Keynote paper presented at the International Leucaena Conference, 1‒3 November 2018, Brisbane, Queensland, Australia.Leucaena has been fed to cattle by the Balinese community in Sumbawa and West Sumbawa districts on Sumbawa Island since the 1980s. However, prior to 2011, this practice was not adopted by the local Sumbawanese farmers. Since then, a model leucaena-based cattle fattening system was developed in Sumbawa and West Sumbawa districts in a collaborative research project between the Assessment Institute for Agricultural Technology (BPTP), University of Mataram and The University of Queensland (UQ) funded by the Australian Centre for International Agricultural Research (ACIAR), followed by a scaling-out project involving collaboration between the University of Mataram and CSIRO (Applied Research and Innovation Systems in Agriculture - ARISA project) funded by DFAT (Department of Foreign Affairs and Trade) promoting public-private partnerships. Further promotion of leucaena-based fattening systems occurred in Dompu, Sumbawa, through a project with the University of Mataram and Massey University funded by the New Zealand Ministry of Foreign Affairs and Trade (MFAT). By the end of October 2018, more than 2,500 farmers on Sumbawa Island were practicing leucaena-based cattle fattening. The main drivers of adoption of cattle fattening with leucaena were: (1) The high growth rates achieved (0.4–0.6 kg/d for bulls fed 100% leucaena and 0.66 kg/d when maize grain was added to the leucaena basal diet) compared with 0.16 kg/d for the traditional system, combined with high profitability; (2) the needs of farmers being met in terms of relevance and cultural appropriateness; (3) field extension staff being well trained and mentored, and respected by the farmers; (4) the local government being highly supportive of leucaena-based cattle fattening; and (5) additional benefits being increased dressing percentage and high carcass quality. The rapid increase in the use of leucaena for cattle fattening in eastern Indonesia is expected to have a significant positive impact on household incomes as well as on regional economic growth.

The relationship between ewe body condition score (BCS) and liveweight (LW) has been exploited previously to predict the former from LW, LW-change, and previous BCS records. It was hypothesized that if fleece weight and conceptus-free liveweight and LW-change, and in addition, height at withers were used, the accuracy of current approaches to predicting BCS would be enhanced. Ewes born in 2017 (n = 429) were followed from 8 mo to approximately 42 mo of age in New Zealand. Individual ewe data were collected on LW and BCS at different stages of the annual production cycle (i.e., prebreeding, at pregnancy diagnosis, prelambing, and weaning). Additionally, individual lambing dates, ewe fleece weight, and height at withers data were collected. Linear regression models were fitted to predict current BCS at each ewe age and stage of the annual production cycle using two LW-based models, namely, unadjusted for conceptus weight and fleece weight (LW alone1) and adjusted (LW alone2) models. Furthermore, another two models based on a combination of LW, LW-change, previous BCS, and height at withers (combined models), namely, unadjusted (combined1) and adjusted for conceptus and fleece weight (combined2), were fitted. Combined models gave more accurate (with lower root mean square error: RMSE) BCS predictions than models based on LW records alone. However, applying adjusted models did not improve BCS prediction accuracy (or reduce RMSE) or improve model goodness of fit (R2) (P > 0.05). Furthermore, in all models, both LW-alone and combined models, a great proportion of variability in BCS, could not be accounted for (0.25 ≥ R2 ≥ 0.83) and there was substantial prediction error (0.33 BCS ≥ RMSE ≥ 0.49 BCS) across age groups and stages of the annual production cycle and over time (years). Therefore, using additional ewe data which allowed for the correction of LW for fleece and conceptus weight and using height at withers as an additional predictor did not improve model accuracy. In fact, the findings suggest that adjusting LW data for conceptus and fleece weight offer no additional value to the BCS prediction models based on LW. Therefore, additional research to identify alternative methodologies to account for individual animal variability is still needed.

Currently, 30–43% of New Zealand sheep farmers breed their ewe lambs, but few retain the offspring as replacements for their flock. No difference in lamb production as a yearling among singletons and twins born to ewe lambs and twins born to mature ewes has been reported, provided the ewe lambs had reached the 60–65% of their likely mature weight prior to breeding at seven to eight months of age. The aim of this experiment was to determine the lamb production from singletons and twins born to ewe lambs and twins born to mature ewes during their first two years of lambing. The experiment included 8-month-old ewes born as twins to mature ewes (M2, n = 135), singletons born to ewe lambs (L1, n = 135), and twins born to ewe lambs (L2, n = 88), bred during the same period to the same rams, over two years. The efficiency of lamb production (total litter weight at weaning divided by the pre-breeding weight of the ewe, for all ewes presented for breeding) after two years of production was not significantly different (p &gt, 0.05) among the groups (0.40 ± 0.02, 0.39 ± 0.02, and 0.39 ± 0.03, for M2, L1, and L2, respectively).

An on-farm study investigated the herbage quality and sward characteristics of plantain-clover mixes and resulting lamb growth rates during late summer and autumn. Three farms were chosen, each with a minimum of 10 ha of plantain-clover mix (Plantago lanceolata ‘Ceres Tonic’, Trifolium pratense, Trifolium repens) used for lamb finishing. Each farm managed the plantain-clover mix area using their routine management without advice or comment from research staff. Throughout autumn, at approximately monthly intervals on the three farms, the pre- and post-grazing herbage masses, botanical composition, herbage quality, plantain dry matter content, secondary chemical composition and lamb growth rates were monitored. A low percentage of clover in the sward, a high percentage of dead stem material and a low crude protein concentration in plantain during dry periods, were identified as the most likely causes of low lamb liveweight gains during autumn. Grazing management during spring that maintains control of plantain stems and encourages the presence of clover, appears to be a key management technique for ensuring high lamb liveweight gains on plantain-clover mixes in late summer and autumn.

Lamb growth to weaning has important implications for the profitability of farms producing lambs for meat. Alternative forage crops such as chicory, plantain, red clover and white clover have the potential to increase lamb growth rates to weaning percentage sold to slaughter at weaning. A study was conducted over two years to examine the growth of single-born lambs offered either pasture or herb-mix (chicory, plantain, white clover and red clover) from birth to weaning. Each year, 80 single-bearing ewes were allocated to either a pasture or herb-clover mix. In 2014, ewes and lambs that were offered the herb-clover mix had greater live weights at weaning than those grazed on ryegrass pasture. In 2015, however, no differences in lamb weaning weight was observed. In both years of the study, lambs reared on the herb clover mix had growth rates in excess of 400 g/day, from birth to weaning.

Background There is increasing demand for colonoscopy quality measures for procedures performed in ambulatory surgery centers. Benchmarks such as adenoma detection rate (ADR) are traditionally reported as static, one-dimensional point estimates at a provider or practice level. Objective To evaluate 6-year variability of ADRs for 370 gastroenterologists from across the nation. Design Observational cross-sectional analysis. Setting Collaborative quality metrics database from 2007 to 2012. Patients Patients who underwent colonoscopies in ambulatory surgery centers. Interventions Colonoscopy. Main Outcome Measurements The number of colonoscopies with an adenomatous polyp divided by the total number of colonoscopies (ADR-T), inclusive of indication and patient's sex. Results Data from 368,157 colonoscopies were included for analysis from 11 practices. Three practice sites (5, 8, and 10) were significantly above and 2 sites (3, 7) were significantly below mean ADR-T, with a 95% confidence interval (CI). High-performing sites had 9.0% higher ADR-T than sites belonging to the lowest quartile ( P P = .001) and site 3 ( P = .021), which were average and low performers, respectively. Limitations Summary-level data, which does not allow cross-reference of variables at an individual level. Conclusion We found performance disparities among practice sites remaining relatively consistent over a 6-year period. The ability of certain sites to sustain their high-performance over 6 years suggests that further research is needed to identify key organizational processes and physician incentives that improve the quality of colonoscopy.

Objective The present study aimed to determine the impact of ewe body condition score (BCS) (over a range of 2.0 to 3.0) and nutritional treatments (consisting of differing herbage masses) during very late pregnancy and lactation and their potential interaction on the performance of twin-bearing ewes and their lambs to weaning. Methods On day 142 of pregnancy, twin-bearing ewes with a BCS of 2.0, 2.5, or 3.0 were allocated to a “Moderate’ or ‘Unrestricted’ nutritional treatment until day 95 of lactation (weaning). The nutritional treatments aimed to achieve average herbage masses of 1,200 to 1,300 kg dry matter (DM)/ha (Moderate) and 1,500 to 1,800 kg DM/ha (Unrestricted). Results There were no three-way interactions between ewe BCS group, nutritional treatment and time for any ewe or lamb parameter. The nutritional treatments had no effect (p>0.05) on lamb birth or weaning weight. Lambs born to Moderate ewes had greater survival and total litter weight at weaning (p1,200 kg DM/ha during very late pregnancy and lactation.

The world wide growth of ecotourism and the need for management of this growth is well documented. Ramsey Island in Pembrokeshire, UK, is examined here in the context of its population of Atlantic Grey Seals. The growth of ecotour boat activity around the island has lead to the development of a voluntary code of conduct for relevant stakeholders, defining limits to their behaviour around key species at the site, including grey seals. Semi-quantitative protocols were developed for recording disturbance and boat behaviour. Data are presented of seal disturbance behaviour at pupping beaches in response to numbers, distance and speed of boats. It is shown that there is a significant correlation between the intensity of disturbance as measured by a disturbing stimulus index (DSI) and disturbance level. Disturbance is measurable at levels of disturbing stimulus that currently would not breach the code recommendations. In eukaryotic cells, the ubiquity of the signaling system of transcription factor nuclear factor-kappa B (NF-κB)/I-kappa B (I-κB) is undisputed. Numerous studies have reported that the NF-κB/I-κB complex plays a pivotal role in regulating gene expression controlling cell differentiation, cell proliferation, inflammation, oncogenesis, and apoptosis. Here we show that NF-κB/I-κB families exist in human platelets, natural anuclear corpuscles derived from megakaryocytes. Moreover, the I-κB kinase (IKK) is present and may phosphorylate I-κB during platelet activation. Coupled with intracellular calcium flux, this leads to I-κB dissociation from the NF-κB/I-κB complex and proteolysis. The NF-κB/I-κB proteins may have function independent of gene regulation in platelets.

Responding to demands for transformed farming practices requires new forms of knowledge. Given their scale and complexity, agricultural problems can no longer be solved by linear transfers in which technology developed by specialists passes to farmers by way of extension intermediaries. Recent research on alternative approaches has focused on the innovation systems formed by interactions between heterogeneous actors. Rather than linear transfer, systems theory highlights network facilitation as a specialized function. This paper contributes to our understanding of such facilitation by investigating the networks in which farmers discuss science. We report findings based on the study of a pastoral farming experiment collaboratively undertaken by a group of 17 farmers and five scientists. Analysis of prior contact and alter sharing between the group's members indicates strongly tied and decentralized networks. Farmer knowledge exchanges about the experiment have been investigated using a mix of quantitative and qualitative methods. Network surveys identified who the farmers contacted for knowledge before the study began and who they had talked to about the experiment by 18 months later. Open-ended interviews collected farmer statements about their most valuable contacts and these statements have been thematically analysed. The network analysis shows that farmers talked about the experiment with 192 people, most of whom were fellow farmers. Farmers with densely tied and occupationally homogeneous contacts grew their networks more than did farmers with contacts that are loosely tied and diverse. Thematic analysis reveals three general principles: farmers value knowledge delivered by persons rather than roles, privilege farming experience, and develop knowledge with empiricist rather than rationalist techniques. Taken together, these findings suggest that farmers deliberate about science in intensive and durable networks that have significant implications for theorizing agricultural innovation. The paper thus concludes by considering the findings' significance for current efforts to rethink agricultural extension.

Compared to the labour-intensive mincing method, autoclaving (pressurised steam cooking) is a fast way of preparing samples for chemical analysis. In addition, autoclaving dissolves fish scales and so enables more homogenous samples to be prepared. Initial method development work has shown that autoclaving at 150 kPa for 4 h is suitable for preparing samples for proximate and fatty acid analyses. However, the effect of autoclaving period and pressure on amino acid recovery has not been measured. A comparative study was made between autoclaving and mincing. The proximate chemical, fatty acid and amino acid composition of silver perch (Bidyanus bidyanus) was measured. The control samples (n = 3) were minced but not autoclaved. Each autoclaved sample (n = 36) was subjected to one of four autoclave periods (1, 2, 3 or 4 h) and one of three pressures (103, 155 or 207 kPa). Compared with mincing, autoclaving decreased nitrogen, increased energy and fat but had a variable effect on fatty acid recovery. Compared with mincing, autoclaving either had no effect on or increased the recovery of all amino acids, except cystine which was marginally reduced. Thus autoclaving appears to offer a superior method for preparing fish samples for amino acid analysis compared with mincing alone. Increasing autoclave pressure from 103 to 207 kPa had no measured effect on flesh composition. Sample processing time in the autoclave can be reduced from 4 to 1 h with silver perch flesh. © 2000 Society of Chemical Industry

The posterior forearm is an excellent donor site for the vascular pedicled cutaneous flaps; yet, there is surprisingly little detailed anatomical information based on clinical decision making. This study was undertaken to evaluate the anatomical basis of the dorsal forearm perforator flaps and to provide anatomical landmarks to facilitate flap elevation. Thirty cadavers were available to perform this anatomical study after arterial injection. Twenty fresh cadavers were injected with a modified lead oxide–gelatin mixture, selected for 3-dimensional reconstruction using special software (MIMICS) and the arterial territory measured with Scion Image. Other ten were injected with red latex preparation, and perforators were identified through dissection. (1) The average number of posterior interosseous artery cutaneous perforators in the dorsal forearm was 5 ± 2, the average diameter was (0.5 ± 0.1) mm, and the pedicle length was (2.5 ± 0.2) cm. The average cutaneous vascular territory was (22 ± 15) cm2. Cutaneous perforators could be found along the line extending from the lateral epicondyle to the radial border of the head of ulna. (2) Dorsal branch of anterior interosseous artery supplied blood to distal third of dorsal forearm; its average diameter was 0.8 mm. The free transplantation of the posterior interosseous perforator artery flaps or rotary flap pedicled by dorsal branch of anterior interosseous artery for defect reconstruction is feasible.

Bone formation and loss are related to the strain imposed on bone by muscle forces. Bone mineral content (BMC) and lean mass (LM) of fetal lambs was determined at day 140 of pregnancy in 8 groups of ewes, which were of either large or small body size, on either high (ad libitum) or maintenance pasture intake from day 21 of pregnancy, or carrying either singletons or twins. BMC and LM (using DXA scanning) of fetal hindquarters/spine were corrected to leg length. BMC and LM were less in twin than singleton groups ( P < 0.001). Large ewes on high intake produced single fetuses with a (group mean) BMC/LM ratio that was higher ( P < 0.002) than that in fetuses of large ewes with singletons on maintenance intake or twins on either high or maintenance intakes, the ratios of which were not different. In single fetuses from small ewes on high intake, the BMC/LM ratio was higher than those from small ewes with singletons on maintenance intake or twins on either high or maintenance intakes, the ratios of which were not different. The ratio was not different in singleton fetuses of ewes on high intake, whether they were large or small. Different fetal environments resulted in a given amount of muscle being associated with a higher or lower bone mass. Dietary intake during pregnancy was more important than maternal size in affecting the ratio. We conclude that intrauterine environmental factors may be important in determining bone mass postnatally, and possibly later in life.

Full platelet activation with serotonin secretion and thromboxane A2 (TxA2) formation induced by a low dose of thrombin receptor agonist peptide (TRAP) or high dose ADP requires platelet aggregation. This requirement can be replaced by pretreatment of platelets with a combination of reagents including: GPIIb/IIIa inhibitors yielding ligand-induced binding sites (LIBS), either arginine–glycine–aspartate–serine (RGDS) peptide or Ro 43-5054, cytochalasin to disrupt actin filaments and crosslinking by a GPIIb/IIIa mAb (pl-62). Crosslinking is required since Fab fragments of pl-62 do not support activation. Engagement of the Fc receptor by the mAb Fc domain is not required for pl-62 augmentation, since it is not blocked by the anti-Fc receptor mAb, IV-3. Another GPIIb/IIIa inhibitor, Ro 44-9883, not yielding LIBS epitopes, serves as a negative control and shows a requirement for LIBS in addition to crosslinking. Focal adhesion kinase tyrosine phosphorylation induced by TRAP is blocked by these GPIIb/IIIa antagonists, but restored by pl-62 crosslinking independent of LIBS induction. Tyrosine phosphorylation of a peptide comigrating with p38 MAP kinase is also inhibited by these antagonists and restored by pl-62 crosslinking. However, p38 MAP kinase activation by low dose TRAP is not affected by these aggregation inhibitors. Tyrosine phosphorylation of a 34-kDa phosphoprotein in the absence of aggregation or TxA2 formation was uniquely augmented by Ro 43-5054 but not Ro 44-9883 under the above activation conditions.

Platelets play a major role in thrombus formation, as well as in the pathogenesis of atherothrombosis. Inhibition of platelet function is now emphasised more than ever for prevention and treatment of almost all vascular diseases, since thrombosis is established as the key pathogenic event causing acute ischaemic coronary and cerebrovascular syndromes. Although acetylsalicylic acid (aspirin) has been shown to reduce the incidence of myocardial infarction and stroke, its effect is weak and more effective antithrombotic agents are required to manage patients at high-risk for recurrent vascular events. Platelet glycoprotein IIb/IIIa receptor (GPIIb/IIIa) blockade represents a significant advance in interventional cardiology and treatment of acute ischaemic syndromes. The past several years have seen the introduction of many platelet GPIIb/IIIa blockers into the clinical arena targeting the unique platelet GPIIb/IIIa receptor for the adhesive proteins, fibrinogen and von Willebrand Factor. Platelet GPIIb/IIIa blockers administered intravenously have proven efficacious in mitigating arterial thrombosis in acute coronary syndromes (unstable angina and non-ST-elevation myocardial infarction) and percutaneous coronary interventions (PCI) such as balloon dilatation and stent implantation. Currently, orally-active platelet GPIIb/IIIa blockers are being developed to provide additional benefits for primary and secondary prevention of thrombosis as chronic treatment, especially in high-risk patients. Lotrafiban (SmithKline Beecham) is a member of the latest generation of orally-active platelet GPIIb/IIIa blockers undergoing Phase III clinical trials to test the relative effectiveness versus other oral platelet inhibitors for ischaemic conditions including unstable angina, restenosis after PCI and stroke. Lotrafiban is converted from an esterified prodrug by plasma and liver esterases to a peptidomimetic of the arginine-glycine-aspartic acid amino acid sequence. This sequence itself mimics the binding site of fibrinogen and von Willebrand Factor to the platelet GPIIb/IIIa receptor. Preliminary results of the clinical trial APLAUD (antiplatelet useful dose) show that lotrafiban is clinically safe and well-tolerated in patients with recent myocardial infarction, unstable angina, transient ischaemic attack (TIA), or stroke when added to aspirin therapy. With lotrafiban, a worldwide large-scale Phase III clinical trial BRAVO (blockage of the GPIIb/IIIa receptor to avoid vascular occlusion) is currently underway. In general, GPIIb/IIIa blockade seems clinically very promising. A number of unresolved issues, however, remain to be elucidated.

Objective: To determine if cytokine responses and lung injury induced by intravenous (iv) lipopolysaccharide (LPS) at 4 hr were enhanced in rats that had been previously subjected to 30 min of total liver ischemia (Pringle's maneuver) followed by 24 hr or 3 days of reperfusion. Background: Many patients with liver trauma require occlusion of hepatic blood flow to control hemorrhage and facilitate repair. A significant number of these patients subsequently develop the systemic inflammatory response syndrome (SIRS) and multiple organ dysfunction (MOD) characterized by the release of cytokines and tissue neutrophil influx. Macrophages, including Kupffer cells, may be activated by ischemic injury and dysregulation of their response to LPS may contribute to the development of SIRS and acute respiratory distress syndrome. Methods: Adult male Sprague–Dawley rats were randomly divided into six groups: three groups received total hepatic ischemia for 30 min and three groups had a sham procedure. Twenty-four hours or 3 days after hepatic ischemia/reperfusion injury, rats were treated with LPS (5 mg/kg) or saline and monitored for 4 hr. We collected serum, bronchoalveolar lavage (BAL) fluid, and lung tissue. Results: Serum and BAL cytokine concentrations were significantly increased by iv LPS; however, hepatic ischemia/reperfusion injury 24 hr or 3 days before iv LPS ameliorated this cytokine response. The LPS-induced pulmonary neutrophil influx and histopathological changes were similar in sham and hepatic ischemia/reperfusion-injured groups. Conclusions: Hepatic ischemia/reperfusion injury significantly attenuated the serum and BAL cytokine concentrations, but did not change pulmonary neutrophil influx or histopathological alterations in response to iv LPS.
