To report a case of popular acantholytic dyskeratosis of the right scrotum. A 58-year-old man complained of pruritic papules on the right scrotum for over 5 years. Dermatologic examination revealed unevenly distributed, millet to green bean sized papules with erosion and maceration, on the right groin. Histopathological changes included hyperkeratosis, parakeratosis, irregular epidermal hyperplasia, acantholysis, intraepidermal vesicles, with a large number of dyskeratotic cells in addition to infiltration of lymphocytes and plasma cells in the dermis. Patient was diagnosed with papular acantholytic dyskeratosis and treated with fractional CO2 laser, and the rashes repeatedly relapsed when followed up 2 years later.

Purpose: The combined use of antibiotics and anti-inflammatory medicine to manage bacterial endotoxin-induced inflammation following injuries or diseases is increasing. The cytokine level produced by macrophages plays an important role in this treatment course. Ciprofloxacin and indomethacin, two typical representatives of antibiotics and anti-inflammatory medicine, are cost-effective and has been reported to show satisfactory effect. The current study aims to investigate the effect of ciprofloxacin along with indomethacin on the secretion of inflammatory cytokines by macrophages in vitro. Methods: Primary murine peritoneal macrophages and RAW 264.7 cells were administrated with lipopolysaccharide (LPS) for 24 h. The related optimal dose and time point of ciprofloxacin or indomethacin in response to macrophage inflammatory response inflammation were determined via macrophage secretion induced by LPS. Then, the effects of ciprofloxacin and indomethacin on the secretory functions and viability of various macrophages were determined by enzyme-linked immunosorbent assay and flow cytometry analysis, especially for the levels of interleukin (IL)-1β, IL-6, IL-10, and tumor necrosis factor (TNF)-α. The optimal dose and time course of ciprofloxacin affecting macrophage inflammatory response were determined by testing the maximum inhibitory effect of the drugs on pro-inflammatory factors at each concentration or time point. Results: According to the levels of cytokines secreted by various macrophages (1.2 × 106 cells/well) after administration of 1 μg/mL LPS, the optimal dose and usage timing for ciprofloxacin alone were 80 μg/mL and 24 h, respectively, and the optimal dose for indomethacin alone was 10 μg/mL. The integration of a two-dimensional (2D) transition metal carbide (MXene) with metal–organic frameworks (MOFs) presents a promising avenue for addressing the limitations of MXene materials in various applications. In this study, we report the fabrication of a novel photocatalyst by in situ-growth self-assembly, where Ti3C2Tx MXene serves as a substrate for immobilizing ZIF-8 MOFs. This composite, Ti3C2Tx@ZIF-8, is further modified with tannic acid (TA) and polyphenol compounds to create a cross-linked network, enhancing the heterogeneous interfaces crucial for efficient photocatalysis. Varying concentrations of tannic acid (10, 20, and 30 mg) were explored to optimize photocatalyst performance. Structural characterization confirms the successful synthesis of Ti3C2Tx@ZIF-8@TA composites, revealing their unique network topology. The prepared ZIF-8, Ti3C2Tx@TA, Ti3C2Tx/ZIF-8@TA10, Ti3C2Tx/ZIF-8@TA20 and Ti3C2Tx/ZIF-8@TA30 photocatalysts were characterized by various techniques (SEM, EDX, FTIR, XRD, TGA, DRS, and BET surface area measurements). Under visible light irradiation, the photocatalyst demonstrates remarkable degradation efficiencies, with RhB, Congo red and methyl orange dyes achieving approximate degradation rates of 87%, 85%, and 79%, respectively, within 60 minutes. Notably, the photocatalyst exhibits low energy consumption, affordability, non-toxicity, and environmental compatibility, underscoring its practical potential for wastewater treatment applications. Human calcitonin (hCT) is an endogenous polypeptide commonly employed in treating bone resorption-related illnesses, but its clinical application is limited due to its high aggregation tendency. Metalloporphyrins are effective in suppressing amyloid fibrillation, positioning them as potential drug candidates for amyloidogenic disorders like Alzheimer's and type 2 diabetes. In this work, we investigated the effects of Fe(III) meso-tetra(4-sulfonatophenyl)porphine chloride (FeTPPS), a highly efficient ONOO− decomposition catalyst, on hCT aggregation. Our findings reveal that FeTPPS effectively precludes hCT fibrillation by stabilizing the monomers and delaying the structural transition from α-helix bundles to β-sheet-rich aggregates. The macrocyclic ring of FeTPPS plays a significant role in disrupting hCT self-associations. Among various porphyrin analogs, those with an iron center and negatively charged peripheral substituents exhibit a stronger inhibitory effect on hCT aggregation. Spectroscopic analyses and computational simulations indicate that FeTPPS binds to hCT's core aggregation region via complexation with His20 in a 1 : 1 molar ratio. Hydrophobic interaction, hydrogen bonding, and π–π stacking with the residues involving Tyr12, Phe19, and Ala26 also contribute to the interactions. Collectively, our study provides a promising approach for developing novel hCT drug formulations and offers theoretical guidance for designing metalloporphyrin-based inhibitors for various amyloidosis conditions.
