Abstract Diarrhea is a common enteric disease in piglets that leads to high mortality and economic losses in swine production worldwide. Antibiotics are commonly used to prevent or treat diarrhea in piglets. However, irrational antibiotic use contributes to the development of resistance in bacteria and antibiotic residues in animal products, threatening public health, while causing gut microbiota dysbiosis and antibiotic-resistant bacterial infection in piglets. Therefore, the quest for alternative products (such as probiotics, prebiotics, organic acids, enzymes, essential oils, medium-chain fatty acids, zinc, and plant extracts) has recently been clearly emphasized through the increase in regulations regarding antibiotic use in livestock production. These antibiotic alternatives could lower the risk of antibiotic-resistant bacteria and meet consumer demand for antibiotic-free food. Several antibiotic alternatives have been proposed, including immunomodulatory probiotics, as candidates to reduce the need for antimicrobial therapy. Many studies have revealed that probiotics can avert and cure bacterial diarrhea by regulating the gut function and immune system of piglets. In this review, we focus on the major pathogenic bacteria causing piglet diarrhea, the research status of using probiotics to prevent and treat diarrhea, their possible mechanisms, and the safety issues related to the use of probiotics. Supplementation with probiotics is a possible alternative to antibiotics for the prevention or treatment of bacterial diarrhea in piglets. Furthermore, probiotics exert beneficial effects on feed efficiency and growth performance of piglets. Therefore, appropriate selection and strategies for the use of probiotics may have a positive effect on growth performance and also reduce diarrhea in piglets. This review provides useful information on probiotics for researchers, pig nutritionists, and the additive industry to support their use against bacterial diarrhea in piglets. Crossbreeding in dairy cattle with exotic breeds continues to be an appealing practice to the dairy farmers of Bangladesh. However, there is limited knowledge regarding the impact of crossbreeding on both the physical attributes and milk quality traits of crossbred cattle in Bangladesh. Therefore, the primary objective of this study was to evaluate the impact of crossbreeding Bangladeshi local cattle with the exotic Holstein breed on their body characteristics and milk quality. To achieve the goal, data pertaining to body traits and milk samples were gathered from a total of 981 cows from 19 dairy farms located in the northwestern region of Bangladesh. A trained evaluator measured body condition score (BCS), udder score, locomotion score, and body conformation traits. Milk yield information was acquired from official records, while milk composition details were determined through milk analysis. Notably, crossbred cows (Holstein × Local cattle) exhibited greater values for wither height (141 vs. 135, cm), body length (157 vs. 153, cm), heart girth (211 vs. 204, cm), BCS (3.69 vs. 3.27), and udder score (3.29 vs. 2.08) than their Bangladeshi local counterparts. Furthermore, crossbred cows produced 42.4% and 35.3% more milk (10.89 vs. 7.65, kg/d) and fat-corrected milk (10.35 vs. 7.54, kg/d) than Bangladeshi local cattle. However, milk from crossbred cows displayed lower fat and protein content, although their somatic cell score (SCS) and energy-corrected milk remained similar. Additionally, milk from crossbred cows exhibited a longer coagulation time when compared to that of Bangladeshi local cattle. In conclusion, crossbred cows (Holstein × Local cattle) had improved body characteristics with greater milk yield than Bangladeshi local cattle; however, lower fat and protein contents in milk with longer coagulation time were noted.

The occurrence of extended-spectrum beta-lactamase (ESBL) producing bacteria is concerning the scientific community since it confers multiple drug resistance (MDR). The study investigated the prevalence of MDR-ESBL-producing Salmonella strains from shrimp in Sylhet region, Bangladesh. A total of 165 shrimp samples were processed from 55 shrimp specimen from different retail shops. The presence of Salmonella was confirmed by standard methods followed by antibiotic susceptibility testing. Isolates exhibiting resistant to third-generation cephalosporin were considered as Salmonella positive isolates which was later proven by the double disc synergy test. Thirty-nine isolates were identified to be Salmonella positive of which eighteen were from seven department stores and twenty-one from the local market. The body parts had the highest rate of positive samples (30.91%), followed by the head (23.63%), and the tail (16.36%). Additionally, isolated Salmonella were resistant to rifampicin and Cefixime but 100% susceptible to co-trimoxazole, ofloxacin, streptomycin, nalidixic acid, and chloramphenicol, while ciprofloxacin showed intermediate resistance. Vancomycin, cephalexin, ampicillin, and colistin, on the other hand, were among the extremely resistant drugs. Finally, while six of the tested isolates demonstrated resistance against the recommended cephalosporin, three of them (7.69%) were Salmonella ESBL positive in the double disc synergy test. The rising incidence of MDR and the developing prevalence of ESBL-positive Salmonella may put a burden on the healthcare system by limiting access to effective antibacterial drugs. ABSTRACT: This study aimed to compare rotational 3-breed crossbred cows of Viking Red, Montbéliarde, and Holstein breeds with purebred Holstein cows for a range of body measurements, as well as different metrics of the cows' productivity and production efficiency. The study involved 791 cows (440 crossbreds and 351 purebreds), that were managed across 2 herds. Within each herd, crossbreds and purebreds were reared and milked together, fed the same diets, and managed as one group. The heart girth, height at withers, and body length were measured, and body condition score (BCS) was determined on all the cows on a single test day. The body weight (BW) of 225 cows were used to develop an equation to predict BW from body size traits, parity, and days in milk, which was then used to estimate the BW of all the cows. Equations from the literature were used to estimate body protein and lipid contents using the predicted BW and BCS. Evidence suggests that maintenance energy requirements may be closely related to body protein mass, and Holstein and crossbred cows may be different in body composition. Therefore, we computed the requirements of net energy for maintenance (NEM) on the basis either of the metabolic weight (NEM-MW: 0.418 MJ/kg of metabolic BW) or of the estimated body protein mass according to a coefficient (NEM-PM: 0.631 MJ/kg body protein mass) computed on the subset comprising the purebred Holstein. On the same day when body measurements were collected, individual test-day milk yield and fat and protein contents were retrieved once from the official Italian milk recording system, and milk was sampled to determine fresh cheese yield. Measures of NEM were used to scale the production traits. Statistical analyses of all variables included the fixed effects of herd, days in milk, parity, and genetic group (purebred Holstein and crossbred), and the herd × genetic group interaction. External validation of the equation predicting BW yielded a correlation coefficient of 0.94 and an average bias of −4.95 ± 36.81 kg. The crossbreds had similar predicted BW and NEM-MW compared with the Holsteins. However, NEM-PM of crossbreds was 3.8% lower than that of the Holsteins, due to their 11% greater BCS and different estimated body composition. The crossbred cows yielded 4.8% less milk and 3.4% less milk energy than the purebred Holsteins. However, the differences between genetic groups were no longer significant when the production traits were scaled on NEM-PM, suggesting that the crossbreds and purebreds have the same productive ability and efficiency per unit of body protein mass. In conclusion, measures of productivity and efficiency that combine the cows' production capability with traits related to body composition and the energy cost of production seem to be more effective criteria for comparing crossbred and purebred Holstein cows than just milk, fat, and protein yields.

Lacticaseibacillus rhamnosus CRL1505 possesses immunomodulatory activities in the gastrointestinal and respiratory tracts when administered orally. Its adhesion to the intestinal mucosa does not condition its beneficial effects. The intranasal administration of L. rhamnosus CRL1505 is more effective than the oral route at modulating immunity in the respiratory tract. Nonetheless, it has not yet been established whether the adherence of the CRL1505 strain to the respiratory mucosa is needed to provide the immune benefits to the host. In this study, we evaluated the role of adhesion to the respiratory mucosa of the mucus-binding factor (mbf) knock-out L. rhamnosus CRL1505 mutant (Δmbf CRL1505) in the context of a Toll-like receptor 3 (TLR3)-triggered innate immunity response. In vitro adhesion studies in porcine bronchial epitheliocytes (PBE cells) indicated that L. rhamnosus Δmbf CRL1505 adhered weakly compared to the wild-type strain. However, in vivo studies in mice demonstrated that the Δmbf CRL1505 also reduced lung damage and modulated cytokine production in the respiratory tract after the activation of TLR3 to a similar extent as the wild-type strain. In addition, the mutant and the wild-type strains modulated the production of cytokines and antiviral factors by alveolar macrophages in the same way. These results suggest that the Mbf protein is partially involved in the ability of L. rhamnosus CRL1505 to adhere to the respiratory epithelium, but the protein is not necessary for the CRL1505 strain to exert its immunomodulatory beneficial effects. These findings are a step forward in the understanding of molecular interactions that mediate the beneficial effects of nasally administered probiotics.

The emergence and spread of antibiotic resistance threat forced to explore alternative strategies for improving the resistance to pathogens in livestock production. Probiotic lactic acid bacteria represent an alternative for this objective. In this study, seven Lactiplantibacillus plantarum strains from porcine colostrum and milk were isolated, identified and characterized in terms of their abilities to modulate immunity in porcine intestinal epithelial (PIE) cells. Then, two potential immunoregulatory strains were studied in terms of their ability to utilize and grow in wakame (Undaria pinnafida). Isolates were identified by 16S rRNA gene and evaluated by studying their interaction with PIE cells. The expressions of peptidoglycan recognition proteins (PGRPs), nucleotide-binding oligomerization domain (NODs), host defense peptides (pBD), and type I interferons (IFNs) were evaluated by RT-qPCR. The strain 4M4417 showed a remarkable capacity to differentially regulate the expression of PGRP1, PGRP3, NOD1, NOD2, and pBD1 in PIE cells. On the other hand, the strain 4M4326 was the most efficient to improve the expression of IFN-α and IFN-β in PIE cells challenged with poly (I:C). Both L. plantarum 4M4326 and 4M4417 were characterized in terms of their ability to utilize wakame. Results demonstrated that both strains efficiently grew in wakame-based broth. Our results suggest that L. planatrum 4M4326 and 4M4417 are interesting candidates to develop immunomodulatory feeds based on wakame utilization. These new immunosynbiotic feeds could help to reduce severity of intestinal infections and improve immune health status in pigs.

In vitro culture models that precisely mirror the porcine respiratory epithelium are needed to gain insight into how pathogens and host interact. In this study, a new porcine bronchial epithelial cell line, designated as PBE cells, was established from the respiratory tract of a neonatal pig. PBE cells assumed a cobblestone-epithelial like morphology with close contacts between the cells when they reached confluence. The PBE cell line was characterized in terms of its expression of pattern recognition receptors (PRRs) and its ability to respond to the activation of the Toll-like receptor 3 (TLR3) and TLR4 signaling pathways, which are key PRRs involved in the defense of the respiratory epithelium against pathogens. PBE cells stimulated with poly(I:C) were able to up-regulate the expression of IFN-β, IFN-λ1 (IL-29), IFN-λ3 (IL-28B), the antiviral factors Mx1, OAS1, and PKR, as well as the viral PRRs RIG-1 and MDA5. The expression kinetics studies of immune factors in PBE cells allow us to speculate that this cell line can be a useful in vitro tool to investigate treatments that help to potentiate antiviral immunity in the respiratory epithelium of the porcine host. In addition, poly(I:C) and LPS treatments increased the expression of the inflammatory cytokines TNF-α, IL-6, IL-8, and MCP-1/CCL2 and differentially modulated the expression of negative regulators of the TLR signaling pathways. Then, PBE cells may also allow the evaluation of treatments that can regulate TLR3- and TLR4-mediated inflammatory injury in the porcine airway, thereby protecting the host against harmful overresponses.

Mesenchymal–epithelial transition (MET) factor is a proto-oncogene encoding tyrosine kinase receptor with hepatocyte growth factor (HGF) or scatter factor (SF). It is found on the human chromosome number 7 and regulates the diverse cellular mechanisms of the human body. The impact of mutations occurring in the MET gene is demonstrated by their detrimental effects on normal cellular functions. These mutations can change the structure and function of MET leading to different diseases such as lung cancer, neck cancer, colorectal cancer, and many other complex syndromes. Hence, the current study focused on finding deleterious non-synonymous single nucleotide polymorphisms (nsSNPs) and their subsequent impact on the protein’s structure and functions, which may contribute to the emergence of cancers. These nsSNPs were first identified utilizing computational tools like SIFT, PROVEAN, PANTHER-PSEP, PolyPhen-2, I-Mutant 2.0, and MUpro. A total of 45359 SNPs of MET gene were accumulated from the database of dbSNP, and among them, 1306 SNPs were identified as non-synonymous or missense variants. Out of all 1306 nsSNPs, 18 were found to be the most deleterious. Moreover, these nsSNPs exhibited substantial effects on structure, binding affinity with ligand, phylogenetic conservation, secondary structure, and post-translational modification sites of MET, which were evaluated using MutPred2, RaptorX, ConSurf, PSIPRED, and MusiteDeep, respectively. Also, these deleterious nsSNPs were accompanied by changes in properties of MET like residue charge, size, and hydrophobicity. These findings along with the docking results are indicating the potency of the identified SNPs to alter the structure and function of the protein, which may lead to the development of cancers. Nonetheless, Genome-wide association study (GWAS) studies and experimental research are required to confirm the analysis of these nsSNPs.

The immunomodulatory properties of exopolysaccharides (EPSs) produced by Streptococcus thermophilus have not been explored in depth. In addition, there are no comparative studies of the functional properties of EPSs produced by streptococci in different food matrices. In this work, EPSs from S. thermophilus SBC8781 were isolated after soy milk (EPS-s) or cow milk (EPS-m) fermentation, identified, and characterized in their abilities to modulate immunity in porcine intestinal epithelial cells. Fresh soy milk and cow milk were inoculated with S. thermophilus SBC8781 (7 log CFU/mL) and incubated at 37 °C for 24 h. The extraction of EPSs was performed by the ethanol precipitation method. Analytical techniques, including NMR, UV-vis spectroscopy, and chromatography, identified and characterized both biopolymer samples as polysaccharides with high purity levels and similar Mw. EPS-s and EPS-m had heteropolysaccharide structures formed by galactose, glucose, rhamnose, ribose, and mannose, although with different monomer proportions. On the other hand, EPS-s had higher quantities of acidic polymer than EPS-m. The biopolymer production of the SBC8781 strain from the vegetable culture broth was 200–240 mg/L, which was higher than that produced in milk, which reached concentrations of 50–70 mg/L. For immunomodulatory assays, intestinal epithelial cells were stimulated with 100 µg/mL of EPS-s or EPS-m for 48 h and then stimulated with the Toll-like receptor 3 agonist poly(I:C). EPS-s significantly reduced the expression of IL-6, IFN-β, IL-8, and MCP-1 and increased the negative regulator A20 in intestinal epithelial cells. Similarly, EPS-m induced a significant reduction of IL-6 and IL-8 expressions, but its effect was less remarkable than that caused by EPS-s. Results indicate that the structure and the immunomodulatory activity of EPSs produced by the SBC8781 strain vary according to the fermentation substrate. Soy milk fermented with S. thermophilus SBC8781 could be a new immunomodulatory functional food, which should be further evaluated in preclinical trials.

Meat from small ruminants is considered a high quality and delicacy product in many countries. Several benefits have been perceived from probiotics as dietary supplements, such as improved carcass weight, color, tenderness, flavor, muscle fiber structure, water-holding capacity, and healthy fatty acid profile of the meat. Thus, the present review focuses on the effect of probiotics on improving the quality of meat from small ruminants. Though many benefits have been associated with the use of probiotics, the findings of all the considered articles are not always consistent, and the mechanisms behind improving meat quality are not appropriately defined. This variability of findings could be due to the use of different probiotic strains, dosage rates, number of days of experiment, nutrition, breed, age, and health status of the animals. Therefore, future research should emphasize specific strains, optimal dose and days of administration, route, and mechanisms for the specific probiotic strains to host. This review provides a comprehensive overview of the use of probiotics for small ruminants and their impact on meat quality.

This study compared the detailed mineral profiles of milk from purebred Holstein (HO) and four generations (F1 to F4) of crossbred cows (CR) derived from a 3-breed rotational crossbreeding system involving Viking Red (VR) and Montbéliarde (MO) sires. Purebred and CR were kept and fed together in the same herd. Individual milk samples were collected once per cow from 120 multiparous cows (40 HO and 20 for each generation of CR from F1 to F4). The milk samples were analysed for macrominerals (Na, Mg, P, S, K, Ca), essential (Cr, Mn, Fe, Cu, Zn, Mo), and environmental microminerals (Li, B, Ba) using inductively coupled plasma-optical emission spectrometry. The coefficients of variation ranged from 11 to 28% for macrominerals and from 17 to 40% for microminerals. The mineral composition of milk from purebred HO and CR were mostly comparable. Conversely, it differed in different breed combinations of CR: VR-sired cows had the greatest macromineral contents, MO-sired cows the lowest, while HO-sired CR was intermediate. The milk from MO-sired crossbreds had lower contents of some microminerals (Mo, Zn, and Ba), while the milk from HO-sired crossbreds had greater contents (Fe and Mo). We conclude that the mineral profile of milk of CR derived from different combinations of VR, MO and HO breeds was comparable to that of milk from HO cows kept in the same herd, and that the variations in the macro- and micromineral profiles were greater among CR than between purebreds and crossbreds.Highlights The mineral composition affects the nutritional and technological properties of milk, and it is influenced by breed effect. Macro- and micromineral compositions of milk from Holstein and crossbred cows from a 3-breed rotational crossbreeding system were mostly comparable. Crossbred cows of different generations showed great variations in the macro- and micromineral profiles.

Research is still being carried out to develop specific medications or vaccinations to fight norovirus, a key contributor to foodborne illness. This study evaluated certain plant-based active chemicals as prospective candidates for such treatments using virtual screening techniques and other computer assessments. Twenty (20) plant metabolites were tested against the norovirus VP1, VP2, P48, and P22 protein domains using the molecular docking method. In terms of the lowest global binding energy, Asiatic acid, avicularin, guaijaverin, and curcumin exhibited the highest binding affinity with all selected proteins. Each viral protein’s essential binding sites with the potential drugs and drug surface hotspots were uncovered. The ADMET (absorption, distribution, metabolism, excretion, and toxicity) analysis was used to further analyze the pharmacological profiles of the top candidates. According to the results, none of the substances showed any adverse consequences that would reduce their drug-like properties. According to the analysis of the toxicity pattern, no detectable tumorigenic, mutagenic, irritating, or reproductive effects of the compounds were discovered. However, among the top four alternatives, curcumin exhibited the highest levels of cytotoxicity and immunotoxicity. These discoveries may open the way for the development of effective norovirus therapies and safety measures. Due to the positive outcomes, we strongly propose more in vivo experiments for the experimental validation of our findings.

Bovine mastitis (BM) is one of the most common diseases of dairy cattle, causing economic and welfare problems in dairy farming worldwide. Because of the predominant bacterial etiology, the treatment of BM is mostly based on antibiotics. However, the antimicrobial resistance (AMR), treatment effectiveness, and the cost of mastitis at farm level are linked to limitations in the antibiotic therapy. These scenarios have prompted the quest for new preventive options, probiotics being one interesting alternative. This review article sought to provide an overview of the recent advances in the use of probiotics for the prevention and treatment of BM. The cellular and molecular interactions of beneficial microbes with mammary gland (MG) cells and the impact of these interactions in the immune responses to infections are revised. While most research has demonstrated that some probiotics strains can suppress mammary pathogens by competitive exclusion or the production of antimicrobial compounds, recent evidence suggest that other probiotic strains have a remarkable ability to modulate the response of MG to Toll-like receptor (TLR)-mediated inflammation. Immunomodulatory probiotics or immunobiotics can modulate the expression of negative regulators of TLR signaling in the MG epithelium, regulating the expression of pro-inflammatory cytokines and chemokines induced upon pathogen challenge. The scientific evidence revised here indicates that immunobiotics can have a beneficial role in MG immunobiology and therefore they can be used as a preventive strategy for the management of BM and AMR, the enhancement of animal and human health, and the improvement of dairy cow milk production.

The objective of this study was to investigate the effects of 3-way rotational crossbreeding scheme of Holstein (HO) cows with Montbéliarde (MO), Viking Red (VR) and HO sires on body and milk quality traits. Cows were purebred HO; F1: (MO × HO; VR × HO); F2: [MO × (VR × HO); VR × (MO × HO)]; F3: {HO × [MO × (VR × HO)]}; HO × [VR x (MO × HO)]}. Data were collected on 745 cows kept in a commercial farm located in Northern Italy. Milk data were acquired from official milk recording and body condition score (BCS) and conformation traits were measured once by trained evaluator. Data were analyzed using a linear model including fixed effects of parity, DIM class and breed combinations. Average body traits: height at withers (HW), body length (BL) and heart girth (HG) of cows were 139,162 and 207 cm respectively. Crossbred cows had shorter HW, BL and HG than pure HO but greater BCS. Milk yield averaged 31.5 kg/d, with fat and protein content close to 3.8 and 3.75%, respectively. Purebred HO produced greater volume of milk than crossbreds but milk from HO had lower protein content. Different combinations of crossbred cows showed similar performance in terms of milk yield traits, but evidenced different body size. The trial is currently in progress to increase cow sample size and widen the number of herds involved.

Summer transhumance to alpine pastures (ALP) is widespread in dairy systems of alpine regions. This study aimed to investigate the effects of transhumance of Brown Swiss cows to ALP on the yield, composition, and coagulation properties of milk (MCP), and on cheese yield (CY). The study involved 12 multiparous cows kept at a mountain lowland permanent farm (PF), which were divided into two equal groups: One remained at the PF, the other was moved to the ALP (1860 m above sea level) from July to September. Every month (June to October), daily milk yield (MY) and body condition score (BCS) were recorded, and individual milk samples (n = 60, 2000 mL each) were collected to assess milk composition, MCP, and CY. Compared with PF, ALP cows had a reduced MY and BCS, which was maintained on return to the PF, greater fat and lower protein contents of milk. Neither MCP nor CY were affected by summer transhumance. In conclusion, summer transhumance did not affect the cheese making efficiency of milk but depressed MY and consequently daily cheese yield, which was nearly 2 kg/d lower for the ALP than the PF cows and was only partially recovered after returning to the PF in autumn.

The use of high grain rations in dairy cows is related to an increase in rumen acidity. This study investigated whether the rumen acidity status affects rumination time (RT), and the production, composition, coagulation properties (MCPs) and cheese yield (CY) of milk. One hundred early-lactating Holstein cows with no clinical signs of disease and fed total mixed rations were used. Rumen fluid was collected once from each cow by rumenocentesis to determine pH and volatile fatty acid (VFA) content. The cows were classified according to the quartile of rumen acidity (QRA), a factor defined by multivariate analysis and associated with VFA and pH. Rumen fluid pH averaged 5.61 in the first quartile and 6.42 in the fourth, and total VFA content increased linearly with increasing rumen acidity. In addition, RT increased as rumen acidity increased, but only in the daily time interval from 08:00 to 12:00. Milk yield linearly decreased as rumen acidity increased, whereas QRA did not affect pH, fat or protein contents of milk. Furthermore, the MCPs, assessed by lactodynamograph, and CY were unaffected by QRA. It is suggested that differences in rumen acidity have little influence on the nutrient content, coagulation properties and CY of milk.

Sustainable livestock production requires reducing competition for food and feed resources and increasing the utilization of food by-products in livestock feed. This study describes the establishment of an anaerobic batch culture model to simulate pig microbiota and evaluate the effects of a food by-product, wakame seaweed stalks, on ex vivo microbial communities. We selected one of the nine media to support the growth of a bacterial community most similar in composition and diversity to that observed in pig donor feces. Supplementation with wakame altered the microbial profile and short-chain fatty acid composition in the ex vivo model, and a similar trajectory was observed in the in vivo pig experimental validation. Notably, the presence of wakame increased the abundance of Lactobacillus species, which may have been due to cross-feeding with Bacteroides. These results suggest the potential of wakame as a livestock feed capable of modulating the pig microbiome. Collectively, this study highlights the ability to estimate the microbiome changes that occur when pigs are fed a specific feed using an ex vivo culture model. [ABSTRACT FROM AUTHOR]
