Natural history collections are essential to a wide variety of studies in biology because they maintain large collections of specimens and associated data, including genetic material (e.g., tissues) for DNA sequence data, yet they are currently under-funded and collection staff have high workloads. With the advent of aggregate databases and advances in sequencing technologies, there is an increased demand on collection staff for access to tissue samples and associated data. Scientists are rapidly developing large DNA barcode libraries, DNA sequences of specific genes for species across the tree of life, in order to document and conserve biodiversity. In doing so, mistakes are made. For instance, inconsistent taxonomic information is commonly taken from different lending institutions and deposited in data repositories, such as the Barcode of Life Database (BOLD) and GenBank, despite explicit disclaimers regarding the need for taxonomic verification by the lending institutions. Such errors can have profound effects on subsequent research based on these mis-labelled sequences in data repositories. Here, we present the production of a large DNA barcode library of reptiles from the National Museum of Natural History tissue holdings. The library contains 2,758 sequences (2,205 COI and 553 16S) from 2260 specimens (four crocodilians, 37 turtles, and 2,219 lizards, including snakes), representing 583 named species, from 52 countries. In generating this library, we noticed several common mistakes made by scientists depositing DNA barcode data in public repositories (e.g., BOLD and GenBank). Our goal is to raise awareness of these concerns and offer advice to avoid such mistakes in the future to maintain accurate DNA barcode libraries to properly document Earth’s biodiversity.

Confusion regarding the identification of Placobdella hollensis (Whitman, 1892) (Hirudinida: Glossiphoniidae) has led to an unclear understanding of the distribution of the species. Two specimens of P. hollensis were collected from Merchants Millpond State Park, Gates County, North Carolina, U.S.A., representing a new geographic distribution record. Specimens were confirmed as P. hollensis by morphological and molecular study. Specimens of P. hollensis from North Carolina, had accessory eyes, 2 thin paramedial dark lines, and 3 pairs of pre-anal papillae. Molecular comparison of cytochrome c oxidase subunit I sequence data revealed a 99.0 to 99.7% similarity to specimens of P. hollensis collected from its type locality (Barnstable County, Massachusetts, U.S.A.). From confirmed specimens of P. hollensis, this report supports the assertion that P. hollensis has an Atlantic coastal distribution.

Confusion exists in the literature concerning the collecting event of the teiid lizard Tiaporus fuliginosus. We investigated the literature and documents stored at the Smithsonian Institution Archives involving the collector of those specimens in an effort to resolve that confusion. We conclude that the type series was collected on the Swan Islands of Honduras by Charles H. Townsend during 1887. We also provide a redescription of that nominal form and show that it is a valid species that should be called Ameiva fuliginosa. We also examined the type series of A. panchlora from Old Providence, Colombia and confirm that its 1950 placement as a junior synonym of A. fuliginosa is correct.

The original description of Crotalus scutulatus (Chordata: Reptilia: Squamata: Viperidae) was published in 1861 by Robert Kennicott, who did not identify a type specimen or a type locality. We review the history of specimens purported to be the type(s) and various designations of type locality. We provide evidence that ANSP 7069 (formerly one of two specimens of USNM 5027) is the holotype and that the appropriate type locality is Fort Buchanan, near present-day Sonoita, in Santa Cruz County, Arizona.

Leptophis santamartensis, known only from Sierra Nevada de Santa Marta, Colombia, is one of the more poorly known species of the genus Leptophis. The characters used for its diagnosis largely overlap with those of other Leptophis, mainly with Leptophis ahaetulla occidentalis, the only other Leptophis known to occur in the Sierra Nevada de Santa Marta. A detailed comparison of L. a. occidentalis with the two known specimens of L. santamartensis leads to the conclusion that the latter should be relegated to the synonymy of the former.

Short-tailed pythons, Python curtus species group, occur predominantly in the Malayan Peninsula, Sumatra, and Borneo. The discovery of an adult female in Mon State, Myanmar, led to a review of the distribution of all group members (spot-mapping of all localities of confirmed occurrence) and an examination of morphological variation in P. brongersmai. The resulting maps demonstrate a limited occurrence of these pythons within peninsular Malaya, Sumatra, and Borneo with broad absences in these regions. Our small samples limit the recognition of regional differentiation in the morphology of P. brongersmai populations; however, the presence of unique traits in the Myanmar python and its strong allopatry indicate that it is a unique genetic lineage, and it is described as Python kyaiktiyo new species.

We demonstrate that USNM 2040 and not UMMZ 3781 (originally part of lot USNM 4500) was most likely the holotype of Tantilla gracilis. The type specimens of Tantilla nigriceps have been lost or destroyed. It is not possible to determine from the original description of Tantilla nigriceps if this name represents what is currently known as T. nigriceps or T. hobartsmithi. In order to attribute the name T. nigriceps firmly to the species as currently recognized, we designate a neotype.

Turtles in Lake Blackshear, Crisp County, Georgia (USA) were evaluated for shell disease during intensive trapping efforts on 8 and 9 May 1990. The disease was most prevalent in Pseudemys concinna (74%) and Trachemys scripta (35%). The degree of necrosis on the carapace was significantly positively correlated with the degree of necrosis on the plastron in T. scripta (rs = 0.50), but not in P. concinna (rs = 0.06). Female T. scripta with lesions were significantly larger than females without lesions. Lesions were not detected on six other species of turtles. Some areas contained multinucleate osteoclasts that were destroying bone. No tumors were detected in soft tissue samples.

Lipids and proteins in the Rathke's gland secretions of the North American mud turtle (Kinosternon subrubrum, Kinosternidae) were analyzed by gas chromatography-mass spectrometry (GC-MS) and SDS-polyacrylamide gel electrophoresis (SDS-PAGE), respectively. Analysis by GC-MS indicates 2,3-dihydroxypropanal and C3–C24 free or esterified fatty acids. Analysis by SDS-PAGE indicates a major protein component with an approximate molecular mass of 60 kDa and minor components ranging from ca. 23 to 34 kDa. The major component of K. subrubrum glandular secretions exhibits a mobility that matches that of the Kemp's ridley sea turtle (Lepidochelys kempi, Cheloniidae), suggesting that these proteins are evolutionarily conserved.

The Chinese coin turtle Chinemys reevesii is widely distributed in eastern Asia from Japan to southern China. Within its range, three taxa have been described which are similar to C. reevesii except for minor differences (Geoclemys grangeri, Geoclemys paracaretta, and Damonia unicolor). Although these variants have been synonymized by some authors, no quantitative studies have been conducted to support or reject their validity. The present study examined 31 characters of specimens throughout the range. Analysis indicates that all variants are within the diagnostic extremes observed in this species. Low interpopulational variation in C. reevesii is due in part to several millennia of human disturbance.
