Abstract

Thrombopoietin (Tpo) is the primary regulator of megakaryocyte and platelet numbers and is required for haematopoetic stem cell maintenance. Tpo functions by binding its receptor (TpoR, a homodimeric Class I cytokine receptor) and initiating cell proliferation or differentiation. Here we characterise the murine Tpo:TpoR signalling complex biochemically and structurally, using cryo-electron microscopy. Tpo uses opposing surfaces to recruit two copies of receptor, forming a 1:2 complex. Although it binds to the same, membrane-distal site on both receptor chains, it does so with significantly different affinities and its highly glycosylated C-terminal domain is not required. In one receptor chain, a large insertion, unique to TpoR, forms a partially structured loop that contacts cytokine. Tpo binding induces the juxtaposition of the two receptor chains adjacent to the cell membrane. The therapeutic agent romiplostim also targets the cytokine-binding site and the characterisation presented here supports the future development of improved TpoR agonists.

Abstract

MLKL is the essential effector of necroptosis, a form of programmed lytic cell death. We have isolated a mouse strain with a single missense mutation, MlklD139V, that alters the two-helix 'brace' that connects the killer four-helix bundle and regulatory pseudokinase domains. This confers constitutive, RIPK3 independent killing activity to MLKL. Homozygous mutant mice develop lethal postnatal inflammation of the salivary glands and mediastinum. The normal embryonic development of MlklD139V homozygotes until birth, and the absence of any overt phenotype in heterozygotes provides important in vivo precedent for the capacity of cells to clear activated MLKL. These observations offer an important insight into the potential disease-modulating roles of three common human MLKL polymorphisms that encode amino acid substitutions within or adjacent to the brace region. Compound heterozygosity of these variants is found at up to 12-fold the expected frequency in patients that suffer from a pediatric autoinflammatory disease, chronic recurrent multifocal osteomyelitis (CRMO).

Abstract

Ustekinumab, a monoclonal antibody to the p40 subunit of interleukin-12 and interleukin-23, was evaluated as an intravenous induction therapy in two populations with moderately to severely active Crohn's disease. Ustekinumab was also evaluated as subcutaneous maintenance therapy.We randomly assigned patients to receive a single intravenous dose of ustekinumab (either 130 mg or approximately 6 mg per kilogram of body weight) or placebo in two induction trials. The UNITI-1 trial included 741 patients who met the criteria for primary or secondary nonresponse to tumor necrosis factor (TNF) antagonists or had unacceptable side effects. The UNITI-2 trial included 628 patients in whom conventional therapy failed or unacceptable side effects occurred. Patients who completed these induction trials then participated in IM-UNITI, in which the 397 patients who had a response to ustekinumab were randomly assigned to receive subcutaneous maintenance injections of 90 mg of ustekinumab (either every 8 weeks or every 12 weeks) or placebo. The primary end point for the induction trials was a clinical response at week 6 (defined as a decrease from baseline in the Crohn's Disease Activity Index [CDAI] score of ≥100 points or a CDAI score150). The primary end point for the maintenance trial was remission at week 44 (CDAI score150).The rates of response at week 6 among patients receiving intravenous ustekinumab at a dose of either 130 mg or approximately 6 mg per kilogram were significantly higher than the rates among patients receiving placebo (in UNITI-1, 34.3%, 33.7%, and 21.5%, respectively, with P≤0.003 for both comparisons with placebo; in UNITI-2, 51.7%, 55.5%, and 28.7%, respectively, with P0.001 for both doses). In the groups receiving maintenance doses of ustekinumab every 8 weeks or every 12 weeks, 53.1% and 48.8%, respectively, were in remission at week 44, as compared with 35.9% of those receiving placebo (P=0.005 and P=0.04, respectively). Within each trial, adverse-event rates were similar among treatment groups.Among patients with moderately to severely active Crohn's disease, those receiving intravenous ustekinumab had a significantly higher rate of response than did those receiving placebo. Subcutaneous ustekinumab maintained remission in patients who had a clinical response to induction therapy. Abstract

The in situ primary production rates and various environmental variables were investigated in the Chukchi Sea during the 3rd RUSALCA expedition, which was conducted in 2012, to identify the current status of primary production. A 13C-15N dual tracer technique was used to measure the daily primary production rates, which ranged from 0.02 to 1.61 gCm-2 d-1 (mean ± SD= 0.42 ± 0.52 gCm-2 d-1). The primary production rates showed large regional differences, with the southern region (0.66 ± 0.62 gCm-2 d-1) producing approximately five times as much as the northern region (0.14 ± 0.10 gCm-2 d-1), which was primarily due to the differences in phytoplankton biomasses induced by regional nutrient conditions. The primary production rates in the Chukchi Sea were averaged using data acquired during the three different RUSALCA expeditions (2004, 2009, and 2012) as 0.33 gCm-2 d-1 (SD= 0.40 gCm-2 d-1), which was significantly lower than previously reported rates. In addition to strong seasonal and interannual variations in primary production, recent decreases in the concentrations of major inorganic nutrients and chlorophyll a could be among the reasons for the recent low primary production in the Chukchi Sea because the primary production is mainly affected by nutrient concentration and phytoplankton biomass. The nutrient inventory and primary production appear to be largely influenced by the freshwater content (FWC) variability in the region due to the significant relationships between FWC, nitrate concentrations (r = 0.54, p < 0.05) and primary production rates (r = 0.56, p < 0.05). Moreover, we found highly significant relationships between the nutrient levels and the primary production rates (r = 0.75, p < 0.001). In conclusion, the primary production in the Chukchi Sea is primarily controlled by nutrient availability which is strongly related to the FWC variability. Our results imply that the predicted increase in freshwater accumulation might cause a decrease in primary production by lowering the nutrient inventory in the euphotic zone of the Chukchi Sea. Abstract

Predictions of the yearly pattern of boiler heat load for the dwellings of an estate served by a group heating scheme are notoriously difficult to fit to metered values. This is because, in conventional theoretical expressions to represent the various heat loss terms, no account is taken of the uncertainties in the values of the various critical factors. A method of allowing for these uncertainties is outlined which enables theoretical predictions to be assigned limits within which measured values of heat load may be expected to lie. 36. In vitro dissolution characteristics of uranium oxides

Abstract

This study compares the in vitro characteristics of laboratory prepared thermochemical intermediates of ADU and uranium oxide produced at Australian uranium mines. All uranium oxide examined by batch dissolution behaved as two discrete fractions; the first a highly soluble (Class D) whose magnitude correlates with the total product sulphate concentration and a second fraction composed of soluble and suspended material. The dissolution of uranium by the batch method appears highly dependent on experimental procedure, invalidating interpretation of in vitro batch procedures in terms of dissolution half-time. It is suggested that the temperature of form ation may determine the degree of dissolution of the second fraction in ionic media. The proportions of the components in saline is predicted by a model given the saturation concentration of the soluble species and the fraction of suspended material.
