Abstract

Our ability to confidently develop appropriate countermeasures for radiations in space in terms of shielding and design of a spacecraft, the mission scenario, or chemoprevention is severely limited by the uncertainties in both the risk itself and the change in that risk with intervention. Despite the fact that the risk of carcinogenesis from exposures of personnel to radiations on long-term missions is considered one of the worst hazards in space, only a limited amount of in-vivo data exist for tumor induction from exposures to protons or energetic heavy ions (HZEs) at lower doses. The most extensive work remains the landmark study. for tumor development in the harderian gland of the mouse. The objective of this study is to characterize the level of risk for tumor induction in another relevant animal model. Subsequent experiments are designed to test the hypothesis that the level of risk can be reduced by pharmaceutical intervention in the promoting and progressing stages of the disease rather than in the initiating stage. The work presented here results from a cooperative effort on the part of investigators from two projects of the Radiation-Effects Team of the National Space Biomedical Research Institute (NSBRI). The collaborating projects are the Core Project which is investigating the risk of carcinogenesis in Sprague-Dawley rats and the Chemoprevention Project which is investigating the ability of Tamoxifen to reduce the number of malignant tumors in the irradiated animals. Research at the cellular and subcellular levels is being conducted in two other projects of the Radiation-Effects Team, Cytogenetics with J. R. Williams as Principal Investigator and Mutations from Repeated DNA Sequences. Results for these other projects also are being presented at this Workshop.

Abstract

Purpose: To study venous injury caused by a prototype percutaneous mechanical thrombolytic device.Methods: Simulated thrombolysis was performed using the device, or the Fogarty balloon catheter (FBC) as control, in the infrarenal inferior vena cava (IVC) of 40 New Zealand white rabbits. Venous injury was evaluated by cavography, Evans blue dye staining, and histology at 0, 1, and 6 weeks postprocedure.Results: Both devices resulted in near complete endothelial denudation acutely. No differences in reendothelialization were noted at any time in the proximal and mid-IVC, but there was significantly greater reendothelialization in the distal IVC in the rabbits treated 6 weeks earlier with the device (p < or = 0.04). Additionally, the inner luminal diameter at necropsy for the 1-week rabbits treated with the FBC was significantly narrower in the distal and middle sections of the IVC when compared with the device (p < or = 0.02 for both segments). There was no luminal diameter difference at 0 or 6 weeks.Conclusion: Based on a rabbit model, venous injury from the device was found to be similar to, and in the distal IVC less than, the routinely used FBC. Abstract

In our first 18 months' clinical experience with embolization in the chest and abdomen using detachable balloons, successful results were obtained in 34 of 38 patients. One and 2-mm detachable silicone balloons, which can occlude vessels 4--8 mm in diameter, were employed. Prolonged balloon inflation was routinely achieved using iso-osmotic iodipamide meglumine as the filler and limiting inflation volumes to experimentally determined maximums. Improvements in introducer catheter design simplified delivery of the balloon into a variety of circulations. Detachable balloons are not suitable for all embolization purposes, and they are frequently used in conjunction with other agents. When used properly, balloons produce a permanent occlusion that is extremely selective and potentially reversible up to a certain point in the procedure. The balloon technique enables the angiographer to occlude vessels at distances of 2--10 mm beyond the introducer catheter, thus avoiding the need for subselective catheterization and minimizing the dangers of inadvertent embolization. 15. A comparison of subcutaneous and intraperitoneal oxytetracycline injection methods for control of infectious disease in the rat

Abstract

After receiving once daily intraperitoneal (i.p.) or subcutaneous (s.c.) injections of oxytetracycline (15 mg/100 g bodyweight) for 7 days, laboratory rats had significantly different oxytetracycline serum levels depending on the method of injection. Regardless of injection method, the biological half-life of the antibiotic was 4·25 h and serum antibiotic levels never fell below therapeutic levels for certain rat pathogens during 24 h post-injection. Severe peritonitis and cellulitis were found in rats whether injected s.c. or i.p. and some animals lost bodyweight. It appeared that either injection method would be an effective treatment for certain infectious diseases of laboratory rats but tissue damage, probably from oxytetracycline's acidity, made both methods unattractive.

Abstract

The cellular mechanisms governing the expression of mononuclear cell vasculitis are poorly understood. For determination of the precise sequence of events in the development of vasculitis in autoimmune MRL/lpr mice, histologic sections from 4-20-week-old mice were evaluated with a panel of cytochemical and immunohistochemical stains. The results show that vascular disease in MRL/lpr mice develops as follows: Thy 1+, Ly 1+, L3T4- T cells assemble around predominantly small-to-medium muscular arteries at approximately 8 weeks of age. At 12 weeks of age, an adventitial inflammatory focus forms, composed of large "reactive" mononuclear inflammatory cells adjacent to hypertrophied vascular smooth muscle cells (VSMCs). Blastic Thy 1+, Ly 1+, L3T4- T cells subsequently infiltrate the tunica media, and selective VSMC karyolysis results. Occasional cytotoxic/suppressor T cells, macrophages, and possibly NK cells are noted primarily distal to the infiltration site. The outer zone of the inflammatory infiltrate is composed of mature B cells and occasional B-cell precursors. These findings suggest that cellular constituents of the immune response mediate mononuclear cell vasculitis in MRL/lpr mice.

Abstract

A new disease of rabbits is described. Following an acute febrile course, animals die or recover by the 11th day postinoculation. The characteristic pathologic finding is multifocal myocardial degeneration and necrosis. The disease can be transmitted by various routes with tissue filtrates or with infectious sera diluted to 10(-6) and passed through 0.1 micron filters. Virus particles with morphologic features characteristic of a coronavirus are present in infectious but not in normal rabbit serums. The antigen(s) in the infectious serums cross-reacts with the 229E and the OC43 strains of human coronavirus. Antigen cross-reacting with the 229E virus is detectable by immunofluorescent staining in frozen sections of heart tissue from sick but not from healthy animals. Animals surviving infection seroconvert to coronavirus specificity, as demonstrated by the presence in convalescent serums of antibody capable of reacting with the 339E virus. Susceptibility to infection has not been demonstrated in mice, hamsters, or guinea pigs, and the virus was not adapted for growth in tissue culture. It is uncertain whether the agent is a natural pathogen of rabbits or a coronavirus contaminant from another species, possibly human. The name rabbit infectious cardiomyopathy is suggested for this disease.

20. Histiocytic sarcoma with a granuloma-like component occurring in a large colony of Sprague-Dawley rats

Abstract

Tumors from a large colony of over 2000 Sprague-Dawley rats were reviewed by a panel of pathologists under the auspices of Universities Associated for Research and Education in Pathology, In the course of this study, a distinct malignancy in 105 rats was segregated and given the name "histiocytic sarcoma." This report describes the histologic features and anatomic distribution of the tumor, along with its incidence by age and sex of the rat. The neoplasm is characterized by a spectrum of histologic patterns, one extreme resembling a sarcomatous process and the other a granulomatous process. The exact morphologic composition varied from animal to animal, as well as in tumors from the same animal. The sarcomatous pattern is composed of sheets of uniform cells, occasionally exhibiting phagocytosis. The granulomatous pattern contains epithelioid histiocytes, central areas of tumor necrosis surrounded by palisading cells, and multinucleate giant cells. The liver is the organ most commonly containing histiocytic sarcoma, both in animals with multiple tumors and in those with involvement of only one organ. Hepatic invasion is typified by expansion of the portal areas and infiltration of the sinusoids by tumor cells. The lung is the second most common site. The distribution of pulmonary tumors is bronchiolocentric and angiocentric. Other common sites included the lymph node and spleen, along with large soft tissue tumors of the mediastinum and retroperitoneum. Although occasional previous reports apparently have described a similar or identical neoplasm under varying names, no large group of tumors of this unique type in rats has been studied and characterized previously.

Abstract

Three 12- to 16-month-old female B6,129 apolipoprotein E-deficient mice were presented for necropsy because of bilateral masses in the gluteal region. Histopathologic examination revealed cholesterol granulomas extending from the superficial dermis to the underlying gluteal muscles. Microscopic granulomas were found on the face, perivaginal tissues, mesenteric lymph nodes, spleen, joints, kidneys, and choroid plexus. Other lesions included severe atherosclerosis of the aortic valves, aorta, pulmonary artery, and renal artery. Here we show that macroscopic cholesterol granulomas can develop in apolipoprotein E-deficient that receive a normal rodent diet.

Abstract

An adult female lesser flamingo (Phoeniconaias minor), caught in the African Rift Valley in 1991 and subsequently housed at the Baltimore Zoo, died of severe visceral gout in 1996. Necropsy revealed a white, moderately firm, nodular lesion, 1 cm in diameter, in the serosal wall of the small intestine. Although it was initially thought to be a tumor or focal granuloma, histologic examination revealed multiple cestodes deeply embedded at the base of the crypts between the intestinal villi, with their massive scolices (up to 3.4 mm in diameter) distending these spaces into multiple diverticulae. The mucosal epithelium surrounding the scolices was severely attenuated. Around the diverticulae, in the submucosa and muscularis, was a mild to moderate lymphocytic reaction and mild fibrosis. The proximity of multiple scolices and extensive invasion of host tissue suggested that the infection occupied a preexisting lesion. The cestodes were cyclophyllids but were distinct from any species previously reported from flamingos. Helminths should be included in differential diagnoses for gastrointestinal nodules in flamingos.
