The bioavailability of small-molecule drugs remains a critical challenge in pharmaceutical development, significantly impacting therapeutic efficacy and commercial viability. This review synthesizes recent advances in understanding and overcoming bioavailability limitations, focusing on key physicochemical and biological factors influencing drug absorption and distribution. We examine cutting-edge strategies for enhancing bioavailability, including innovative formulation approaches, rational structural modifications, and the application of artificial intelligence in drug design. The integration of nanotechnology, 3D printing, and stimuli-responsive delivery systems are highlighted as promising avenues for improving drug delivery. We discuss the importance of a holistic, multidisciplinary approach to bioavailability optimization, emphasizing early-stage consideration of ADME properties and the need for patient-centric design. This review also explores emerging technologies such as CRISPR-Cas9-mediated personalization and microbiome modulation for tailored bioavailability enhancement. Finally, we outline future research directions, including advanced predictive modeling, overcoming biological barriers, and addressing the challenges of emerging therapeutic modalities. By elucidating the complex interplay of factors affecting bioavailability, this review aims to guide future efforts in developing more effective and accessible small-molecule therapeutics. Objective: Eugenol (EG) is a significant ingredient in clove oil that is commonly used as a tooth pain reliever. However, topical application of this phenol has been linked to a number of serious negative consequences. Methods: The goal of this study was to position EG into the coumarin nucleus, yielding a precursor and ten variants labeled (EGCOU1–EGCOU10). To validate their molecular structures, the FT-IR, 1H, 13C NMR, and mass spectra were analyzed. The microbiological study was conducted against many pathogens using a growth medium-dilution approach. These include four aerobic bacteria, four anaerobic bacteria, and two fungi. On the other hand, the biosafety study explored the effect of the experimental coumarins on two non-pathogenic aerobic bacteria, three normal cellular populations, and lab-treated human blood. Results and Discussion: The results of the former study indicated that the experimental coumarins demonstrated outstanding antimicrobial activity compared with the standards used, with minor variances. The most efficacy was discovered against the test fungi, followed by aerobic bacteria, and lastly anaerobic ones, with (EGCOU7) having a preferential impact. The biosafety investigation found that the experimental coumarins were extremely biocompatible with the test nonpathogenic bacteria, normal cells, and human blood erythrocytes. Conclusions: Given the above findings, the author concluded that EG transferred into biosafe wide-spectrum antimicrobial agents through its incorporation within coumarin backbones. Among them, the ones with three substituted groups on the off-side aromatic ring showed higher activity than those with one or two groups. Furthermore, among the prior compounds, the one containing di-meta chlorides and mono-para methoxy exhibited the best benefits. Sophoridine is a quinolizidine alkaloid isolated from Sophora flavescens and Sophora alopecuroides. Its medical activities include anticancer, anti-inflammation, etc., but little pesticide research. To improve the agricultural value of sophoridine, a series of 15-chloro-18-alkyl (benzyl) oxyiminomethyne derivatives were designed and synthesized through structural modification at C-14 and C-15 positions. Compound 4i (R=4-NO2Bn) showed good acaricidal activity against female adults of Tetranychus cinnabarinus. Its 72 h LC50 value was 0.408 mg/mL, and its acaricidal activity was 8.5 times that of sophoridine (LC50=3.487 mg/mL). At the concentration of 1 mg/mL, compound 4f (R=4-BrBn) had the best insecticidal activity against Mythimna separata, with a 35 d corrected mortality rate of 62.3%. BACKGROUND: The threats to the safety of humans and the environment and the resistance of agricultural chemicals to plant pathogenic fungi and bacteria highlight an urgent need to find safe and efficient alternatives to chemical fungicides and bactericides. In this study, a series of Berberine (BBR) derivatives were designed, synthesized and evaluated for in vitro and in vivo antimicrobial activity against plant pathogenic fungi and bacteria. RESULTS: Bioassay results indicated that compounds A11, A14, A20, A21, A22, A25, A26, E1, E2, E3, Z1 and Z2 showed high inhibitory activity against Sclerotinia sclerotiorum and Botrytis cinerea. Especially, A25 showed a broad spectrum and the highest antifungal activity among these compounds. Its EC50 value against Botrytis cinerea was 1.34 μg mL−1. Compound E6 possessed high inhibitory activity against Xanthomonas oryzae and Xanthomonas Campestris, with MIC90 values of 3.12 μg mL−1 and 1.56 μg mL−1. A Topomer CoMFA model was generated for 3D‐QSAR studies based on anti‐B. cinerea effects, with high predictive accuracy, showed that the addition of an appropriate substituent group at the para‐position of benzyl of BBR derivatives could effectively improve the anti‐B. cinerea activity. In addition, compound A25 could significantly inhibit the spore germination of Botrytis cinerea at low concentration, and compound F4 exhibited remarkable curative and protective efficiencies on rice bacterial leaf blight. The electrochemical performance of double perovskite oxides as cathode material for intermediate-temperature solid oxide fuel cells can be enhanced through structural modifications. The electrochemical performance of Sr2CoNbO6-δ-Sm0.2Ce0.8O2-δ (SCNO-SDC) composite structurally assembled using various techniques, one-pot, electrospinning, and mixing have been studied to understand the structural change and performance relationship. The nanofiber composite exhibited superior cell performance of 0.996 W/cm2 peak power density and 0.079 Ω cm2 polarisation resistance compared to one-pot and the mixed composite, which yielded 0.831 W/cm2 and 0.0908 Ω cm2, and 0.706 W/cm2 and 0.206 Ω cm2, respectively, at a temperature of 700°C. The work emphasises a successful approach to modifying the structure of the SCNO-SDC cathode by identifying the rate-determining process to produce superior electrode materials. The improved electrochemical performance of the structurally modified cathode composite is a result of its cohesive interfacial interaction, which significantly improves the kinetics of the electrode reaction and stability. Iridoid glycosides are a class of chemical structures with various pharmacological activities and have excellent research potential. However, the poor stability, water solubility, and oral bioavailability limit their practical application and clinical research. In this review, we systematically summarize the structural modifications of iridoid glycosides and attempt to demonstrate the structure-activity relationship between chemical modifications on iridoid skeleton and glycosidic bond, noting that some derivatives exhibit satisfactory pharmacological activities. This review aims to provide valuable assistance for further research and clinical application of derivatives, to provide ideas for the design and synthesis of novel iridoid glycosides, and to provide a research basis for developing new drugs with higher activity in the future. Deep eutectic-like solvents (DESs) are ideal green solvents for achieving depolymerization of kraft lignin under mild conditions. In this study, the impact of changes in chemical structures in hardwood kraft lignin (HKL), softwood kraft lignin (SKL), hydroxypropyl-modified hardwood kraft lignin (PHKL), and acetylated hardwood kraft lignin (AHKL) on depolymerization was investigated. The results indicated that structural modification can effectively adjust the depolymerization reaction of lignin. Under the depolymerization conditions (150 °C, 3 h, CHCl:p-TSOH), the depolymerization extent of HKL was 44.8%, which was 9.4% higher than that of SKL (35.4%). It was found that after hydroxypropyl modification of phenolic hydroxyl in HKL, the depolymerization extent of PHKL decreased to 29.5%, confirming that phenolic hydroxyl was the key functional group promoting lignin depolymerization. Acetylation modification of HKL can effectively increase the lignin depolymerization extent from 44.8% to 54.2%. The experimental results of lignin model compound addition confirmed that acetylation modification can effectively inhibit condensation reactions. The GPC analysis showed that the average molecular weight (Mw) of depolymerized oil was less than 800 g/mol and had a narrow polydispersity index (Mw/Mn < 1.2), making it an important precursor for high-value applications such as the preparation of aviation fuel.

In recent years, exopoly saccharides (EPS) produced by microorganisms have garnered attention due to their functional properties such as improving food quality, regulating intestinal microbiota, antioxidative effects, anticancer properties, immunoprevention, and treatment of cardiovascular diseases. Currently, research on microbial EPS (extracellular polymeric substances) in China is still in its early stages and requires further exploration. This paper comprehensively reviewed the latest advancements in the classification, biosynthesis, structural modification, functional properties, and applications of EPS produced by microorganisms both domestically and internationally. This study also focused on analyzing the biosynthetic pathways and structural features of different types of EPS, as well as the strategies for functional modification through physical, chemical, and biological methods. The aim is to provide new references and guidance for research and development in relevant fields, thereby promoting the application and industrialization of EPS in the food, pharmaceutical, and industrial sectors.

Dermatan sulfate and chondroitin sulfate are dietary supplements that can be utilized as prophylactics against thrombus formation. Low-molecular-weight dermatan sulfate (LMWDS) is particularly advantageous due to its high absorbability. The enzymatic synthesis of low-molecular-weight dermatan sulfates (LMWDSs) using chondroitin B lyase is a sustainable and environmentally friendly approach to manufacturing. However, the industrial application of chondroitin B lyases is severely hampered by their low catalytic activity. To improve the activity, a semi-rational design strategy of engineering the substrate-binding domain of chondroitin B lyase was performed based on the structure. The binding domain was subjected to screening of critical residues for modification using multiple sequence alignments and molecular docking. A total of thirteen single-point mutants were constructed and analyzed to assess their catalytic characteristics. Out of these, S90T, N103C, H134Y, and R159K exhibited noteworthy enhancements in activity. This study also examined combinatorial mutagenesis and found that the mutant H134Y/R159K exhibited a substantially enhanced catalytic activity of 1266.74 U/mg, which was 3.21-fold that of the wild-type one. Molecular docking revealed that the enhanced activity of the mutant could be attributed to the formation of new hydrogen bonds and hydrophobic interactions with the substrate as well as neighbor residues. The highly active mutant would benefit the utilization of chondroitin B lyase in pharmaceuticals and functional foods.

The extraction and purification methods of polysaccharides from edible fungi determine their structure, which in turn affects their biological activity, and is the basis of research on edible fungi polysaccharides. Some specific structural features make the biological activity of edible mushroom polysaccharides significantly enhanced. Therefore, researchers have used physical, biological and chemical methods to modify the molecular structure of edible mushroom polysaccharides to improve their biological activity. The review introduced the extraction, purification and structural modification methods of edible fungi polysaccharides. The advantages and disadvantages of extraction, purification of polysaccharides from edible fungi, and the effects of chemical modification on their structure and biological activity were summarized. The importance of appropriate extraction and purification methods for obtaining the target polysaccharides and the importance of chemical modification methods for changing the molecular structure and improving the functional properties and biological activities of polysaccharides from edible fungi were prospected.

BACKGROUND RESULTS CONCLUSION Sesame paste faces issues with poor colloidal stability during storage, thereby affecting product quality and consumer experience. This study aimed to modify the proteins in sesame paste through stone milling and investigated the differences in stability produced in this environment, with the goal of addressing this issue.As the number of grinding times increased from one to three, the median diameter of sesame paste significantly decreased from 85 to 74 μm (P < 0.05), and the centrifugal oil separation rate dropped from 9.05% to 6.82%. Rheological measurements indicated an increase in the flow behavior index (n) from 0.51 to 0.61. Confocal laser scanning microscopy results revealed a more uniform co‐distribution of protein and oil when ground thrice. The β‐sheet content of the protein in sesame paste increased from 52.92% to 56.34%, with enhancements in surface hydrophobicity, hydrophobic interactions and emulsification of protein. When the number of grinding times increased to five, the particle size of the sesame paste was further reduced and the β‐sheet content of the protein decreased to 51.00%, while the oil separation rate increased to 7.78%.Stone milling induces structural modifications in proteins, which in turn alter the internal structure of sesame paste, resulting in varying levels of oil separation at different grinding times. Among them, sesame paste ground thrice showed a 25% reduction in the oil separation rate and experienced minimal oil separation over 120 days, making it suitable for practical production. © 2024 Society of Chemical Industry. Inhibition of SIRT3 triggered differentiation of multiple myeloma (MM) cells. In discovery of potent SIRT3 inhibitors for cancer differentiation therapy, structural modification was performed on the previously developed lead compound S27. A total of 49 compounds divided into two series were designed and synthesized. In the enzyme inhibitory assay, several molecules (A7, A13, B15, and B26) exhibited potent SIRT3 inhibitory activity and selectivity. Significantly, representative compounds, especially A7, promoted differentiation of MM cells from cancer phenotype to normal cells, accompanied by increased expression of antigen CD49e, human immunoglobulin light chain λ‐IgLG and κ‐IgLG. Additionally, molecule A7 reversed growth factor IL‐6 induced MM cell proliferation, improved the antiproliferative activity of Ixazomib and increased the apoptotic rate of MM cells treated with Ixazomib. Lupeol is a natural pentacyclic triterpenoid with a wide range of biological activities. To enhance the antitumor efficacy of lupeol, eight lupeol-3-succinate derivatives and seven lupeol-3-maleate derivatives were designed and synthesized. The new compounds were evaluated for their cytotoxic activities against human lung cancer A549 cell line, human liver cancer HepG2 cell line, and human breast cancer MCF-7 cell line. All compounds were found to show better cytotoxic activity in vitro than lupeol. Among them, lup-20(29)-en-3β-yl 4-oxo-4-(pyrrolidin-1-yl)butanoate displayed remarkable cytotoxic activity against lung cancer A549 cells, with an IC50 of 2.33 μM, and potently induced apoptosis in lung cancer A549 cells. The mechanism of action of lup-20(29)-en-3β-yl 4-oxo-4-(pyrrolidin-1-yl)butanoate can be subjected to further evaluation. A series of novel betulin-28-isatin hydrazone derivatives was prepared, which were evaluated for their anticancer activity. Among them, 3β-hydroxy-lup-20(29)-ene-28-[(5-bromo-1H-indole-2,3-dione)-3-yl]hydrazone exhibited the most potent anticancer activity against A549 cells, with an IC50 value of 6.95 µM, being 3.85-fold more potent than its parental compound betulin. Meanwhile, it also exhibited a negligible anti-proliferative activity against the human normal GES-1 cells, indicating its significant selectivity in inhibiting the proliferation of cancer cells than normal cells. The further virtual screening and mechanism studies suggested that this compound exerts its anticancer activity through the inhibition of PI3K-Akt signaling pathway. BACKGROUND: The natural product paeonol is a rich and sustainable natural bioresource, and its derivatives have various unique biological efficacy. As is well known, Schiff bases are a class of organic compounds with a wide range of biological activities, including anti‐fungal, insecticidal, anti‐viral, and nematicidal. RESULTS: To discover biorational natural product‐based pesticides, nine intermediates (2–10), 12 sulfonylhydrazones (11a–11c, 12a–12c, 13a–13c, and 14a–14c) and 20 benzylidene hydrazones (18a–18r, 19a, and 20a) were synthesized by structural modification of paeonol, and their structures were characterized by proton nuclear magnetic resonance (1H NMR), carbon‐13 (13C) NMR, and high‐resolution mass spectrometry (HRMS). The stereochemical configurations of compounds 14a, 18d, and 18r were unambiguously confirmed by single‐crystal X‐ray diffraction. Furthermore, bioactivities of these compounds as anti‐oomycete, anti‐fungal, and nematicidal agents against three serious agricultural pests, Phytophthora capsici, Fusarium graminearum, and Heterodera glycines were evaluated. Among all tested compounds: (i) compound 7 exhibited promising anti‐oomycete against Phytophthora capsici, with a half maximal effective concentration (EC50) value of 15.81 mg L−1; (ii) compounds 2, 7, 10, and 19a displayed promising anti‐fungal against F. graminearum, with EC50 values of 12.22, 14.72, 23.39, and 33.10 mg L−1, respectively; (iii) ten compounds (12a–12c, 14c, 18g–18j, 18m, and 19a) showed significant nematicidal activity against H. glycines, with median lethal concentration (LC50) values all less than 30.00 mg L−1. Especially for compound 18g, its LC50 value is the smallest, at 12.65 mg L−1. CONCLUSION: The research results indicate that introducing nitro groups at the C5 position of paeonol, or introducing halogens at both C5 and C3 positions, can significantly enhance its biological activity against Phytophthora capsici, F. graminearum, and H. glycines. © 2024 Society of Chemical Industry. A series of novel lupeol-3-urea/thiourea derivatives were designed, synthesized and evaluated for their in vitro anticancer activity against human lung cancer A549, human hepatocellular carcinoma HepG2 and human breast cancer MCF-7 cell lines. The antitumor activities of all compounds were higher than that of parent Lupeol. Among them, lup-20(29)-en-3β-yl-piperazinecarboxylate-(2,4-fluorophenyl)urea showed the strongest antitumor activity against A549 cell lines, with an IC50 of 3.22 μM, which is tenfold than that of Lupeol. Therefore, lup-20(29)-en-3β-yl-piperazinecarboxylate-(2,4-fluorophenyl)urea can be used as a new lead compound for the development of more potent antitumor drugs. [ABSTRACT FROM AUTHOR]

The research on boron/nitrogen (B/N)-based multiresonance thermally activated delayed fluorescence (MR-TADF) emitters has been a prominent topic due to their narrowband emission and high luminous efficiency. However, devices derived from the common types of narrowband TADF materials often experience an efficiency roll-off, which could be ascribed to their relatively slow triplet–singlet exciton interconversion. Since inserting the heavy Se atom into the B/N scheme has been a proven strategy to address the abovementioned issues, herein, extensive density functional theory (DFT) and time-dependent DFT (TD-DFT) simulations have been employed to explore the effects of the structural modification on a series of structurally modified selenium-doped derivatives. Furthermore, the two-layered ONIOM (QM/MM) model has been employed to study the pressure effects on the crystal structure and photophysical properties of the pristine CzBSe. The theoretical results found that the introduced tert-butyl units in Cz-BSeN could result in a shorter charge transfer distance and smaller reorganization energy than the parent CzBSe. In contrast to directly incorporating the o-carborane (Cb) unit to CzBSe, incorporating the bridged phenyl units is important in order to achieve narrowband emissions and high luminous efficiency. The lowest three triplet excited states of CzBSe, Cz-BSeN and PhCb-BSeN all contribute to their triplet–singlet exciton conversions, resulting in a high utilization of triplet excitons. The pressure has an evident influence on the photophysical properties of the aggregated CzBSe and is favored for obtaining narrowband emissions. Our work is promised to provide a feasible strategy for designing selenium-doped derivatives with narrowband emissions and rapid triplet–singlet exciton interconversions. [ABSTRACT FROM AUTHOR]

This study monitored the spatial and temporal fluctuations in the riverine ecosystem of Noa Dihing, Arunachal Pradesh, over three years (October 2020 to December 2023). The habitat quality of the riparian ecosystem, along with key physicochemical parameters (such as temperature, pH, dissolved oxygen, and others), organic matter composition (OMC), substrate composition, and fish species diversity, was assessed seasonally. The habscore and riverine habitat quality index (RHQI) were recorded the highest in the upstream regions (8.00 and 0.85, respectively), while OMC was the highest in the downstream (7.51). Fish biodiversity was evaluated through indices like the Shannon-Wiener diversity index, which peaked at 3.60 in the post-monsoon, and the Margalef richness index, which reached 6.61 in the same season. Simpson's dominance index showed its highest value in winter (0.7), while the evenness index was the greatest in the post-monsoon (0.90). Altogether, 46 fish species from 5 orders were recorded, with Cyprinids being the dominant group. The study utilized Duncan's multiple range test (DMRT) (P≤ 0.05) to confirm the importance of water quality in shaping fish assemblages. Principal component analysis (PCA) on physicochemical parameters explained over 60% of the total variance, with eigenvalues greater than 1.0. Species such as those from the genera Balitora, Garra, Sisor, and Oreichthys exhibited notable structural adaptations to their environment, while species like Pseudoecheneis and Glyptothorax showed thoracic modifications, and Chagunius, Garra, and Paracanthocobitis bore suctorial discs for adhesion. The emergence of natural products has provided extremely valuable references for the treatment of various diseases. Cucurbitacin B, a tetracyclic triterpenoid compound isolated from cucurbitaceae and other plants, is the most abundant member of the cucurbitin family and exhibits a wide range of biological activities, including anti-inflammatory, anti-cancer, and even agricultural applications. Due to its high toxicity and narrow therapeutic window, structural modification and dosage form development are necessary to address these issues with cucurbitacin B. This paper reviews recent research progress in the pharmacological action, structural modification, and application of cucurbitacin B. The enmein-type diterpenoids are a class of anticancer ent-Kaurane diterpnoids that have received much attention in recent years. Herein, a novel 1,14-epoxy enmein-type diterpenoid 4, was reported in this project for the first time. A series of novel enmein-type diterpenoid derivatives were also synthesized and tested for anticancer activities. Among all the derivatives, compound 7h exhibited the most significant inhibitory effect against A549 cells (IC50 = 2.16 µM), being 11.03-folds better than its parental compound 4. Additionally, 7h exhibited relatively weak anti-proliferative activity (IC50 > 100 µM) against human normal L-02 cells, suggesting that it had excellent anti-proliferative selectivity for cancer cells. Mechanism studies suggested that 7h induced G0/G1 arrest and apoptosis in A549 cells by inhibiting the PI3K/AKT/mTOR pathway. This process was associated with elevated intracellular ROS levels and collapsed MMP. In the following study, a series of new lupeol-3-carbamate derivatives were synthesized, and the structures of all the newly derived compounds were characterized. The new compounds were screened to determine their anti-proliferative activity against human lung cancer cell line A549, human liver cancer cell line HepG2, and human breast cancer cell line MCF-7. Most of the compounds were found to show better anti-proliferative activity in vitro than lupeol. Among them, obvious anti-proliferation activity (IC50 = 5.39~9.43 μM) was exhibited by compound 3i against all three tumor cell lines. In addition, a salt reaction was performed on compound 3k (IC50 = 13.98 μM) and it was observed that the anti-proliferative activity and water solubility of compound 3k·CH3I (IC50 = 3.13 μM), were significantly enhanced subsequent to the salt formation process. The preliminary mechanistic studies demonstrated that apoptosis in HepG2 cells was induced by compound 3k·CH3I through the inhibition of the PI3K/AKT/mTOR pathway. In conclusion, a series of new lupeol-3-carbamate derivatives were synthesized via the structural modification of the C-3 site of lupeol, thus laying a theoretical foundation for the design of this new anticancer drug. All‐solid‐state Li‐metal battery (ASSLB) represents advantageous energy storage system for automotive applications. For ASSLB, inorganic solid electrolyte is essential in determining safety and cycling performance. However, significant challenges persist in practical construction of ASSLB with optimized electrolyte. Specifically, electrolyte's structural instability influencing its electrochemical performance remains critical issue within typical operating temperatures for ASSLB in electric vehicles. Herein, this challenge is fundamentally addressed by substituting trace amount of lithium with cadmium, which lacks crystal field stabilization energy. This strategy of atomic interaction modification has induced electrolyte's structural distortion and electronic alteration by deliberately introducing disorder at local lithium sites. Li symmetric cell with cadmium‐substituted antiperovskite solid electrolyte exhibits outstanding critical current density of 11.5 mA cm−2 (5.75 mAh cm−2) and excellent stability for 3000 h at 10.0 mA cm−2 (5.0 mAh cm−2). Sufficient utilization of visible‐light generated charge carriers in proton reduction reactions is of great significance for the development of effective solar‐fuel technologies. Achieving simultaneous bulk rapid transfer and surface efficient extraction of charge carriers is still very challenging. Herein, it is found for the first time ammonium persulfate (APS) can significantly influence polymerization processes of C3N4 (CN) from melamine to poly (heptazine imide) (PHI) under the simultaneous oxygen doping and etching effect of SO42−. PHI with high crystallinity, porous structure, and in‐situ oxygen doping was therefore obtained through one‐step APS‐assisted salt strategy. Benefiting from sufficient visible‐light absorption and upshifted conduction band originating from regulated electronic structure and optimized morphology through APS modification, the as‐prepared PHI achieved a H2 evolution activity of 3274.23 μmol h−1 g−1 (λ > 420 nm), which is appropriately 148 and 19 times that of conventional and crystalline CN. Seawater electrolysis presents a viable route for sustainable large‐scale hydrogen production, yet its practical application is hindered by several technical challenges. These include the sluggish kinetics of hydrogen evolution, poor stability, cation deposition at the cathode, electrode corrosion, and competing chloride oxidation at the anode. To overcome these obstacles, the development of innovative electrocatalysts is crucial. Transition metal phosphides (TMPs) have emerged as promising candidates owing to their superior catalytic performance and tunable structural properties. This review provides a comprehensive analysis of recent progress in the structural engineering of TMPs tailored for efficient seawater electrolysis. We delve into the catalytic mechanisms underpinning hydrogen and oxygen evolution reactions in different pH conditions, along with the detrimental side reactions that impede hydrogen production efficiency. Several methods to prepare TMPs are then introduced. Additionally, detailed discussions on structural modifications and interface engineering tactics are presented, showcasing strategies to enhance the activity and durability of TMP electrocatalysts. By analyzing current research findings, our review aims to inform ongoing research endeavors and foster advancements in seawater electrolysis for practical and ecologically sound hydrogen generation. Inhibition of SIRT3 exhibited potency in triggering leukemic cell differentiation. In discovery of potent SIRT3 inhibitors for cancer differentiation therapy, structural modification was performed on the previously developed lead compound P6. A total of 33 compounds were designed and synthesized. In the enzyme inhibitory assay, several molecules S18, S26, S27 and T5 showed potent SIRT3 inhibitory activity with IC50 value of 0.53, 1.86, 5.06, and 2.88 μM, respectively. Moreover, the tested compounds exhibited SIRT3 inhibitory selectivity over SIRT1 and SIRT2. Compounds S27 and T5 were potent in inhibition the growth of MM1.S and RPMI‐8226 cells in the in vitro antiproliferative test. Significantly, representative compounds, especially S27 and T5, promoted differentiation of tested MM cells in the cellular morphological evaluation, accompanied by increasing the expression of differentiation antigen CD49e and human immunoglobulin light chain lambda and kappa. Additionally, molecule S18 without antiproliferative potency itself, showed significant inhibitory activity against growth factor IL‐6 induced RPMI‐8226 cell proliferation. Collectively, potent SIRT3 selective inhibitors with MM cell differentiation potency were developed for further discovery of anticancer drugs. [ABSTRACT FROM AUTHOR]

This paper systematically reviews the research advancements in indium oxide (In2 O3)-based gas-sensitive materials and focuses on their potential applications in environmental monitoring and safety. It provides an in-depth analysis of the structure, properties and gas sensing mechanisms of indium oxide, detailing the impact of various synthesis methods (such as hydrothermal and chemical vapor deposition) on the microstructure and performance of the materials. Additionally, this paper discusses the applications of modification techniques, including element doping, material loading and semiconductor compositing in enhancing gas sensitivity. It also analyzes current research challenges and future directions, offering significant perspectives and directions for further research and application of indium oxide-based gas-sensitive materials. Tea is a daily drink for most people, and one of its major ingredients, epigallocatechin-3-gallate (EGCG), has been widely recognized as a potent antioxidant with diverse biological activities. However, its low stability and bioavailability hinder its further clinical applications. In this study, we designed and synthesized a novel EGCG-valine derivative 4 by replacing the gallic acid with a valine moiety in four steps. The structural elucidation of derivative 4 was performed using NMR, IR, mass, and UV spectroscopies. Additionally, the physicochemical properties of 4 were predicted by SwissADME, showing improved drug-like parameters and intestinal absorption compared to the parent compound EGCG. High thermostability and catalytic efficiency of α-amylase are required for the industrial applications. To improve catalytic parameters including Michaelis constant, maximum velocity, turnover number, enzyme half-life, and the catalytic efficiency of α-amylase, it was treated alone and/or simultaneously with Ca2+ ions (12.5 and 25 mM) and/or ultrasound (USN) (64.5 W, 25 + 40 kHz). The structural analysis of the enzyme (the enzyme's secondary structure and conformation) was carried out using Fourier transform infrared (FTIR) spectroscopy and circular dichroism (CD) techniques. Moreover, the thermal properties of the enzyme were evaluated using differential scanning calorimetry (DSC). USN + 12.5 mM Ca2+ and 25 mM Ca2+, changed the maximum velocity (+ 6.5% and + 37.4%), turnover number (+ 6.6% and + 38.2%), enzyme half-life (+ 51.6% and + 113.7%), and catalytic efficiency (+ 18.1% and -3.3%) of α-amylase. The energy barriers of thermo-inactivation and inactivation enthalpy of α-amylase exposed to USN + 25 mM Ca2+ were increased by 25% and 5.8%, respectively, without affecting entropy of inactivation, showing how enzyme's thermal stability was improved after the treatments. The increase in midpoint temperature (35%) and α-helix (14%) values as well as the decrease in the β-sheet structure (39%) of the α-amylase after applying USN + Ca2+ (25 mM) confirmed the thermal stability enhancement of α-amylase after the treatment compared to control.
