Abstract

High-performance liquid chromatography method for anti-asthmatic β2-agonist drug bambuterol, its process-related impurities and its major degradation products was developed and validated using quality by design concept. A 3³ full factorial design was employed to study the effect of three independent factors, namely, ratio of organic modifiers in mobile phase, pH of the buffer and flow rate of the mobile phase. The responses considered were retention time of the last peak and resolution of poorly separated peaks (drug and PR-4 and drug and DP-3). The optimum conditions for separation were determined with the aid of design of experiments. The optimized ternary solvent composition was a mixture of 10 mM ammonium acetate buffer (pH 6.0), methanol and acetonitrile in the ratio of 90:5: 5 (v/v/v) in solvent reservoir A and 10:45:45 (v/v/v) in solvent reservoir B. The separation of the analytes was achieved by using a gradient method. The predictability criteria of the optimized method demonstrated good correlation between observed and predicted response. The method was validated for specificity, linearity, accuracy, precision and robustness in compliance with the International Conference on Harmonization guidelines Q2R1. [ABSTRACT FROM AUTHOR]

Abstract

An efficient one-pot synthesis of 1,2,4-oxadiazol-3- amines from simple starting materials, isothiocyanates, amidines/ guanidines, and hydroxylamine, is described. The reaction is facilitated by metallic-thiophile-assisted desulfurization of in situ formed amidino- or guanidinothiourea to give chemoselectively Nhydroxyguanidine intermediates that give exclusively various 1,2,4-oxadiazol-3-amines in good to excellent yields. Abstract

Based on principles of pharmacophore delineation and drug designing, compounds containing diketofunctionallity namely 1,2-bis[5-thiazolyl]ethane-1,2-diones were designed and synthesized as antiinflammatory agents. The compounds were evaluated in carrageenan-induced rat-paw edema method. G-3, G-6, G-17, G-20, G-23, G-22, L-708 and 906 showed good antiinflammatory activity. In addition as diketo functionality containing compounds are reported to have HIV-1 integrase inhibitory property, and these compounds contains diketo functionality, so these compounds were screened in assay for HIV-1 integrase inhibition. Abstract

Spring maize area has emerged as a niche market in South Asia. Production of maize during this post-rainy season is often challenged due to heat stress. Therefore, incorporating heat stress resilience is an important trait for incorporation in maize hybrids selected for deployment in this season. However, due to the significant genotype × environment interaction (GEI) effects under heat stress, the major challenge lies in identifying maize genotypes with improved stable performance across locations and years. In the present study, we attempted to identify the key weather variables responsible for significant GEI effects, and identify maize hybrids with stable performance under heat stress across locations/years. The study details the evaluation of a set of prereleased advanced maize hybrids across heat stress vulnerable locations in South Asia during the spring seasons of 2015, 2016 and 2017. Using factorial regression, we identified that relative humidity (RH) and vapor pressure deficit (VPD) as the two most important environmental covariates contributing to the large GEI observed on grain yield under heat stress. The study also identified reproductive stage, starting from tassel emergence to early grain-filling stage, as the most critical crop stage highly susceptible to heat stress. Across-site/year evaluation resulted in identification of six high yielding heat stress resilient hybrids.

Abstract: Careful reaction of equimolar amounts of 1, 3-dichloroacetone and the amidinothioureas (4), (5), (6) or (10) has given the (2-amino-5-thiazolyl) chloromethyl ketones (7), (8), (9) and (11) respectively in 20 to 40% yields. The presence of excess amidinothiourea in the reaction mixture leads to the bis thiazolyl ketones (3).
