Abstract

The association of varicoceles with infertility is well established, but the exact effect of varicoceles on semen quality among patients with infertility is still poorly known. The study aimed to examine the prevalence of varicoceles among Chinese men with infertility and to examine the factors associated with semen quality. This was a cross-sectional study of 5447 male patients treated for infertility at the Affiliated Hospital of Guangdong Medical University from October 2012 to December 2015. The patients were divided on the basis of the presence of varicoceles. Examinations of the amount of semen and sperm morphology were performed according to seminal parameter detection methods recommended by the World Health Organization. 19. Paris polyphylla 26triggers G2/M phase arrest and induces apoptosis in HepG2 cells via inhibition of the Akt signaling pathway

Abstract

Objectives Paris polyphylla 26(PP-26) is a monomer purified from Paris polyphylla, which has traditionally been used as an antimicrobial, hemostatic, and anticancer agent in China. The anti-proliferation effect and underlying molecular mechanism of PP-26 were investigated in vitro.Methods The effects of PP-26 on various tumor cells were detected by MTT assay. PP-26-affected cell cycle and cell cycle-related proteins in HepG2 cells were detected by flow cytometry and western blotting, respectively. Apoptosis in response to PP-26 was assessed by Hoechst 33258 staining and flow cytometry. PP-26-affected apoptosis-related proteins and Akt signaling were detected by western blotting. The inhibitory effect of PP-26 on HepG2 cells, when combined with 5-fluorouracil (5-FU), was also assessed.Results PP-26 inhibited proliferation of HepG2 cells in a dose-dependent manner by triggering G2/M-phase arrest. Moreover, PP-26 induced apoptosis of HepG2 cells. Expression levels of apoptosis proteins caspase 9, caspase 3, PARP, Bcl-2, Bcl-xL, and Mcl-1 were downregulated, while the expression level of apoptosis protein Bax was upregulated. Expression levels of p-Akt, p-GSK-3β, and p-Foxo3 were downregulated. Combination with PP-26 enhanced 5-FU inhibition of HepG2 cell proliferation.Conclusions PP-26 triggers G2/M-phase arrest and induces apoptosis in HepG2 cells via inhibition of the Akt signaling pathway.

Abstract

The aim of this study was to discuss the causes and preventions of sacromatoid renal cell carcinoma (SRCC) after renal allograft from donation after citizens death (DCD). It was reported on a 45-year-old male patient had undergone renal transplantation in other hospital because of chronic renal insufficiency uraemia in August of 2014. Two months later, lesions found in the renal parenchyma in the right iliac fossa via the color doppler ultrasound. The area of renal transplantation was bulged with tenderness. The renal dysfunction and microscopic haematuria were found. The review color doppler ultrasound and CT showed that the allograft kidney is enlarged rapidly. On 22nd December 2014, a surgical exploration was performed. An irregular and hard mass within the parenchyma of engrafted kidney was found, which was considered as malignant. The nephrectomy of the renal allograft was performed in the operation. First postoperative day, his condition was deteriorating rapidly. Finally, the patient died of cardiac functional failure. The tumor was pathologically diagnosed as sacromatoid renal cell carcinoma (SRCC). SRCC found in the donor allograft kidney is highly malignant and poorly prognosis. The repeated imaging investigations are necessary to exclude the hidden trouble of tumors, such as SRCC. Postoperative immunosuppressive regimens should be modified and the screening program of renal tumor for the early control and prevention should be strengthen.

Abstract

Purpose: To explore the regulatory roles of microRNA-140 and SOX4 in prostate cancer (PCa) tissues and paracancerous tissues, and their underlying mechanism., Methods: MicroRNA-140 expressions in PCa tissues, paracancerous tissues and PCa cell lines were detected by quantitative real-time polymerase chain reaction (qRT-PCR). Proliferation, apoptosis and cell cycle of PCa cells after altering expressions of microRNA-140 and SOX4 were detected by MTT assay and flow cytometry, respectively. The regulatory effect of microRNA-140 on SOX4 was detected by Western blot and qRT-PCR. The binding condition of microRNA-140 on SOX4 was verified by luciferase reporter gene assay., Results: MicroRNA-140 was downregulated in PCa tissues compared to paracancerous tissues. In particular, lower expression of microRNA-140 was found in PCa with Grade I+II compared to Grade III+IV. In vitro, microRNA-140 expression was negatively correlated with proliferative and invasive abilities, while positively correlated with apoptosis of PCa cells. MicroRNA-140 promoted cell cycle arrest in G0/G1 phase. SOX4 expression was inhibited by microRNA-140 overexpression in PCa cells., Conclusions: Downregulated microRNA-140 promotes proliferation and cell cycle arrest, but inhibits apoptosis of PCa cells. MicroRNA-140 inhibits PCa development via degrading SOX4.

Abstract

Objective: To investigate the expressions of Engrailed-2 (EN2) and β-catenin in bladder urothelial carcinoma and explore their significance., Methods: Sixty bladder urothelial carcinoma samples of different grades and stages and 10 normal bladder mucosal tissues were examined for expressions of EN2 and β-catenin proteins and mRNA using immunochemistry, Western blotting and RT-PCR. RESULTS Compared to normal bladder mucosa, bladder urothelial carcinoma tissues showed significantly increased expressions of EN2 and β-catenin proteins (P<0.05), and the high-grade carcinoma tissues exhibited significantly stronger expressions than the low-grade ones (P<0.05); the expressions of the proteins increased also significantly with advanced pathological stages of bladder urothelial carcinoma (P<0.05). The expressions of EN2 and β-catenin mRNAs showed a consistent pattern of changes with their protein expressions., Conclusion: The expressions of EN2 and β-catenin are significantly increased in bladder urothelial carcinoma. EN2 may contribute to the development and progression of bladder urothelial carcinoma by activating Wnt/β-catenin signal pathway.

30. [Effects of cryopreservation on expression of connective tissue growth factor in rat kidney].

Abstract

Objective: To investigate the effect of cryopreservation (CP) on the expression of connective tissue growth factor (CTGF) in the renal tubular epithelial cells., Methods: A total of 40 male Wistar rats (weighing 230-250 g) were used in this study. En bloc removal with in situ cooling both kidneys and hypertonic citrate adenine preservation solution were adopted. The rat kidney was be preserved 0, 12, 24, 36 and 48 hours at 0-4 degrees C (n=8), respectively. The expression of CTGF of renal tubular epithelial cells was detected by using immunohistochemistry and in situ hybridization analysis., Results: The expression of CTGF was less in CP 0 hour group and CP 12 hours group, the positive unit (PU) values of CTGF protein were 5.91 +/- 2.30 and 5.57 +/- 2.40 (P > 0.05), respectively, and the PU values of CTGF mRNA were 6.24 +/- 2.79 and 6.51 +/- 2.43 (P > 0.05), respectively. The PU values of CTGF protein increased at CP 24 hours group (10.25 +/- 2.92), CP 36 hours group (14.31 +/- 2.83) and CP 48 hours group (18.11 +/- 3.94, P < 0.05), respectively, and the PU values of CTGF mRNA increased at CP 24 hours group (15.24 +/- 3.95), CP 36 hours group (19.20 +/- 4.73) and CP 48 hours group (23.09 +/- 4.40, P < 0.05), respectively; showing significant differences when compared with CP 0 hour group and CP 12 hours group (P < 0.05)., Conclusion: CTGF expression may increase with severe cold ischemia injury, and might play an important role in regeneration and repair of renal tubular epithelial cell injury.
