In this work, we developed a series of novel 5-[3-(4-chlorophenyl)-substituted-1,3-dimethylpyrimidine-2,4,6(1H,3H,5H)-trione derivatives 4(a-e)via a one-pot multicomponent reaction. The structures of the compounds were confirmed using analytical and spectroscopic techniques. Also, the synthesized compounds were screened for their anti-diabetic activity, cytotoxicity and in silico studies. The activity results suggested that the compound 4e exhibited least IC50 values of 0.055 ± 0.002 µM, 0.050 ± 0.002 µM and 0.009 ± 0.001 µM for α-amylase, α-glucosidase and cytotoxicity respectively. Further, in silico molecular docking results revealed that all the obtained compounds effectively interacted with exo-β-D-glucosaminidase and P38 MAP kinase proteins with good binding energies. In that, 4e compound established the least binding energy of −9.6 and −9.1 kcal/mol, respectively. Moreover, our synthesized compounds were subjected to ADME studies, which suggested that all the synthesized compounds obeyed all five rules with good bioavailability and were suitable as drug leads against anti-diabetic and anticancer treatment. Recently, there had been a massive group of people, who were being rapidly affected by melanoma. Melanoma is a form of skin cancer that develops on the skin's surface layer. This is primarily caused due to excessive skin exposure to UV radiation and severe sunburns. Thus, the early detection of melanoma can aid us to cure it completely. This paper intends to introduce a new melanoma detection framework with four main phases viz. segmentation, feature extraction, optimal feature selection, as well as detection. Initially, the segmentation process takes place to the input skin image via Fuzzy C-Means Clustering (FCM) approach. From the segmented image ( Im seg ) , some of the features such as Gray Level Run Length Matrix (GLRM), Local Vector Pattern (LVP), Local Binary Pattern (LBP), Local Directional Pattern (LDP) and Local Tetra Pattern (LTrP) are extracted. As the extracted features (F) suffered from the issue of "curse of dimensionality", this paper utilizes optimization to select optimal features, which makes the detection more precise. As a novelty, a new hybrid algorithm Particle-Assisted Moth Search Algorithm (PA-MSA) is introduced that hybridizes the concept of Moth Search Algorithm (MSA) and Particle Swarm Optimization (PSO), respectively. For the classification process, the optimally chosen features (F opt ) are fed as input, where Deep Convolution Neural Network (DCNN) is used. Abstract Rotational shifting cultivation (RSC) is commonly practiced in northern Thailand for upland rice cultivation, primarily for household consumption. However, the potential health risks from heavy metal contamination in these soils have not been thoroughly explored. This study aimed to evaluate the contamination of six heavy metals (Arsenic (As), Mercury (Hg), Cadmium (Cd), Lead (Pb), Copper (Cu), and Chromium (Cr)) in upland rice across RSC fields with varying fallow periods and assess the associated health risks from rice consumption. Four RSC fields with 5, 6, 10, and 12-year fallow periods were examined. The RSC-6Y and RSC-12Y fields were used for upland rice cultivation in 2022, while the RSC-5Y and RSC-10Y fields were cultivated in 2023. The geo-accumulation index (Igeo) was calculated, and translocation factors (TF) were assessed for the transfer of heavy metals from soil to straw (TFStraw/Soil), straw to grain (TFGrain/Straw), and soil to grain (TFGrain/Soil). The results indicated that after burning vegetation in the RSC fields, the highest concentrations of Pb, Cr, and Hg were found in the ash. In RSC soils, Cu, Cr, As, Pb, and Hg levels were below reference standards, with Cd undetected. In rice grains, the order of concentration was Pb > Cu > Cr > As, with Hg and Cd undetected. Pb levels in rice grains exceeded the safety threshold. Igeo values indicated no contamination to moderate contamination across sites, with negative Igeo values for Cr and Cu, and zero values for Cd. Abstract White spot syndrome virus (WSSV) is an exceptionally harmful virus that generally causes high levels of mortality in cultured shrimp. Attempts at viral suppression have been made to control the disease and have achieved limited efficiency. Recent advances in genome editing technology using CRISPR/Cas9 have led to potential innovations to prevent or treat many viral diseases. In this study, a CRISPR/Cas9 system was applied to WSSV genome cleavage to suppress WSSV infection in shrimp. The U6 promoter sequence was identified. A chimeric DNA vector consisting of the shrimp U6 promoter with gRNA expression sequences specific to two sites of the WSSV genome and the WSSV ribonucleotide reductase promoter with the Cas9 DNA sequence in pAC-sgRNA-Cas9 was constructed. The expression of gRNAs specific to the WSSV genome and Cas9 was determined in primary cultured hemocyte cells and in shrimp tissue via RT‒PCR. The efficacy of CRISPR/Cas9-WSSV for WSSV genome cleavage was determined in vitro and against WSSV-infected Penaeus vannamei. The reaction of synthetic gRNAs and recombinant Cas9 was able to cleave WSSV DNA amplicons, and shrimp that received CRISPR/Cas9-WSSV presented significantly lower WSSV DNA. In addition to interfering with viral DNA propagation, CRISPR/Cas9-WSSV encapsulated with IHHNV-VLP also stimulated an immune-related gene response. Treatment with CRISPR/Cas9-WSSV against WSSV challenge resulted in a significantly longer survival period. This finding has led to the development and application of a CRISPR/Cas9 system for WSSV infectious disease control, which could be used for managing shrimp aquaculture in the future.

Abstract Background Skin commensal bacteria (Staphylococcus epidermidis) can help defend against skin infections, and they are increasingly being recognized for their role in benefiting skin health. This study aims to demonstrate the activities that Myristica fragrans Houtt. seed extracts, crude extract (CE) and essential oil (EO), have in terms of promoting the growth of the skin commensal bacterium S. epidermidis and providing metabolites under culture conditions to disrupt the biofilm formation of the common pathogen Staphylococcus aureus. Methods The culture supernatant obtained from a co-culture of S. epidermidis with M. fragrans Houtt. seed extracts in either CE or EO forms were analyzed using gas chromatography-mass spectrometry (GC-MS) and liquid chromatography tandem mass spectrometry (LC-MS/MS), in silico investigations, and applied to assess the survival and biofilm formation of S. aureus. Results The combination of commensal bacteria with M. fragrans Houtt. seed extract either CE or EO produced metabolic compounds such as short-chain fatty acids and antimicrobial peptides, contributing to the antimicrobial activity. This antimicrobial activity was related to downregulating key genes involved in bacterial adherence and biofilm development in S. aureus, including cna, agr, and fnbA. Conclusion These findings suggest that using the culture supernatant of the commensal bacteria in combination with CE or EO may provide a potential approach to combat biofilm formation and control the bacterial proliferation of S. aureus. This may be a putative non-invasive therapeutic strategy for maintaining a healthy skin microbiota and preventing skin infections.

Purpose – The current research investigates the gender difference in the attitudes of current employees and job seekers (or “future” employees) to payroll cost reduction methods (downsizing vs cutting pay). Design/methodology/approach – Two studies were conducted. In Study 1, we analyzed a secondary dataset (survey data) of 2,139 employees in Ireland. In Study 2, we conducted an online experiment on 384 people in the US. Findings – Study 1 reveals that, for males, downsizing survivors' commitment and job satisfaction levels are higher than those of employees whose pay is cut. In contrast, there were no differences in the commitment and job satisfaction levels between survivors of downsizing and pay-reduced employees for females. The analysis in Study 2 indicates that females are more attracted to organizations that utilize pay cuts over downsizing to overcome financial difficulties. In contrast, males demonstrated no differences in job-seeker attraction outcomes to organizations that chose either of these two payroll cost reduction methods. Thus, the results of the two studies indicate that females, compared to males, form less negative (or more favorable) attitudes toward pay cuts over downsizing. Practical implications – The findings urge organizations to consider gender differences and develop relevant mitigation plans when one method must be chosen to reduce payroll costs. Originality/value – The outcomes of this research indicate that the selection of a payroll cost reduction method may lead to a disparate impact on gender composition in an organization.
