Science capital consists of science-related cultural and social capital and science-related behaviours and practices. This study aims to clarify the core dimensions of science capital among Finnish parents through consideration of how parents' educational degree, profession, age and residential area are associated with their science capital. To test this, a survey was conducted with a sample of 740 parents in Finland aged 26 to 69 years. Exploratory factor analysis was used to explore the science capital dimensions and confirmatory factor analysis supported the final seven-factor model. Differences in parents' responses, including by sociodemographic background, were measured using an analysis of variance (ANOVA) test. The core dimension of cultural capital focuses on the valuing of science in society. Behaviour and practices consist of everyday and informal learning in community spaces, and social capital is strongly linked to future affinity and science identity. Parents' residential areas and educational degrees seem to have the largest effects on different dimensions. However, a parent's level of education, profession or residential area do not seem to affect their engagement in everyday science activities with their children. These results provide support to discuss inequalities among families, and ways to increase family science capital.

Abstract To understand the determinants of inhaled aerosol particle distribution and targeting in the lung, knowledge of regional deposition, lung morphology and regional ventilation, is crucial. No single imaging modality allows the acquisition of all such data together. Here we assessed the feasibility of dual-energy synchrotron radiation imaging to this end in anesthetized rabbits; both in normal lung (n = 6) and following methacholine (MCH)-induced bronchoconstriction (n = 6), a model of asthma. We used K-edge subtraction CT (KES) imaging to quantitatively map the regional deposition of iodine-containing aerosol particles. Morphological and regional ventilation images were obtained, followed by quantitative regional iodine deposition maps, after 5 and 10 minutes of aerosol administration. Iodine deposition was markedly inhomogeneous both in normal lung and after induced bronchoconstrition. This study, conducted in Finnish private and Montessori child care centres, aimed at investigating the development of the phonological awareness (PA) of two- to five-year-old preschool children within a six-month period in relation to emerging letter knowledge and reading skills. The children (N = 72) performed five phonological tasks and a vocabulary test, a letter knowledge test and a reading skills test twice; in the beginning, when knowing no letters, and at the end of the study. The results show that the children's performance in the different phonological tasks improved over a six-month period. Detecting word structure or blending phonemes were easier than producing rhymes or twisting. The children who had learned to recognize letters performed better in phonological tasks than the children who had not learned any letters, but children able to read precociously exceeded all. Our results appear to indicate that learning letters does not require previous PA ability but affects PA.

The present study is the first part of a longitudinal research project investigating whether children become more aware of phonemes or rhyming when they learn letters or letter sounds or even begin to read, and if so how. For the present paper, the phonological awareness of young children aged 2-6 years was analyzed, particularly their auditory knowledge of the phonemic structure of words as well as their knowledge of rhyme. Altogether, 110 children from Finnish day-care centres performed seven phonological tasks and a vocabulary size test. The results showed a clear age-dependent improvement in phonological awareness. Blending words of phonemes, separating rhyming words from each other and correcting words by substituting the wrong phoneme with the right one were found to be easier tasks for 2- to 5-year-old children than producing rhymes, twisting words or detecting an initial letter.

Safe biodegradable “green” alternatives with minimal environmental and health risks have received widespread research interest. Thirty different kinds of bio-based flocculants (modified starches, modified celluloses, native chitosan, and lignin-based flocculant) were pre-tested using a simple jar test for the examination of the applicability of new organic flocculants in papermaking and in conditioning of waste activated sludge from the pulp and paper industry. Three starch-based and two cellulose-based polymers were chosen for further flocculation and filtrations tests. Key optimization parameters for the polymer were identified as the increasing of molecular weight and nitrogen content. The starch-based polymer had the best performance in both applications, but in neither of the cases did it function as well as the commercial polyacrylamide-based polymers. The importance of the molecular weight came up in the experiments. The developed starch-based polymer was cationic and had the charge density used in industry. On the other hand, although cationic flocculants are the most used in sludge conditioning, also anionic and non-ionic polymers are needed, depending on the characteristics of the sludge to be flocculated. Overall action of the tailored polymers was also studied in order to predict their potential as papermaking retention and dewatering aids.

Oxidative polymerization of two isolated lignans, secoisolariciresinol, and secoisolariciresinol diglucoside, as well as the lignan macromolecule, by a high redox potential Trametes hirsuta laccase was studied with different analytical methods. The reactivity of laccase with the different compounds was studied by an oxygen consumption measurement. The polymerization of laccase-treated lignans was evidenced by size exclusion chromatography, reversed phase - high performance liquid chromatography, and matrix-assisted laser desorption/ionisation - time of flight mass spectrometry. The data showed that the selected substrates could be oxidised by laccase. Secoisolariciresinol and secoisolaricires-inol diglucoside were polymerized by laccase to a similar extent. The lignan macromolecule reacted to a lesser extent. Polymerization of the macromolecule proceeded mainly via its secoisolariciresinol diglucoside moieties. Furthermore, it was shown that ferulic acid can be linked to polymerized secoisolariciresinol via decarboxylation by laccase. This investigation showed that lignans can be enzymatically modified by Trametes hirsuta laccase.

In this study the oxidative polymerization of different lignins, i.e. Flax Soda lignin, Spruce EMAL, and Eucalyptus Dioxane lignin by Trametes hirsuta laccase was compared. Initially the structures of the different lignins were compared by Fourier transform infrared spectroscopy. The reactivity of laccase with the different types of lignins in the absence of mediators was examined and verified by oxygen consumption measurements. The molecular weight distributions of treated and untreated lignins were determined by two different size exclusion chromatography methods. Furthermore, the potential of matrix-assisted laser desorption/ionisation-time of flight-mass spectroscopy for determination of the absolute molecular weights of the different lignins was evaluated. The data showed that all the technical lignins could be activated and polymerized by laccase to different degrees. The efficiency as indicated by measurements of the degree of polymerization was found to increase in the order of Spruce EMAL < Eucalyptus Dioxane lignin < Flax Soda lignin. Overall, this data supplies foundations for using enzymes more efficiently in the enzymatic upgrading of lignin.

BackgroundThe acquired component of complex traits is difficult to dissect in humans. Obesity represents such a trait, in which the metabolic and molecular consequences emerge from complex interactions of genes and environment. With the substantial morbidity associated with obesity, a deeper understanding of the concurrent metabolic changes is of considerable importance. The goal of this study was to investigate this important acquired component and expose obesity-induced changes in biological pathways in an identical genetic background.Methods and findingsWe used a special study design of "clonal controls," rare monozygotic twins discordant for obesity identified through a national registry of 2,453 young, healthy twin pairs. A total of 14 pairs were studied (eight male, six female; white), with a mean +/- standard deviation (SD) age 25.8 +/- 1.4 y and a body mass index (BMI) difference 5.2 +/- 1.8 kg/m(2). Sequence analyses of mitochondrial DNA (mtDNA) in subcutaneous fat and peripheral leukocytes revealed no aberrant heteroplasmy between the co-twins. However, mtDNA copy number was reduced by 47% in the obese co-twin's fat. In addition, novel pathway analyses of the adipose tissue transcription profiles exposed significant down-regulation of mitochondrial branched-chain amino acid (BCAA) catabolism (p < 0.0001). In line with this finding, serum levels of insulin secretion-enhancing BCAAs were increased in obese male co-twins (9% increase, p = 0.025). Lending clinical relevance to the findings, in both sexes the observed aberrations in mitochondrial amino acid metabolism pathways in fat correlated closely with liver fat accumulation, insulin resistance, and hyperinsulinemia, early aberrations of acquired obesity in these healthy young adults.ConclusionsOur findings emphasize a substantial role of mitochondrial energy- and amino acid metabolism in obesity and development of insulin resistance.

Abstract Background Secretion stress is caused by compromised folding, modification or transport of proteins in the secretory pathway. In fungi, induction of genes in response to secretion stress is mediated mainly by the unfolded protein response (UPR) pathway. This study aims at uncovering transcriptional responses occurring in the filamentous fungi Trichoderma reesei exposed to secretion stress and comparing these to those found in the yeast Saccharomyces cerevisiae. Results Chemostat cultures of T. reesei expressing human tissue plasminogen activator (tPA) and batch bioreactor cultures treated with dithiothreitol (DTT) to prevent correct protein folding were analysed with cDNA subtraction and cDNA-amplified fragment length polymorphism (AFLP) experiments. ESTs corresponding to 457 unique genes putatively induced under secretion stress were isolated and the expression pattern of 60 genes was confirmed by Northern analysis. Expression of these genes was also studied in a strain over-expressing inositol-requiring enzyme 1 (IREI) protein, a sensor for the UPR pathway. To compare the data with that of S. cerevisiae, published transcriptome profiling data on various stress responses in S. cerevisiae was reanalysed. The genes up-regulated in response to secretion stress included a large number of secretion related genes in both organisms. In addition, analysis of T. reesei revealed up regulation of the cpc1 transcription factor gene and nucleosomal genes. The induction of the cpcA and histone gene H4 were shown to be induced also in cultures of Aspergillus nidulans treated with DTT. Conclusion Analysis of the genes induced under secretion stress has revealed novel features in the stress response in T. reesei and in filamentous fungi. We have demonstrated that in addition to the previously rather well characterised induction of genes for many ER proteins or secretion related proteins also other types of responses exist.

Many lung disease processes are characterized by structural and functional heterogeneity that is not directly appreciable with traditional physiological measurements. Experimental methods and lung function modeling to study regional lung function are crucial for better understanding of disease mechanisms and for targeting treatment. Synchrotron radiation offers useful properties to this end: coherence, utilized in phase-contrast imaging, and high flux and a wide energy spectrum which allow the selection of very narrow energy bands of radiation, thus allowing imaging at very specific energies. K-edge subtraction imaging (KES) has thus been developed at synchrotrons for both human and small animal imaging. The unique properties of synchrotron radiation extend X-ray computed tomography (CT) capabilities to quantitatively assess lung morphology, and also to map regional lung ventilation, perfusion, inflammation and biomechanical properties, with microscopic spatial resolution. Four-dimensional imaging, allows the investigation of the dynamics of regional lung functional parameters simultaneously with structural deformation of the lung as a function of time. This review summarizes synchrotron radiation imaging methods and overviews examples of its application in the study of disease mechanisms in preclinical animal models, as well as the potential for clinical translation both through the knowledge gained using these techniques and transfer of imaging technology to laboratory X-ray sources.
