Abstract

Advancements in wearable bioanalytical microsystems have enabled diurnal and (semi)continuous monitoring of physiologically-relevant indices that are accessible through probing sweat. To deliver an undistorted and physiologically-meaningful interpretation of these readings, tracking the sweat secretion rate is essential, because it allows for calibrating the biomarker readings against variations in sweat secretion and inferring the bodys hydration/electrolyte homeostasis status. To realize an autonomous wearable solution with intrinsically high signal-to-noise ratio sweat rate sensing capabilities, here, we devise a digitized microbubble detection mechanism-delivered by a hybrid microfluidic/electronic system with a compact footprint. This mechanism is based on the intermittent generation of microliter-scale bubbles via electrolysis and the instantaneous measurement of their time-of-flight (and thus, velocity) via impedimetric sensing. In this way, we overcome the limitations of previously proposed sweat rate sensing modalities that are inherently susceptible to non-targeted secretion characteristics (pH, conductivity, and temperature), constrained by volume, or lack system integration for autonomous on-body operation. By deploying our solution in human subject trials, we validate the utility of our solution for seamless monitoring of exercise- and iontophoretically-induced sweat secretion profiles.

Abstract

Background Obesity is an established risk factor for hypertension. Although obesity-induced gut barrier breach leads to the leakage of various microbiota-derived products into host circulation and distal organs, the roles of microbiota in mediating the development of obesity-associated adrenomedullary disorders and hypertension have not been elucidated. We seek to explore the impacts of microbial DNA enrichment on inducing obesity-related adrenomedullary abnormalities and hypertension. Methods and Results Obesity was accompanied by remarkable bacterial DNA accumulation and elevated inflammation in the adrenal glands. Gut microbial DNA containing extracellular vesicles (mEVs) were readily leaked into the bloodstream and infiltrated into the adrenal glands in obese mice, causing microbial DNA enrichment. In lean wild-type mice, adrenal macrophages expressed CRIg (complement receptor of the immunoglobulin superfamily) that efficiently blocks the infiltration of gut mEVs. In contrast, the adrenal CRIg+ cell population was greatly decreased in obese mice. In lean CRIg-/- or C3-/- (complement component 3) mice intravenously injected with gut mEVs, adrenal microbial DNA accumulation elevated adrenal inflammation and norepinephrine secretion, concomitant with hypertension. In addition, microbial DNA promoted inflammatory responses and norepinephrine production in rat pheochromocytoma PC12 cells treated with gut mEVs. Depletion of microbial DNA cargo markedly blunted the effects of gut mEVs. We also validated that activation of cGAS (cyclic GMP-AMP synthase)/STING (cyclic GMP-AMP receptor stimulator of interferon genes) signaling is required for the ability of microbial DNA to trigger adrenomedullary dysfunctions in both in vivo and in vitro experiments. Restoring CRIg+ cells in obese mice decreased microbial DNA abundance, inflammation, and hypertension. Conclusions The leakage of gut mEVs leads to adrenal enrichment of microbial DNA that are pathogenic to induce obesity-associated adrenomedullary abnormalities and hypertension. Recovering the CRIg+ macrophage population attenuates obesity-induced adrenomedullary disorders.

Abstract

The contribution of altered mitochondrial Ca2+ handling to metabolic and functional defects in type 2 diabetic (T2D) mouse hearts is not well understood. In this study, we show that the T2D heart is metabolically inflexible and almost exclusively dependent on mitochondrial fatty acid oxidation as a consequence of mitochondrial calcium uniporter complex (MCUC) inhibitory subunit MCUb overexpression. Using a recombinant endonuclease-deficient Cas9-based gene promoter pulldown approach coupled with mass spectrometry, we found that MCUb is upregulated in the T2D heart due to loss of glucose homeostasis regulator nuclear receptor corepressor 2 repression, and chromatin immunoprecipitation assays identified peroxisome proliferator-activated receptor α as a mediator of MCUb gene expression in T2D cardiomyocytes. Upregulation of MCUb limits mitochondrial matrix Ca2+ uptake and impairs mitochondrial energy production via glucose oxidation by depressing pyruvate dehydrogenase complex activity. Gene therapy displacement of endogenous MCUb with a dominant-negative MCUb transgene (MCUbW246R/V251E) in vivo rescued T2D cardiomyocytes from metabolic inflexibility and stimulated cardiac contractile function and adrenergic responsiveness by enhancing phospholamban phosphorylation via protein kinase A. We conclude that MCUb represents one newly discovered molecular effector at the interface of metabolism and cardiac function, and its repression improves the outcome of the chronically stressed diabetic heart.

Abstract

En el presente artículo, se realiza un análisis empírico, acerca del objeto que se estudia, el objetivo que se propone, es caracterizar el estado inicial de la alfabetización informacional en la preparación para el empleo de los recién graduados de la carrera de Ingeniería en Ciencias Informáticas, en la Universidad de las Ciencias Informáticas en Cuba, a partir de la aplicación de los métodos de investigación científicos utilizados (empíricos y estadísticos) entre estos, el análisis documental, la observación científica, la triangulación para contrastar los resultados, los instrumentos que fueron utilizados para la recolección de datos, son las encuestas y entrevistas aplicadas a tutores y graduados que laboran en la universidad como docentes, los que permitieron arrojar a los resultados alcanzados, en los cuales, se logró determinar las dimensiones e indicadores del objeto de estudio que se analiza, operacionalizando la variable dependiente de la investigación (la alfabetización informacional en la preparación para el empleo) y su correspondiente parametrización. Esto posibilitó medir cada uno de los indicadores, en los instrumentos aplicados, corroborando la situación que presentan los recién graduados de la universidad, en relación a la alfabetización informacional. Proceso que desarrolla competencias informacionales en cualquier profesional, el cual posibilita, una eficiente gestión de la información y del conocimiento científico.

Abstract

Technology has supposed a profound paradigm shift in human evolution, following Haraway’s cyborg metaphor we have forged a profound psycho-social rapport machines. This connectivity has also brought changes in crime patterns and fostered the development of cybercrime. From a criminological perspective, this work aims to explore the role of Per-Olof Wikströms Situational Action Theory in explaining cybercrime by including Syke and Matza’s neutralisation techniques in its formulation. The SAT-RI (SAT- Revised for the Internet) takes into consideration the interaction between cyber-crime propensity (based essentially in moral perceptions), the internet, neutralisation techniques (cognitive scripts used as protection against blame) and self-control. The theory was tested by using a mixed methods design that includes an online survey (N=709) and case studies (N=20) stemming from interviews with law enforcement agents. Once the data was analysed, it was demonstrated that individuals with low self-control tend to have higher cybercrime propensity and are more prone to justify their acts by using adequate neutralisations. In addition, there are differences in the perceptions of cybercriminals by law enforcement agents depending on whether they are fraudsters, child sex abusers, sex abusers or hackers. The resulting theory can be useful in terms of prevention, as it can help design programmes that focus on the different stages of the cybercrime process (self-control, propensity or neutralisation). Also, the thesis calls for a more anthropological conception of cybercriminology called cyborg criminology.
