With the continuous development of informatization, digital video is increasing sharply every moment. Multimedia information represented by video streams has gradually become the main form of information media in the field of information processing. This paper proposes a method of directly extracting I-frames in the compressed domain, which overcomes the shortcomings of traditional video retrieval algorithms that decompress video files before processing. From the point of view of the scene, it discusses how to synthesize the scene summary using I frames to obtain the smallest computer-aided unit in the video. The new model promotes the information transformation of art design activities, provides excellent theoretical tools for the extension of digital network to art design, and creatively brews a digital network learning platform based on network sharing. The new model applies the advantages of computer technology to the design of art categories, promotes the sound and orderly development of art design, and improves the professional level of art designers and producers. The new model was tracked and analyzed by means of work research and judgment, and the results showed that the effect of computer-aided art design and production model design was significant.

The state of charge (SOC) is one of the main indexes of the lithium-ion battery, which affects the practice range of new energy vehicles and the safety of the battery. Nevertheless, the value of SOC cannot be measured directly. At present, the algorithm for estimating the state of charge is not very satisfactory. The multilayer perceptron algorithm designed during this paper encompasses a sensible impact on state estimation. During this paper, the multilayer network is designed to estimate the charged state of lithium batteries from the three-layer artificial neural network to the eleven-layer artificial neural network. After preprocessing the dataset and comparing several activation functions, the ten-layer fully connected neural network is the most efficient to estimate the SOC. In order to prevent over-fitting of the multilayer perceptron algorithm, the two techniques of the BatchNormalization layer and Dropout layer work together to inhibit over-fitting. At the same time, the accuracy of extended Kalman filter, long and short memory network, and recurrent neural network are compared. The multilayer perceptron network designed during this paper has the highest accuracy. Finally, in the open dataset, both the training and test errors achieve good results. The algorithm developed in this paper has made some progress in SOC estimation.

Breast cancer is the most prevalent cancer among women, and diagnosis and treatment represent a substantial challenge due to the lack of adequate molecular targets. It has been shown that long noncoding RNAs (lncRNAs) serve pivotal roles in regulating gene expression in tumors. The roles of long intervening noncoding RNA (Linc)-OIP5 has been demonstrated in different types of cancer; however, its function in breast cancer has not been determined. In the present study, expression of Linc-OIP5, YAP1 (Hippo signaling component) and JAG1 (Notch signaling component) in breast cancer cells with different degrees of malignancy were determined. To assess whether Linc-OIP5 regulated the malignant biological behaviors of MDA-MB-231 cells, its expression was knocked down using a specific small interfering RNA (siRNA), and cell proliferation was determined using a CCK-8 assay, apoptosis was evaluated using an Annexin V-FITC apoptosis detection kit, migration was assessed using a wound healing and transwell migration assays, and cell invasion examined using a transwell invasion assays. The effect of Linc-OIP5 knockdown on YAP1 and JAG1 expression was quantified using reverse transcription-quantitative PCR and immunoblotting. Cell proliferation, migration and invasion were reduced, while apoptosis was increased in MDA-MB-231 cells transfected with Linc-OIP5-specific siRNA. Mechanistic investigations showed that Linc-OIP5 knockdown downregulated YAP1 and JAG1 expression. The results of the present study suggest that Linc-OIP5 affects the malignant biological behaviors of MDA-MB-231 cells, at least partly through its effects on YAP1/JAG1 signaling. Whilst there are a number of mechanisms underlying the pathogenesis of breast cancer, the results of the present study highlight Linc-OIP5 as a potential therapeutic target in breast cancer.

Diabetic patients often have a heightened risk of cardiomyopathy, even in the absence of traditional risk factors such as hypertension and atherosclerotic coronary artery disease. Diabetic cardiomyopathy is characterized by a typical cardiomyopathy specific to diabetes, the pathogenesis of which has yet to be fully elucidated. As a well-documented oncogenic long noncoding RNA (lncRNA), metastasis-associated lung adenocarcinoma transcript 1 (MALAT1) has been implicated in a variety of pathological processes, including diabetic complications. This study aimed to evaluate the functional roles of MALAT1 in the pathogenesis of diabetic cardiomyopathy. Spontaneously diabetic (db/db) C57BL/Ks mice were employed to establish diabetic cardiomyopathy models in vivo and high glucose (HG)-cultured mouse cardiomyocytes for myocardial damage models in vitro. Mouse left ventricular volume and function were evaluated by echocardiography, while the myocyte cross-sectional area was calculated to evaluate the degree of myocardial hypertrophy. TUNEL staining and flow cytometric analysis were performed to evaluate myocardial damage and cardiomyocyte apoptosis. Silencing of MALAT1 was found to attenuate cardiac dysfunction and inhibit cardiomyocyte apoptosis in db/db mice and HG-cultured mouse cardiomyocytes. MALAT1 recruited the histone methyltransferase EZH2 to the miR-22 promoter region and inhibited its expression. EZH2 induced an increased in the expression of ATP-binding cassette transporter A1 (ABCA1), which was identified to be a target gene of miR-22. Silencing of EZH2 was found to improve cardiac function and prevent cardiomyocyte apoptosis in db/db mice and HG-cultured mouse cardiomyocytes in the presence of MALAT1, suggesting that MALAT1 mediated myocardial damage by recruiting EZH2 to the miR-22 promoter. Taken together, this study's findings provide evidence confirming our hypothesis, suggesting the involvement of MALAT1 in the processes of cardiac function and cardiomyocyte apoptosis via the EZH2/miR-22/ABCA1 signaling cascade, which has potential therapeutic implications for the understanding of diabetic cardiomyopathy.

Diabetic cardiomyopathy (DCM) is one of the cardiovascular complications of diabetes mellitus independent of hypertension, coronary disease, and other heart diseases. The development of DCM is multifactorial and hard to detect at an early stage. Long non-coding RNA metastasis-associated lung adenocarcinoma transcript 1 (Malat1) is emerging as a regulator of DCM, the underlying mechanism of its role in DCM has not been elaborated yet. In this study, we established a mouse DCM model via streptozocin injection as evidenced by cell hypertrophy and cell apoptosis of myocardial tissue, and found that Malat1 expression was upregulated in the myocardium in DCM mice. Meanwhile, elevated expression of pro-apoptotic factors p53, p21, cleaved caspase 3, cleaved caspase 9 and BAX, and down-regulation of anti-apoptotic BCL-2 were observed in DCM myocardium. We further investigated the effect of Malat1 on cardiomyocytes under high glucose condition by silencing Malat1 with its specific short-hairpin RNA. Like in vivo, expression of Malat1 in cardiomyocytes was notably raised, remarkable cell apoptosis and changes in apoptosis-related factors were also observed following high glucose treatment. Besides, we validated that Malat1 acted as a sponge of miR-181a-5p. Inhibition of miR-181a-5p could, at least partially, abolish Malat1 knockdown-induced alteration in cardiomyocytes. In addition, p53, a critical regulator of apoptosis, was validated to be a downstream target of miR-181a-5p. In summary, our findings reveal that Malat1 knockdown attenuates high glucose-induced cardiomyocyte apoptosis via releasing miR-181a-5p, and this mechanism may provide us with new diagnosis target of DCM.

Quasars are essential for astrometric in the sense that they are spatial stationary because of their large distance from the Sun. The European Space Agency (ESA) space astrometric satellite Gaia is scanning the whole sky with unprecedented accuracy up to a few muas level. However, Gaia's two fields of view observations strategy may introduce a parallax bias in the Gaia catalog. Since it presents no significant parallax, quasar is perfect nature object to detect such bias. More importantly, quasars can be used to construct a Celestial Reference Frame in the optical wavelengths in Gaia mission. In this paper, we compile the most reliable quasars existing in literatures. The final compilation (designated as Known Quasars Catalog for Gaia mission, KQCG) contains 1843850 objects, among of them, 797632 objects are found in Gaia DR1 after cross-identifications. This catalog will be very useful in Gaia mission.

The formation of EMT and EMT-induced CSC-like phenotype is crucial for the metastasis of tumor cells. The stromal cell-derived factor-1 (SDF-1) is upregulated in various human carcinomas, which is closely associated with proliferation, migration, invasion and prognosis of malignancies. However, limited attention has been directed towards the effect of SDF-1 on epithelial to mesenchymal transition (EMT) or cancer stem cell (CSC)-like phenotype formation in breast cancer cells and the related mechanism. In the present study, we screened MCF-7 cells with low SDF-1 expression level for the purpose of evaluating whether SDF-1 is involved in EMT and CSC-like phenotype formation in MCF-7 cells. The Lunar-based Ultraviolet Telescope (LUT) is an astronomical instrument aboard Chang'e 3, the lunar probe of China's Lunar Exploration Program that successfully landed on the northern part of the Moon's Mare Imbrium (340.4884E, 44.1214N) in late 2013. LUT is charting an ultraviolet map of the plane of the Milky Way and is also providing long-term light variability monitoring for a sample of RR Lyrae stars. However, the principal goal of the computer-controlled landing of the probe was a safe descent to a stable resting-place, and therefore, the precise orientation of LUT was never a priority. For this reason, at least theoretically, touch-down could have occurred anywhere and, for LUT, at any attitude, which would make the pointing and tracking of the wanted celestial objects practically impossible. Moreover, to reduce the data transmission load, the whole frame of every exposure could not be downloaded: only the image data containing the objects can make it to the ground; also, in order to save on electricity, the telescope does not usually track objects, which means that targets' accurate positions and velocities (within the focal plane CCD) are both needed. This paper presents the astrometric solution devised to solve these problems: feasibility is first shown with experiments done from Earth, and then confirmed with actual LUT observations from the Moon's surface.

Diabetic cardiomyopathy (DCM) is defined as ventricular dysfunction occurring independently of a recognized cause such as hypertension or coronary artery disease. Liver X receptor α (LXRα), a subtype of ligand-activated transcription factors LXRs, has been considered as a potential pharmacological target in the pathogenesis of cardiovascular and metabolic diseases. However, the potential mechanism of how LXRα is regulated in cardiomyocytes is still unclear. This study investigated the effect of activating LXRα with GW3965 on cardiomyocyte apoptosis and its upstream regulator in glucose-induced H9C2 cells. Our data indicated that GW3965 up-regulated the expression of LXRα, inhibited cardiomyocyte apoptosis, and altered the apoptosis-related proteins in glucose-induced H9C2 cells. In addition, GW3965 restored the mitochondrial membrane potential level and decreased the ROS production induced by glucose. Moreover, LXRα was confirmed as a direct target of microRNA-1 (miR-1) that was involved in cardiomyocyte apoptosis of DCM, and overexpression of miR-1 abrogated the inhibiting effect of GW3965 on glucose-induced apoptosis in H9C2 cells. This study highlights an important role of LXRα in the development of DCM and brings new insights into the complex mechanisms involved in the pathogenesis of DCM.

Subdwarf A-type stars (sdAs) are objects that have hydrogen-rich spectra with surface gravity similar to that of hot subdwarf stars but effective temperature below the zero-age horizontal branch (ZAHB). They are considered to be metal-poor main sequence (MS) stars or extremely low-mass white dwarfs (ELM WDs). In this work, using the stellar evolution code, Modules for Experiments in Stellar Astrophysics (MESA), we investigate the sdAs formed both by the evolution of (pre-)ELM WDs in double degenerate systems (DDs) and metal-poor main sequence with the single evolution models. We find that both of the evolutionary tracks of ELM WDs and metal-poor MSs can explain the observation properties of sdAs. However, the proportion between these two populations are uncertain. In this work, we adopt the method of binary population synthesis of both ELM WDs in the disk and metal-poor MS in the halo to obtain the populations of ELM WDs and metal-poor MSs at different stellar population ages and calculate their proportions. We find that the proportion of metal-poor MSs to sdA population for a stellar population of 10 Gyr is $\sim$98.5 percent, which is consistent with the conclusion that most sdAs ($>$95 percent) are metal-poor MSs. We report the distances of molecular clouds at high Galactic latitudes (|b|>10$^\circ$) derived from parallax and G band extinction (A$_{\rm G}$) measurements in the second Gaia data release, Gaia DR2. Aided by Bayesian analyses, we determined distances by identifying the breakpoint in the extinction A$_{\rm G}$ towards molecular clouds and using the extinction A$_{\rm G}$ of Gaia stars around molecular clouds to confirm the breakpoint. We use nearby star-forming regions, such as Orion, Taurus, Cepheus, and Perseus, whose distances are well-known to examine the reliability of our method. By comparing with previous results, we found that the molecular cloud distances derived from this method are reliable. The systematic error in the distances is approximately 5%. In total, 52 molecular clouds have their distances well determined, most of which are at high Galactic latitudes, and we provide reliable distances for 13 molecular clouds for the first time., Accepted by A&A. 13 pages, 9 figures

The effects of temperature and humidity on the emergence patterns of diamondback moth, Plutella xylostella (L.) (Lepidoptera: Plutellidae), were studied at four temperatures (20, 25, 28, and 33 degrees C),three relative humidities (50, 70, and 90%) and a photoperiod of 14:10 (L:D) h. Both sexes emerged primarily in the late afternoon to early morning, and the peaks of emergence varied depending on temperature and humidity. Females emerged significantly earlier than males. Incubation at 33 degrees C and 90% RH had a significant effect on the emergence rate, but no significant interaction was found between temperature and humidity. Emergence duration was 25.3 h for both sexes at 90% RH, with emergence duration at 33 degrees C lower than the other treatments. The sex ratios of diamondback moth under different temperature and humidity treatments were approximately 1:1, and there were no significant effects of temperature and humidity or interactions between these two factors.

Context. Quasars are spatially stationary, and they are essential objects in astrometry when defining reference frames. However, the census of quasars is far from complete. Mid-infared colors can be used to find quasar candidates because AGNs show a peculiar appearance in mid-infrared color, but these methods are incapable of separating quasars from AGNs. Aims. The aim of our study is to use astrometric and mid-infrared methods to select quasars and get a reliable quasar candidates catalog. Methods. We used a near-zero proper motion criterion in conjuction with WISE (all-sky Wide-field Infrared Survey Explorer) [W1-W2] color to select quasar candidates. The [W1-W2] color criterion is defined by the linear boundary of two samples: LAMOST DR5 quasars, which serve as the quasar sample, and LAMOST DR5 stars and galaxies, which serve as the non-quasar sample. The contamination and completeness are evaluated. Results. Studies were conducted to investigate whether endothelial progenitor cells (EPCs) derived from CD34(+) cells could differentiate to endothelial- and smooth muscle-like cells, respectively. Differentiation was induced by either basic fibroblast growth factor (bFGF) or platelet-derived growth factor-BB (PDGF-BB). Further, the contribution of these cells to cooperative blood vessel formation was examined. Immunofluorescence microscopy and Western blot analysis were used to assess vascular smooth muscle (SMC) or endothelial (EC) cell differentiation. SMC (α-SM actin and calponin) or EC (CD31 and von Willebrand factor, vWF) markers showed significant expression. In contrast, expression of the stem/progenitor cell marker CD133 gradually decreased. To quantify the percentage of positive cells of smooth muscle cell marker and endothelial cell marker, flow cytometric analysis was performed, and the results demonstrated that up to 83.76% of PDGF-BB treated-cells were positive for α-SMA marker and up to 89.27% of bFGF-treated cells were positive for vWF marker. To assess functional properties of the endothelial- and smooth muscle-like cells and to demonstrate an association of the two cell types in vascular structures, we evaluated the capacity of these cells to form angiogenesis-like networks on Matrigel. Both endothelial-like cells plated in isolation and co-cultured with smooth muscle-like cells showed formation of vascular networks on Matrigel while smooth muscle-like cells alone did not. Co-culture of fluorescently labeled endothelial- (PKH26, red fluorochrome) and smooth muscle-like cells (PKH67, green fluorochrome) revealed that these cells formed cooperative networks of tubular structures on Matrigel. Collectively, our findings suggest that EPCs can differentiate into both smooth muscle- and endothelial-like cells and contribute to the development of cooperative vascular networks. Notably, this differentiation could also provide a stable foundation for maintenance of the newly formed vascular networks.

The aim of the present study was to investigate the protective effects of T0901317 (T0), a potent agonist of liver X receptors (LXRs), on high glucose-induced oxidative stress and apoptosis in H9c2 cardiac cells. Exposure of H9c2 cells to high glucose alone, not only caused a significant increase in apoptosis and reactive oxygen species (ROS) generation, but also led to a decrease in mitochondrial membrane potential (ΔΨm), release of cytochrome c, decrease in Bcl-2, increase in Bax expression and the activation of caspase-3, caspase-9, poly (ADP-ribose) polymerase (PARP) and nuclear factor (NF)-κB. However, pretreatment with T0 effectively decreased apoptosis, reduced the levels of ROS, abrogated ΔΨm, inhibited cytochrome c release and NF-κB activation, increased Bcl-2 and decreased Bax expression. In conclusion, our data suggest that T0 exerts protective effects against high glucose-induced apoptosis in H9C2 cardiac muscle cells via inhibition of ROS production, mitochondrial death and NF-κB activation.

Hypoxia-inducible factor-1α (HIF-1α) and hepatocyte growth factor (HGF) play important roles in postnatal neovascularization. However, the interaction of these two pathways is not fully understood. The present study utilized CoCl(2) treated-endothelial progenitor cells (EPCs) (EPCs exposure to CoCl(2) are under mimic hypoxia) to examine the expressions of HIF-1α and HGF and futher to assess whether or not the inhibitor (2-methoxyestradiol [2ME2]) of HIF-1α decrease the HGF expression. In addition, to investigate the effects of HGF on the proliferation and tube formation of EPCs under mimic hypoxia, EPCs were transfected with NK4 (HGF antagonist) plasmid and exposed to CoCl(2), then the proliferation of these EPCs was assayed by MTS and the tube formation capacity of these EPCs on Matrigel was detected. The analysis indicated that CoCl(2) treatment induced HIF-1α expression of EPCs, and futher promoted HGF expression. While after 2ME2 was used in CoCl(2) treated-EPCs, HGF expression was markedly inhibited compared with non-pretreated EPCs with 2ME2, which also showed that HGF expression in EPCs was mediated by HIF-1α. Further, the results showed that after EPCs were transfected with NK4 in spite of being exposed to CoCl(2), their proliferation activity and tube formation capacity were weakened, which in turn indicated that HGF could promote the proliferation and the tube formation of EPCs, and this process might be regulated by HIF-1α.

The aims of this study were to assess the effects and potential mechanisms of parthenolide on the expression of vascular endothelial growth factor (VEGF), interleukin 8 (IL-8) and matrix metalloproteinase 9 (MMP-9) in human breast cancer cell line MDA-MB-231. After incubation with different concentrations of parthenolide for 24 h, MDA-MB-231 cells were collected, and the expressions of VEGF, IL-8 and MMP-9 were measured by real-time PCR and Western blot. The secretions of VEGF, IL-8 and MMP-9 in culture supernatant of MDA-MB-231 cells were then measured with ELISA assays. The NF-κB DNA-binding activity of breast cancer cells treated with parthenolide was analyzed using electrophoretic mobility assays. The real-time PCR and Western blot data showed that the expressions of VEGF, IL-8 and MMP-9 were significantly inhibited by parthenolide at both transcription level and protein level in MDA-MB-231 cells. ELISA results also confirmed these effects at a secretion level. The electrophoretic mobility assay results demonstrated that parthenolide can inhibit NF-κB DNA-binding activity of the breast cancer cells. Hence, the expression of VEGF, IL-8 and MMP-9 may be suppressed by parthenolide through the inhibition of NF-κB DNA-binding activity in MDA-MB-231 cells.

Diabetic cardiomyopathy (DCM) is a progressive disease of the heart muscle and the third most common cause of heart failure. In the present study, we evaluated the effects of bone marrow‑derived endothelial progenitor cell (EPC) transplantation on the development of DCM in a streptozotocin (STZ)-induced diabetic rat model. Ex vivo generated, characterized and cultivated rat EPCs were identified by flow cytometry of their surface markers. EPCs were transplanted intravenously into rats through the tail vein 6 weeks after they were challenged with STZ and the rats were sacrificed 4 weeks later. Before sacrifice, left ventricular (LV) catheterization was performed to evaluate the cardiac function. Myocardium sections were stained with Masson's trichrome staining to investigate myocardial collagen contents. Fibrosis-, apoptosis- and oxidative stress-related gene expressions were analyzed by western blot analysis. Transplantation of EPCs alleviated the impaired cardiac function associated with diabetes and decreased the collagen volume in diabetic myocardium resulting in improved cardiac function. Furthermore, EPC transplantation decreased the expression of type I collagen, Bax, caspase-3 and p67phox, while increasing the expression of Bcl-2 and manganese superoxide dismutase (MnSOD). Taken together, our results suggest that transplantation of EPCs improved cardiac function in the rat DCM model, likely through inhibition of cardiomyocyte apoptosis and attenuating myocardial fibrosis.

Mesenchymal stem cells have become a very attractive source of cell implantation for neural tissue engineering. The ideal stem cells for transplantation should be easily obtained, and should rapidly proliferate in vitro and have low immunogenicity. The purpose of this study was to investigate the regenerative potential of adipose-derived stem cells (ADSC) on peripheral nerve repair. ADSCs were isolated from rat adipose tissue and cultured until adherent cells became morphologically homogeneous with a fibroblast-like shape, and transplanted with acellular nerve allografts (ANAs) into rat models with a 10 mm gap of transected sciatic nerve defect. After cell transplantation, we found that ADSC implantation improved functional recovery of exercise behavior and increased wet weight ratio of the anterior tibial muscle. In the electrophysiological testing, we found that the percentage of activated fibers was higher in the ADSC-implanted animals as evidenced by the increase of nerve conduction velocity and amplitude. Histological examination revealed that the number of nerve fibers, axonal diameter and myelin thickness were significantly higher in the ADSC-implanted animals compared to the control. In addition, we demonstrated that the progression of the regenerative process after ADSC implantation was accompanied by elevated expression of neurotrophic factors at both the early and later phase. Taken together, these results suggest that ADSCs can promote the repair of peripheral nerve injury, and the combination of ADSC and ANA transplantation is a new therapeutic method for long distant peripheral nerve defects. Our data also provide evidence indicating the strong association of neurotrophic factor production to the regenerative potential of implanted ADSCs.

The present study was designed to investigate the myocardial expression of liver X receptor alpha (LXRα) in a streptozotocin (STZ)-induced diabetic rat model. Immunohistochemical staining, quantitative real-time RT-PCR, and Western blot analysis were used to determine the expression of LXRα in the myocardium of STZ-induced diabetic rats. The myocardial expression of LXRα target genes, long-chain acyl-CoA synthetase 3 (ACSL3), fatty acid transporter protein (FAT/CD36), ATP-binding cassette transporter A1 (ABCA1), and ABCG1 were also detected. Bisulfite sequencing analysis was employed to examine the methylation status of the CpG island at the LXRα promoter region in the myocardium of STZ-induced diabetic rats. We found that LXRα mRNA and protein expression in the left ventricles, right ventricles, and atria of diabetic rats were gradually increased during the progression of diabetic cardiomyopathy (DCM). The mRNA expression levels of ACSL3 and FAT/CD36 and the protein expression levels of ABCA1 and ABCG1 were also markedly increased in different heart chambers of diabetic rats. SMC (α-SM actin and calponin) or EC (CD31 and von Willebrand factor, vWF) markers showed significant expression. In contrast, expression of the stem/progenitor cell marker CD133 gradually decreased. To quantify the percentage of positive cells of smooth muscle cell marker and endothelial cell marker, flow cytometric analysis was performed, and the results demonstrated that up to 83.76% of PDGF-BB treated-cells were positive for α-SMA marker and up to 89.27% of bFGF-treated cells were positive for vWF marker. To assess functional properties of the endothelial- and smooth muscle-like cells and to demonstrate an association of the two cell types in vascular structures, we evaluated the capacity of these cells to form angiogenesis-like networks on Matrigel. Both endothelial-like cells plated in isolation and co-cultured with smooth muscle-like cells showed formation of vascular networks on Matrigel while smooth muscle-like cells alone did not. Co-culture of fluorescently labeled endothelial- (PKH26, red fluorochrome) and smooth muscle-like cells (PKH67, green fluorochrome) revealed that these cells formed cooperative networks of tubular structures on Matrigel. Collectively, our findings suggest that EPCs can differentiate into both smooth muscle- and endothelial-like cells and contribute to the development of cooperative vascular networks. Notably, this differentiation could also provide a stable foundation for maintenance of the newly formed vascular networks.
