5. Mechanisms Underlying Mu Opioid Receptor Effects on Parallel Fiber-Purkinje Cell Synaptic Transmission in Mouse Cerebellar Cortex

Abstract

μ-opioid receptors (MOR) are widely expressed in the brain, varying in density in different areas. Activation of MORs underlies analgesia, euphoria, but may lead to tolerance, dependence, and ultimately opioid addiction. The Purkinje cell (PC) is the only efferent neuron in the cerebellar cortex and receives glutamatergic synaptic inputs from the parallel fibers formed by the axons of granule cells. Studies have shown that MORs are expressed during the development of cerebellar cells. However, the distribution of MOR and their effects on PF-PC synaptic transmission remain unclear. To examine these questions, we used whole-cell patch clamp recordings and pharmacological methods to determine the effects and mechanisms of MOR activation on synaptic transmission at PF-PC synapses. The MOR-selective agonist DAMGO significantly reduced the amplitude and area under the curve (AUC) of PF-PC evoked (e) EPSCs, and increased the paired-pulse ratio (PPR).DAMGO-induced inhibitory effects on PF-PC eEPSCs and PPR were abolished by MOR specific blocker CTOP. Further, DAMGO significantly reduced the frequency of PF-PC mEPSCs, but had no obvious effect on their amplitude, suggesting a presynaptic site of action. The DAMGO-induced reduction in the frequency of PF-PC mEPSCs also was blocked by CTOP. A protein kinase A (PKA) inhibitor PKI added in the pipette solution did not affect the inhibitory effects on PF-PC mEPSCs induced by DAMGO. Both the PKA inhibitor K5720 and MEK inhibitor U0126 in artificial cerebrospinal fluid (ACSF) prevented the inhibitory effects of DAMGO on PF-PC mEPSCs. These findings reveal that MORs are expressed in presynaptic PF axon terminals, where DAMGO can activate presynaptic MORs to inhibit PF-PC synaptic transmission by regulating the release of glutamate. G-protein-dependent cAMP-PKA signaling pathway may be involved in this process.

Abstract

Background: Primary Sjögren syndrome (pSS) is a progressive autoimmune disease that primarily affects exocrine glands. The clinical presentation of pSS may vary from an asymptomatic condition to severe skin symptom, resulting in a difficult and challenging diagnosis and treatment. Methods and results: Here, we report a 47-year-old Chinese woman who lost vision in the right eye for 7 days. She had been misdiagnosed with primary optic neuritis for 3 months. After 3 months, the results of immunohistochemistry, salivary gland scintigraphy, and antibody tests proved the diagnosis of pSS. After an IV methylprednisolone treatment for 3 days (1.0 g/d), her final visual and perimetry outcome were satisfactory. A review of the relevant English literature based on PubMed encompassing dates up to July 2016 has been discussed. Conclusion: Our finding and the literature review suggest that an early treatment may be beneficial but long-term disease may cause permanent irreparable damage.

13. Preparation of Non-Grinding Long Afterglow SrAl2O4: Eu2+, Dy3+ Material by Microwave Combustion Method

Abstract

The non-grinding long afterglow material SrAl 2 O 4 :Eu 2+ , Dy 3+ was prepared by combustion method in home microwave oven directly, after dispersant, frother, comburent, and mineralizer were added into the reacting system. XRD analysis showed that the powders were nearly pure SrAl 2 O 4 phase with few other phases, and the size of the grain was 41.1 nm. Fluorescence spectrum results indicated that there were 2 excitation peaks located at 345 and 400 nm, and the emission peak located at 516 nm, afterglow lasted up to 30 min or more. The microwave combustion method has advantages of less time, low temperature and no grinding process, and the material made by the method has good luminescent property.

39. Preparation and Luminescence Properties of Dy3+ Doped Sr3Lu(PO4)3 as a New Single Phase White Light Emitting Phosphor

Abstract

The novel white light emitting phosphors Sr3Lu(1-x)Dyx(PO4)3 (x = 0.02, 0.04, 0.06, 0.08, 0.10) were synthesized via a conventional high-temperature solid-state reaction method at 1250°C. The excitation spectrum indicated that these phosphors had a strong absorption in near UV region in the range from 260 to 460 nm. And under 350 nm excitation, the blue emission at 483nm (4F9/2→6H15/2) and yellow emission at 576 nm (4F9/2→6H13/2) were observed in the phosphors, respectively. We studied the effect of different doping concentrations of Dy3+ activator on the luminescence properties and found that the optimum doping concentration is 6 mol%. According to the Dexter’s theory, this quenching behavior is ascribed to be electric d-d interaction. Furthermore, the chromaticity coordinate (x = 0.28, y = 0.32) of Sr3Lu (PO4)3:0.06Dy3+ phosphor was very close to the “ideal white” (x = 0.33, y = 0.33) in the chromaticity diagram. Our results indicate that the Sr3Lu (PO4)3:Dy3+ phosphor can serve as a promising candidate for single-component white light emitting phosphor under near UV excitation.

40. Up-Conversion Luminescence Properties of Ho3+ Doped and Yb3+/Ho3+ Co-Doped Y2WO6 Phosphors

Abstract

In this article, Ho3+ doped and Yb3+/ Ho3+ co-doped Y2WO6 phosphors were successfully prepared via high temperature solid method. Their structures were investigated with X-ray diffraction, the up-conversion (UC) excitation from a 980 nm diode laser, and the developed phosphor shown two UC emission bands in the visible region 540 and 643 nm. Based on the result, the intensity of the frequency up-conversion emission was enhanced significantly through co-doping with Yb3+ ions in the Y2WO6: Ho3+ phosphor. The dependence of emission intensity on the pump power indicated that the up-conversion emission was a two-photon process. At last, the energy level diagrams was discussed. The results show that Y2WO6 is a promising host material for UC phosphors.

41. Synthesis and Luminescence Properties of a Blue-Emitting Sr3NaLu(PO4)3F:Eu2+ Phosphor

Abstract

A novel blue-emitting Sr3NaLu (PO4)3F:Eu2+ phosphor for white light-emitting diodes (w-LEDs) have been successfully prepared via the solid-state reaction. The crystal structure and phase of the prepared phosphors were investigated using X-ray diffraction (XRD). The band gap of Sr3NaLu (PO4)3F was estimated to be about 5.79 eV from the diffuse reflection spectrum. The Sr3NaLu (PO4)3F:0.05Eu2+ phosphor exhibits an intense and broad excitation band ranging from 200 to 450 nm in the near ultraviolet region and produces a bright blue emission band with the CIE chromaticity coordinates of x = 0.133 and y = 0.148 under the excitation of 365 nm, which were ascribed to the 4f-5d transitions of Eu2+. Moreover, the fluorescence decay curves were also investigated in detail. All the results suggest that Sr3NaLu (PO4)3F:Eu2+ might be a promising blue-emitting phosphor used in w-LEDs.
