Body temperature is an important parameter for assessing animal health and physiological function. An iButton thermometer was used to measure the body temperatures of three Chinese pangolins (Manis pentadacyla) and three Sunda pangolins (M. javanica). The body temperature of Chinese pangolins was 33.2 ± 0.95°C (30.6–34.2°C, n = 3) and that of Sunda pangolins was 32.8 ± 0.48°C (31.7–34.2°C, n = 3). The daily variation in body temperature was 1.0–2.9°C in three Chinese pangolins and 1.2–1.9°C in three Sunda pangolins. The body temperature of Sunda pangolins tended to rise at night, with the highest body temperatures occurring mostly at night. This study provides valuable body temperature data for Chinese and Sunda pangolins and lays the foundation for future research on their mechanisms of body temperature regulation., The body temperatures of Chinese pangolin and Sunda pangolin were 33.2 ± 0.95°C (30.6–34.2°C) and 32.8 ± 0.48°C (31.7–34.2°C). Pangolins are of conservation concern as one of the most heavily poached, yet least understood mammals. The Sunda pangolin (Manis javanica) in particular is a critically endangered species. Here, we investigate the behaviour of these pangolins, for the first time, using a battery of cognitive tasks based on a manipulation of available sensory information. In an object-choice task in which only one of two containers was baited with food, the pangolins were able to find the food with olfactory information alone (N = 2), but not with visual or acoustic information alone (N = 1). The single subject tested on all three domains was further tested on how he used smell to find food by providing him with an opportunity to find it from a controlled distance or by using scent trails as a guide. The results suggest that our subject may have the capacity to exploit scent trails left by prey which can be tracked to a final source, though we found no evidence to suggest that he had the ability to initiate hunts based on distant prey odors. Despite the small sample size, this is the first controlled experiment to investigate pangolin foraging behaviour and cognition, which may have implications for the future protection of pangolin habitat based on the location of prey species.

The expression of animal digestive enzymes reflects important dietary adaptations. The pangolin, also known as scaly anteater, is a specialized myrmecophage that consumes mainly ants and termites, but its digestive enzymes have not been fully investigated. Therefore, in this study, we used shotgun proteomic analysis to examine the protein components of the saliva and intestinal juice of a Sunda pangolin (Manis javanica) that died shortly after being rescued. The intestinal juice contained greater variety of digestive enzymes, including α-amylase, maltase-glucoamylase, α,α-trehalase, sucrase-isomaltase, pepsin A, trypsin, pancreatic endopeptidase E, carboxypeptidase A1, carboxypeptidase B, dipeptidyl-peptidase 4, and pancreatic triacylglycerol lipase. The digestive enzymes identified in the saliva were maltase-glucoamylase and trypsin, and chitinase which was also found in the intestinal juice. Compared with other animals, the Sunda pangolin has less intestinal protease diversity and lacks key digestive enzymes, such as chymotrypsin and pancreatic elastase. The expression profile of the digestive enzymes of the Sunda pangolin reveals animal's adaptation to a diet consisting mainly of ants and termites. Our results will facilitate the preparation of artificial food for rescued pangolins and for those in captivity for conservation breeding efforts.

SummaryDestruction of tropical rainforests reduces many unprotected habitats to small fragments of remnant forests within agricultural matrices. To date, these remnant forest fragments have been largely disregarded as wildlife habitat, and little is known about mammalian use of these areas in Sumatra. Here, we conducted camera trap surveys (2285 trap-nights) within Bukit Barisan Selatan National Park and five surrounding remnant forest fragments during 2010–2013 and used species composition metrics to compare use. We found 28 mammal species in the protected forest and 21 in the fragments. The fragments harboured a subset of species found in the protected forest and several species not observed in the protected forest. Critically endangered species such as Sunda pangolin (Manis javanica) and Sumatran tiger (Panthera tigris sumatrae) were found in the forest fragments, along with species of conservation concern such as marbled cat (Pardofelis marmorata) and Asiatic golden cat (Pardofelis temminckii). The biodiversity found within the fragments suggests that these small patches of remnant forest may have conservation value to certain mammal species and indicates the importance of further research into the role these habitats may play in landscape-level, multispecies conservation planning.

The Sunda Pangolin Manis javanica is naturally present in Gaya Island, Sabah, but its population status is largely unknown. Results from a recent survey using camera traps indicated the presence of a few individuals, who were strictly nocturnal. There is a strong indication that the population in Gaya Island is in danger of local extinction. There is also an urgent need to conduct an in-depth study to gather scientific information for further conservation action.

The distribution of a species can help guide the protection activities in their natural habitat. Conversely, the lack of information on this distribution makes the protection strategy of this species difficult. The research was conducted in Way Canguk Research Station, Bukit Barisan Selatan National Park from January until March 2018. The purposes of this research were to create a distribution prediction map of Sunda pangolin (Manis javanica) and estimating the environment variables that most influenced the probability of the distribution. Fourteen points of camera trap coordinates were used for presence data with nine types of environment variables such as elevation, slope, understorey, canopy cover, distance from roads, distance from rivers, distance from villages, food source, and distance from the threat. The result of maxent showed an Area Under the Curve (AUC) value of 0.909 categorized as very good. The highest probability of Sunda pangolin distributions was in the Pemerihan Resort and Way Haru Resort area, while the dominant environmental variables included the distance from the village, the canopy cover, and the distance from threat with the value 47.7; 25.85; and 15.8%, respectively. Prediction maps and environment variables can help to identify the population of Sunda pangolin in the wild and can provide input for the national parks to prioritize protection areas for Sunda pangolin from the increased poaching.

Native to Southeast Asia, the Sunda pangolin (Manis javanica) is critically endangered largely because of poorly regulated wildlife trade, consumptive practices, and use in traditional Chinese medicine. Efforts to rescue and rehabilitate animals confiscated from the illegal trade are complicated by a general lack of knowledge surrounding the normal health and disease processes unique to the species. To provide clinical reference intervals for normal health states of Sunda pangolins, biochemical parameters were determined from rescued individuals in Vietnam that had undergone a 14-day observation period and met a set of criteria for release back into the wild. Blood samples were collected from 42 apparently healthy Sunda pangolins while anesthetized or awake. Packed cell volume (PCV) and total solids (TS) were determined manually, and serum biochemistry values were determined in-house with a benchtop analyzer. Additional biochemical and mineral parameters not included in the primary panel were determined from a subset of 10 pangolins through an external diagnostic laboratory. Overall reference intervals were calculated for PCV and TS (n = 29) and for standard serum biochemistry parameters (n = 42). Females and males demonstrated significant variation with respect to body mass, potassium (K+), and phosphorus, whereas age was a significant source of variation in alkaline phosphatase. Seasonal variation in glucose (GLU), creatinine (CRE), total proteins, sodium, calcium, and K+ was also observed. Comparisons between anesthetized and awake pangolins demonstrated significant variation in GLU, CRE, and K+. The parameters determined in this study can serve as a clinical reference for ex situ Sunda pangolin conservation efforts. In the context of wildlife rehabilitation, serial bloodwork allows for continued monitoring of patient health and should inform decision making regarding release readiness and timing.

Summary The Sunda pangolin (Manis javanica) is one of the most trafficked animal species globally, and is listed as Critically Endangered by the IUCN. There is limited information on reproductive biology or pathology of this species. We now document the clinical and pathological features of pyelonephritis and cystic endometrial hyperplasia in one of these animals. Ultrasonographic examination revealed pathological changes in the kidneys and uterus. On histopathological examination, there was marked interstitial infiltration of macrophages, neutrophils and lymphocytes in kidney tissue, fibrinonecrotic ureteritis and mild endometrial hyperplasia. This first report of these urogenital lesions in this species will be valuable for the conduct of health and reproductive assessments of the Sunda pangolin, which inform conservation and ex-situ management of this species.

The use of genome-wide genetic markers is an emerging approach for informing evidence-based management decisions for highly threatened species. Pangolins are the most heavily trafficked mammals across illegal wildlife trade globally, but critically endangered Sunda pangolins (Manis javanica) have not been widely studied in insular Southeast Asia. We used > 12,000 single nucleotide polymorphic markers (SNPs) to assign pangolin seizures from illegal trade of unknown origin to possible geographic sources via genetic clustering with pangolins of known origin. Our SNPs reveal three previously unrecognized genetic lineages of Sunda pangolins, possibly from Borneo, Java and Singapore/Sumatra. The seizure assignments suggest the majority of pangolins were traded from Borneo to Java. Using mitochondrial markers did not provide the same resolution of pangolin lineages, and to explore if admixture might explain these differences, we applied sophisticated tests of introgression using > 2000 SNPs to investigate secondary gene flow between each of the three Sunda pangolin lineages. It is possible the admixture which we discovered is due to human-mediated movements of pangolins. Our findings impact a range of conservation actions, including tracing patterns of trade, repatriation of rescue animals, and conservation breeding. In order to conserve genetic diversity, we suggest that, pending further research, each pangolin lineage should as a precaution be protected and managed as an evolutionarily distinct conservation unit.

The Vulnerable fishing cat Prionailurus viverrinus faces a perilous future in South-east Asia. It was last sighted in Cambodia in 2003. We deployed 16 camera traps at four sites in southern Cambodia during January–May 2015 to determine if the fishing cat was still present in the country. Eight photograph/video captures of fishing cats were recorded from the mangroves in Peam Krasop Wildlife Sanctuary and one from Ream National Park, but there were no records from Botum Sakor National Park or Prey Nup. A number of other globally threatened species were also photographed in Peam Krasop Wildlife Sanctuary: the Sunda pangolin Manis javanica, the hog deer Axis porcinus and the large-spotted civet Viverra megaspila. We learnt of the killing of an alleged fishing cat at the Sanctuary in July 2015 in retaliation for raiding fishing nets. Illegal hunting and capture of fishing cats for the wildlife trade were reported by local informants at all sites. We provide photographic and video evidence of the fishing cats and highlight the importance of Cambodia's mangroves for threatened species conservation.

Background The gut microbiomes of mammals are closely related to the diets of their hosts. The Sunda pangolin (Manis javanica) is a specialized myrmecophage, but its gut microbiome has rarely been studied. Methods Using high-throughput Illumina barcoded 16S rRNA amplicons of nine fecal samples from nine captive Sunda pangolins, we investigated their gut microbiomes. Results The detected bacteria were classified into 14 phyla, 24 classes, 48 orders, 97 families, and 271 genera. The main bacterial phyla were Firmicutes (73.71%), Proteobacteria (18.42%), Actinobacteria (3.44%), and Bacteroidetes (0.51%). In the PCoA and neighbor-net network (PERMANOVA: pangolins vs. other diets, weighted UniFrac distance p < 0.01, unweighted UniFrac distance p < 0.001), the gut microbiomes of the Sunda pangolins were distinct from those of mammals with different diets, but were much closer to other myrmecophages, and to carnivores, while distant from herbivores. We identified some gut microbiomes related to the digestion of chitin, including Lactococcus, Bacteroides, Bacillus, and Staphylococcus species, which confirms that the gut microbiome of pangolins may help them to digest chitin. Significance The results will aid studies of extreme dietary adaption and the mechanisms of diet differentiation in mammals, as well as metagenomic studies, captive breeding, and ex situ conservation of pangolins.

The case in the key biodiversity area of Sumatera named Batang Angkola indicate clearly that the trend of deforestation effected to the biodiversity lost within past few years. The objective of this research is to identify the remind wild animal in Batang Angkola Forest, as an effort to reduce the wildlife conflict, the result were significantly showed that protected forest Batang Angkola has some of the list of animals that also apart of IUCN, as tiger, pangoline, sunda pangolin and sun bear. There are 450 animals photos including 21 types of mammals, some animals that registered in the IUCN or International Union or Conservation Nature, as a part of the animals captures by the camera traps in Batang Angkola listed as endangered, endangered and critical endangered, threated and vulnerable. The analysis provides an occurrence probability for respective species, which is a proxy for abundance, that is allowing population trends to be tracked and also a Relative Abundance Indices measure for respective capture species. The high number of endangered and critical species illustrated that the forest contribution to conserving the Sumatera key biodiversity area significantly needed. Many of the wildlife animals are highly sensitive to the forest fragmentation.

Although tropical forests are among the most species-rich ecosystems on earth, 42% of mammal species in tropical forests are endangered because of overhunting and/or unsustainable exploitation. Camera-trap surveys have shown that natural saltlicks can be used to determine mammalian fauna, especially medium to large endangered species in tropical forests; establishment of camera traps, however, is time and effort intensive. Furthermore, the photographic range and detectable size of species are often restricted. Environmental DNA (eDNA) metabarcoding is a powerful approach that might provide a better way to study terrestrial animals in tropical forests. In this study, we examined whether eDNA from natural saltlicks comprehensively represented species composition in a Bornean tropical forest. We collected 100–150-mL water samples from natural saltlicks in Sabah, Malaysian Borneo. We constructed amplicon libraries for MiSeq sequencing using eDNA extracted from the water samples. Six endangered species were detected using this method, including Bornean orangutan (Pongo pygmaeus), Bornean banteng (Bos javanicus lowi), Asian elephant (Elephas maximus), Sunda pangolin (Manis javanica), sambar deer (Rusa unicolor) and bearded pig (Sus barbatus). However, most small and minor species were not detected, with low sequence identity (80–96%). Therefore, we propose that more species of tropical forest mammals should have their sequences deposited in DNA databases. This study is the first to report the endangered mammals of a tropical forest detected using eDNA from natural saltlicks.

Pangolins (Manis spp.) are myrmecophagous mammals with a wild diet of termites and ants. Diets are not yet readily acceptable by all pangolins and lack soil and chitin compared to their wild diet. This study aimed to assess the usefulness of including soil and chitin in pangolin diets by measuring changes in digestibility, food mean retention time, faecal scoring and body weight changes. Two male and two female Sunda pangolins (Manis javanica) from Night Safari (Singapore) were fed their typical gruel-type diets, with soil and chitin added in various amounts throughout 10 different treatments. Diet intake was calculated daily and faecal boluses were collected for digestibility analysis. Chromium oxide was added to the diet to measure mean retention time four times per animal per treatment. Faecal consistency scores were recorded daily where 1 is firm and 5 is liquid. Every animal was weighed weekly. Adding soil or chitin to the diet had various effects on the apparent digestibility of organic matter, crude protein and fiber with little effects on crude fat or calcium. Soil had a general positive influence on organic matter and fiber digestion. The addition of chitin and soil together had the stronger effects. Only at 25% inclusion of soil did the faecal score begin to improve. The overall maintenance energy requirements for adult M. javanica was 79.28 Kcal/kg BW0.75 /day, similar to other myrmecophagous mammals. The usage of soil and chitin in pangolin diets may be helpful in controlling weights while preventing behaviours associated with hunger.

Despite being heavily exploited, pangolins (Pholidota: Manidae) have been subject to limited research, resulting in a lack of reliable population estimates and standardised survey methods for the eight extant species. Camera trapping represents a unique opportunity for broad-scale collaborative species monitoring due to its largely nondiscriminatory nature, which creates considerable volumes of data on a relatively wide range of species. This has the potential to shed light on the ecology of rare, cryptic and understudied taxa, with implications for conservation decision-making. We undertook a global analysis of available pangolin data from camera trapping studies across their range in Africa and Asia. Our aims were (1) to assess the utility of existing camera trapping efforts as a method for monitoring pangolin populations, and (2) to gain insights into the distribution and ecology of pangolins. The Sunda pangolin (Manis javanica) is a quite remarkable and distinctive mammal species with its diet specialization of ants and termites, so it plays an important role in forest protection and maintaining ecological balance. But now the wild population of M. javanica has become declining dramatically due to over hunting and it has been listed as a critically endangered species by IUCN. In order to aid the conservation of this species, we report the development of 21 novel single nucleotide polymorphism (SNP) markers based on high-throughput sequencing in this study. The results show that observed heterozygosity and expected heterozygosity varied from 0.0333 to 0.9000 and from 0.0333 to 0.5172, respectively. The polymorphic information content and the minor allele frequency ranged from 0.0320 to 0.3750 and from 0.0168 to 0.5000, respectively. Only two loci showed significant deviation from Hardy–Weinberg equilibrium, but no significant linkage disequilibrium existed after Bonferroni correction. These SNP markers may provide a valuable tool for population genetic analysis, natural resource conservation and captive breeding of M. javanica.

The first authenticated record of the co-endangered pangolin tick (Amblyomma javanense) is reported from the Republic of Singapore, based on specimens of males and females collected from the Sunda pangolin (Manis javanica). The biology of the species is reviewed and steps for its conservation are outlined to ensure its continued survival across its range.

Understanding reproductive behavior is important for the conservation of endangered species, but research on the reproductive behavior of Sunda pangolins (Manis javanica Desmarest, 1822) is still very scarce. In this study, we used focal animal sampling and all-occurrence recording by an infrared monitor to observe the behaviors of two Sunda pangolins during a 5-day breeding period at the Pangolin Research Base for Artificial Rescue and Conservation Breeding of South China Normal University (PRB-SCNU). The behavioral characteristics and breeding strategies were analyzed, and the results were discussed together with information on other captive Sunda pangolin pairs at PRB-SCNU. The results found that there was no obvious estrus behavior in the captive female, while the male could exhibit sexual excitement and courtship behavior after a brief introduction period. Repeated copulation continued over many days after the female accepted the courtship. The average duration of copulatory behavior was 248.9 ± 148.7 s (n = 25), and all copulation occurred between 20:00 and 08:00 hr in the natural day-night photoperiod. The mating position of Sunda pangolins was lateral-ventral and was classified as the ninth or eleventh pattern under both Dewsbury's and Dixson's classification systems. This study can provide scientific guidance for the captive breeding and management of Sunda pangolins and other pangolin species, which is of great significance for ex situ conservation tactics.

The observations of Sunda pangolin reproductive parameters in this paper were based on the wild-caught animals and those that had spent time in captive environments, however, when analyzing the results, we did not consider differences in terms of breeding habits between the two. Still, this research has led to an increase in knowledge of the breeding habits of the Sunda pangolin. Our results suggest that there is no breeding season or season of parturition for the Sunda pangolin, which breeds all year round. We estimated the gestation period in this species to be around six months. Sexual maturity occurred at one year old or as early as six-seven months old in some individuals, and requires further investigation. Each Sunda pangolin in this study gave birth to one offspring at a time. The sex ratio at birth was 0.875:1 (♀:♂) (n = 15); and the weaning age was estimated at four months with a weight of 1.19 ± 0.50 kg (n = 3), which concurs with recent research. Findings in this study will contribute to future analyses of population dynamics, species conservation, and both in situ and ex situ management of the Sunda pangolin. Despite this contribution, further studies are needed on the reproductive parameters of Sunda pangolin.

The Sunda pangolin (Manis javanica Desmarest, 1822) is a Critically Endangered species. Given that this species lacks effective policies for in situ conservation and prevention of poaching in the wild, ex situ conservation and a captive breeding program are urgently needed to save this species from extinction. However, techniques for the maintenance and captive breeding of pangolins have not been well developed. In June 2010, we established the Pangolin Research Base for Artificial Rescue and Conservation Breeding of South China Normal University (PRB-SCNU). To date, a total of 34 Sunda pangolin cubs have been born at PRB-SCNU, 26 of which were captive bred, indicating great progress in keeping and breeding the Sunda pangolin. The techniques for maintenance—including housing, transitioning to an artificial diet, husbandry, and captive breeding—are described in this paper. The purpose of this manuscript is to share our successful experiences and techniques in the maintenance, management, and captive breeding of Sunda pangolins, and to provide guidance and a reference for the captive care and management of Sunda pangolins and other pangolin species.

It was 15,246 nucleotides with four nucleotides substitutions, which resulted in four changes of amino acid that were found only in PIV5-GD18, which further broadens the PIV5 infection host spectrum and will aid in our understanding of the complete genome sequence of PIV5 in different hosts.

Two morphologically similar, but phylogenetically unrelated Eimeria species from ancient mammals, African Tree Pangolin Phataginus tricuspis and Sunda Pangolin Manis javanica (Pholidota: Manidae), from two distant biogeographic realms (Afrotropical and Oriental), are characterized and compared morphologically and molecularly. Phylogenetic analyses produced an unstable topology. However, while precise position of the two Eimeria species from pangolins could not be firmly established due to the lack of related taxa, it is evident that they are not closely related and do not fall into any of the so far recognized eimerian lineages. Moreover, an eimerian found in P. tricuspis is described as a new species Eimeria nkaka n. sp., based on morphology of oocysts, endogenous developmental stages and sequence data.

Here we present the first field report on the ecology of a single female Sunda pangolin and her young in their natural habitat on an offshore island of Singapore. Observations of this event were made possible by the use of radio-telemetry and infrared-triggered camera traps. Only one offspring was recorded from this birth event and the period of maternal care was approximately 3 to 4 mo. A total of 3 natal dens were used throughout the monitoring period, and hollows of large trees (>50 cm diameter at breast height, DBH) were associated with all dens. The 100 and 95% home range estimated by minimum convex polygons is 6.97 and 5.63 ha, respectively. The daily active period of the pangolin was 127 ± 13.1 min, and peak activity levels were between 03:00 and 06:00 h. The findings of this report are discussed in relation to the conservation challenges confronting this species.

International audience; Despite being protected by both international and national regulations, pangolins are threatened by illegal trade. Here we report mitochondrial DNA identification and haplotype richness estimation, using 239 pangolin scale samples from two confiscations in Hong Kong. We found a total of 13 genetically distinct cytochrome c oxidase I (COI) haplotypes in two confiscations (13 and ten haplotypes respectively, with ten shared haplotypes between confiscations). These haplotypes clustered in two distinct clades with one clade representing the Sunda pangolin (Manis javanica). The other clade did not match with any known Asian pangolin sequences, and likely represented a cryptic pangolin lineage in Asia. By fitting sample coverage and rarefaction/regression models to our sample data, we predicted that the total number of COI haplotypes in two confiscations were 14.86 and 11.06 respectively, suggesting that our sampling caught the majority of haplotypes and that we had adequately characterized each confiscation. We detected substantial sequence divergence among the seized scales, likely evidencing that the Sunda pangolins were harvested over wide geographical areas across Southeast Asia. Our study illustrates the value of applying DNA forensics for illegal wildlife trade monitoring.

We use discrete morphological characters in a statistical framework to reassess the taxonomic status of the Palawan pangolin Manis culionensis relative to the Sunda pangolin M. javanica. We recommend that the 15 species-level traits previously proposed in the literature to distinguish the 2 pangolins be replaced by 5 newly defined diagnostic characters related to skull and external scales. Our study supports species-level partition between the Palawan and Sunda pangolins at a frequency of expected polymorphism threshold fixed to 0.10. Isolation through sea level rising (approximately 800,000–500,000 years ago) of proto-Palawan pangolins coming from Borneo through Early Pleistocene land bridges might have promoted the speciation of M. culionensis, a Palawan endemic species to be considered of high conservation concern.
