures en disques sur films avec différents paramètres géométriques et différentes configurations d'espaceurs ont été étudiées de manière systématique par des expériences et des simulations. Les modèles circulaires de cavité FP (Fabry-Pérot) ont été appliqués pour décrire les modes de plasmon dans les cavités MIM. Les modes de la cavité ont été divisés en modes de bord et modes de surface, ce qui montre différentes variations avec la gravure humide, les performances de détection RI et SERS. La dépendance des modes de plasmon de la cavité en fonction de la périodicité, du rayon de la cavité et de sa hauteur a été étudiée. La raison des différentes raies présentes dans les spectres optiques en champ lointain a été révélée. L'influence de différents espaceurs sur la localisation des 'points chauds' et des longueurs d'onde de résonance a été systématiquement étudiée. Les 'points chauds' des modes dans la cavité ont été révélés par gravure humide. Les sensibilités RI des structures MIM ont été minutieusement étudiées. Des EF (facteur d'amélioration) SERS notables ont été atteints en sous-gravant. Des pics EF attribuables à différents ordres de modes de plasmon dans la cavité ont été observés à la fois dans les expériences et les simulations. Une comparaison de performance SERS a été faite entre le dimère de disque sur film et les dimères de disque verticaux.

Surface-enhanced Raman spectroscopy (SERS) is widely used as a non-intrusive and label-free tool to identify the molecular spectral fingerprints in pharmacology, biology, etc. This thesis presents a SERS sensor based on the hybrid waveguide made of metallic (Au/Al) slots and dielectric (Si3N4) strips and a method to improve the SERS-detection capacity by placing metallic nanoparticles into the sensor’s slot. The theoretical investigation of the sensor is mainly conducted by the 3D finite-difference time-domain method (3D-FDTD) which provides the broadband electromagnetic response of metallic nanostructures. The enhancement factors in the sensor’s slot are estimated based on the |E|4-approximation. The experiments are mainly the fabrication of metallic slots, which is conducted by the electron beam lithography (EBL), and the characterization of the SERS-detection capacity of the sensors. The results show that the Raman signals given out by the SERS sensors are detectable. Metallic nanoparticles, which are located in the sensor’s slot, can improve dramatically the SERS-detection capacity. Surface plasmon polariton (SPP) and Localized surface plasmon (LSP) have attracted numerous researchers due to their high technological potential. Recently, strong attention was paid to the potential of SPP and LSP combinations by investigating metallic nanoparticles (NPs) on top of metallic thin films. Several studies on such systems have shown the coupling and hybridization between localized and delocalized modes. In this work, we propose a full systematic study on coupled NP/film systems with Au NPs and Au films. We investigate both experimentally and theoretically the influence of an ultra-thin SiO2 dielectric spacer layer, as well as the evolution of the plasmonic modes as the spacer thickness increases. We show that coupled systems exhibit enhanced optical properties and larger tunability compared to uncoupled systems. We also compare these results with those measured for coupled interfaces using graphene as a non-dielectric sub-nanometer spacer. Introducing graphene adds complexity to the system. We show that such coupled systems also exhibit enhanced optical properties and larger tunability of their spectral properties compared to uncoupled systems as well as unexpected optical behavior. We explain this behavior by evidencing graphene doping by metallic NPs, which can be a first step towards graphene based optoelectronic devices. After establishing a deep understanding of coupled systems we perform both SERS and RI sensing measurements to validate the high potential of these plasmonic interfaces; Les plasmons polaritons de surface (SPP) et les plasmons localisés de surface (LSP) font l’objet de nombreuses investigations du fait de leur fort potentiel technologique. Récemment, une attention particulière a été portée à des systèmes supportant ces deux types de résonances en déposant des nanoparticules (NPs) métalliques sur des films minces métalliques. Plusieurs études ont mis en évidence le couplage et l’hybridation entre modes localisés et délocalisés. Cependant, une compréhension en profondeur des propriétés optiques et du potentiel de ces interfaces est toujours manquante. Nous avons mené ici une étude de systèmes NPs/film couplés. Nous avons étudié à la fois expérimentalement et théoriquement l’influence d’une couche séparatrice ultra-mince en SiO2 ainsi que l’évolution des différents modes plasmoniques pour différentes épaisseurs. Nous avons ainsi mis en lumière que de tels systèmes couplés offrent des propriétés optiques exaltées et une large accordabilité spectrale. Nous avons aussi cherché à diminuer l’épaisseur de la couche séparatrice vers le cas ultime monoatomique en utilisant le graphène. Du fait du caractère non-diélectrique de celui-ci, nous avons mis en évidence un comportement optique inattendu de la résonance plasmonique. Nous avons expliqué celui-ci par la mise en évidence du dopage du graphène par les NPs, ce qui est un premier pas en direction de dispositifs optoélectroniques à base de graphène. Enfin, après avoir amélioré notre compréhension théorique de ces systèmes, nous avons évalué leur potentiel comme capteurs SERS ou LSP

Surface Enhanced Raman Spectroscopy (SERS) is a popular method that amplifies weak Raman signals from Raman-active analyte molecules making use of certain specially-prepared metallic surfaces. The main challenge in SERS is to design and fabricate highly repeatable, predictable, and sensitive substrates. There are many fabrication methods that strive to achieve this goal, which are briefly summarized in this paper. The E-beam lithography method is proposed to be superior to the mentioned techniques. In this paper, we review how EBL can be utilized in the preparation of SERS substrates and we discuss the contributions to the field by the Ozbay group. (C) 2015 Elsevier B.V. All rights reserved.

The recent advances in nanofabrication techniques have allowed for the emergence of novel sensing approaches. Amongst these various approaches, Surface Enhanced Raman Spectroscopy (SERS) via the use of plasmonic substrates has received wide-spread attention due to its many interesting proper-ties. In fact, plasmonic substrates enhance the Ra-man signal up to 12 orders of magnitude, paving the path for single molecule detection. Nevertheless, the sensitivity of this technique is strongly affected by the physical and structural properties of the metallic nanoparticles (MNPs). Thus, the mastering of the MNPs fabrication is a major challenge for various nanotechnological applications.In this context, we have developed a novel approach for the fabrication of organized NMPs through poly-mer self-assembly. The fabrication technique con-sists on controlling the physical interactions which occur during the fabrication through a nanophase separation in the polymer solution. This results in a nanostructuring of the polymer and a strong self-organization of the metallic precursor which is rapidly reduced into the MNPs. Experimental investigations of the different physical and chemical processes in play allow for a better understanding of the various keystone parameters of the nanostructuring as well as for determining their influences on the dimensions and optical response of MNPs. Finally, the fabricated plasmonic substrate demonstrated SERS limits of detection down to 10-13 M; Les avancées récentes de la nanofabrication ont permis de faire émerger un nouveau champ de recherche, celui des nanocapteurs. La sensibilité du capteur dépend des propriétés optiques des Nanoparticules Métalliques (NPMs) qui sont liées aux propriétés physiques et structurales de ces dernières. Ainsi, la maîtrise de la fabrication de NPMs est un réel défit pour des multiples applications nanotechnologiques. Dans ce contexte, nous avons développé une approche originale de fabrication de NPMs organisées par auto-assemblage de polymère. Il s’agit d’introduire de manière contrôlée des interactions physiques qui se manifestent lors de la fabrication par une nano-séparation de phase au sein du matériau. Ceci se traduit par un nanstructuration du polymère et une auto-organisation très spécifique du précurseur métallique qui se transforme spontanément en NPMs. Les investigations expérimentales en considérant les différents facteurs physico-chimiques impliqués, nous ont permis d’identifier les paramètres clés de cette structuration et de hiérarchiser leur influence sur les dimensions structurales et la réponse optique des NPMs. Finalement, la capacité du nanocapteur à détecter de faibles traces (

Driven by the interest in finding ultrasensitive sensors devices, reliable surface enhanced Raman scattering (SERS) based substrates are fabricated. Silver and gold nanoparticles are two of the best candidates for SERS substrates where Ag nanoparticles exhibit large enhancing ability in Raman intensity while Au nanostructures are stable in biological systems. Hence, combining the two metals in bimetallic nanostructures appeared to be a promising approach in order to sum the merits of Au surface properties and Ag enhancing ability. Thermal annealing of thin metallic films is used as a simple and relatively inexpensive technique to elaborate homogenous and reproducible Ag/Au bimetallic nanoparticles SERS substrates with high enhancing ability. The fabricated nanoparticles proved their enhancing stability even after one year of fabrication. Manipulating the composition of Ag/Au bimetallic NPs resulted in tuning the Localized Surface Plasmon Resonance (LSPR) over the whole visible spectrum, where the substrates are characterized with higher SERS enhancement when they exhibit LSPR closer to the Raman excitation wavelength. Additionally, bimetallic nanoparticles patterns with different size, composition and lattice constants have been conducted by electron beam lithography. The systematic study of their interesting plasmonic and SERS enhancing properties revealed maintenance in the LSPR-SERS relation by changing the nanoparticle size; Afin de développer des capteurs ultrasensibles des substrats fiables pour la diffusion Raman exaltée de surface (SERS) ont été fabriqués. Le recuit thermique des couches métalliques minces est utilisé comme une technique simple et peu coûteuse. Cette dernière permet d’élaborer des substrats homogènes et reproductibles de nanoparticules bimétalliques or-argent ayant un facteur d’exaltation importante. Ces nanoparticules gardent leurs propriétés d’exaltation même après une année de fabrication. En jouant sur la composition de nanoparticules bimétalliques il est possible d’avoir une résonance de plasmons de surface localisés (LSPR) sur tout le spectre visible. Ces substrats sont caractérisés par une exaltation SERS supérieure lorsque la résonance plasmon est plus proche de la longueur d'onde d'excitation Raman. En outre, les nanoparticules bimétalliques de différentes tailles, compositions ont été réalisés par lithographie électronique. Ultra sensitive sensors are required to probe very low concentrations of molecules. It has been shown that plasmonic nano-sensors could play this role. Nevertheless, even after thirteen years of research, a lot of questions remain unanswered.We will try to answer them in this study: what happens when a single molecule is adsorbed on a nanoparticle surface? In a monolayer? And what happens for the next layer of molecules? Can we easily enhance the sensitivity and the readability of sensors? We demonstrate experimentally a singular behavior when the quantity of molecules in the near-field region is very low, typically in the zeptogram level. To measure the low quantity of matter, different techniques to enhance the signal are studied: integration of sensor on axicon micro-lenses of Fabry-Perot like nano-cavities. We developed axicon micro-lenses to increase the intensity of unique nanoparticle signal. They redistribute the electromagnetic field into a Bessel beam with low numerical aperture, allowing an easy collection in far field. Nano-cavities have been designed to decrease the damping and refine the plasmonic resonance to increase the readability of the sensors. All these studies have the same target: to detect in-situ disease markers at very low concentrations in order to treat the patients before the first symptoms; Venir sonder de faibles quantités de molécules nécessite des capteurs ultra-sensibles. Il a été démontré que les capteurs plasmoniques pouvaient remplir ce rôle. Toutefois, même après trente ans de recherches, beaucoup de questions restent sans réponses. Dans cette étude nous tentons d'y répondre : que se passe-t-il lorsqu'une molécule s'adsorbe sur la surface d'une nanoparticule ? Lorsqu'une monocouche de molécule s'adsorbe ? Et que se passe-t-il pour les molécules suivantes ? Peut-on améliorer simplement la sensibilité et la lisibilité des nano-capteurs plasmoniques? Nous démontrons expérimentalement un comportement singulier lorsque la quantité de molécules dans le champ proche des nanoparticules est très faible, typiquement de quelques zeptogrammes. Afin de mesurer cette infime quantité de matière, des solutions d'amplification des signaux sont étudiées comme l'intégration de capteurs sur des micro-lentilles axicon, ou encore sur des nano-cavités de type Fabry-Perot. Nous avons développé les micro-lentilles axicon afin de palier la faible intensité du signal émanant de nanoparticules uniques. Elles ont pour but de redistribuer le champ électromagnétique, en faisceau de Bessel de faible ouverture numérique, donc facilement mesurable. Les nano-cavités optiques ont, quant à elles, étaient développées afin de diminuer l'amortissement des résonances plasmon et ainsi affiner les résonances et augmenter la lisibilité des capteurs.Toutes ces études ont un même but : détecter in-situ les marqueurs de maladies à des concentrations infinitésimales afin de traiter les patients avant les premiers symptômes

The fabrication of substrates for Surface Enhanced Raman Scattering (SERS) applications matching the needs for high sensitive and reproducible sensors remains a major scientific and technological issue. We correlate the morphological parameters of silver (Ag) nanostructured thin films prepared by sputter deposition on flat silicon (Si) substrates with their SERS activity. A maximum enhancement of the SERS signal has been found at the Ag percolation threshold, leading to the detection of thiophenol, a non-resonant Raman probe, at concentrations as low as 10-10M, which corresponds to enhancement factors higher than 7 orders of magnitude. To gain full control over the developed nanostructure, we employed the combination of in-situ time-resolved microfocus Grazing Incidence Small Angle X-ray Scattering with sputter deposition. This enables to achieve a deepened understanding of the different growth regimes of Ag. Thereby an improved tailoring of the thin film nanostructure for SERS applications can be realized. © 2014 AIP Publishing LLC.

Stable, high-index facet Re nanoparticles have been grown by a solid state synthetic method, negating the need for solutions or surfactants to control seeding, supracrystallization and NP shape. By using mixtures of K[ReO4] and the cyclic triphosphazene [NP(O2C12H8)]3, high-index facet nanoparticles and nanocrystals [similar]3 nm in size can be seeded and grown from drop-cast films and powders due to phase demixing of the metallopolymer. NP dispersions are formed directly within a carbon support that liquefies, allowing NP coarsening and ripening, and the eventual formation of a solidified graphitic support filled with crystals. Successful growth of mesoscale supracrystals of Re also occurs from ripening of nucleated NP seeds, incubated within a solidified and partially dewetted solid support that patterns the surface. The supported Re NP dispersions also exhibit surface enhanced Raman scattering within a graphitic matrix.

During my thesis, various subjects of research were tackled. I started by making nanofabrication of Raman active surface for enhancement -SERS- (gold nanowires arrays) by e-beam lithography. Then, I carried out a study about SERS efficiency according to the position of plasmon resonance (controlled by nanowires dimensions) for various wavelengths in the visible range. All the studies give a similar conclusion: to obtain a Raman enhanced maximum, the plasmon resonance have to be closed to the Raman wavelength.For the second part of my PhD thesis, I have worked on gold nano-tips fabrication and the implementation of a tip-enhanced Raman scattering set-up (TERS). Thus, I have developed a new process, based on an electrochemical attack, to produce reproducible gold tips for apertureless scanning near-field microscopy (SNOM) and TERS experiments. I have done a study about the depolarization properties of gold nano-tips. Our results show depolarization factor between 5 and 30 % that depends on the shape, the roughness and the material of the tip used and the excitation polarization state. Important consequences of this depolarization effect were underline in TERS experiments on Silicon crystal. It was bringing to light the importance to consider weight of the depolarization factor in the enhancement Raman during polarization TERS experiments., L'objectif de cette thèse est l'étude de nanostructures d'or ayant un rapport de forme important (nanofils, pointes...), assimilables à des nanoantennes optiques. Ainsi, j'ai tout d'abord étudié la diffusion Raman exaltée de surface (SERS) de réseaux de nanofils d'or de forme et de taille contrôlées produits par lithographie électronique. Je me suis notamment intéressé à l'influence des plasmons de surface localisés sur l'efficacité SERS de ces nanofils. Nous avons ainsi observé les plasmons de surface d'ordre supérieur jusqu'à 7. Nous avons alors relié l'exaltation SERS à la position de la résonance plasmon et déterminé l'efficacité des ordres supérieurs. Nous avons également mis en évidence que le maximum d'exaltation est obtenu pour une résonance plasmon proche de la longueur d'onde de diffusion Raman et ceci pour deux excitations laser différentes.Ensuite, j'ai monté une expérience de Raman en champ proche (ou TERS) et développé une technique reproductible de fabrication de pointes en or. Puis, j'ai effectué une étude quantitative des propriétés de dépolarisation des pointes métalliques utilisées en a-SNOM et en TERS. Nos résultats montrent des facteurs de dépolarisation entre 5 et 30% qui varient en fonction de la polarisation de la lumière incidente et de la forme de la pointe. Les conséquences importantes de ce phénomène de dépolarisation ont été mises en évidence dans des expériences TERS sur du Silicium cristallin; On montre que la dépolarisation doit être prise en compte pour une estimation correcte de l'exaltation induite par la pointe.

Using a recently obtained (general) formula for the interaction energy between an excited and a ground-state atom (Sherkunov Y 2007 Phys. Rev. A 75 012705), we consider the interaction energy between two such atoms near the interface between two media. We demonstrate that under the circumstances of the resonant coupling of the excited atom to the surface polariton mode of a vacuum-medium system the nonretarded atom*-atom interaction energy can be enhanced by (several) orders of magnitude in comparison with the van der Waals interaction energy of the two isolated atoms., Comment: 8 pages, 5 figures, local-field corrections included and improved presentation

Raman spectroscopy is rapidly finding favour for applications in the life science because of the ease with which it can be used to extract significant data from tissue and cells. However, the Raman effect is an inherently weak effect, which hinders the analysis of low concentration analytes. Raman sensitivity can be improved via the surface enhanced Raman scattering (SERS) effect. In SERS, Raman spectra are dramatically amplified when a molecule is adsorbed onto nano-roughened noble metal surfaces such as silver and gold. The degree of enhancement enables single-molecule detection, which offers the potential for the unambiguous identification of analytes at concentrations that are useful in both a forensic and a chemical biology context. Here we discuss some of the practical applications of SERS to both low-level narcotic detection, and how this can be applied to chemical biology. International audience; As a first step towards near-field Raman, we chose to study surface enhanced Raman scattering (SERS)-active substrates to cope with the weakness of Raman scattering (small cross-section and low concentration). We concentrated our work on localized surface plasmon (LSP) since they turned out to play a great part in SERS and we put forward the relation between LSP resonance and Raman enhancement. Roughness of our samples is controlled either by annealing process or electron-beam lithography (EBL); this latter technique proved to best suit to our study. Substrates are characterized by extinction spectroscopy which determines the LSP resonance and then Raman spectrum of a probe molecule, trans-1,2-bis(4-pyridyl)ethylene (BPE) is recorded. We show that maximum of enhancement is obtained when the LSP resonance is red-shifted (50 nm) compared to the excitation laser line (632.8 nm).

Raman spectra from single DNA molecules in their natural aqueous environment are presented. A DNA molecule that is anchored between two optically trapped dielectric beads is suspended in a solution with nanosized silvercolloid particles. The nonspecific binding of the metal to the DNA enhances the Raman scattering that is excited by a near-infrared beam. A Raman spectrum is first recorded followed by a force-extension curve that verifies the presence of a single DNA molecule.

Hybrid plasmonics has given rise to increasing interest in the context of the interaction between metal nano-objects and other materials. By benefiting from each of its constituents, hybrid nanostructures are commonly adopted in studies and optimization of biological and chemical sensors, nanoparticle with high plasmon resonance tunability, and nano-emitters. This PhD thesis presents a hybrid nanostructure of photopolymer/metal nanoparticle that is used as a near-field characterizing tool and as an anisotropic nano-emitter.The fabrication of this hybrid nanostructure is a near-field imprinting process based on nanoscale photopolymerization. This technique, compared with traditional near-field characterization methods, provides not only the image of the field distribution, but also enables quantification of the surface plasmon properties with sub-5nm resolution and reproduction of the exponential decay of the near-field.Under dipolar mode plasmon, the photopolymer was created anisotropically in the vicinity of the metal nanoparticle. With high concentration of dye molecules trapped in the polymer, the hybrid nano-emitter displays surface enhanced fluorescence and Raman signal that is dependent on the incident polarization. To our knowledge, this is the first achievement of the anisotropic nano-emitter based on the inhomogeneous distribution of the active molecule; La plasmonique hybride est un sujet d’actualité qui exploite des interactions physiques entre nano-objets métalliques et d’autres nanomatériaux. En bénéficiant des propriétés de chacun de leurs constituants, les nanostructures hybrides sont utilisées dans de nombreuses applications comme la détection d’espèces bio-chimiques. Dans cette thèse, nous présentons une nouvelle nanostructure hybride polymère/metal qui est non seulement utilisée comme nano-émetteur anisotrope qui s’avère aussi être un outil puissant de caractérisation du champ proche optique.La fabrication de cette nouvelle nanostructure est basée sur une approche de par photopolymérisation à l’échelle nanométrique. Cette technique, en comparaison aux méthodes traditionnelles de caractérisation, ne fournit pas seulement l’image de la distribution du champ, mais permet aussi des mesures quantitatives des plasmons de surface avec une résolution sub -5nm, incluant une description fine de la décroissance exponentielle des ondes évanescentes impliquées.A l’aide du mode plasmon dipolaire, une distribution anisotrope de matériau organique est intégrée dans le voisinage de la nanoparticule métallique. Avec une haute concentration de molécules de colorant dans le polymère, l’intensité des signaux de fluorescence et Raman du nano-émetteur hybride dépend de la polarisation incidente. À notre connaissance, il s’agit de la première réalisation d’un nano-émetteur dont le milieu à gain présente une distribution spatiale complexe le rendant sensible à la polarisation
