The study aimed to determine the potential of the infrared (IR) spectrophotometric technique for measuring the content of sulphanilamide with the sulfonamide group. The study aimed to obtain the IR spectra of sulfanilamide and use the -SO 2 band at 1114.37 for the quantitative assay, determining its area under the curve (AUC). The study gives an alternative approach to existing analytical techniques that require vast amounts of organic solvents, which are costly and can be toxic, thus impacting the environment and increasing the analysis cost. The study evaluated the method's whiteness and greenness by utilizing the Complex green analytical procedure index, analytical GREEness calculator and Red Green Blue algorithm tool. The linierity was found to be 5 to 30 µg/ml. The present study has developed an infrared (IR) spectroscopic method that employs a straightforward sample preparation technique in methanol. The IR spectroscopic method's linearity range was determined to be 5-30 µg/ml. The p-value was 0.001 at 95 % confidence level assuring better recovery. This method is evaluated according to the Q2R1 ICH guideline. It is applicable to routine quality control analysis without pre-extraction using green IR spectroscopy. In conclusion, the study demonstrated that IR spectrophotometric techniques can quantify sulfanilamide while reducing the use of organic solvents, contributing to the green-and-white analytical chemistry approach. Analysis of real objects based on surface-enhanced Raman spectroscopy (SERS) often utilizes new SERS substrates and/or complex analysis procedures, and they are optimized for only the determination of a single analyte. Moreover, analysis simplicity and selectivity are often sacrificed for maximum (sometimes unnecessary) sensitivity. Consequently, this trend limits the versatility of SERS analysis and complicates its practical implementation. Thus, we have developed a universal, but simple SERS assay suitable for the determination of structurally related antibiotics (five representatives of the sulfanilamide class) in complex objects (human urine and saliva). The assay involves only mixing of acidified analyzed solution with co-activating agent (polydiallyldimethylammonium chloride - PDDA) and SERS substrate (standard colloidal silver nanoparticles). Acidification promotes the generation of SERS spectra with maximum similarity and intensity, which is explained by the favorable enhancement of the protonated sulfanilamide moiety (a structurally similar part of the studied antibiotics) as a result of its strong electrostatic interaction with the SERS-active surface. Meanwhile, the addition of PDDA improves analysis selectivity by reducing background signal from body fluids, enabling to simplify sample pretreatment (dilution for urine; mucin removal and dilution for saliva). Therefore, the assay allows for rapid (≤10 min), precise, and accurate class-specific determination of sulfanilamides within concentration ranges suitable for non-invasive therapeutic drug monitoring in urine (40-600 μM) and saliva (10-30 μM). This study employed the use of a microfluidic paper-based analytical device (µPAD) to determine the concentration of nitrite in pork and enhanced the limit of detection by analyzing the coffee-ring effect. The µPAD was fabricated by designing and embedding wax channels onto the cellulose-based filter paper through printing and subjecting the paper to heat treatment to allow wax penetration. Nitrite concentration was determined by monitoring the colorimetric reaction that occurred between nitrite and the added Griess reagent. The limit of detection of this device for nitrite in pork was determined to be 19.2 mg kg -1 by analyzing the inner-chamber reaction, while it could be as low as 1.1 mg kg -1 if the coffee-ring region was analyzed. The overall analysis could be completed within 15 min. Primaquine (PMQ), an important anti-malarial drug, has been recommended by the World Health Organization (WHO) for the treatment of life-threatening infections caused by P. vivax and ovale. However, PMQ has unwanted adverse effects that lead to acute hemolysis in patients with glucose-6-phosphate dehydrogenase (G6PD) deficiency. There is a need to develop simple and reliable methods for PMQ determination with the purpose of dosage monitoring. In early 2019, we have reported an UV-Vis and naked-eye based approach for PMQ colorimetric quantification. The detection was based on a Griess-like reaction between PMQ and anilines, which can generate colored azo products. The detection limit for direct measurement of PMQ in synthetic urine is in the nanomolar range. Moreover, this method has shown great potential for PMQ quantification from human serum samples at clinically relevant concentrations. In this protocol, we will describe the technical details regarding the syntheses and characterization of colored azo products, the reagent preparation, and the procedures for PMQ determination.

The direct photo-transformation of widely used antibiotics, including Tetracycline (TTC), chlortetracycline (CTC), sulfamethoxazole (SMX) and sulfamethazine (SMZ) were quantified for surface water by using artificial UV irradiation. The photolysis rate is directly proportional to the overlap between the absorption spectrum of the solution and the spectrum of the terrestrial sunlight. Increasing overlap fraction of Tetracycline (TC) group than Sulfanomide (Sulfa) group, the transformation of TC group is certified much faster than the sulfa group. The speciation of TC and Sulfa group antibiotics are pH-dependent and consequently influence its light adsorption spectrum. And the toxicity of the four target antibiotics along the photo-transformation was assessed. In field aquatic environment, a temporal- and spatial half-life model described the behavior of the antibiotics in water column of victoria harbour could be validated by using experimentally obtained quantum yield with the target field meteorological data. The modeling results indicated the photolysis rate of different kind of antibiotics varied differently along season, daily time and water depth. Summer, midday and surface layer of water body would be the time- and space-highlight spot in which the phototransformation are the dominant process for antibiotics concentration depletion. Seasonal variety would be enhanced for sulfa-group kind antibiotics, which having only tail overlapped with irradiation spectrum. Daily averaged half-lives of TC group were relatively stable, while the sulfa group antibiotics were found to vary from about 300 to 750 h, dependent on the seasonal change., (Copyright © 2019 Elsevier B.V. Bicarbonate, phosphate, chloride ions, and humic substances are among the constituents most widely present in natural waters. These non-target constituents can greatly affect the efficiency of advanced oxidation processes used for water decontamination due to their capacity to interfere with the adsorption of the target compounds on the surface of TiO 2 , absorb photons, scavenge hydroxyl radicals (·OH), and generate photochemical reactive intermediates. In this work, the effect of these constituents on the degradation of sulfaclozine (SCL) was monitored in three different AOPs systems: UV/TiO 2 , UV/K 2 S 2 O 8 , and UV/TiO 2 /K 2 S 2 O 8 . It was shown that bicarbonate (HCO 3 - ) and phosphate (HPO 4 2- ) ions enhanced the degradation of SCL in UV/TiO 2 and UV/TiO 2 /K 2 S 2 O 8 systems whereas the addition of humic substances influenced these rates with a much smaller extent. On the other hand, the degradation rate of SCL in the UV/K 2 S 2 O 8 system was not affected by the presence of HCO 3 - and HPO 4 2- but was inhibited in the presence of humic substances. In addition, the different mechanisms that can take place in the presence of these constituents were discussed and the degradation rate enhancement in presence of HCO 3 - and HPO 4 2- was attributed to the formation of new reactive species such as carbonate (CO 3 ·- ) and hydroxyl (·OH) radicals activated by TiO 2 holes (h + ). In the presence of chloride (Cl - ) and nitrate (NO 3 - ) ions, an enhancement of SCL adsorption on the surface of TiO 2 was observed. Finally, a comparative study of the degradation of SCL in river water and ultrapure water was reported.

Understanding antibiotic biodegradation is important to the appreciation of their fate and removal from the environment. In this research an Isotope Ratio Mass Spectrometry (IRMS) method was developed to evaluate the extent of biodegradation of the antibiotic, sulphanilamide, in contaminated groundwater. Results indicted an enrichment in δ 13 C of 8.44‰ from -26.56 (at the contaminant source) to -18.12‰ (300m downfield of the source). These results confirm reductions in sulphanilamide concentrations (from 650 to 10mg/L) across the contaminant plume to be attributable to biodegradation (56%) vs. other natural attenuation processes, such as dilution or dispersion (42%). To understand the controls on sulphanilamide degradation ex-situ microcosms assessed the influence of sulphanilamide concentration, redox conditions and an alternative carbon source. Results indicated, high levels of anaerobic capacity (~50% mineralisation) to degrade sulphanilamide under high (263mg/L), moderate (10mg/L) and low (0.02mg/L) substrate concentrations. The addition of electron acceptors; nitrate and sulphate, did not significantly enhance the capacity of the groundwater to anaerobically biodegrade sulphanilamide. Interestingly, where alternative carbon sources were present, the addition of nitrate and sulphate inhibited sulphanilamide biodegradation. These results suggest, under in-situ conditions, when a preferential carbon source was available for biodegradation, sulphanilamide could be acting as a nitrogen and/or sulphur source. These findings are important as they highlight sulphanilamide being used as a carbon and a putative nitrogen and sulphur source, under prevailing iron reducing conditions., (Copyright © 2017. Veterinary antibiotics such as sulfonamides are frequently applied in livestock farming worldwide. Due to poor absorption in the animal gut and/or reversible metabolization sulfonamides are excreted in considerable amounts and can subsequently be detected in liquid manure. As manure is utilized for soil fertilization, sulfonamides can enter the environment via this pathway. Water samples taken below an agriculture field in Lower Saxony revealed the permanent entrance of sulfamethazine into groundwater and concentrations up to 100 ng L -1 were determined. During a long-term lysimeter study, nine sulfonamides were applied to two different soil types by using fortified liquid manure. Divert mobilities were found with sulfamethazine und sulfamethoxazole showing the highest detection frequency in water samples taken below both bedrock and sandy soil. Four years after the last application of fortified manure, sulfonamides were still detectable in the leachate. Based on analyses of manure and fermentation residue samples, a permanent input of sulfonamides to the soil can be excluded. Thus, the positive findings must be caused by the antibiotics once applied. Two approaches for sulfonamides (SA's) determination, based on carbonic anhydrase enzyme inhibition have been investigated in this work. In the first method, nine different SA's have been screened using simultaneous multiphotometric measurement in multiwell plates. The sulfanilamide (SAD) showed significant inhibition compared to other sulfonamides. The carbonic anhydrase (CA) kinetic interactions reveal noncompetitive binding of SAD. Interferences from other inhibitors with enzyme were studied and the results showed very good selectivity toward SAD. In the second approach, an electrochemical enzyme inhibition biosensor, based on CA entrapped in a carbon paste electrode using carbon black nanoparticles and solid paraffin, was successfully applied to SAD measurements. Results from the quantitative analysis of SAD are discussed in terms of detection limit, linear range and sensitivity using multiphotometric and biosensor-based methods The biosensor developed was successfully applied to the determination of SAD at submicromolar levels and it is recommended for application for in situ analysis., (Copyright © 2016 Elsevier Inc. The present work demonstrates the first automated enrichment approach for antibiotics in milk using specific DNA aptamers. First, aptamers toward the antibiotic sulfanilamide were selected and characterized regarding their dissociation constants and specificity toward relevant antibiotics via fluorescence assay and LC-MS/MS detection. The performed enrichment was automated using the KingFisherDuo and compared to a manual approach. Verifying the functionality, trapping was realized in different milk matrices: (i) 0.3% fat milk, (ii) 1.5% fat milk, (iii) 3.5% fat milk, and (iv) 0.3% fat cocoa milk drink. Enrichment factors up to 8-fold could be achieved. Furthermore, it could be shown that novel implementation of a magnetic separator increases the reproducibility and reduces the hands-on time from approximately half a day to 30 min.

Sorption experiments of sulfanilamide (SAA) on well-characterized samples of soil size-fractions were combined with the modeling of SAA-soil-interaction via quantum chemical calculations. Freundlich unit capacities were determined in batch experiments and it was found that they increase with the soil organic matter (SOM) content according to the order fine silt > medium silt > clay > whole soil > coarse silt > sand. The calculated binding energies for mass-spectrometrically quantified sorption sites followed the order ionic species > peptides > carbohydrates > phenols and lignin monomers > lignin dimers > heterocyclic compounds > fatty acids > sterols > aromatic compounds > lipids, alkanes, and alkenes. SAA forms H-bonds through its polar centers with the polar SOM sorption sites. In contrast dispersion and π-π-interactions predominate the interaction of the SAA aromatic ring with the non-polar moieties of SOM. Moreover, the dipole moment, partial atomic charges, and molecular volume of the SOM sorption sites are the main physical properties controlling the SAA-SOM-interaction. Further, reasonable estimates of the Freundlich unit capacities from the calculated binding energies have been established. A sensitive and highly selective stability-indicating gradient HPLC method was developed and validated for simultaneous determination of clorsulon (CLO) and ivermectin (IVM) in the presence of their degradation products. The drugs were subjected to different stress conditions, including acid and alkaline hydrolysis, oxidative, thermal, and photolytic forced degradation. The robustness of the proposed method was assessed using the Plackett-Burman experimental design, the factors affecting system performance were defined, and nonsignificant intervals for the significant factors were determined. The separation was carried out on a ZORBAX SB phenyl analytical column (250 × 4.6 mm id, 5 μm particle size), with gradient elution utilizing 10 mM sodium dihydrogen phosphate and acetonitrile as mobile phase. UV detection was performed for CLO and IVM at 254 nm over a concentration range of 4-140 and 5-50 μg/mL, respectively, with mean percentage recoveries of 99.90 ± 1.30 and 98.59 ± 1.16%, respectively. The proposed method was successfully applied to a pharmaceutical dosage form containing the investigated drugs. The results were statistically compared with the official HPLC methods, and no significant differences were found.

A sensitive method followed by capillary electrophoresis with on-line perconcentration and capacitively coupled contactless conductivity detection (CE-C(4)D) was evaluated as a novel approach for the determination of three sulfanilamide artificial sweeteners (acesulfame-K, sodium saccharin and sodium cyclamate) in beverages. The on-line preconcentration technique, namely field-amplified sample injection, coupled with CE-C(4)D were successfully developed and optimized. The separation was achieved within 10 min under the following conditions: an uncoated fused-silica capillary (45 cm × 50 μm i.d., Leff=40 cm), 20 mmol L(-1) HAc as running buffer, separation voltage of -12 kV, electrokinetic injection of -11 kV × 8 s. The detection limits of acesulfame-K, sodium saccharin and sodium cyclamate were 4.4, 6.7 and 8.8 μg L(-1), respectively. The relative standard deviation varied in the range of 3.0-5.0%. Animal manure application is a main spreading route of veterinary antibiotics in soil and groundwater. The sorption and leaching behavior of five commonly used sulfonamides in five typical soil and soil/manure mixtures from China were investigated in this study. Results showed that the empirical Freundlich equation fits well the sorption behavior of selected sulfonamides (r(2) was between 0.803 and 0.999, 1/n was between 0.68 and 1.44), and pH and soil organic carbon (OC) were the key impact factors to sorption and leaching. Addition of manure was found to increase the Kd values of sulfonamides in five different soils, following the rules that the more polar substances, the more increased extent of sorption after manure amendment (5.87 times for sulfadiazine with Log Kow = -0.09, and 2.49 times for sulfamethoxazole with Log Kow = 0.89). When the simulated rainfall amount reached 300 mL (180 mm), sulfonamides have high migration potential to the groundwater, especially in the soil with low OC and high pH. However, manure amendment increased the sorption capacity of sulfonamides in the top layer, thus it might play a role in decreasing the mobility of sulfonamides in soils. The systematic study would be more significant to assess the ecological risks and suggest considering the influence of manure amendment for the environmental fate of antibiotics.

In salting-out liquid-liquid extraction (SALLE) technique, water-miscible organic solvents are used for extraction of polar analytes from saline solutions. In this study, for the first time, a coupled 1-mL syringes system was utilized to perform a miniaturized SALLE method. Sulfanilamide antibiotic was extracted and determined via the developed method followed by high performance liquid chromatography-ultraviolet detection (HPLC-UV). The extraction process was carried out by rapid shooting of acetonitrile as extraction solvent (syringe B) into saline aqueous sample solution (syringe A), and then the shooting was repeated several times at a rate of 1 cycles(-1). Thereby, an extremely large contact surface area was created between phases and led to a rapid equilibrium and mass transfer. In order to improve the efficiency of the method, the effect of extraction solvent (type and volume), shooting times, salt concentration, and pH on the extraction efficiency was investigated. The best performance of the method was achieved with 250 µL of acetonitrile, salt concentration of 250 mg mL(-1), pH of 7, and shooting times of 5. The linear dynamic range was 0.001-10 µg mL(-1) with the determination coefficient of 0.9999. The relative standard deviation (RSD; n=3, C=5 µg mL(-1)), and the limit of detection (LOD) were 1.55% and 0.3 ng mL(-1), respectively. This study focused on the detection and validation of the residues of the four veterinary drugs, mebendazole, clorsulon, diaveridine, and tolfenamic acid, using high performance liquid chromatography (HPLC) and an ultraviolet (UV) detector. Utilizing C18 column as a stationary phase and applying appropriate mobile phases to each analysis according to the properties of the analytes, target compounds in food samples were successfully detected and separated within 15-50 min. Additionally, in order to optimize detection, liquid-liquid extraction (LLE) and purification steps were established to minimize the endogenous peaks and their interferences. The method was validated through testing of linearity, accuracy, precision, the limit of detection (LOD) and the limit of quantification (LOQ). The LOQ levels of the four drugs were lower than the maximum residual limit, and the coefficient of determination (R(2) ) was over 0.99. The recovery results ranged from 82.3-105.2%, 79.3-83.3%, 79.4-86.0%, and 81.7-88.5% with relative standard deviations lower than 20% for mebendazole, clorsulon, diaveridine, and tolfenamic acid, respectively, corresponding to the CODEX guideline. The residue depletion profile of sulfachlorpyrazine was studied in healthy broilers after oral administration of Sulfatyf®, given at a dose of 50 mg sulfachlorpyrazine (SCP) per kg body weight once daily for 3 consecutive days. Twenty-five medicated broilers were slaughtered on the 5th, 10th, 14th and 16th day of post-medication, and muscle, fat with skin, liver and kidney tissues were sampled and analysed using an high-performance liquid chromatography (HPLC) method with satisfactory recovery (74.5% ± 2.9%) and specificity. The estimated values of LOD and LOQ were 18.40 and 55.70 ng ml(-1), respectively. On the 5th and 10th day of post-medication, the concentrations of SCP residue in all samples examined were higher than the corresponding MRLs established by the European Union and the highest SCP concentrations were measured in muscle. A maximum withdrawal time of 14 days was calculated to ensure consumer safety.

The purpose of this study was determination of sulphonamide residues (sulphanilamide, sulphadiazine, sulphathiazole, sulphamerazine, sulphamethazine, sulphamethoxazole, and sulphadimethoxine) in cattle meat by the Charm II technique and the validation of sulphonamide levels by high performance liquid chromatography with fluorescence detector (HPLC-FLD). Of 157 meat samples, 9 samples (5.73%) were found positive by the Charm II method. To make quantitative confirmation of sulphonamide content of positive samples, HPLC-FLD was used and four samples were confirmed as positive. In HPLC analysis, the limit of detection (LOD) was in the range 8-15 μg/kg and the limit of quantification (LOQ) was 13-25 μg/kg. Average recoveries of sulphonamides ranged from 44.6% to 81% with relative standard deviations below 6% (n=6). In conclusion, we consider that the results obtained in field screening by only using the Charm II system, as is common practice in Turkey and worldwide, are inadequate and thus the results should be confirmed by sensitive systems like HPLC., (Copyright © 2012 Elsevier Ltd. A new, rapid, and sensitive method was proposed for the determination of sulfonamide residues in milk and chicken muscle samples by microchip electrophoresis with laser-induced fluorescence detection. Separation of fluorescamine-labeled sulfonamides was accomplished by using a buffer containing 5 mmol/L boric acid and 1% (w/v) polyvinyl alcohol (PVA). The pH, amount of PVA, and concentration of boric acid in the running buffer were found to have great influence on the separation. By optimizing these conditions, the separation of four sulfonamides, sulfamethazine, sulfamethoxazole, sulfaquinoxaline, and sulfanilamide, was achieved within 1 min with limits of detection (S/N = 3) of 0.2-2.3 μg/L, which are well below the maximum residue limit. The proposed method also exhibited very good repeatability; the relative standard deviations for both within-day and between-day measurements were ≤3.0%. With a simplified sample pretreatment protocol, fast determination of sulfonamides in real samples was successfully performed with standard addition recoveries of 93.3-100.8 and 82.9-92.3%, respectively.

A rapid, simple and sensitive spectrophotometric method for the determination of some sulphanilamides is described. The method is based on the formation of blue coloured disazo dyes by the diazotization of sulphonamides viz. sulphanilamide (SA), sulphamerazine (SMR), sulphamethazine (SMZ), sulphadimethoxine (SDM), sulphamethoxazole (SMX), sulphadiazine (SDA), sulfathiazole (STZ), sulphaguanidine (SGN), sulphamonomethoxine (SMM), sulphamethoxypyridazine (SMP) in 0.5M hydrochloric acid media at ice bath followed by the azocoupling reaction with acid monoazo dye Tropaeolin O (TrO) at pH=10.5. Formed products are stable for 10h at room temperature. Effective molar absorptivities at absorbance maxima 595nm for disazo dyes were ∼10(4)M(-1)cm(-1). Stoichiometric ratios of the components of disazo dyes were determined by means of mole ratio and continuous variations methods. Linear ranges for sulphanilamides determination were 0.4-14.0μgml(-1). 1. A physiologically-based pharmacokinetic model was developed for the purpose of describing the relationship between plasma concentration of drugs and their deposition into eggs. 2. By incorporating the physiology of egg formation into the model, the transfer of drugs into the egg albumen and yolk could be described using rate constants. 3. The model was used to describe concentrations in albumen and yolk of sulphanilamide, sulphaquinoxaline and pyrimethamine as a function of time using datasets from the literature. 4. The model could be used as a tool to obtain an insight into those properties of a drug which are responsible for the amount of residue in eggs, and could help in the design of critical studies for determining withdrawal periods for eggs.

Sulfadimethoxine was subjected to different International Conference on Harmonisation prescribed stress conditions (hydrolysis, oxidation and photolysis). A stability-indicating high-performance liquid chromatography method was developed for analysis of the drug in the presence of its major degradation products. It involved a Knauer Eurospher C18 thermostated column at 25°C, and 9.57 mM phosphate buffer (pH adjusted to 2.0 with orthophosphoric acid)-acetonitrile (70:30 v/v) as mobile phase. The mobile phase flow rate and sample volume injected were 1.2 mL/min and 20 μL respectively. The selected wavelength for the determination was 248 nm. The method was validated for its linearity, precision, accuracy, specificity and selectivity, and then applied to the assay of sulfadimethoxine in pharmaceutical formulations. The results of the study show that sulfadimethoxine is highly sensitive to basic hydrolysis and oxidation. The mechanisms and schemas of hydrolytic, oxidative and photolytic degradation are also studied. The method developed, which separates all of the most degradation products, is simple, accurate, precise and specific. Sulfachlorpyrazine (SCP) is currently used to treat coccidian infections in turkeys; however, there is no information available about the withdrawal period necessary for the turkey to be safe for human consumption. A high performance liquid chromatography method with ultraviolet-visible light detection was adapted and validated for the determination of SCP in turkey tissues. The procedure is based on isolation of the (SCP sodium) compound from edible turkey tissues (muscles, liver, kidneys, and fat with skin) with satisfactory recovery (72.80 +/- 1.40) and specificity. The residue depletion of SCP in turkeys was conducted after a dose of 50 mg/kg body weight/day had been administrated orally for 3 days. After treatment has been discontinued residue concentrations were detected in tissues on the 7th day. The highest SCP concentrations were measured in muscles. Based on the results presented in this study, it could be assumed that a withdrawal period of 21 days, before medicated turkeys could be slaughtered, would be sufficient to ensure consumer safety.

A HPLC method to determine and quantify sulphachloropyrazine residues from broilers and turkeys is reported. This procedure permitted sulphachloropyrazine to be separated from muscle tissue, liver, kidneys and fat with skin after extraction with dichloromethane under slightly acidic conditions. The analytical methodology showed a high specificity and sensitivity and an adequate precision and accuracy with a limit of quantification of 56 ng mL(-1). The peak area showed a linear relationship with a concentration over the range 50-750 ng mL(-1) for sulphachloropyrazine standard solutions. Recovery dates were also satisfactory with values between 69.7 and 77.5%.

A modification of AOAC Method 973.31 is proposed to improve the extraction efficiency of nitrite from cured meat samples and its subsequent quantification based on the diazotization-coupling reaction of sulfanilamide with N-(1-naphthyl)ethylenediamine dihydrochloride (NED). The various experimental parameters were thoroughly investigated. A 5 g meat sample was mixed with 400 mL water; the pH of the mixture was adjusted to 5.5 +/- 0.3 and allowed to stand for 2 h on a water bath at 80 degrees C, with occasional shaking for the complete extraction of nitrite. After quantitative filtration, an aliquot was mixed with chloroacetic-chloroacetate buffer, pH 1.80 +/- 0.05, sulfanilamide, and NED, and the absorbance of the resulting azodye was recorded at 540 nm against water as a reference. Following the recommended procedure, a linear calibration graph was obtained for up to 0.8 microg/mL NO2(-), with a correlation coefficient of 0.9996 and a detection limit (based on the 3 Sb-criterion) of 5.6 ng/mL NO2(-). The proposed method was conveniently applied to various cured meat samples and was validated by comparison with the original AOAC method and by recovery experiments that gave quantitative results (94-98%) with convenient reproducibility. Statistical analysis of the analytical data could not detect any systematic error and revealed the high accuracy and precision of the proposed method.

A procedure for rapid quantitative analysis of pharmaceutical powders is described. Powdered samples were measured in a rotating cell in order to avoid sub-sampling problems by increasing the irradiated area. Quantitative determination of sulfathiazole and sulfanilamide, using a simple univariate calibration model is proposed. Even though both antibacterials are of the same chemical family (sulfonamides), the richness of structural information contained in the Raman spectra allowed their determination using the area of two selected bands (1255 and 1629 cm(-1) for sulfathiazole and sulfanilamide, respectively). Relative standard deviation (R.S.D.) values (n=10) of 3.35% and 3.46% for sulfathiazole and sulfanilamide, respectively, demonstrate the good reproducibility of the measurement technique with the rotating cell. The method was successfully applied to the analysis of synthetic mixtures and commercial pharmaceutical powders. The procedure is suitable to be applied to pharmacopoeial uniformity of content testing of batches.

A method for the determination of 12 sulfonamides (SAs) (sulfanilamide, sulfamonomethoxine, sulfacetamide, sulfamethoxazole, sulfadiazine, sulfisoxazole, sulfathiazole, sulfadi-methoxine, sulfamerazine, sulfaquinoxaline, sulfamethazine, sulfanitran) in cosmetics was developed by ultra performance liquid chromatography with photodiode array detector (UPLC-PDA). The chromatographic column used was Acquity UPLC BEHC C18 (50 mm x 2. 1 mm, 1. 7 microm) and the mobile phase was acetonitrile-0. 1% formic acid aqueous solution. A gradient elution program was utilized for the separation and determination. After liquid-liquid extraction, SAs were separated and detected by UPLC-PDA. The qualification analysis was done by using retention time and spectrum, and the quantification was based on the detection wavelength of 268 nm. The limits of qualification (S/N = 3) and quantification (S/N = 10) for 12 SAs were 1 microg/g and 2 -3 microg/g, respectively. The correlation coefficient of linear calibration curve was over 0. 999 7 within the SAs concentration range of 1 - 25 mg/L (except sulfanitran 0. 5 - 12. 5 mg/L). At the spiked levels of 40 and 8 microg (except sulfanitran 20 and 4 microg), the average recoveries for 12 SAs were 86. 8% - 98. 1% and 80. 1% - 96. 9%, respectively. Relative standard deviations were less than 10%. Routine tests show that the method is simple, fast, and has a good separation efficiency. It can be routinely used for the determination of these SAs in cosmetics.

Realistic estimation of sorption parameters is essential to predict long-term herbicide availability in soils and their contamination of surface water and groundwater. This study examined the temporal change of an effective partition coefficient Kd(eff) for the herbicides simazine, diuron, and oryzalin from a 0.12 ha field experiment during 7 vineyard growing seasons. Kd(eff) is the ratio of solvent extractable herbicide concentrations in the top soil (0-2 cm) to the average concentrations in runoff water and is considered to assess the effective availability of herbicides to runoff transfer. Kd(eff) increased largely with aging time since application, from values similar to those of the literature (determined in 24 h batch conditions, Kd(ref)), up to 88, 164, and 30 times these initial values for simazine, diuron, and oryzalin respectively. The seasonal variation of Kd(eff) values between years and compounds could be adequately described by a unique model, taking into account the cumulative rainfall since application and Kd(ref) of each compound. This simple model was able to represent the influence of the soil moisture content and its changes in the different biological and physicochemical processes that may contribute to the (bio)available, sorbed, or entrapped state of any of the studied herbicides with aging time under Mediterranean climate.

Nine selected antibiotics in the Victoria Harbour of Hong Kong and the Pearl River at Guangzhou, South China, were analyzed using high-performance liquid chromatography-electrospray ionization tandem mass spectrometry. The results showed that the concentrations of antibiotics were mainly below the limit of quantification (LOQ) in the marine water of Victoria Harbour. However, except for amoxicillin, all of the antibiotics were detected in the Pearl River during high and low water seasons with the median concentrations ranging from 11 to 67 ng/L, and from 66 to 460 ng/L, respectively; and the concentrations in early spring were about 2-15 times higher than that in summer with clearer diurnal variations. It was suggested that the concentrations of antibiotics in the high water season were more affected by wastewater production cycles due to quick refreshing rate, while those in the low water season may be more sensitive to the water column dynamics controlled by tidal processes in the river.

Use of an RP-C(12) analytical column with a mobile phase at pH 4.5 enabled excellent chromatographic separation and quantification of sulfadiazine (SDZ), sulfamerazine (SMR), and sulfamethazine (SMZ) in edible calf and pig tissue (muscle and liver). Tissue samples were extracted with acetonitrile, centrifuged without further clean-up, and analyzed by LC-DAD. The proposed conditions are useful for a rapid and reliable screening of the SDZ, SMR, and SMZ content of calf and pig tissue at concentrations lower than the maximum residue level (MRL) (LOQ between 27 and 55 ppb). Separation of some possible SMZ metabolites should be further investigated.

Field studies monitoring herbicide pollution in the vineyards of Burgundy (France) have revealed that drinking water reservoirs are contaminated with several pre-emergence herbicides. An assessment of the leaching of two such herbicides, diuron and oryzalin, was therefore performed using lysimeters, under outdoor conditions, from May 2001 to May 2002. Four vineyard soils from Vosne-Romanée (Burgundy) were chosen along a topolithosequence: a rendosol and three calcosols. After 673 mm of rainfall, greater amounts of diuron than oryzalin were measured in percolates: respectively 0.10-0.84% and 0.02-0.43% of applied herbicide, depending on soils. Measurements for diuron metabolites detected greater amounts of DCPMU than DCPU in the percolates: respectively 0.05-0.13% and 0-0.04% of the applied diuron. At the end of the monitoring period, more residues of diuron than oryzalin were recovered in the soil profiles: respectively 4.6-9% and 1.4-4.4%. The oryzalin residues were found mainly in the upper 10 cm of soil columns, whereas diuron residues were present in the whole core. The mobility of both oryzalin and diuron seems fairly well-related to soil organic carbon content; the mobility of diuron is also related to soil texture (sand and coarse material contents). Under such experimental conditions, this study confirms that diuron leaching, and therefore potential groundwater contamination, is greater than that of oryzalin.

This manuscript reports the usefulness of the determination of sulphathiazole (ST) using photochemically induced fluorescence (RTPF) and RTPF coupling with first derivative (D1-RTPF), and the determination of sulphanilamide (SAN) by meaning first derivative of the emission spectrum. By irradiating 5 min, with intense UV radiation, sulphathiazole, in ethanol:water 20:80 (v/v) solutions at pH 4.5-5.0, show fluorescence emission at 342 nm (lambdaex=251 nm). Under these conditions, a linear relation, fluorescence intensity-ST concentration, was found between 0.23 and 3.00 microg mL(-1) of ST. The method was applied for determining ST in a pharmaceutical drug. ST was also determined in honey by using the D1-RTPF signal, applying the standard addition method, and measuring at 324.8 nm. Under the same experimental conditions of pH and solvent, a fluorimetric method for determining SAN in presence of ST is proposed. Calibration graphs for SAN determination were established using the amplitude of the first derivative of the emission spectrum measured at 324.4 nm, as the analytical signal. This method has been applied to determining SAN in a pharmaceutical formulation.

The aerobic biodegradability of four antimicrobials (sulfamethazine, sulfamethoxazole, sulfathiazole, and trimethoprim) was investigated in sewage collected at four treatment stages (primary treatment, activated sludge treatment, aerobic nitrification process, and after disinfection of treated sewage) of a municipal sewage treatment plant. The biodegradability tests were conducted in aerated batch reactors by spiking the sewage with 20 microg/L of each of the test substance. Concentration profiles of the assayed compounds were monitored during a 54-d period using liquid chromatography/electrospray ionization/mass spectrometry. Substantial differences in the degradation curves were observed between trimethoprim and the three sulfonamides. The behavior of the latter was characterized by a general biodegradability in the primary and secondary treatment. The highest degradation rates were obtained in the sewage from the activated sludge treatment, where no adaptation phase was observed. On the other hand, the onset of biodegradation in the sewage from the primary treatment was preceded by a lag phase ranging from 10 to 15 d. In contrast, trimethoprim displayed high resistance to microbial degradation in the sewage from the primary treatment and the activated sludge treatment. However, primary degradation of this compound was completed within only 3 d in the sewage from the nitrification process.

Soil contamination from pharmaceuticals is an emerging problem, though quantitative data on their microbial effects are lacking. Thus, nine pharmaceutical antibiotics were tested for their effects on the microbial iron(III) reduction in six different topsoils. Complete dose-response curves were obtained and best-fit by sigmoidal Logit, Weibull, Box-Cox Logit, and Box-Cox Weibull equations (r2 0.73-1.00). The derived effective doses (ED [micromol/kg soil]) for the different antibiotics increased in the order (average ED50 in parentheses) chlortetracycline (53) < sulfadimethoxine (58) < oxytetracycline (170) < sulfadiazine (190) < sulfadimidine (270) = tetracycline (270) < sulfapyridine (430), though no effect was found for sulfanilamide and fenbendazole at doses up to 5,800 and 3,300 micromol/kg, respectively. Due to a strong soil adsorption, especially of the tetracyclines, the corresponding effective concentrations in the soil solution (EC50), derived from sorption experiments, were considerably smaller and ranged from 0.004 micromol/L (chlortetracycline) to 120 micromol/L (sulfapyridine). The effects of the antibiotics were governed by soil sorptive properties, especially the concentration of soil organic matter. The microbial inhibition was influenced indirectly by the soil pH, which affects the ionization status of the amphoteric antibiotics.

An experimental study was conducted in a 91.4-ha Mediterranean vineyard catchment in southern France to characterize the fate and transport of oryzalin in runoff water and thus to assess the risk of contamination of surface waters. Oryzalin concentrations in soil were monitored on two fields, one no-till and one tilled from March 1998 to March 2000. Concentrations in solution and on solid phase of runoff water were measured at the outlets of both fields and the catchment. The droughts in the two summer periods reduced the dissipation of oryzalin and increased its field half-life up to 35 days. Consequently, oryzalin was detected throughout the year in runoff water, with maximum dissolved concentrations > 600 microg l(-1) at the field scale. Oryzalin transport essentially occurred in solution. At the no-till field, seasonal losses were 2.29% and 1.89% of the applied amount in 1998 and 1999, respectively. The corresponding values at the tilled field were 1.56% and 0.29%, since tillage reduced total losses by reducing surface runoff. At the catchment scale, oryzalin concentrations were smaller than those at the field scale, due to dilution effects and staggering of application. Large part of the overland flow from the fields reinfiltrated in the ditches before reaching the outlet of the catchment. As a result, seasonal oryzalin losses were <0.2% of the applied amount.

Two derivative spectrophotometric (ratio derivative spectra and algorithm bivariate calibration) and a chemometric methods (partial least squares, PLS) are proposed for the simultaneous determination of binary mixtures in tablet analysis and dissolutions tests, without prior separation. These approaches are successfully applied to quantify trimethoprim (TMP) combined with sulfamethoxazole (SMX) or sulfamethazine (SMZ) or sulfafurazole (SFZ) using the information in the absorption spectra of appropriate solutions. Beer's law was obeyed in the concentration range of 0.98-17.5 microg/ml for TMP, 0.95-17.2 microg/ml for SMX, 1.16-17.5 microg/ml for SMZ and 0.97-17.4 microg/ml for SFZ. The first derivative (1D) bivariate algorithm method involves the use of four calibration curves: two for each compound at two different wavelengths, selected by Kaiser's method. Similarly, the first derivative ratio spectrophotometry employs the linear relationship between the ratio spectra of the analytes and the concentration range. The results were compared with those obtained by PLS multivariate calibration. The calibration models from PLS were pre-treated by orthogonal signal correction and evaluated by cross-validation using the 'SIMCA-P 9' software. Synthetic mixtures of TMP and sulfonamides were used in five different sets for the validity of the calibrations. Mean recoveries for derivative ratio, derivative bivariate and PLS methods were found to be between 99.7% and 102.0% for TMP, 99.4% and 100.2% for SMX, 99.3% and 101.0% for SMZ and 98.1% and 102.3% for SFZ. The calibrations of the three methods were successfully applied to the assaying and dissolution of placebo and commercial tablets without any prior separation. More than 85% of TMP, SMX and SMZ were dissolved within 15 min. For SFZ, only 85% of the compound was dissolved after 60 min. In this study, the three spectrophotometric methods can be satisfactorily used for the quantitative analysis and for dissolution tests of multicomponent dosage forms.

The photochemical fate of five sulfa drugs with varying five-membered heterocyclic substituents (sulfamethoxazole, sulfisoxazole, sulfamethizole, sulfathiazole, and sulfamoxole) was investigated in aqueous solution. The rate of direct photolysis of these compounds is dependent upon the identity of the heterocyclic R group as well as the pH of the solution. Matrix deconvolution methods were employed to determine the absorption spectrum and photolysis rate of each protonation state (cationic, neutral, and anionic). From these data, quantum yields for direct photodegradation were calculated for each protonation state of the sulfa drugs. The quantum yields calculated range from <0.005 for the neutral state of sulfamethizole to 0.7 +/- 0.3 for the protonated state of sulfisoxazole. The protonation state that is most photoreactive varies among the sulfa drugs and cannot be attributed to the rate of photon absorption. Products arising from the direct photolysis of the sulfa drugs were also investigated. For all the compounds, sulfanilic acid was observed as a common product. The singlet oxygen quenching rates of the sulfa drugs were determined by laser flash photolysis and range from (2 +/- 1) x 10(4) M(-1) s(-1) for sulfamethoxazole to (3.0 +/- 0.7) x 10(8) M(-1) s(-1) for sulfamoxole. Reaction of the sulfa drugs with hydroxyl radical is not modulated by the R group, and the rate constants are all near the bimolecular diffusion-controlled limit of 10(10) M(-1) s(-1). The photodegradation of the sulfa drugs in natural water samples of Lake Josephine (St. Paul, MN) and Lake Superior was attributed solely to direct photolysis. This study indicates thatthese similarly structured antibiotics will be subject to a wide range of photodegradation rates with sulfathiazole degrading relatively quickly, sulfisoxazole and sulfamethizole degrading moderately, and sulfamethoxazole degrading much more slowly.

A number of antibacterial drugs (antibiotics) like sulfonamides, tetracyclines and streptomycin are used for the treatment of bacterial diseases in beehives. Yet, the finding of sulfanilamide residues in some 15 Swiss honeys out of some 350 samples could not be explained by such apicultural practice. Bees occasionally collect nectar from meadows treated with the herbicide asulam. Such honey is not only contaminated by asulam, but also by its degradation product sulfanilamide. This is the first report that the use of a herbicide causes the appearance of residues of an antibacterial active metabolite belonging to the category of sulfonamide drugs in food. The relevance of this finding lies in the fact that the use of the herbicide asulam might cause unacceptable residue levels of sulfanilamide in a product fbr human consumption.

High-performance liquid chromatography (HPLC) combined with inductively coupled plasma mass spectrometry (ICPMS) has been studied as a means for the detection of carbon to provide a 'universal' method for detecting organic compounds in chromatographic eluents. Carbon is particularly difficult to ionise and the amount of carbon present in normal chromatographic systems leads to high backgrounds, making detection a challenge. Novel separation approaches were therefore employed, using either entirely aqueous eluents (at temperatures of 60 and 160 degrees C, dependent on the column used) to eliminate the organic modifier completely, or isotopically enriched solvents. For the aqueous eluents, detection limits for sulphanilamide were found to be 2.26 microg, corresponding to 1.13 micromol (0.47 micromol of carbon), injected on a conventional 4.6 mm i.d. column. The use of a narrow bore column with highly isotopically enriched 12C-methanol (99.95 atom%) as organic modifier for the mobile phase enabled the detection of 86 micromol for 13C-triple-labelled caffeine and 79 micromol for 13C-double-labelled phenacetin. The sensitive detection of 12C-compounds with 13C-enriched methanol as organic modifier proved impractical due to a lower level of isotopic enrichment (99 atom%) of this solvent, with the residual 12C-methanol resulting in significant interference., (Copyright 2004 John Wiley & Sons, Ltd.)

A sensitive, rapid, and simple spectrophotometric method is described for the determination of sulfa drugs. The method is based on the formation of a red-colored product by the diazotization of sulfonamides such as sulfathiazole (SFT), sulfadiazine (SFD), sulfacetamide (SFA), sulfamethoxazole (SFMx), sulfamerazine (SFMr), sulfaguanidine (SFG), and sulfamethazine (SFMt), followed by complexation with dopamine in the presence of molybdate ions in (1 + 1) H2SO4 medium. Absorbance of the resulting red product is measured at 490-510 nm, and the product is stable for 2 days at 27 degrees C. Beer's law is obeyed in the concentration range of 0.04-8.0 microg/mL at the wavelength of maximum absorption. The method was used successfully for the determination of some sulfonamides in tablets and eye drops. Common excipients used as additives in pharmaceuticals do not interfere in the proposed method. The method offers the advantages of simplicity, rapidity, and sensitivity without the need for extraction or heating. The limits of detection and quantitation were calculated for SFT, SFD, SFA, SFMx, SFMr, SFG, and SFMt.

An HPLC method for the determination of sulfacetamide sodium and sulfanilamide in Shao Tang Ling ointment was developed. The chromatographic conditions were: column, Spherisorb C18, mobile phase, methanol-0.25% (V/V)-acetic acid solution (7:93); detector UV-257 nm. The average recoveries were 98.9% (n = 5), RSD = 0.64% for sulfacetamide sodium and 99.3% (n = 5), RSD = 1.58% for sulfanilamide. The analytical method is simple, quick, accurate and reproducible.

Healthy gilts and market-ready hogs were administered a single intramuscular (IM) injection of Borgal, a commercial formulation of trimethoprim-sulfadoxine (TMP-SDX), once or twice daily. The objectives were to determine if a newly-developed high-performance liquid chromatographic (HPLC) method would be suitable for measuring the residual concentrations of TMP in the plasma of these live animals, and to determine if the administration of this veterinary drug would leave measurable residues in their plasma and tissues at slaughter. Plasma and tissue concentrations of SDX and TMP from these animals were determined over a period of 14 d using thin-layer chromatography/densitometry (TLCD), and the newly-developed HPLC method, respectively. The lowest detectable limit (LDL) for SDX in plasma and tissue was 20 ppb by TLCD. The HPLC method had a LDL of 5 ppb for TMP in plasma and tissue. Both methods were then used to provide baseline data on the absorption and depletion of TMP and SDX from these healthy animals. It was observed that both TMP and SDX were readily absorbed into the blood and tissues, but TMP was eliminated much faster than SDX. No TMP residues were detected in the plasma of any of the gilts at and beyond 21 h after drug administration. Also, no TMP residues were detected in the plasma of any of the market-ready hogs 24 h after drug administration at either the label dose or twice the label dose. Sulfadoxine residues at concentrations above the maximum residue limit (MRL) of 100 ppb were, however, detected in the plasma, muscle, kidney, liver, and injection sites of hogs slaughtered 1 and 3 d after a single IM administration at the label dose. Although SDX residues were still detectable in the lungs, kidney, liver and plasma of some hogs 10 d after administration of the label dose and twice the label dose, these were below the MRL. Postmortem examination revealed necrosis and inflammation at the injection sites, but no visible deposits of the injected drug.

First-derivative and second-derivative spectrofluorimetric methods have been developed for the analysis of sulphanilamide (SAN) in milk, using an ethanol-water (60:40, v/v) medium. Linear calibration plots were obtained over a concentration range of 30-120 ppb, with correlation coefficients greater than 0.997. Relative standard deviations were within the range 1.6-3.6%. Limits of detection were 0.7-2.7 ppb. Recoveries of 90-102% were obtained. The derivative fluorescence methods are simple, rapid and sensitive and there is no interference from fluorescent impurities in the determination of low levels of SAN in milk.

A stability-indicating, quality control analysis method was developed and validated for the diuretic drug substances hydrochlorothiazide (HCTZ) and chlorothiazide (CTZ). Micellar electrokinetic capillary chromatography employing the anionic detergent sodium dodecyl sulfate at 30 mM in 20 mM sodium borate buffer pH 9.5 was utilized to separate and quantify the active drug substance HCTZ from CTZ and the common impurity, 4-amino-6-chloro-1,3-benzenedisulfonamide (DSA). A 100 microns I.D. uncoated fused-silica capillary was necessary to provide the sensitivity, i.e. 1 microgram/ml, for quantification of the DSA impurity. In this study, the linearity, precision, selectivity, accuracy, reproducibility and limit of quantitation for the method were investigated for HCTZ, CTZ and DSA. As the first validation of a drug substance method by capillary electrophoresis in this laboratory, unusual care was taken to insure reliability and ruggedness with multiple instruments, capillaries and analysts. Precision and reproducibility in the range of 1% R.S.D. was achieved by controlling subtle injection factors. These included minimizing the time in which the capillary ends were not immersed in buffer or sample during the injection process and minimizing the number of assays for each anode or inlet buffer vial. Stacking induced by differences in ionic strength between sample and capillary buffer was reduced by using a sample buffer concentration similar to that of the capillary buffer. Although stacking accomplished by using lower sample buffer concentrations increased sensitivity, reproducibility was decreased. Achievement of the 1% R.S.D. precision level means that many quality control assays for drugs with good absorbance characteristics can be validated with HPLC reproducibility and CE efficiency. These micellar electrokinetic capillary chromatography methods conform to the USA and European Pharmacopoeial validation guidelines.

High-performance liquid chromatographic methods were developed for the determination of azosemide and its metabolite, M1, in human plasma and urine and rabbit blood and tissue homogenates. The methods involved deproteinization of the biological samples: 2.5 volumes of acetonitrile were used for the determination of azosemide and 1 volume of saturated Ba(OH)2 and ZnSO4 for that of M1. A 50-microliters aliquot of the supernatant was injected onto a C18 reversed-phase column in each instance. The mobile phases employed were 0.03 M phosphoric acid-acetonitrile (50:40, v/v) for azosemide and 0.03 M phosphoric acid/0.2 M acetic acid-acetonitrile (83:17, v/v) for M1. The flow-rate was 1.5 ml/min in both instances. The column effluent was monitored by ultraviolet detection at 240 and 236 nm for azosemide and M1, respectively. The retention times for azosemide and M1 were 6.0 and 8.3 min, respectively. The detection limits for both azosemide and M1 in both human plasma and urine were 50 ng/ml. The coefficients of variation of the assay were generally low (below 11.0%) for plasma, urine, blood and tissue homogenates. No interferences from endogenous substances or other diuretics tested were observed.

Simultaneous determination of 7 penicillins, 3 tetracyclines, 5 aminoglycoside antibiotics, 5 macrolide antibiotics, 2 polyether antibiotics, chloramphenicol and 13 drugs was evaluated. The 36 agents in meat were extracted with 10% trichloroacetic acid. The extracted solutions were adsorbed on an Amberlite XAD-2 resin column and an activated carbon resin column and then eluted with methanol and 0.01N HCl-methanol, respectively. The eluted solutions were concentrated to dryness and trimethylsilylated with pyridine, N, 0-bis (trimethylsilyl) acetamide, N-trimethylsilylimidazole and trimethylchlorosilane. They were then measured by gas chromatography with a flame-ionization detector. The recovery for almost all of the 36 compounds from the original meat specimens was more than 82%. The limits of detection for most of the 36 agents using this method were equivalent to the original individual methods of analysis.

In this paper, we optimized the solvent systems of sulfa drugs and five traditional Chinese medicines for thin layer chromatography by means of uniform design method. The optimal system for the eleven sulfa drugs is benzene-n-butanol-n-hexane (5.71:2.86:1.43). The optimal systems for Mu Xiang and Jiu Li Xiang are all absolute chloroform. The optimal systems for Liang Mian Zhen and San Cha Ku are all acetonitrile-ethyl ether-methyl chloride-cyclohexane (0.05:0.3:0.1:0.55). The optimal system for Jin Yin Hua is ethyl acetate-methyl chloride-formic acid (1:1:1). The method has been born out to be theoretical, simple and practical. In our opinion, it is an effective systematic method.

Third-order and fourth-order derivative spectra have been used for determining sulphathiazole, sulphanilamide and their binary mixtures. The method is suitable for 1-22 micrograms ml-1 of sulphathiazole and for 0.5-14 micrograms ml-1 of sulphanilamide and can be applied for determining the sulphonamides in pharmaceuticals.

Three simple and sensitive spectrophotometric methods are described for the determination of sulphathiourea. The methods are based on the interaction of this thioamide with either iodine, tetracyanoethylene (TCNE) or copper nitrate to give the corresponding charge-transfer or metal-ion complexes, which can be measured at 363, 350 and 353 nm, respectively. In each case, a 1:1 complex was formed. Beer's law is obeyed for each procedure in a concentration range of 1-150 micrograms ml-1. The proposed procedures can be applied to the determination of sulphathiourea in its pharmaceutical formulations.
