Thromboembolic conditions are the most common cause of death in developed countries. Anticoagulant therapy is the treatment of choice, and heparinoids and warfarin are the most adopted drugs. Sulphated polysaccharides extracted from marine organisms have been demonstrated to be effective alternatives, blocking thrombus formation by inhibiting some factors involved in the coagulation cascade. In this study, four acidic glycan fractions from the marine sponge Sarcotragus spinosulus were purified by anion-exchange chromatography, and their anticoagulant properties were investigated through APTT and PT assays and compared with both standard glycosaminoglycans and holothurian sulphated polysaccharides. Moreover, their topographic localization was assessed through histological analysis, and their cytocompatibility was tested on a human fibroblast cell line. A positive correlation between the amount of acid glycans and the inhibitory effect towards both the intrinsic and extrinsic coagulation pathways was observed. The most effective anticoagulant activity was shown by a highly charged fraction, which accounted for almost half (about 40%) of the total hexuronate-containing polysaccharides. Its preliminary structural characterization, performed through infrared spectroscopy and nuclear magnetic resonance, suggested that it may consist of a fucosylated chondroitin sulphate, whose unique structure may be responsible for the anticoagulant activity reported herein for the first time.

In some Porifera (Demospongiae: Keratosa), prototypes of the connective system are almost exclusively based on collagenic networks. We studied the topographic distribution, spatial layout, microtraits, and/or morphogenesis of these collagenic structures in Ircinia retidermata (Dictyoceratida: Irciniidae). Analyses were carried out on a clonal strain from sustainable experimental mariculture by using light and scanning electron microscopy. Histology revealed new insights on the widely diversified and complex hierarchical assemblage of collagenic structures. Key evolutionary novelties in the organization of sponge connective system were found out. The aquiferous canals are shaped as corrugate-like pipelines conferring plasticity to the water circulation system. Compact clusters of elongated cells are putatively involved in a nutrient transferring system. Knob-ended filaments are characterized by a banding pattern and micro-components. Ectosome and outer endosome districts are the active fibrogenetic areas, where exogenous material constitutes an axial condensation nucleus for the ensuing morphogenesis. The new data can be useful to understand not only the evolutionary novelties occurring in the target taxon but also the morpho-functional significance of its adaptive collagenic anatomical traits. In addition, data may give insights on both marine collagen sustainable applied researches along with evolutionary and phylogenetic analyses, thus highlighting sponges as a key renewable source for inspired biomaterials. Therefore, we also promote bioresources sustainable exploitation with the aim to provide new donors of marine collagen, thereby supporting conservation of wild populations/species., (© 2022 The Authors. Eight new species of Duplominona (Platyhelminthes, Proseriata, Monocelididae) are described from the Pacific coast of Panama. They differ from their congeners in the detailed morphology of hard structures associated with the copulatory organ. Duplominona basidilatata n. sp. has a cirrus provided with 5-6 rows of triangular spines, 3-8 μm long, with a large, flat, poorly sclerotized basis. D. hystricina n. sp. has 10-12 rows of needle-shaped spines, 3.5-15 μm long, with a swollen basis. The cirrus of D. hyperhystricina n. sp. is provided with 20-25 rows of slender spines 1.5-9 μm long, with a recurved distal tip. In D. veracruzensis n. sp., cirrus spines increase abruptly in size, from 1.5-2 μm to 6-7 μm. D. uniserta n. sp. has a very long seminal vesicle and a small cirrus, provided with one girdle of hook-shaped spines, 3-5 μm long. D. macrodon n. sp. has one girdle of large, triangular spines, 8-18 μm long. Both D. trimera n. sp. and D. pseudotrimera n. sp. have a tripartite tail, and their cirrus is provided with a stylet. In D. trimera n. sp., the stylet is surrounded by 15-20 rows of spines, 6.5-10 μm long, while D. pseudotrimera n. sp. has 6-8 rows of large spines, 7-22 μm long. D. uniserta n. sp. and D. aduncospina Curini-Galletti, 2019 from the Caribbean coast of Panama have few rows of morphologically nearly identical spines, and are possible candidates as trans-isthmian geminate species. The presence of species with a tripartite tail on both sides of the Isthmus of Panama suggests the possibility of further geminate species pairs; however, no support could be obtained on the basis of the morphology of their hard structures. Five of the eight new species of Duplominona have been found in a single locality, and the diversity of genus along the Pacific coast of Panama may be far higher than present contribution suggests.

Nine new species of Duplominona and one new Pseudominona (Platyhelminthes, Proseriata, Monocelididae) are described from the Caribbean coast of Panama and from Puerto Rico.                Duplominona aduncospina n. sp.; D. terdigitata n. sp.; D. pusilla n. sp.; D. bocasana n. sp. (from Puerto Rico) can be distinguished from the numerous congeneric species based on fine details of the sclerotized structures of the copulatory organ. Duplominona aduncospina n. sp. is characterised by a cirrus provided with 3-4 rows of recurve spines, 2-3 μm long. D. terdigitata n. sp. shows a tripartite tail, and needle-shaped cirrus spines, 1.5-9 μm long. Cirrus spines of D. pusilla n. sp. are scale-like, 1.5-3 μm long. D. bocasana n. sp. has triangular spines, 1.5-6 μm long. D. dissimilispina n. sp. has needle-shaped spines, 3.5-15 μm long. D. chicomendesi n. sp. has a small cirrus, with few, strongly curved spines, 2-7.5 μm long. D. macrocirrus n. sp. has a large cirrus, provided with spines 3-9 μm long, with the longest spines placed medially. D. diademata n. sp. has a cirrus with two separate spiny areas, with spines 7-13 μm long. D. puertoricana n. sp. has a very long cirrus, with poorly sclerotised proximal spines, and distal spines to 6 μm long. A taxonomic key of the genus Duplominona is provided. Pseudominona cancan n. sp. from Panama differs from P. dactylifera from Bermuda, the only species known in the genus so far, for its shorter cirrus and fewer, triangular spines 3-5 μm long, and for the position of the vagina, close to mouth. A specimen attributed to P. dactylifera collected in Puerto Rico is described. Distribution of the new species suggests a complete separation of Panamanian and Puerto Rican proseriate fauna, confirming previous reports of restricted ranges and high endemicity of mesopsammic Platyhelminthes.

An annotated checklist is presented of the triclad fauna of the island of Sardinia, Italy, including an update of their geographic distribution and new records for several species. All three families of European freshwater planarians are represented on the island, viz. Dugesiidae (3 genera, 9 species), Planariidae (3 genera, 2 species), and Dendrocoelidae (1 genus, 3 species), inhabiting surface and subterranean waters. Besides freshwater species, over the last decades also land planarians (Geoplanidae) have been recorded.

Available evidence strongly suggests that alternative, Earth-bound explanations should be sought first for the occurrence of a single bipolar planarian flatworm returning from space travel. Double-headed worms have been amply documented as arising under experimental conditions as well as spontaneously in stock cultures of planarians.

In this paper we report a new species of Dugesia of the family Dugesiidae from Madagascar, representing the fourth species of freshwater planarian known from this global biodiversity hotspot. In some respects the new species is aberrant, when compared with its congeners, being characterized by a head with smoothly rounded auricles, a peculiar course of the oviducts, including the presence of a common posterior extension, and by the asymmetrical openings of the vasa deferentia at about halfway along the seminal vesicle. Further, it is characterized by a ventral course of the ejaculatory duct with a terminal opening, very long spermiducal vesicles and unstalked cocoons. Its diploid chromosome complement with 18 chromosomes represents an uncommon feature among fissiparous species of Dugesia.

A new species of the genus Dugesia is described from the Lake Ohrid region in the western part of the Balkan Peninsula, forming the first fully documented species description for this genus in the Ohrid area. The morphological species delimitation is supported by complementary molecular, karyological, and cytogenetic data available from the literature. Therefore, species delineation is based on a truly integrative approach. Further, a short account on the degree of freshwater planarian endemicity in the Ohrid region is provided.

Background: Schmidtea mediterranea (Platyhelminthes, Tricladida, Continenticola) is found in scattered localities on a few islands and in coastal areas of the western Mediterranean. Although S. mediterranea is the object of many regeneration studies, little is known about its evolutionary history. Its present distribution has been proposed to stem from the fragmentation and migration of the Corsica-Sardinia microplate during the formation of the western Mediterranean basin, which implies an ancient origin for the species. To test this hypothesis, we obtained a large number of samples from across its distribution area. Using known and new molecular markers and, for the first time in planarians, a molecular clock, we analysed the genetic variability and demographic parameters within the species and between its sexual and asexual populations to estimate when they diverged., Results: A total of 2 kb from three markers (COI, CYB and a nuclear intron N13) was amplified from ~200 specimens. Molecular data clustered the studied populations into three groups that correspond to the west, central and southeastern geographical locations of the current distribution of S. mediterranea. Mitochondrial genes show low haplotype and nucleotide diversity within populations but demonstrate higher values when all individuals are considered. The nuclear marker shows higher values of genetic diversity than the mitochondrial genes at the population level, but asexual populations present lower variability than the sexual ones. Neutrality tests are significant for some populations. Phylogenetic and dating analyses show the three groups to be monophyletic, with the west group being the basal group. The time when the diversification of the species occurred is between ~20 and ~4 mya, although the asexual nature of the western populations could have affected the dating analyses., Conclusions: S. mediterranea is an old species that is sparsely distributed in a harsh habitat, which is probably the consequence of the migration of the Corsica-Sardinia block. This species probably adapted to temperate climates in the middle of a changing Mediterranean climate that eventually became dry and hot. These data also suggest that in the mainland localities of Europe and Africa, sexual individuals of S. mediterranea are being replaced by asexual individuals that are either conspecific or are from other species that are better adapted to the Mediterranean climate.

Planarians of the genus Dugesia have a worldwide distribution with high species diversity in the Mediterranean area. In this area, populations with a triploid karyotype that reproduce by fissiparity are exceptionally frequent, outnumbering the sexual populations. This situation poses interesting questions, such as the age of these asexual lineages, whether they all belong to the same species or whether the triploidization event is recurrent, and what factors (climatic, geographical, historical...) explain the prevalence of these asexual forms. However, asexual populations cannot be assigned to a species due to the lack of copulatory apparatus--the main structure used in species identification. In this study, we have developed a DNA barcoding method, based on COI and ITS-1 sequences, which allows the assignment of the fissiparous forms to sexual species. At the same time, phylogenetic analysis from species of the western Mediterranean have unveiled the presence of species with highly differentiated populations alongside species with a wide distribution and almost no genetic variation. The roles of habitat instability, dispersal capacity and human activities are briefly discussed.
