24. Engineering Titanium–Hydroxyapatite Nanocomposite Hydrogels for Enhanced Antibacterial and Wound Healing Efficacy

Abstract

External factors often lead to predictable damage, such as chemical injuries, burns, incisions, and wounds. Bacterial resistance to antibiotics at wound sites underscores the importance of developing hydrogel composite systems with inorganic nanoparticles possessing antibacterial properties to treat infected wounds and expedite the skin regeneration process. In this study, a promising TiO2–HAp@PF-127@CBM inorganic and organic integrated hydrogel system was designed to address challenges associated with bacterial resistance and wound healing. The synthesized TiO2–hydroxyapatite (HAp) nanocomposites were coated with an FDA-approved PluronicF-127 polymer and combined with a carbomer hydrogel (CBM) to accomplish the final product. The synthesized nanoparticles exhibit enhanced biocompatibility against L929 and HUVECs and cell proliferation effects. To mitigate oxidative stress caused by TiO2-induced reactive oxygen species in dark environments for effective antibacterial effects, HAp promotes cell proliferation, expediting wound skin layer formation. CBM binds to inorganic nanoparticles, facilitating their gradual release and promoting wound healing. The reduced inflammation and enhanced tissue regeneration observed in the TiO2–HAp@PF-127@CBM group suggest a favorable environment for wound repair. These results align with prior findings highlighting the biocompatibility and wound-healing properties of titanium-HAp-based materials. The ability of the TiO2–HAp@PF-127@CBM hydrogel dressing to promote granulation tissue formation and facilitate epidermal regeneration underscores its potential for promoting antibacterial effects and wound healing applications.

37. Preparation, Swelling, and Drug Release Studies of Chitosan-based Hydrogels for Controlled Delivery of Buspirone Hydrochloride

Abstract

Background: Buspirone is used for the management of depression and anxiety disorders. Due to its short half-life and low bioavailability, it requires multiple daily doses and is associated with some side effects.Aim: This study aimed to develop chitosan-based hydrogels as drug-controlled release carriers.Objective: The objective of this study is to prepare chitosan-based hydrogels as controlled release carriers in order to overcome the side effects of buspirone HCl and improve patients' compliance and their life quality.Methods: Polymer chitosan was polymerized with two monomers, acrylic acid and itaconic acid, to synthesize pH-sensitive hydrogel. The Fourier transform infrared spectroscopy (FTIR), thermogravimetric analysis (TGA) and differential scanning calorimetry (DSC) analysis were performed to confirm the structure formation and thermal stability. Water penetration capability and loading of the drug were performed by porosity and drug loading studies. The swelling and dissolution tests were performed to analyze the pH-sensitive nature of the developed hydrogels.Results: FTIR, TGA, and DSC demonstrated that the chitosan-based hydrogels were successfully prepared. An increase in water penetration and drug loading into the hydrogel network was seen with the high incorporation of chitosan, acrylic acid, and itaconic acid. The swelling and dissolution studies revealed that prepared hydrogel offered the greatest swelling and drug release at a high pH of 7.4. The swelling and drug release from the hydrogel were affected by the concentrations of the incorporated contents. A controlled release of the drug was achieved by using chitosan-based hydrogel as a delivery carrier compared to commercial tablets of buspirone.Conclusion: The results showed that the developed chitosan-based hydrogel can be considered one of the most suitable drug carrier systems for the controlled delivery of buspirone.

Abstract

Objective of the research was to potentiate solubility of Rosuvastatin by nanogels synthesized by free radical polymerization. Surface morphology and structural compatibility were studied through scanning electron microscopy (SEM) and Fourier Transform Infrared (FTIR) spectroscopy. Zeta sizer was used for particle size analysis. Swelling, in-vitro release and solubility studies were accomplished in different media. Similarly, results of in-vitro release of nanogels and commercially available tablets of Rosuvastatin were compared in different dissolution media. Biocompatibility study was performed in healthy rabbits for safety profile. FTIR spectroscopy confirmed the presence of corresponding peaks of respective functional groups of individual constituents and that of the drug in unloaded and loaded nanogels. SEM images depicted the fluffy appearance of nanogels with small pores inside the structure. Particle size of the optimized formulation was in the range of 217.70 ± 2.23 for unloaded and 257.33 ± 2.11 for loaded nanogels. Percent drug loading, in-vitro drug release and solubility profiles were also satisfactory. In comparison with commercially available tablet Rovista®, in-vitro release profiles of synthesized nanogels showed better response. Moreover, synthesized nanogels were also biocompatible. It was concluded that the solubility of Rosuvastatin was potentially enhanced by nanogels and such formulation can also be used for other poorly soluble drugs. [ABSTRACT FROM AUTHOR]

Abstract

The Liu estimator is used to get precise estimatesby introducing bootstrap technique to reduce the problem of multicollinearity in Poisson regression model. In the presence of multicollinearity, the variance of maximum likelihood estimator (MLE) becomes overstated and theinference based on MLEdoes not remain trustworthy. In this article, we proposed some new Poisson bootstrap Liu and ridge estimators. The proposed estimators are then compared with the non-bootstrap Liu and ridge estimators. Based on mean-squared error criterion, the simulation study revealed showed that the proposed estimators showed efficient results as compared to other existing estimators. Abstract

The main objective of the current research work was to fabricate polyethylene glycol-based hydrogels for the controlled delivery of Ketorolac. Polyethylene glycol was chemically cross-linked with acrylic acid using 2-hydroxyethyl methacrylate as a cross-linker with ammonium persulfate and sodium metabisulphite as initiators. To determine the efficiency of the synthesized system, it was subjected to various structural characterization, such as Fourier transform infrared spectroscopy, Differential scanning calorimetry, Thermogravimetric analysis, and Scanning electron microscopy. All the results obtained from structural characterizations confirmed a well-cross-linked, thermally stable, and a porous system. Swelling and in-vitro release studies performed in pH 1.2 and 7.4 showed pH responsiveness as more swelling and drug release were observed in pH 7.4 as compared to pH 1.2. The drug release was considered as non-Fickian transport from kinetic modeling and partially controlled by the viscoelastic relaxation of hydrogel. Sol-gel fraction depicted that gel content was increased as the concentrations of the polymer, monomer, and cross linker were increased. Similarly, the results obtained from stability studies confirmed a stable synthesized system. Abstract

Neural Sign Language Translation (SLT), which is an important cross-modal task to bridge the communication gap between the deaf and the hearing people, has attracted great attention in the field of artificial intelligence, computer vision, and multimedia, etc. Although some great progress has been achieved recently, current neural SLT models still suffer from translation errors caused by under-consideration of non-manual features such as facial expressions, which can carry critical information during communication among the deaf. This paper aims to enhance the traditional neural SLT models by highlighting the facial expression information in the CNN-based sign video representing part. Two novel schemes have been proposed. The first scheme is based on a Multi-stream Architecture, which extracts and represents the facial expression information in an additional stream and aggregates it with the information from the main stream. The second scheme is a pre-trained scheme based on Regions of Interest (RoIs), which first trains a multi-region detection module for recognizing the features of faces and bodies and then transfers the pre-trained parameters to the module in SLT model. To validate the proposed models, we conducted the experiments upon the publicly available SLT benchmark dataset: RWTH-PHOENIX-Weather-2014T. Experimental results showed that both the above-mentioned schemes can improve the performance of SLT models. Especially, the RoIs-based scheme can achieve an improvement up to 1.6+ BLEU-4 score gains, while the multi-stream scheme quantitatively analyzed the importance of the face mainly through flexible components, providing a sufficient theoretical basis for the RoIs-based scheme.
