Abstract

SUMMARY: The jasmonic acid (JA) signaling pathway is involved in the plant response to drought stress. JA and other hormones synergistically regulate the drought response in plants. However, the molecular mechanism underlying this synergism remains poorly defined. In the present study, transcriptome analyses of guard cells and quantitative PCR experiments revealed that MYC2 negatively regulated the negative regulator of ABA signaling, SlPP2C1, and the type‐B response regulator in the cytokinin pathway, SlRR26, and this negative regulation was direct. SlRR26 overexpression reduced drought tolerance in transgenic tomatoes, whereas slrr26cr lines were more tolerant to drought. SlRR26 negatively modulated reactive oxygen species levels in stomata and stomatal closure through RobhB. Moreover, SlRR26 overexpression counteracted JA‐mediated stomatal closure, suggesting that SlRR26 played a negative role in the JA‐mediated drought response. These findings suggest that MYC2 plays a key role in JA‐regulated stomatal closure under drought stress by inhibiting SlPP2C1 and SlRR26. [ABSTRACT FROM AUTHOR]

33. Effect of high pressure processing of pork (Longissimus dorsi) on changes of protein structure and water loss during frozen storage

Abstract

Protein and water modification in pork Longissimus dorsi (LD) pressurized at 200–400 MPa (5 min, 4 °C) followed by storage at −20 °C for up to 84 days were analyzed by complementary methodologies. Changes in proteins depended to a larger extent on storage time than pressure levels. High pressure (HP) improved drip loss by 35% after 84 days of storage. However, the meat's ability to retain water upon mechanical force (expressible moisture) was unaffected by both HP treatment and storage. No free water was present in the meat (due to drip loss), only immobile water and a larger fraction of water entrapped in the myofibrillar network were present. The water mobility after thawing was not affected as such by HP treatment and frozen storage. HP treatment and frozen storage results in a highly dynamic protein-water system, whereby protein and water molecules compete for H-bonding sites, consequently affecting the water binding.

Abstract

To assess the contamination of lettuce with norovirus in Nanjing city (China), two hundred and eighty one lettuce samples were collected between 2017 and 2018 from production sites (preharvest) and retail outlets (postharvest) and analyzed. The presence of norovirus was confirmed in 26.9% (71/264) of lettuce samples, with norovirus genogroup I detected more frequently (20.1%) than norovirus genogroup II. Norovirus was detected less frequently in lettuce samples from supermarkets (4.9%) than in samples from greengrocers or farmer markets (36.5%) and was detected both in field-grown (28.9%) and in hydroponic lettuce samples (30%). The norovirus occurrence and levels varied markedly by seasons. Abstract

The history of traceability reveals that nomadic herders as early as 1000 BCE marked livestock with irons and ear incisions in order to protect against thefts. Nowadays, we build traceability systems to document the origin of foods, and in order to ensure safer foods when tracking and recalling products. A holistic traceability system includes, as a minimum, identification elements, databases and an information flow. The animal identification elements refers to body marks, ear tags, Radio-Frequency Identification (RFID) tags, retina image recognition, or DNA fingerprinting. The product identification refers to barcodes (EAN UCC, PLU, and GS1), 2D barcodes (QR, VC, and DM) and RFID or Electronic Product Code (EPC). The present review describes existing and upcoming traceability technologies for farm animals and their products, to update the common methods for information collection and data inquiry, with the view to expound traceability policies and regulations between developed and developing countries. The benefits of the new technologies and their practical limitations are also discussed.

Abstract

Body measurement is an important technique to acquire the livestock body parameters for precision agriculture and effective management of livestock. Traditionally, the livestock body parameters are often manually measured, which suffers from several drawbacks, including unstable accuracy, low efficiency, difficulty in measuring complex parameters, and potentially negative influence on animal welfare. Considering this, this paper presents a 3D surface reconstruction and body size measurement system based on multi-view RGB-D cameras. We use Kinect depth cameras to obtain the point clouds of the freely walking pigs from three different views (i.e., upper-view, left-view and right-view). The registration parameters are obtained by a rectangular cuboid, which can be used to reconstruct three local point clouds. Thereafter, the distribution of the point cloud projection in different directions is used to locate the measuring positions, which then contribute to the precise measurement of several key parameters, such as body length, body height, body width and abdominal girth. Moreover, the polar coordinate transform is developed to improve the measurement accuracy of the abdominal girth. The experiments performed on forty pigs have shown that our average relative errors in the body length, height, width and abdominal girth are 2.97%, 3.35%, 4.13% and 4.67%, respectively, which demonstrate the feasibility and accuracy of the proposed system.

Abstract

This study investigates the possible mechanism of the protective effects of blueberry anthocyanins on human embryonic liver L-02 cells. Results of the WST-8 method showed that different concentrations of blueberry anthocyanins protected the human embryonic liver L-02 cells against CCl4-induced injury in a dose-dependent manner with IC50 at 6.2×g/L. Cell clone formation inhibition assay demonstrated that cell clones increased with increased concentration of anthocyanins. Propidium iodide (PI) staining analysis showed that anthocyanins can decrease cell cycle in the G1 phase. DNA ploidy analysis showed a weakened percentage of hypodiploid cells in a dose-dependent manner. Moreover, Annexin V-FITC/PI staining analysis showed a dose-dependent effect of anthocyanins on late apoptotic and necrotic cells. 47. Effects of postharvest nitric oxide and 24-Epibrassinolide treatments on cultivated blueberry (Vaccinium corymbosumL.): From flavor and antioxidant properties to edibility evaluation

Abstract

Blueberry is a tender and perishable berry. This study identified the effects of nitric oxide (NO) and 2,4-Epibrassinolide (EBR) on the flavor, antioxidant properties, and edible value of blueberry. The results indicated that 1.0 mmol·L-1sodium nitroprusside (SNP, a NO donor) and 0.8 mg·L-1EBR groups showed high sourness and low sweetness in taste. SNP preserved the content of volatile flavor compounds on 8 d, while EBR preserved that on 16 d. The principal component analysis (PCA) plot from the flavor well differentiated between the treatment and control blueberry. Moreover, the biplot shows the correlation between volatile flavor compounds and taste perception. From the perspective of antioxidant properties, SNP and EBR inhibited the accumulation of H2O2, which showed a negative correlation with total phenolic (TPC), total flavonoid content (TFC), and ascorbic acid (AsA) content. SNP and EBR increased enzyme activities, such as catalase (CAT), peroxidase (POD), superoxide dismutase (SOD), ascorbic acid peroxidase (APX), and glutathione reductase (GR), antioxidant activity, including 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging and ferric ion-reducing antioxidant power (FRAP), and the gene expression levels of VcSOD, VcAPX,and VcGR. The random forest model showed that AsA content, weight loss rate, decay incidence, TFC, TPC, Methyl-2-methylbutyrate, POD activity, and DPPH radical scavenging activity played critical roles in the edible value. Collectively, NO and EBR preserved the flavor, increased antioxidant capacity, and maintained postharvest quality.

48. Allergenic risk assessment of enolase leaked from Saccharomyces cerevisiaeunder pressurization

Abstract

High hydrostatic pressure (HHP) is widely employed in food sterilization and processing, but it can release the allergenic proteins into the food matrix, raising the risk of sensitization. In this research, the leaked allergen enolase from Saccharomyces cerevisiaeafter HHP-treatment was characterized and quantified by ELISA, Western-blot, and LC-MS/MS. MS2 results indicated that the amounts of leaked enolase following untreated, 300 MPa, and 600 MPa were respectively 0.0291, 0.0816, and 0.229 μg/mL. This illustrated a progressive release of the allergen under HHP. Furthermore, Molecular docking experiments demonstrated that enolase binds ARD1, GMP1 and TDH3 during the leakage process, affecting its allergenicity. The implications of these findings are noteworthy for comprehending the safety hazards of allergens in the context of HHP processing and for advocating the adoption of HHP technology in the food sector.
