These applications include measurements of spontaneous motions/deformations generated by heart motion; pulsations from phenomena such as blood flow (intracardiac, intravascular, and carotid); heart, blood vessel, and liver motion; and motion from artificial sources such as motions/deformations generated by applying static compression/stretching forces, vibration or acoustic radiation forces (breast and liver). For arbitrary orthogonal coordinate systems obtained using arbitrary transducer types (eg, linear, convex, sector, arc, or radial array types, or single aperture types with a mechanical scan), several lateral modulation (LM) methods (eg, scanning with plural crossed or steered beams over a region of interest) have been developed that can be used with new echo imaging methods for tissue displacement/deformation measurements. Specifically, by using such beamforming methods, in addition to highly accurate displacement vector and lateral displacement measurements, LM echo imaging with a high lateral carrier frequency and a high lateral resolution has been developed. Another new beamforming method, referred to as “a steering angle (ASTA) method,” ie, scanning with a defined steering angle, is also described. In addition to conventional non-steered-beam scanning (ie, a version of ASTA) and conventional steered-beam scanning with a variable steering angle (eg, sector, arc, radial scan), a simple, single-beam scanning method also permits the use of LM, which yields an accurate displacement vector measurement with fewer calculations than the original LM methods. This is accomplished by using a previously developed spectra frequency division method (SFDM). However, the lateral carrier frequency and the measurement accuracy acquired by using such a single-beam scanning method are lower than those achieved with the original LM scanning methods and should be increased (ie, by using a quasi-LM method). In this report, the effectiveness of the use of the new SFDMs is verified with experiments on agar phantoms, in which conventional non-steered, focused single-beam transmission/reception scanning is performed together with high-speed non-steered single plane-wave transmission and non-steered, focused single-beam reception scanning using a linear array-type transducer. For comparison, the original LMs, with their respective transmissions of crossed, steered focused beams and plane waves are also performed. Because the use of rectangular apodization functions (ie, no apodization) yields a larger bandwidth in a lateral direction than the effective use of parabolic functions with the original LM method, it is shown that disregarding the lateral low-frequency spectra yields useful quasi-LM echo imaging with a high lateral frequency, and further significantly increases the measurement accuracy of a displacement vector. In addition, when no apodization is used with the original version of LM, disregarding the low-frequency lateral spectra is effective. In addition, the interchangeability of cosine and sine modulations performed after completing beamforming can also be used for single-beam scanning as well as for the original LM scanning method. Specifically, the cosine and sine modulations, respectively, are used for LM and quasi-LM imaging and displacement vector measurements. It is concluded that the appropriate use of the new SFDMs with simple single-beam scanning or with simple plural crossed-beam scanning with no apodization can achieve almost the same accuracy as the original LM scanning method using plural crossed beams with the effective apodization. Another new application of SFDM is also described: an incoherent superposition of the frequency-divided spectra reduces speckles. Previously developed lateral modulation (LM) methods with a multidimensional autocorrelation method (MAM) or multidimensional Doppler method (MDM) and a steering angle method (ASTA) with lateral Doppler method produced accurate displacement vector and lateral displacement measurements, respectively. Such measurements cannot be obtained using only a conventional Doppler technique. Another new method has also been reported, using multiple crossed beams (MCBs) to obtain high-accuracy displacement vector measurements; that is, a displacement vector is synthesized using accurately measured axial displacements with previously developed multidimensional displacement measurement methods, including the one-dimensional autocorrelation method (1D AM) with a multidimensional moving average (MA), together with conventional rotation processing of global echo data or a coordinate system (ie, a global echo rotation referred to as r method) by the negative value of the steering angles used in beamforming. However, in real-world applications, directivities of transmission and reception apertures, scattering, reflection, and attenuation affect the direction and properties of US beams used for conventional axial displacement measurements employing beamforming methods such as a conventional nonsteered, steered, or secta beam, and they also affect ASTA and MCB methods. In this report, to improve accuracy in the measurements of an arbitrary directional displacement and a displacement vector using any beamforming methods, a spatial resolution in a beam angle (BA) is generated. For instance, for a two-dimensional (2D) Cartesian coordinate system, this is obtained by calculating the arctangent of the ratio of the axial and lateral instantaneous frequencies or the first moments of local spectra. On the basis of the 1D AM with a multidimensional MA, the local displacement in the beam direction is accurately measured by dividing the local instantaneous phase change by the instantaneous frequency calculated in the beam direction, and an arbitrary directional displacement can be measured (axial, lateral, radial, and so forth), which is done with or without a rotation of local echo data. These are respectively referred to as BA and BAr (BA + local rotation) methods or, specifically, as the 1D AMBA (1D AM + BA) and the 1D AMBAr (1D AM + BAr). Also, it is theoretically shown that the 1D AM with MCB but no echo rotation (ie, 1D AMBA with MCB) is equivalent to the most accurate MAM with LM, and that the 1D AMBA with ASTA can also provide a lateral Doppler measurement. Through agar phantom experiments, in addition to steered spherical focusing beams for both transmission and reception, measurement accuracies with all of the new methods are also evaluated for rapid scanning beamforming such as using one or plural, steered or nonsteered plane wave transmissions, and steered or nonsteered spherical focusing beam receptions. For comparisons with the 1D AMBA and the 1D AMBAr, the spectra frequency division method (SFDM) used in the MAM is also used instead of the 1D AM. All for the measurements of an axial displacement with axial compression and nonsteering, a lateral displacement with lateral compression and ASTA and a 2D displacement vector with lateral compression and MCBs, BA methods based on the 1D AM and SFDM approaches achieve more accurate measurements with significantly fewer calculations than the corresponding BAr or r methods (ie, real time). BA methods do not yield dead region data in a region of interest and they do not yield any measurement errors because of an approximate interpolation of echo data or of measured motion/deformation data. However, the measurement accuracies of one-directional displacements are significantly lower than those of displacement vectors, because of the practical three-dimensional deformation or motion (eg, about a half standard deviation of the lateral displacement measurement). In terms of required processing, the MAM or the MDM with LM is better than methods with MCB (ie, real time). The proper combination of the SFDM with LM or ASTA will yield more accurate measurements in a trade-off with increasing the number of calculations. This is achieved by superimposing crossed, steered beams, which is referred to as “lateral modulation” (LM). This type of beamforming achieves almost the same accuracy in lateral displacement measurements as in axial displacement measurements, ie, displacement vector measurements. The steering angle (ASTA) can also be used instead of LM, which uses only beams with a steering angle. In this report, the accuracy of the displacement vectors and lateral displacement measurements for LM and ASTA were evaluated using simulations and agar phantom experiments. The parameters used were direction of a displacement vector, steering angles, and rotation angle for the coordinate system. Changes in the steering angle and rotation angle of the coordinate system permit control of frequencies in the respective directions of the coordinate axes. As shown, when performing a simple ASTA for displacement vector measurement, a spectra frequency division should be performed using a previously developed multidimensional autocorrelation or Doppler method instead of block-matching methods. In this version of ASTA, the combination of nonsteering and rotation of the coordinate system is also effective, because the lateral bandwidth does not decrease. In such a case, transmission of a laterally wide wave will also be effective, particularly for three-dimensional measurement/imaging using a two-dimensional array transducer. ASTA can also be used for accurate lateral displacement measurements. Although a proper beam steering and/or a proper coordinate rotation improves the measurement accuracy achieved with LM and ASTA, all measurement accuracies obtained with ASTA are lower than with LM.Keywords: lateral modulation, single steering angle, coordinate system rotation, spectra frequency division

Chikayoshi Sumi, Kunio ShimizuDepartment of Information and Communication Sciences, Faculty of Science and Technology, Sophia University, Chiyoda, Tokyo, JapanAbstract: Comparison of the measurement accuracy of elasticity was performed through agar phantom experiments, ie, two-dimensional (2D) strain tensor components and 2D shear modulus reconstruction obtained by previously developed multidimensional displacement vector measurement methods on the basis of autocorrelation and Doppler methods (2D AM and 2D DM) and the corresponding 1D methods. The multidimensional methods yield more accurate and stable results than the corresponding 1D methods. BACKGROUND: Using a rabbit cornea model, our recent study demonstrated that Chen Medium (CM), an isotonic media enriched with nonlactate-generating high-energy substrates, is very effective for organ preservation. In the present study, the efficacy of CM is further evaluated with human corneas METHODS: The effectiveness of CM and Optisol for preserving the endothelial integrity of human corneas in vitro was evaluated by scanning electron microscopy. Clinical efficacy was evaluated in a total of 83 patients: 10 patients with keratoconus grafted randomly with either CM- or Optisol-stored cornea of the same donor, and 73 patients with various conditions grafted with CM-stored corneas. After surgery, visual acuity and quality of the graft were monitored for up to 4.6 years. RESULTS: The scanning electron microscopic study revealed that after 11-day storage at 4 degrees C, the CM-stored cornea had only marginal disruptive changes, 9.4+/-1.1%, in endothelial cells, as opposed to 42.4+/-4.6% of the Optisol-stored cornea. All 78 CM-stored corneas, including 67 with 12.2- to 17.7-hr death-to-storage time, 3-7.6 days of storage time, and initial marginal quality before storage, were successfully transplanted. These grafts were thin and clear, with an excellent epithelial integrity and without significant changes in endothelial cell density. Five Optisol-stored corneas were also successfully grafted; one of them, however, was edematous for about 4 weeks, and all the grafts were slightly thicker with substantial endothelial cell loss. CONCLUSION: Using a cornea model, present and recent studies show that CM is very effective for preserving tissue viability and endothelial integrity. Previous study revealed that CM-stored tissues maintained high levels of ATP and metabolic function, with suppression of lactate formation and accumulation. Thus, these findings support the concept that preservation of tissue viability is closely associated with the ability of the tissues to retain metabolic activity, to generate ATP efficiently, and to prevent acidosis effectively during storage.

Naïve dialecticism refers to a set of East Asian lay beliefs characterized by tolerance for contradiction, the expectation of change, and cognitive holism. In five studies, the authors examined the cognitive mechanisms that give rise to global self-concept inconsistency among dialectical cultures. Contradictory self-knowledge was more readily available (Study 1) and simultaneously accessible (Study 2) among East Asians (Japanese and Chinese) than among Euro-Americans. East Asians also exhibited greater change and holism in the spontaneous self-concept (Study 1) and inconsistency in their implicit self-beliefs (Study 3). Cultural differences in self-concept inconsistency were obtained when controlling for alternative explanatory variables, including self-criticism (Study 4) and self-concept certainty (Studies 2 and 3) and were fully mediated by a direct measure of dialecticism (Study 5). Naïve dialecticism provides a comprehensive theoretical framework for understanding these cultural differences and the contradictory, changeable, and holistic nature of the East Asian self-concept.

Studies on the intact avascular cornea reveal two types of lactate effluxes: exogenous glucose-elicited and spontaneous. The former type exhibits characteristics resembling the proton-lactate symport system previously found in tumor cells and erythrocytes, including an enhanced lactate efflux at a higher extracellular pH and in the presence of H+ and K+ ionophores, and an inhibition by mersalyl with subsequent lactate accumulation in the tissue and cessation of glycolytic activity. The latter type occurs immediately following the incubation of freshly isolated cornea in a medium containing no exogenous glucose, with a rate about 10 times that of exogenous glucose-elicited lactate efflux. It is insensitive to 10 m m iodoacetate and lacks the characteristics of the proton-lactate symport system. Findings reveal that about 50% of corneal glucose utilization occurs in the epithelium, with the stroma and endothelium sharing the other 50% approximately equally. Of the glucose utilized, the lactate formation to pyruvate oxidation rate ratios are approximately 1:1 in the epithelium, 2:1 in the stroma, and 1:2 in the endothelium. About 79% of total tissue lactate is formed in the epithelium and stroma, and in vivo, this is probably pumped into the stromal extracellular space (about 90% of total tissue volume) via the proton-lactate symport system, with spontaneous release into the aqueous humor via a simple diffusion process. The H+ and K+ ionophores facilitate lactate efflux at the expense of the cellular pyruvate pool, without significant effect on the glucose uptake and glycolytic activity. These findings suggest that the ionophore-mediated lactate efflux favors the reduction of low pyruvate concentration in the tissue, rather than parallel increases in glycolytic activity.

We provide a basis for automated single-cell sorting based on optical trapping and manipulation using human peripheral blood as a model system. A counterpropagating dual-beam optical-trapping configuration is shown theoretically and experimentally to be preferred due to a greater ability to manipulate cells in three dimensions. Theoretical analysis performed by simulating the propagation of rays through the region containing an erythrocyte (red blood cell) divided into numerous elements confirms experimental results showing that a trapped erythrocyte orients with its longest axis in the direction of propagation of the beam. The single-cell sorting system includes an image-processing system using thresholding, background subtraction, and edge-enhancement algorithms, which allows for the identification of single cells. Erythrocytes have been identified and manipulated into designated volumes using the automated dual-beam trap. Potential applications of automated single-cell sorting, including the incorporation of molecular biology techniques, are discussed.
