Abstract

Background: Humans are exposed to tens of thousands of chemical substances that need to be assessed for their potential toxicity. Acute systemic toxicity testing serves as the basis for regulatory hazard classification, labeling, and risk management. However, it is cost- and time-prohibitive to evaluate all new and existing chemicals using traditional rodent acute toxicity tests. In silica models built using existing data facilitate rapid acute tox- icity predictions without using animals. Objkctivks: The U.S. Interagency Coordinating Committee on the Validation of Alternative Methods (ICCVAM) Acute Toxicity Workgroup organ- ized an international collaboration to develop in silica models for predicting acute oral toxicity based on five different end points: Lethal Dose 50 (LD50 value, U.S. Environmental Protection Agency hazard (four) categories, Globally Harmonized System for Classification and Labeling hazard (five) categories, very toxic chemicals [LD50 (LD50 ≤ 50 mg/kg)], and nontoxic chemicals (LD50 > 2,000 mg/kg). Mkthods: An acute oral toxicity data inventory for 11,992 chemicals was compiled, split into training and evaluation sets, and made available to 35 participating international research groups that submitted a total of 139 predictive models. Predictions that fell within the applicability domains of the submitted models were evaluated using external validation sets. These were then combined into consensus models to leverage strengths of individual approaches. Results: The resulting consensus predictions, which leverage the collective strengths of each individual model, form the Collaborative Acute Toxicity Modeling Suite (CATMoS). CATMoS demonstrated high performance in terms of accuracy and robustness when compared with in viva results. Discussion: CATMoS is being evaluated by regulatory agencies for its utility and applicability as a potential replacement for in viva rat acute oral toxicity studies. Abstract

Locoweeds cause significant livestock poisoning and economic loss in the western United States. The toxicity of Embellisia sp. fungi isolated from locoweed was compared with locoweed toxicity using the rat as a model. Rats were fed diets containing locoweed, fungus and alfalfa, or alfalfa. Locoweed- and fungus-fed rats consumed swainsonine-containing food at approximately 1.3 mg*[kg.sup.-1]*[d.sup.-1], gained less weight (P = 0.001) and ate less than controls. Swainsonine is the principal agent responsible for inducing locoism in animals. The concentrations of alkaline phosphatase and aspartate aminotransferase enzymes were greater (P < 0.05) in serum of locoweed- and fungus-fed rats compared with control rats. Similar intracellular vacuolation was observed in renal, pancreatic, and hepatic tissues of rats that consumed either locoweed or fungus. Rats that ate locoweed or Embellisia fungi displayed indistinguishable toxicity symptoms. The Embellisia fungi from locoweed can induce toxicity without the plants. Abstract

Vitamin A is an essential nutrient and is well established to be crucial for regulating hematopoiesis, epithelial integrity, vision, and embryogenesis. In humans, vitamin A deficiency is associated with increased risk of morbidity, mortality, and a decreased immune response to vaccines. However, despite being the first vitamin discovered, little is known about its role in these outcomes in dairy cows. Dairy cows experience a significant decrease in serum vitamin A concentrations at the time of calving when cows are most at risk for disease. Previous study findings show conflicting results as to the effects of vitamin A and diseases such as mastitis in dairy cows. However, many studies with negative results are limited by their small sample size and short study period. Abstract

CYP enzyme induction is a sensitive biomarker for phenotypic metabolic competence of in vitro test systems; it is a key event associated with thyroid disruption, and a biomarker for toxicologically relevant nuclear receptor-mediated pathways. This paper summarises the results of a multi-laboratory validation study of two in vitro methods that assess the potential of chemicals to induce cytochrome P450 (CYP) enzyme activity, in particular CYP1A2, CYP2B6, and CYP3A4. The methods are based on the use of cryopreserved primary human hepatocytes (PHH) and human HepaRG cells. The validation study was coordinated by the European Union Reference Laboratory for Alternatives to Animal Testing of the European Commission’s Joint Research Centre and involved a ring trial among six laboratories. The reproducibility was assessed within and between laboratories using a validation set of 13 selected chemicals (known human inducers and non-inducers) tested under blind conditions. The ability of the two methods to predict human CYP induction potential was assessed. Chemical space analysis confirmed that the selected chemicals are broadly representative of a diverse range of chemicals. The two methods were found to be reliable and relevant in vitro tools for the assessment of human CYP induction, with the HepaRG method being better suited for routine testing. Recommendations for the practical application of the two methods are proposed.

Abstract

Numerical modeling of the nonreactive mixing processes associated with a lobed fuel injector in a coflowing air stream is presented. The lobed fuel injector is a device which generates strong streamwise vorticity, producing locally high strain rates which can enhance the molecular mixing of reactants while delaying ignition in a controlled manner. Vortex element modeling is used to simulate flow field evolution and fuel element mixing characteristics for this lobed fuel injector. Quantitative predictions for vorticity generation and qualitative results for streamwise rollup compare well qualitatively with recent experimental investigations of this flow field [Smith et al, Phys. Fluids 9, 667 (1997)]. Parametric studies of the effects of lobe amplitude-to-wavelength ratio, lobe angle, and lobe shape for given flow conditions suggest that geometrical features may be optimized to enhance mixing and control reaction processes.

Abstract

Despite the controversy over abortion rights, there is a clear public consensus in the United States that abortion should be legal in cases of rape and incest. The Utah and Louisiana antiabortion laws enacted in 1991 purport to grant an exception for cases of rape and incest, but the victim must comply with onerous procedural conditions in order to meet the statutory requirements of the exception. The author examines the statutory conditions that rape victims must comply with in order to satisfy Louisiana&apos;s antiabortion rape exception and concludes that in practice these conditions will impede rather than ensure most victims&apos; access to legal abortion. The Author argues that corroboration requirements are imposed by legislatures to alleviate their mistrust of a woman&apos;s report of rape and that it is unfair to burden victims with such requirements. The Author concludes that given the prevalence of rape in our society, women must be ensured access to legal abortion without unrealistic restrictions.
