Elektroeğirme; nano/mikro skaladaki fiberlerin üretilmesinde kullanılan, tek adımlı, uygun maliyetli ve çok yönlü bir tekniktir. Bu teknikle üretilen fiberler, yüksek yüzey alanı/ hacim oranı, fleksibilite, yüksek gözeneklilik dâhil olmak üzere birçok eşsiz fonksiyonlara sahip olmaları sayesinde biyomedikal uygulamalar başta olmak üzere birçok alanda ilgi görmüştür. Elektroeğrilmiş fiberler hücrelerin tutunması, gelişmesi ve çoğalmasını destekleyici gözenekli yapısıyla ekstra selüler matrisi (ECM) başarılı bir şekilde taklit edebilmektedir. Elektroeğirme tekniğinin sağladığı önemli bir avantaj da geniş yelpazedeki doğal ve sentetik polimerlerin ilaçlar ve/veya biyoaktif maddelerle elektroeğrilebilmesidir. Bu çalışmada yanıklar ve diyabete bağlı ülserler gibi yara iyileşme profili kötü seyreden yaralarda kullanılmak üzere yara iyileşmesini hızlandırıcı biyoaktif maddeler içeren nanofiber yara örtüleri üretilmiştir. Biyoaktif maddeler olarak keratin ve hyaluronik asit hayvansal kaynaklardan ekstrakte edilmiş ve karakterizasyon çalışmaları gerçekleştirilmiştir. Üretilen keratin ve hyaluronik asit eş eksenli polikaprolakton ve polietilen oksit polimerlerinin çekirdek yapısına dahil edilerek iki farklı teknik yardımıyla (emülsiyon ve eş eksenli elektroeğirme) elektroeğrilmiştir. Keratin ve hyaluronik asidin bu yapılara ayrı ayrı ve birlikte katılarak üretildiği tüm fiberlerin morfolojileri, kimyasal bağ yapıları, termal davranışları ve mekanik dayanımları incelenmiştir. İçeriklerin ve iki farklı tekniğin fiber yapılar üzerindeki etkileri ayrıntılı karakterizasyon testleri yapılarak araştırılmıştır.--------------------Electrospinning is a one-step, cost-effective and versatile technique that is used at the production of nanofibers. The nanofibers produced with this technique have high surface area / volume ratio, flexibility, and high porosity. These features make nanofibers favorable at many fields, especially for biomedical applications. Electrospun nanofibers can successfully mimic extracellular matrix (ECM) thanks to their porous structure that supports cell adhesion, development, and proliferation. Another major advantage of the electrospinning technique is that it enables the electrospinning of a wide range of natural and synthetic polymers with drugs and bioactive components. In the present study, a nanofiber dressing was produced which contains curative bioactive substances that accelerate the healing of wounds and is likely to be beneficial in wounds such as burns and diabetes-related ulcers. Keratin and hyaluronic acid as bioactive substances were produced by extracting from animal sources and characterized. The produced keratin and hyaluronic acid were electrospun with the help of two different techniques by incorporating into the core structure of coaxial polycaprolactone and polyethylene oxide polymers. The morphology, chemical bonding structures, thermal behaviour and mechanical strength of all nanofibers which were produced by adding keratin and hyaluronic acid to these structures separately and together, were investigated. The effects of the substances and the two techniques were compared by detailed characterization tests.
