[Objective] In the context of promoting energy structure transformation and accelerating the construction of a new energy system, clarifying the energy input strategies and their influencing factors across different regions is of great significance for formulating region-specific energy policies and orderly advancing regional renewable energy integration. [Methods] Based on a game theory framework, this study theoretically deduced the energy input strategies and influencing factors of different regions in the context of energy structure transformation. It also conducted empirical tests corresponding to the implementation effects of wind power subsidy policies in different regions of China and the progress of renewable energy integration in Europe. [Results] (1) Under the fossil fuel energy scenario, regions exhibited non-cooperative game behavior, where regional energy input strategies were characterized by non-cooperative games, displaying path dependency and influenced by the scale of capital and labor factors in the region. (2) Under the renewable energy scenario, regions engaged in cooperative games, with energy input strategies reflecting cooperative games, influenced by the unit cost of renewable energy, the scale of capital and labor factors, and the differences in the scale of capital and labor factors within the region. (3) When there are larger disparities in the scale of capital and labor factors among different regions within an area, lower costs of renewable energy, and a higher overall scale of capital and labor factors, the area is suitable for prioritizing the renewable energy integration, which is more conducive to the orderly achievement of energy efficiency improvement and energy structure transformation. [Conclusion] Dynamically adjusting energy input strategies, emphasizing the alignment of renewable energy deployment with regional factor endowments, and leveraging the leading role of central cities are crucial strategies for promoting China’s energy structure transformation. The findings of this study provide policy insights for the orderly advancement of renewable energy integration and the construction of a new energy system.

The subtype of ischemic stroke is valuable for highly effective interventions and treatments and essential for the prognosis of ischemic stroke. Background: Epileptogenesis and glioma growth have a bidirectional relationship. We hypothesized people with gliomas can benefit from the removal of epileptic tissue and that tumor-related epileptic activity may signify tumor infiltration in peritumoral regions. We investigated whether intraoperative electrocorticography (ioECoG) could improve seizure outcomes in oncological glioma surgery, and vice versa, what epileptic activity (EA) tells about tumor infiltration., Methods: We prospectively included patients who underwent (awake) ioECoG-assisted diffuse-glioma resection through the oncological trajectory. The IoECoG-tailoring strategy relied on ictal and interictal EA (spikes and sharp waves). Brain tissue, where EA was recorded, was assigned for histopathological examination separate from the rest of the tumor. Weibull regression was performed to assess how residual EA and extent of resection (EOR) related to the time-to-seizure recurrence, and we investigated which type of EA predicted tumor infiltration., Results: Fifty-two patients were included. Residual spikes after resection were associated with seizure recurrence in patients with isocitrate dehydrogenase (IDH) mutant astrocytoma or oligodendroglioma (HR = 7.6[1.4-40.0], P -value = .01), independent from the EOR. This was not observed in IDH-wildtype tumors. All tissue samples resected based on interictal spikes were infiltrated by tumor, even if the MRI did not show abnormalities., Conclusions: Complete resection of epileptogenic foci in ioECoG may promote seizure control in IDH-mutant gliomas. The cohort size of IDH-wildtype tumors was too limited to draw definitive conclusions. Interictal spikes may indicate tumor infiltration even when this area appears normal on MRI. Knowledge of the tumor microenvironment (TME) in patients with early lung cancer, especially in comparison with the matched adjacent tissues, remains lacking. To characterize TME of early-stage lung adenocarcinoma, we performed RNA-seq profiling on 58 pairs of minimally invasive adenocarcinoma (MIA) tumors and matched adjacent normal tissues. MIA tumors exhibited an adaptive TME characterized by high CD4 + T cell infiltration, high B-cell activation, and low CD8 + T cell infiltration. The high expression of markers for B cells, activated CD4 + T cells, and follicular helper T (Tfh) cells in bulk MIA samples and three independent single-cell RNA-seq datasets implied tertiary lymphoid structures (TLS) formation. Multiplex immunohistochemistry staining validated TLS formation and revealed an enrichment of follicular regulatory T cells (Tfr) in TLS follicles, which may explain the lower CD8 + T cell infiltration and attenuated anti-tumor immunity in MIA. Cancer immunotherapy targeting co-inhibitory pathways by checkpoint blockade shows remarkable efficacy in a variety of cancer types. However, only a minority of patients respond to treatment due to the stochastic heterogeneity of tumor microenvironment (TME). Recent advances in single-cell RNA-seq technologies enabled comprehensive characterization of the immune system heterogeneity in tumors but posed computational challenges on integrating and utilizing the massive published datasets to inform immunotherapy. Here, we present Tumor Immune Single Cell Hub (TISCH, http://tisch.comp-genomics.org), a large-scale curated database that integrates single-cell transcriptomic profiles of nearly 2 million cells from 76 high-quality tumor datasets across 27 cancer types. All the data were uniformly processed with a standardized workflow, including quality control, batch effect removal, clustering, cell-type annotation, malignant cell classification, differential expression analysis and functional enrichment analysis. TISCH provides interactive gene expression visualization across multiple datasets at the single-cell level or cluster level, allowing systematic comparison between different cell-types, patients, tissue origins, treatment and response groups, and even different cancer-types. In summary, TISCH provides a user-friendly interface for systematically visualizing, searching and downloading gene expression atlas in the TME from multiple cancer types, enabling fast, flexible and comprehensive exploration of the TME., (© The Author(s) 2020.
