- Abstract
Background: Advanced prostate cancer etiology is poorly understood. Few studies have examined associations ofanthropometric factors (e.g. early adulthood obesity) with advanced prostate cancer risk. Patients and methods: We carried out pooled analyses to examine associations between body fatness, height, andprostate cancer risk. Among 830 772 men, 51 734 incident prostate cancer cases were identified, including 4762advanced (T4/N1/M1 or prostate cancer deaths) cases, 2915 advanced restricted (same as advanced, but excludinglocalized cancers that resulted in death) cases, 9489 high-grade cases, and 3027 prostate cancer deaths. Coxproportional hazards models were used to calculate study-specific hazard ratios (HR) and 95% confidence intervals(CI); results were pooled using random effects models. Results: No statistically significant associations were observed for body mass index (BMI) in early adulthood for advanced,advanced restricted, and high-grade prostate cancer, and prostate cancer mortality. Positive associations were shown forBMI at baseline with advanced prostate cancer (HR ¼ 1.30, 95% CI ¼ 0.95e1.78) and prostate cancer mortality (HR ¼1.52, 95% CI ¼ 1.12e2.07) comparing BMI 35.0 kg/m2 with 21e22.9 kg/m2. When considering early adulthood andbaseline BMI together, a 27% higher prostate cancer mortality risk (95% CI ¼ 9% to 49%) was observed for men withBMI <25.0 kg/m2 in early adulthood and BMI 30.0 kg/m2 at baseline compared with BMI <25.0 kg/m2 in earlyadulthood and BMI <30.0 kg/m2 at baseline. Baseline waist circumference, comparing 110 cm with <90 cm, andwaist-to-hip ratio, comparing 1.00 with <0.90, were associated with significant 14%e16% increases in high-gradeprostate cancer risk and suggestive or significant 20%e39% increases in prostate cancer mortality risk. - Abstract
Background: Increased vitamin B6 catabolism related to inflammation, as measured by the PAr index (the ratio of 4-pyridoxic acid over the sum of pyridoxal and pyridoxal-5'-phosphate), has been positively associated with lung cancer risk in two prospective European studies. However, the extent to which this association translates to more diverse populations is not known. Materials and methods: For this study, we included 5323 incident lung cancer cases and 5323 controls individually matched by age, sex, and smoking status within each of 20 prospective cohorts from the Lung Cancer Cohort Consortium. Cohort-specific odds ratios (ORs) and 95% confidence intervals (CIs) for the association between PAr and lung cancer risk were calculated using conditional logistic regression and pooled using random-effects models. Results: PAr was positively associated with lung cancer risk in a dose-response fashion. Comparing the fourth versus first quartiles of PAr resulted in an OR of 1.38 (95% CI: 1.19-1.59) for overall lung cancer risk. The association between PAr and lung cancer risk was most prominent in former smokers (OR: 1.69, 95% CI: 1.36-2.10), men (OR: 1.60, 95% CI: 1.28-2.00), and for cancers diagnosed within 3years of blood draw (OR: 1.73, 95% CI: 1.34-2.23). Conclusion: Based on pre-diagnostic data from 20 cohorts across 4 continents, this study confirms that increased vitamin B6 catabolism related to inflammation and immune activation is associated with a higher risk of developing lung cancer. Moreover, PAr may be a pre-diagnostic marker of lung cancer rather than a causal factor.

- Abstract
Quantifying the genetic correlation between cancers can provide important insights into the mechanisms driving cancer etiology. Using genome-wide association study summary statistics across six cancer types based on a total of 296,215 cases and 301,319 controls of European ancestry, here we estimate the pair-wise genetic correlations between breast, colorectal, head/neck, lung, ovary and prostate cancer, and between cancers and 38 other diseases. We observed statistically significant genetic correlations between lung and head/neck cancer (rg = 0.57, p = 4.6 × 10−8), breast and ovarian cancer (rg = 0.24, p = 7 × 10−5), breast and lung cancer (rg = 0.18, p =1.5 × 10−6) and breast and colorectal cancer (rg = 0.15, p = 1.1 × 10−4). We also found that multiple cancers are genetically correlated with non-cancer traits including smoking, psychiatric diseases and metabolic characteristics. Functional enrichment analysis revealed a significant excess contribution of conserved and regulatory regions to cancer heritability. Our comprehensive analysis of cross-cancer heritability suggests that solid tumors arising across tissues share in part a common germline genetic basis.

- Abstract
Background: There is observational evidence suggesting that high vitamin D concentrations may protect against lung cancer. To investigate this hypothesis in detail, we measured circulating vitamin D concentrations in prediagnostic blood from 20 cohorts participating in the Lung Cancer Cohort Consortium (LC3). Patients and methods: The study included 5313 lung cancer cases and 5313 controls. Blood samples for the cases were collected, on average, 5 years before lung cancer diagnosis. Controls were individually matched to the cases by cohort, sex, age, race/ethnicity, date of blood collection, and smoking status in five categories. Liquid chromatography coupled with tandem mass spectrometry was used to separately analyze 25-hydroxyvitamin D2 [25(OH)D2] and 25-hydroxyvitamin D3 [25(OH)D3] and their concentrations were combined to give an overall measure of 25(OH)D. We used conditional logistic regression to calculate odds ratios (ORs) and 95% confidence intervals (CIs) for 25(OH)D as both continuous and categorical variables. Results: Overall, no apparent association between 25(OH)D and risk of lung cancer was observed (multivariable adjusted OR for a doubling in concentration: 0.98, 95% CI: 0.91, 1.06). Similarly, we found no clear evidence of interaction by cohort, sex, age, smoking status, or histology. Conclusion: This study did not support an association between vitamin D concentrations and lung cancer risk.

14. Interseismic coupling on the main Himalayan thrust

- Abstract
We determine the slip rate and pattern of interseismic coupling on the Main Himalayan Thrust along the entire Himalayan arc based on a compilation of geodetic, interferometric synthetic aperture radar, and microseismicity data. We show that convergence is perpendicular to the arc and increases eastwards from 13.3 ± 1.7 mm/yr to 21.2 ± 2.0 mm/yr. These rates are comparable to geological and geomorphic estimates, indicating an essentially elastic geodetic surface strain. The interseismic uplift rate predicted from the coupling model closely mimics the topography, suggesting that a small percentage of the interseismic strain is permanent. We find that the fault is fully locked along its complete length over about 100 km width. We don't find any resolvable aseismic barrier that could affect the seismic segmentation of the arc and limit the along-strike propagation of seismic ruptures. The moment deficit builds up at a rate of 15.1 ± 1 × 10^(19) N m/yr for the entire length of the Himalaya.

The presence of multiple (5-100) colorectal adenomas suggests an inherited predisposition, but the genetic aetiology of this phenotype is undetermined if patients test negative for Mendelian polyposis syndromes such as familial adenomatous polyposis (FAP) and MUTYH-associated polyposis (MAP). We investigated whether 18 common colorectal cancer (CRC) predisposition single-nucleotide polymorphisms (SNPs) could help to explain some cases with multiple adenomas who phenocopied FAP or MAP, but had no pathogenic APC or MUTYH variant. No multiple adenoma case had an outlying number of CRC SNP risk alleles, but multiple adenoma patients did have a significantly higher number of risk alleles than population controls (P = 5.7 × 10-7). The association was stronger in those with ≥ 10 adenomas. The CRC SNPs accounted for 4.3% of the variation in multiple adenoma risk, with three SNPs (rs6983267, rs10795668, rs3802842) explaining 3.0% of the variation. In FAP patients, the CRC risk score did not differ significantly from the controls, as we expected given the overwhelming effect of pathogenic germline APC variants on the phenotype of these cases. More unexpectedly, we found no evidence that the CRC SNPs act as modifier genes for the number of colorectal adenomas in FAP patients. In conclusion, common colorectal tumour risk alleles contribute to the development of multiple adenomas in patients without pathogenic germline APC or MUTYH variants. This phenotype may have 'polygenic' or monogenic origins. The risk of CRC in relatives of multiple adenoma cases is probably much lower for cases with polygenic disease, and this should be taken into account when counselling such patients. © 2015 Macmillan Publishers Limited All rights reserved.

- Abstract
The Himalayan mountain range has been the locus of some of the largest continental earthquakes, including the 2015 magnitude 7.8 Gorkha earthquake. Competing hypotheses suggest that Himalayan topography is sustained and plate convergence is accommodated either predominantly on the main plate boundary fault, or more broadly across multiple smaller thrust faults. Here we use geodetic measurements of surface displacement to show that the Gorkha earthquake ruptured the Main Himalayan Thrust fault. The earthquake generated about 1 m of uplift in the Kathmandu Basin, yet caused the high Himalaya farther north to subside by about 0.6 m. We use the geodetic data, combined with geologic, geomorphological and geophysical analyses, to constrain the geometry of the Main Himalayan Thrust in the Kathmandu area. Structural analyses together with interseismic and coseismic displacements are best explained by a steep, shallow thrust fault flattening at depth between 5 and 15 km and connecting to a mid-crustal, steeper thrust. We suggest that present-day convergence across the Himalaya is mostly accommodated by this fault—no significant motion on smaller thrust faults is required. Furthermore, given that the Gorkha earthquake caused the high Himalayan mountains to subside and that our fault geometry explains measured interseismic displacements, we propose that growth of Himalayan topography may largely occur during the ongoing post-seismic phase.

34. Millenary Mw> 9.0 earthquakes required by geodetic strain in the Himalaya

- Abstract
The Himalayan arc produced the largest known continental earthquake, the Mw≈ 8.7 Assam earthquake of 1950, but how frequently and where else in the Himalaya such large‐magnitude earthquakes occur is not known. Paleoseismic evidence for coseismic ruptures at the front of the Himalaya with 15 to 30 m of slip suggests even larger events in medieval times, but this inference is debated. Here we estimate the frequency and magnitude of the largest earthquake in the Himalaya needed so that the moment released by seismicity balances the deficit of moment derived from measurements of geodetic strain. Assuming one third of the moment buildup is released aseismically and the earthquakes roughly follow a Gutenberg‐Richter distribution, we find that Mw> 9.0 events are needed with a confidence level of at least 60% and must return approximately once per 800 years on average. Both endogenous factors (genomic variations) and exogenous factors (environmental exposures, lifestyle) impact the balance of reactive oxygen species (ROS). Variants of the ND3 (rs2853826; G10398A) gene of the mitochondrial genome, manganese superoxide dismutase ( MnSOD; rs4880 Val16Ala) and glutathione peroxidase ( GPX-1; rs1050450 Pro198Leu), are purported to have functional effects on regulation of ROS balance. In this study, we examined associations of breast and prostate cancer risks and survival with these variants, and interactions between rs4880-rs1050450, and alcohol consumption-rs2853826. Nested case-control studies were conducted in the Breast and Prostate Cancer Cohort Consortium (BPC3), consisting of nine cohorts. The analyses included over 10726 post-menopausal breast and 7532 prostate cancer cases with matched controls. Logistic regression models were used to evaluate associations with risk, and proportional hazard models were used for survival outcomes. We did not observe significant interactions between polymorphisms in MnSOD and GPX-1, or between mitochondrial polymorphisms and alcohol intake and risk of either breast (p-interaction of 0.34 and 0.98, respectively) or prostate cancer (p-interaction of 0.49 and 0.50, respectively). We observed a weak inverse association between prostate cancer risk and GPX-1 Leu198Leu carriers (OR 0.87, 95% CI 0.79-0.97, p = 0.01). Overall survival among women with breast cancer was inversely associated with G10398 carriers who consumed alcohol (HR 0.66 95% CI 0.49-0.88). Given the high power in our study, it is unlikely that interactions tested have more than moderate effects on breast or prostate cancer risk. 40. Maternal Fumonisin Exposure as a Risk Factor for Neural Tube Defects.

- Abstract
Fumonisins are mycotoxins produced by the fungus F. verticillioides, a common contaminant of maize (corn) worldwide. Maternal consumption of fumonisin Bl-contaminated maize during early pregnancy has recently been associated with increased risk for neural tube defects (NTDs) in human populations that rely heavily on maize as a dietary staple. Experimental administration: of purified fumonisin to mice early in gestation also results in an increased incidence of NTDs in exposed offspring. Fumonisin inhibits the enzyme ceramide synthase in de novo sphingolipid biosynthesis, resulting in an elevation of free sphingoid bases and depletion of downstream glycosphingolipids. Increased sphingoid base metabolites (i.e., sphinganine-l-phosphate) may perturb signaling cascades involved in embryonic morphogenesis by functioning as ligands for sphingosine-1-P (S1P) receptors, a family of G-proteincoupled receptors that regulate key biological processes such as cell survival/proliferation, differentiation and migration. Fumonisininduced depletion of glycosphingolipids impairs expression and function of the GPI-anchored folate receptor (Folr1), which may also contribute to adverse pregnancy outcomes. NTDs appear to be multifactorial in origin, involving complex gene-nutrientenvironment interactions. Vitamin supplements containing folic acid have been shown to reduce the occurrence of NTDs, and may help protect the developing fetus from environmental teratogens. Fumonisins appear to be an environmental risk factor for birth defects, although other aspects of maternal nutrition and genetics play interactive roles in determining pregnancy outcome. Minimizing exposures to mycotoxins through enhanced agricultural practices, identifying biomarkers of exposure, characterizing mechanisms of toxicity, and improving maternal nutrition are all important strategies for reducing the NTD burden in susceptible human populations. The folate receptor (FR) binds physiologic folates with nmol/L affinities and is expected to play an important role in transporting serum folates into cells that express this receptor. Although it has been shown that FR expression increases when extracellular levels of folate are low, whether this receptor is regulated in response to altered cellular requirements for folates or by intracellular levels of this vitamin has not been investigated. In this study, FR levels, FR function and cellular folate levels were measured in cells with different growth rates to investigate FR regulation of this receptor under conditions in which cellular requirements for folate are altered. These experiments used cells that endogenously express FR (JAR, Caco-2 and MA-104) and cells stably transfected with this receptor (FRGPI-16 and FRTM-8). FR function decreased as cellular growth slowed in four of the five cell lines examined. Although cellular folate levels also decreased as cells reached confluence, the total amount of cellular folate in the culture remained constant, suggesting the depleted cellular folate was because of the cell partitioning its pool throughout cell division, not because of decreased FR function. Conversely, there was an inverse association with FR levels and cell growth (r = -0.998 to -0.999, P < 0.05) in cells endogenously expressing FR, with a significant increase in the percentage of total FR located in an intracellular compartment as growth slowed. These results suggest FR function is regulated by cellular requirements for folates but not in response to changing FR levels or cellular levels of this vitamin. [ABSTRACT FROM AUTHOR]

43. Clostridium septicum alpha toxin uses glycosylphosphatidylinositol-anchored protein receptors.

- Abstract
The alpha toxin produced by Clostridium septicum is a channel-forming protein that is an important contributor to the virulence of the organism. Chinese hamster ovary (CHO) cells are sensitive to low concentrations of the toxin, indicating that they contain toxin receptors. Using retroviral mutagenesis, a mutant CHO line (BAG15) was generated that is resistant to alpha toxin. FACS analysis showed that the mutant cells have lost the ability to bind the toxin, indicating that they lack an alpha toxin receptor. The mutant cells are also resistant to aerolysin, a channel-forming protein secreted by Aeromonas spp., which is structurally and functionally related to alpha toxin and which is known to bind to glycosylphosphatidylinositol (GPI)-anchored proteins, such as Thy-1. We obtained evidence that the BAG15 cells lack N-acetylglucosaminyl-phosphatidylinositol deacetylase-L, needed for the second step in GPI anchor biosynthesis. Several lymphocyte cell lines lacking GPI-anchored proteins were also shown to be less sensitive to alpha toxin. On the other hand, the sensitivity of CHO cells to alpha toxin was increased when the cells were transfected with the GPI-anchored folate receptor. We conclude that alpha toxin, like aerolysin, binds to GPI-anchored protein receptors. Evidence is also presented that the two toxins bind to different subsets of GPI-anchored proteins.

The folate receptor, like many glycosylphosphatidylinositol-anchored proteins, is found associated with membrane domains that are insoluble in Triton X-100 at low temperature and that are enriched in cholesterol and sphingolipids. Depletion of cellular cholesterol has been shown to inhibit vitamin uptake by this receptor (Chang, W. -J., Rothberg, K. G., Kamen, B. A., and Anderson, R. G. W. (1993) J. Cell Biol. 118, 63-69), suggesting that these domains regulate this process. In this study, the importance of sphingolipids for folate receptor function was investigated in Caco-2 cells using fumonisin B1, a mycotoxin that inhibits the biosynthesis of these lipids. The folate receptor-mediated transport of 5-methyltetrahydrofolate was almost completely blocked in cells in which sphingolipids had been reduced by approximately 40%. This inhibition was dependent on the concentration and duration of the treatment with the mycotoxin and was mediated by the sphingolipid decrease. Neither receptor-mediated nor facilitative transport was inhibited by fumonisin B1 treatment, indicating that the effect of sphingolipid depletion was specific for folate receptor-mediated vitamin uptake. A concurrent loss in the total amount of folate binding capacity in the cells was seen as sphingolipids were depleted, suggesting a causal relationship between folate receptor number and vitamin uptake. These findings suggest that dietary exposure to fumonisin B1 could adversely affect folate uptake and potentially compromise cellular processes dependent on this vitamin. Furthermore, because folate deficiency causes neural tube defects, some birth defects unexplained by other known risk factors may be caused by exposure to fumonisin B1.

- Abstract
Many eukaryotic cell surface proteins are bound to the cell membrane by a glycosylphosphatidylinositol (GPI) anchor. Several genes involved in GPI anchor biosynthesis have been cloned using complementation of mutant mammalian cell lines and yeasts that are defective in its biosynthesis pathway. However, the gene involved in the second step of this pathway, in which N-acetylglucosaminyl-phosphatidylinositol (GlcNAc-PI) is N-deacetylated to form glucosaminyl (GlcN)-PI, has not been cloned. In this study, we established a GPI anchor-deficient mutant of Chinese hamster ovary (CHO) cells defective in the second step. Complementation analysis with the known GPI anchor mutant cells demonstrated that it belonged to the same complementation group as the CHO cell mutant G9PLAP.85. Using the new mutant, we cloned a rat gene termed PIG-L (for phosphatidylinositol glycan class L) that is involved in this step. PIG-L encodes a 252-amino acid, endoplasmic reticulum membrane protein, most of which is in the cytoplasmic side. This orientation of PIG-L protein is consistent with the notion that the second step of GPI anchor biosynthesis occurs on the cytoplasmic side of the endoplasmic reticulum.

The effects of long-chain (sphingoid) bases on the phorbol ester-dependent differentiation of HL-60 cells were investigated since these molecules are potent inhibitors of protein kinase C (Hannun, Y. A., Loomis, C. R., Merrill, A. H., Jr., and Bell, R. M. (1986) J. Biol. Chem. 261, 12604-12609). After 24 h, low concentrations of sphinganine (1-5 microM blocked both cell adherence and the inhibition of growth in response to phorbol 12-myristate 13-acetate, as measured by cell number and acid phosphatase activity. Sphinganine and sphingosine decreased adherence by 50% at 1-3 microM; other long-chain bases were effective in parallel to their inhibition of protein kinase C. Sphinganine decreased the binding of [3H]phorbol dibutyrate by the phorbol receptor of HL-60 cells, protein kinase C, and inhibited the response of HL-60 cells to dioctanoylglycerol, a cell permeable activator of this enzyme. Long-chain base uptake by HL-60 cells was demonstrated with [3-3H]sphinganine and within 1-3 days much had been converted to ceramides. By day 3, most of the cells had recovered the ability to adhere and exhibited macrophage characteristics, whereas cells in suspension did not differentiate. The level of free sphinganine in HL-60 cells was determined to be 12.3 +/- 1.2 pmol/10(6) cells. These results establish that sphingoid bases inhibit protein kinase C in HL-60 cells and may function physiologically as negative effectors of this enzyme.

47. Oxygen dependence of adrenal cortex cholesterol side chain cleavage. Implications in the rate-limiting steps in steroidogenesis.

The oxygen dependence of cholesterol side chain cleavage to form pregnenolone was measured, using both purified phospholipid vesicle-reconstituted cytochrome P-450scc and rat adrenal mitochondria. At saturating cholesterol and nonlimiting electron supply (via NADPH-adrenodoxin reductase and adrenodoxin) the Km(O2) is low (4 microM). Limitations in the availability of both cholesterol and reductant caused elevations in the observed Km(O2). Pregnenolone synthesis was measured in mitochondria from variously pretreated rats, using a phospholipid-cholesterol dispersion as the source of exogenous substrate. In mitochondria obtained from ether-stressed rats (which elevates adrenocorticotropic hormone) two phases of malate-supported pregnenolone production are seen, a rapid (first 2 min) highly oxygen-dependent phase (Km = 150 microM) and a slow (2-10 min) relatively oxygen-independent phase (Km less than 10 microM). Comparison of side chain cleavage rates with mitochondrial 11 beta-hydroxylation rates at various oxygen concentrations suggests that the rapid phase is limited by the availability of reducing equivalents. In cycloheximide-pretreated ether-stressed rats, only a linear slow rate of pregnenolone production was seen (about 25% of the rate of the slow phase in the ether-stressed group), while in mitoplasts from both groups only a linear rapid rate was seen. Data are consistent with the proposal (Privalle, C. T., Crivello, J. F., and Jefcoate, C. R. (1983) Proc. Natl. Acad. Sci. U. S. A. 80, 702-706) that the adrenocorticotropic hormone-regulated cycloheximide-inhibitable rate of cholesterol side chain cleavage is limited by the rate of cholesterol transfer from outer to inner mitochondrial membranes.
