Neurodegenerative diseases are characterized by progressive cell loss leading to disruption of the structure and function of the central nervous system. Amyotrophic lateral sclerosis (ALS) was among the first of these disorders modeled in patient-specific iPSCs, and recent findings have translated into some of the earliest iPSC-inspired clinical trials. Focusing on ALS as an example, we evaluate the status of modeling neurodegenerative diseases using iPSCs, including methods for deriving and using disease-relevant neuronal and glial lineages. We further highlight the remaining challenges in exploiting the full potential of iPSC technology for understanding and potentially treating neurodegenerative diseases such as ALS.

Item does not contain fulltext, Tumour budding in colorectal cancer, defined as single tumour cells or small clusters containing four or fewer tumour cells, is a robust and independent biomarker of aggressive tumour biology. On the basis of published data in the literature, the evidence is certainly in favour of reporting tumour budding in routine practice. One important aspect of implementing tumour budding has been to establish a standardised and evidence-based scoring method, as was recommended by the International Tumour Budding Consensus Conference (ITBCC) in 2016. Further developments have aimed at establishing methods for automated tumour budding assessment. A digital approach to scoring tumour buds has great potential to assist in performing an objective budding count but, like the manual consensus method, must be validated and standardised. The aim of the present review is to present general considerations behind the ITBCC scoring method, and a broad overview of the current situation and challenges regarding automated tumour budding detection methods.

Contains fulltext : 244608.pdf (Publisher’s version ) (Closed access), Aberrant inflammation in the CNS has been implicated as a major player in the pathogenesis of human neurodegenerative disease. We developed a new approach to derive microglia from human pluripotent stem cells (hPSCs) and built a defined hPSC-derived tri-culture system containing pure populations of hPSC-derived microglia, astrocytes, and neurons to dissect cellular cross-talk along the neuroinflammatory axis in vitro. We used the tri-culture system to model neuroinflammation in Alzheimer's disease with hPSCs harboring the APP(SWE)+/+ mutation and their isogenic control. We found that complement C3, a protein that is increased under inflammatory conditions and implicated in synaptic loss, is potentiated in tri-culture and further enhanced in APP(SWE)+/+ tri-cultures due to microglia initiating reciprocal signaling with astrocytes to produce excess C3. Our study defines the major cellular players contributing to increased C3 in Alzheimer's disease and presents a broadly applicable platform to study neuroinflammation in human disease.

- Abstract
The International Society for Stem Cell Research (ISSCR) presents its 2016 Guidelines for Stem Cell Research and Clinical Translation (ISSCR, 2016). The 2016 guidelines reflect the revision and extension of two past sets of guidelines (ISSCR, 2006; ISSCR, 2008) to address new and emerging areas of stem cell discovery and application and evolving ethical, social, and policy challenges. These guidelines provide an integrated set of principles and best practices to drive progress in basic, translational, and clinical research. The guidelines demand rigor, oversight, and transparency in all aspects of practice, providing confidence to practitioners and public alike that stem cell science can proceed efficiently and remain responsive to public and patient interests. Here, we highlight key elements and recommendations in the guidelines and summarize the recommendations and deliberations behind them.

A small prototype of gamma-ray detector suitable for both imaging and spectroscopy, based on a single CsI(Tl) scintillator coupled to a two-dimensional Silicon Drift Detector (SDD) array, has been developed. The determination of the position of interaction is performed by applying the centroid method to the signals from the single SDD units, while the measurement of the y photon energy is performed by summing together these signals. This first prototype has been developed in order to evaluate the performances of a CsI(Tl)-SDD detection scheme in view of the realization of larger scintillation cameras for nuclear medicine. The low electronics noise of the SDD allows obtaining good performances both in position and in energy resolution. A position resolution of the order of 0.6 mm and an energy resolution of about 17 % FWHM at 122 keV have been measured.

23. L’éducation pour la santé : une activité intégrée aux programmes du projet Equal-Reset

Les déterminants majeurs de la non-réinsertion et de la récidive sont la mauvaise santé mentale, une faible estime de soi, mais surtout le manque d’attitudes positives envers le travail telles que la motivation à avoir un emploi, la responsabilité à l’égard des résultats professionnels. Dans le cadre du projet Equal-reset (Réinsertion Économique et Sociale par l’Éducation et le Travail des détenus), nous avons tenté de montrer qu’il existait un lien entre santé mentale et valeur accordée au travail. Méthodes Parmi les 62 détenus présents au Centre Pénitentiaire de Givenich au moment de l’enquête, 52 ont été volontaires. Variables mesurées à l’aide d’un questionnaire : (1) la santé mentale à l’aide du General Health Questionnaire (GHQ-12 items) (Goldberg 1972), (2) la valeur attribuée au travail avec l’échelle de centralité du rôle professionnel (Meyers & Houssemand 2007) ; plus le score est élevé plus l’investissement accordé au travail dans la vie est important. Résultats Les détenus représentent une population jeune avec une surreprésentation des 25-34 ans. Une majorité d’entre eux a déjà eu une expérience professionnelle de plus de 24 mois, mais a un niveau d’éducation scolaire inférieur. Plus les détenus attribuent de l’importance au travail, moins ils ont de détresse psychique. Par rapport à un niveau d’instruction scolaire supérieur, un niveau inférieur favorise la dépression. Discussion Plus les détenus accordent de l’importance au travail, moins ils paraissent en souffrance psychologique ; les moins instruits étant les plus concernés. Développer des habitudes positives envers le travail dans le sens d'une motivation accrue pour la tâche est un moyen pour favoriser leur réadaptation et leur insertion dans la société, mais cela représente aussi des activités structurantes qui permettent aux détenus pendant le temps de la détention de garder ou de se reconstruire un équilibre mental. L’innovation du projet EQUAL-RESET a été de concevoir un dispositif comportant : • un accompagnement personnalisé assuré par un case manager du Service Psycho-Socio-Educatif selon la méthodologie de l’Empowerment Case Management ; • trois programmes d’accompagnement : Emploi & Employabilité, Enseignement & Formation, Développement Personnel (activités d’éducation pour la santé). Un parcours individualisé et un soutien psychologique sont recommandés pour aider les détenus à mieux exploiter leur potentiel dans les programmes d’accompagnement proposés.

This note describes the systematic study made of the resolution of the LHCb VELO telescope used in the Autumn 99 test-beam. Measurements were made of the resolution for each region of both the LHCb {\it R} and $\Phi$ vertex detectors. The best value obtained for a 40$\mu$m pitch detector was 5.8 $\pm$ 0.5 $\mu$m, which is comparable to the 6.0 $\mu$m prediction of the Technical Proposal. At larger pitches, it was found that the resolutions deviate from those given in the Technical Proposal e.g. for an 80 $\mu$m detector a resolution of \mbox{15.4 $\pm$ 0.3 $\mu$m} is compared to the Technical Proposal value of 10 $\mu$m.

Data of a beam test in Autumn 1999 have been analysed to determine the pulse shape of the signal as read out by an SCTA chip from a detector prototype for the LHCb Vertex Locator. The measurements have been performed at different bias voltages and distinguishing non-irradiated and irradiated regions on the detector. The results are interpreted in terms of the signal overspill that remains for the subsequent readout after 25 ns, and an optimal working point is determined, considering both the signal amplitude and the overspill fraction.

Direct photon production cross sections obtained in high statistics $\bar{p}p$ and $pp$ collisions at $\sqrt{s}=24.3$ GeV at the CERN SPS are used in a next-to-leading order QCD analysis. From the ( stat.)~\pm0.0033\,(syst.)~^{+0.0077}_{-0.0034} \,(t cross section difference $\sigma(\bar{p}p~\to~\gamma X)~-~\sigma(pp~\to~\gamma X)$ and quark distributions measured in deep inelastic scattering, a determination of the strong coupling constant, $\alpha_s$, is performed via a measurement of \ms. This measurement yields a value \ms\ = $210 \pm 22 \,(stat.) \pm 44 \,(syst.) ^{+105}_{-36} \,(theo.)$ MeV. The corresponding value of $\alpha_s$ expressed at $M^2_Z$ is $\alpha_s(M^2_Z)=0.1112~\pm0.0016\

- Abstract
Les déterminants majeurs de la non-réinsertion et de la récidive sont la mauvaise santé mentale, une faible estime de soi, mais surtout le manque d’attitudes positives envers le travail telles que la motivation à avoir un emploi, la responsabilité à l’égard des résultats professionnels. Dans le cadre du projet Equal-reset (Réinsertion Économique et Sociale par l’Éducation et le Travail des détenus), nous avons tenté de montrer qu’il existait un lien entre santé mentale et valeur accordée au travail. Méthodes Parmi les 62 détenus présents au Centre Pénitentiaire de Givenich au moment de l’enquête, 52 ont été volontaires. Variables mesurées à l’aide d’un questionnaire : (1) la santé mentale à l’aide du General Health Questionnaire (GHQ-12 items) (Goldberg 1972), (2) la valeur attribuée au travail avec l’échelle de centralité du rôle professionnel (Meyers & Houssemand 2007) ; plus le score est élevé plus l’investissement accordé au travail dans la vie est important. Résultats Les détenus représentent une population jeune avec une surreprésentation des 25-34 ans. Une majorité d’entre eux a déjà eu une expérience professionnelle de plus de 24 mois, mais a un niveau d’éducation scolaire inférieur. Plus les détenus attribuent de l’importance au travail, moins ils ont de détresse psychique. Par rapport à un niveau d’instruction scolaire supérieur, un niveau inférieur favorise la dépression. Discussion Plus les détenus accordent de l’importance au travail, moins ils paraissent en souffrance psychologique ; les moins instruits étant les plus concernés. Développer des habitudes positives envers le travail dans le sens d'une motivation accrue pour la tâche est un moyen pour favoriser leur réadaptation et leur insertion dans la société, mais cela représente aussi des activités structurantes qui permettent aux détenus pendant le temps de la détention de garder ou de se reconstruire un équilibre mental. - Abstract
A skeletal muscle is composed of motor units, each consisting of a motoneuron and the muscle fibers it innervates. The input to the motor units is formed of electrical signals coming from higher motor centers and propagated to the motoneurons along a network of nerve fibers. Because of its complexity, this network still escapes actual direct observations. The present model describes the steady state activation of a muscle, i.e., of its motor units. It incorporates the network as an unknown quantity and, given the latter, predicts the input-force relation (activation curve) of the muscle. Conversely, given a suitable activation curve, our model enables the recovery of the network. This step is performed by using experimental data about the activation curve, and the whole activation process of a muscle can then be theoretically investigated. In this way, this approach provides a link between the macroscopic (activation curve) and microscopic (network) levels. From a mathematical viewpoint, solving the preceding inverse problem is equivalent to solving an integral equation of a new type.

- Abstract
The correct identity and functional capacity of transplanted dopamine (DA) neurons derived in vitro from embryonic stem (ES) cells is a critical factor for the development of an ES cell-based replacement therapy for Parkinson's disease. We transplanted primate Cyno-1 ES cells differentiated in vitro for 4 (progenitor ES cells) or 6 (differentiated ES cells) weeks, or control fetal primate cells into the striatum of hemi-parkinsonian rats. Partial behavioral recovery in amphetamine-induced rotation was correlated with the number of ES-derived tyrosine hydroxylase-positive (TH +) neurons in the grafts (r = 0.5, P < 0.05). Post mortem analysis of ES-derived grafts revealed TH + neurons with mature morphology, similar to fetal DA neurons, and expression of midbrain transcription factors, such as Engrailed (En) and Nurr-1. While the total number of TH + neurons was not different between the two groups, TH/En co-expression was significantly higher (> 90%) in grafts from differentiated ES cells than in grafts derived from progenitor cells (< 50%), reflecting a more heterogeneous cellular composition. Within the grafts there was an overlap between ES-derived TH + axonal arbors and clusters of primate ES-derived striatal neurons expressing brain factor 1 (Bf-1, Foxg1) and DA and cAMP-regulated phosphoprotein (DARPP-32). Such overlap was never observed for other regional transcription factors that define neighboring forebrain domains in the developing brain, such as Nkx2.1 (medial ganglionic eminence), Nkx2.2 (pallidal and diencephalic progenitors) or Pax6 (dorsal telencephalic progenitors). Despite the heterogeneity of ES-derived graft cell composition, these results demonstrate normal phenotypic specification, conserved natural axonal target selectivity and functionality of DA neurons derived from primate ES cells. © The Authors (2006).

- Abstract
Dopamine (DA) neurons can be derived from human and primate embryonic stem (ES) cells in vitro. An ES cell-based replacement therapy for patients with Parkinson's disease requires that in vitro-generated neurons maintain their phenotype in vivo. Other critical issues relate to their proliferative capacity and risk of tumor formation, and the capability of migration and integration in the adult mammalian brain. Neural induction was achieved by coculture of primate parthenogenetic ES cells (Cyno-1) with stromal cells, followed by sequential exposure to midbrain patterning and differentiation factors to favor DA phenotypic specification. Differentiated ES cells were treated with mitomycin C and transplanted into adult immunosuppressed rodents and into a primate (allograft) without immunosuppression. A small percentageof DA neurons survived in both rodent and primate hosts for the entire term of the study (4 and 7 months, respectively). Other neuronal and glial populations derived from Cyno-1 ES cells showed, in vivo, phenotypic characteristics and growth and migration patterns similar to fetal primate transplants, and a majority of cells (>80%) expressed the forebrain transcription factor brain factor 1. No teratoma formation was observed. In this study, we demonstrate long-term survival of DA neurons obtained in vitro from primate ES cells. Optimization of differentiation, cell selection, and cell transfer is required for functional studies of ES-derived DA neurons for future therapeutic applications. ©AlphaMed Press.
