Abstract

Due to the increasing need of new treatment options against bacterial lung infections, novel antimicrobial peptides (AMPs) are under development. Local bioavailability and less systemic exposure lead to the inhalation route of administration. Combining AMPs with nanocarriers (NCs) into nanosystems (NSs) might be a technique for improved results. An air-liquid interface (ALI) in vitro inhalation model was set up including a human alveolar lung cell line (A549) and an optimized exposure system (P.R.I.T.® ExpoCube®) to predict acute local lung toxicity. The approach including aerosol controls (cupper-II-sulfate and lactose) delivered lowest observable adverse effect levels (LOAELs). Different combinations of AMPs (AA139, M33) and NCs (polymeric nanoparticles (PNPs), micelles and liposomes) were tested under ALI and submerged in vitro conditions. Depending on the nature of AMP and NCs, packing of AMPs into NSs reduced the AMP-related toxicity. Large differences were found between the LOAELs determined by submerged or ALI testing with the ALI approach indicating higher sensitivity of the ALI model. Since aerosol droplet exposure is in vivo relevant, it is assumed that ALI based results represents the more significant source than submerged testing for in vivo prediction of local acute lung toxicity. In accordance with the current state-of-the-art view, this study shows that ALI in vitro inhalation models are promising tools to further develop in vitro methods in the field of inhalation toxicology.

Abstract

Staphylococcus aureus is considered to be an extracellular pathogen. However, survival of S. aureus within host cells may provide a reservoir relatively protected from antibiotics, thus enabling long-term colonization of the host and explaining clinical failures and relapses after antibiotic therapy. Here we confirm that intracellular reservoirs of S. aureus in mice comprise a virulent subset of bacteria that can establish infection even in the presence of vancomycin, and we introduce a novel therapeutic that effectively kills intracellular S. aureus. This antibodyantibiotic conjugate consists of an anti-S. aureus antibody conjugated to a highly efficacious antibiotic that is activated only after it is released in the proteolytic environment of the phagolysosome. The antibodyantibiotic conjugate is superior to vancomycin for treatment of bacteraemia and provides direct evidence that intracellular S. aureus represents an important component of invasive infections., Author(s): Sophie M. Abstract

Since the inception of affinity chromatography 30 years ago (1), it has developed into a powerful tool mainly for the purification of proteins. It is based on the reversible formation of a tight binding complex between a ligand, immobilized on an insoluble matrix and a substance, the ligate, to be isolated from the solution. Typically the ligate is adsorbed by a column with the immobilized ligand, whereas noninteracting substances are washed off. By changing the elution conditions, the ligate can be released in a highly purified form. Some researchers argue that this procedure is based on specific extraction rather than by chromatography, which should rely on the differential migration of various substances. Regardless of the definitions, it is clear that traditional affinity chromatography exploits high affinity or avidity (binding constant (Ka) > 105/M) between the interacting molecules, which will result in an effective adsorption of the ligate. In this context the distinction between affinity and avidity is important: Whereas affinity describes the interaction in an individual binding site, avidity describes the multivalent binding between multiple binding sites of the ligand and ligate, respectively. High binding strength is required to achieve efficient adsorption, whereas weaker interactions will not produce adequate binding and therefore insufficient specificity will be acquired. This statement that strong specific binding is a prerequisite for the successful isolation of an interacting molecule has been in a nutshell the consensus of affinity chromatography.
