The ongoing pandemic of the respiratory disease COVID-19 is caused by the SARS-CoV-2 (SCoV2) virus. SCoV2 is a member of the Betacoronavirus genus. The 30 kb positive sense, single stranded RNA genome of SCoV2 features 5'- and 3'-genomic ends that are highly conserved among Betacoronaviruses. These genomic ends contain structured cis-acting RNA elements, which are involved in the regulation of viral replication and translation. Structural information about these potential antiviral drug targets supports the development of novel classes of therapeutics against COVID-19. The highly conserved branched stem-loop 5 (SL5) found within the 5'-untranslated region (5'-UTR) consists of a basal stem and three stem-loops, namely SL5a, SL5b and SL5c. Both, SL5a and SL5b feature a 5'-UUUCGU-3' hexaloop that is also found among Alphacoronaviruses. The stem-loop (SL1) is the 5'-terminal structural element within the single-stranded SARS-CoV-2 RNA genome. It is formed by nucleotides 7–33 and consists of two short helical segments interrupted by an asymmetric internal loop. This architecture is conserved among Betacoronaviruses. SL1 is present in genomic SARS-CoV-2 RNA as well as in all subgenomic mRNA species produced by the virus during replication, thus representing a ubiquitous cis-regulatory RNA with potential functions at all stages of the viral life cycle. We present here the 1H, 13C and 15N chemical shift assignment of the 29 nucleotides-RNA construct 5_SL1, which denotes the native 27mer SL1 stabilized by an additional terminal G-C base-pair.

The SARS-CoV-2 virus is the cause of the respiratory disease COVID-19. As of today, therapeutic interventions in severe COVID-19 cases are still not available as no effective therapeutics have been developed so far. Despite the ongoing development of a number of effective vaccines, therapeutics to fight the disease once it has been contracted will still be required. Promising targets for the development of antiviral agents against SARS-CoV-2 can be found in the viral RNA genome. The 5′- and 3′-genomic ends of the 30 kb SCoV-2 genome are highly conserved among Betacoronaviruses and contain structured RNA elements involved in the translation and replication of the viral genome. The 40 nucleotides (nt) long highly conserved stem-loop 4 (5_SL4) is located within the 5′-untranslated region (5′-UTR) important for viral replication. 5_SL4 features an extended stem structure disrupted by several pyrimidine mismatches and is capped by a pentaloop. Here, we report extensive 1H, 13C, 15N and 31P resonance assignments of 5_SL4 as the basis for in-depth structural and ligand screening studies by solution NMR spectroscopy.

Proteome integrity is crucial for cellular homeostasis and adaptation to stress conditions such as hypoxia. One mechanism for rapid adaptation of the proteome in response to changing environmental signals is alternative splicing. In addition to generating different protein isoforms, alternative splicing is also capable of controlling total protein levels by the regulated synthesis of non-productive mRNA isoforms. The hypoxia-induced isoform E of the tumor suppressor MAX is produced by retention and translation of the last intron. This leads to an alternative C-terminus that harbors a potent C-degron, the isoE degron. Strikingly, the isoE degron represents a universal protein degradation signal that is not only functional in mammalian cells, but also in yeast and even in bacteria. Essential for efficient protein decay is a conserved (F/W)xxW motif. Degradation of isoE tagged proteins is mediated by the proteasome in eukaryotes and Lon protease in bacteria. Thus, the isoE degron is a broadly applicable and highly efficient tool in protein analyses.

Adenylate/uridylate-rich elements (AREs) are the most common cis-regulatory elements in the 3′-untranslated region (UTR) of mRNAs, where they fine-tune turnover by mediating mRNA decay. They increase plasticity and efficacy of mRNA regulation and are recognized by several ARE-specific RNA-binding proteins (RBPs). Typically, AREs are short linear motifs with a high content of complementary A and U nucleotides and often occur in multiple copies. Although thermodynamically rather unstable, the high AU-content might enable transient secondary structure formation and modify mRNA regulation by RBPs. We have recently suggested that the immunoregulatory RBP Roquin recognizes folded AREs as constitutive decay elements (CDEs), resulting in shape-specific ARE-mediated mRNA degradation. However, the structural evidence for a CDE-like recognition of AREs by Roquin is still lacking. We here present structures of CDE-like folded AREs, both in their free and protein-bound form. Moreover, the AREs in the UCP3 3′-UTR are additionally bound by the canonical ARE-binding protein AUF1 in their linear form, adopting an alternative binding-interface compared to the recognition of their CDE structure by Roquin. Strikingly, our findings thus suggest that AREs can be recognized in multiple ways, allowing control over mRNA regulation by adapting distinct conformational states, thus providing differential accessibility to regulatory RBPs.

The current pandemic situation caused by the Betacoronavirus SARS-CoV-2 (SCoV2) highlights the need for coordinated research to combat COVID-19. A particularly important aspect is the development of medication. In addition to viral proteins, structured RNA elements represent a potent alternative as drug targets. The search for drugs that target RNA requires their high-resolution structural characterization. Using nuclear magnetic resonance (NMR) spectroscopy, a worldwide consortium of NMR researchers aims to characterize potential RNA drug targets of SCoV2. Here, we report the characterization of 15 conserved RNA elements located at the 5' end, the ribosomal frameshift segment and the 3'-untranslated region (3'-UTR) of the SCoV2 genome, their large-scale production and NMR-based secondary structure determination. The NMR data are corroborated with secondary structure probing by DMS footprinting experiments. The close agreement of NMR secondary structure determination of isolated RNA elements with DMS footprinting and NMR performed on larger RNA regions shows that the secondary structure elements fold independently. The NMR data reported here provide the basis for NMR investigations of RNA function, RNA interactions with viral and host proteins and screening campaigns to identify potential RNA binders for pharmaceutical intervention.

This thesis analyses interviews with 13 guerrilla combatants originally conducted in early 1997, while I was a journalist with the CERIGUA news agency covering the Guatemalan armed conflict, the Peace Accords and the demobilization and reintegration into civil society of Guatemalan National Revolutionary Unity (URNG) fighters. The work gathers the testimonies of the guerrillas, including their motivations for joining the insurgency, experiences in the guerrilla, and feelings regarding the end of the armed conflict and their pending reintegration into Guatemalan society. I compare what the URNG combatants expressed in these interviews with other research regarding the 36-year conflict and especially studies documenting the experience of demobilized URNG members a decade or more after their reintegration into Guatemalan civil society. Based on this research, the thesis argues that, contrary to what is promoted in some Disarmament, Demobilization and Reintegration (DDR) literature, in the Guatemalan experience collective reintegration proved more successful than individualized reintegration, and should have been provided to a much large number of former combatants. In this research, successful reintegration is interpreted as economic and social well-being, as well as the political and social engagement of the former combatants in broader Guatemalan society, particularly engagement aimed at addressing the factors that originally gave rise to the armed conflict. Special attention is paid to these criteria in the reintegration of female former combatants.

As the major structural constituent of the cytoskeleton, microtubules (MTs) serve a variety of biological functions that range from facilitating organelle transport to maintaining the mechanical integrity of the cell. Neuronal MTs exhibit a distinct configuration, hexagonally packed bundles of MT filaments, interconnected by MT-associated protein (MAP) tau. Building on our previous work on mechanical response of axonal MT bundles under uniaxial tension, this study is focused on exploring the compression scenarios. Intracellular MTs carry a large fraction of the compressive loads sensed by the cell and therefore, like any other column-like structure, are prone to substantial bending and buckling. Various biological activities, e.g., actomyosin contractility and many pathological conditions are driven or followed by bending, looping, and buckling of MT filaments. The coarse-grained model previously developed in our lab has been used to study the mechanical behavior of individual and bundled in vivo MT filaments under uniaxial compression. Both configurations show tip-localized, decaying, and short-wavelength buckling. This behavior highlights the role of the surrounding cytoplasm and MAP tau on MT buckling behavior, which allows MT filaments to bear much larger compressive forces. It is observed that MAP tau interconnections improve this effect by a factor of two. The enhanced ability of MT bundles to damp buckling waves relative to individual MT filaments, may be interpreted as a self-defense mechanism because it helps axonal MTs to endure harsher environments while maintaining their function. The results indicate that MT filaments in a bundle do not buckle simultaneously implying that the applied stress is not equally shared among the MT filaments, that is a consequence of the nonuniform distribution of MAP tau proteins along the bundle length. Furthermore, from a pathological perspective, it is observed that axonal MT bundles are more vulnerable to failure in compression than tension.

Microtubule bundles cross-linked by tau protein serve a variety of neurological functions including maintaining mechanical integrity of the axon, promoting axonal growth, and facilitating cargo transport. It has been observed that axonal damage in traumatic brain injury leads to bundle disorientation, loss of axonal viability, and cognitive impairment. This study investigates the initial mechanical response of axonal microtubule bundles under uniaxial tension using a discrete bead-spring representation. Mechanisms of failure due to traumatic stretch loading and their impact on the mechanical response and stability are also characterized. This study indicates that cross-linked axonal microtubule bundles in tension display stiffening behavior similar to a power-law relationship from nonaffine network deformations. Stretching of cross-links and microtubule bending were the primary deformation modes at low stresses. Microtubule stretch was negligible up to tensile stresses of ∼1 MPa. Bundle failure occurred by failure of cross-links leading to pull-out of microtubules and loss of bundle integrity. This may explain the elongation, undulation, and delayed elasticity of axons following traumatic stretch loading. More extensively cross-linked bundles withstood higher tensile stresses before failing. The bundle mechanical behavior uncovered by these computational techniques should guide future experiments on stretch-injured axons.

The social and economic contribution of franchising to many Western economies is widely reported (Alon, 2004). Franchising is a business model in which a franchisee is granted the right to engage in offering, selling, or distributing goods or services under a marketing system that has been designed by the franchisor (Justis and Judd, 2004). The franchisor permits the franchisee to use the franchisor’s trade name and advertising (Mendelsohn, 2004). The Australian franchising sector has achieved consistent growth since the 1980s, with franchising activity currently contributing approximately (12 percent) of the country’s gross domestic product (Frazer et al., 2008). However, research has revealed that franchising systems may be unable to expand at the rate they desire owing to difficulties in obtaining franchisee finance and a shortage of suitable franchisee applicants (Price Waterhouse Coopers, 2009; Frazer et al., 2008). There are currently 56,200 business format franchised units operating in Australia (Frazer et al., 2008), accounting for approximately (5 percent) of all small business in Australia (ABS, 2009). Though this appears to suggest that franchising has the potential to expand, prior research has shown that the lack of available suitable franchisee candidates may constrain system growth and sector expansion (Frazer et al., 2008). Previous research shows that this is not a recent phenomenon (Knight, 1986; McGuire, 1971) and may be reliant upon how franchisors classify attributes that are desirable in potential incumbent franchisees (Frazer and McCosker, 1999; Hedricks and Kiefer, 2005; Michael, 2003). However, there appears to be a gap in our current understanding of how these impediments to franchise growth may be overcome. In addition to acquiring valuable insights into current recruitment practice, the primary purpose of this research is to sustain informed decision-making in the future through a study of the fundamental and under-researched question of how franchisors may overcome the shortage of suitable franchisees. On this basis, a thorough enquiry into possible solutions to the problem associated with a shortage of suitable franchisees is warranted. Hence, the following research questions are posed. Research question 1: “What are desirable franchisee attributes?” Research question 2: “How can franchisors overcome the perceived shortage of suitable franchisees”? This research focuses on theory development by integrating known concepts with new research and emerging marketing techniques that may be applied to this problem.

This paper explores the impact of different forms of political leadership created through the Local Government Act 2000. The concept of facilitative leadership is used to explore how leaders use powers and abilities to draw citizens and other stakeholders into a shared vision for the locality, and enable local councils and other actors to shape places and improve service performance. The paper also reflects on possible implications for proposals in the 2006 Local Government White Paper. It is an interim output from the five year evaluation of council constitutions.

This report presents findings from a 2005 survey of councillors, officers and stakeholders in 40 local authorities and is part of the ongoing evaluation of new council constitutions. The survey was designed to see whether the new arrangements were perceived to have achieved their objectives. Overwhelmingly, respondents express the view that the new arrangements provide a structure for effective political leadership while the evidence on the new arrangements having enhanced democratic legitimacy is less convincing. Encouragingly, the majority of officers and stakeholders (and about 50 per cent of councillors) are confident that checks and balances are in place to support transparent decision-making.

SARS‐CoV‐2 contains a positive single‐stranded RNA genome of approximately 30 000 nucleotides. Within this genome, 15 RNA elements were identified as conserved between SARS‐CoV and SARS‐CoV‐2. By nuclear magnetic resonance (NMR) spectroscopy, we previously determined that these elements fold independently, in line with data from in vivo and ex‐vivo structural probing experiments. These elements contain non‐base‐paired regions that potentially harbor ligand‐binding pockets. Here, we performed an NMR‐based screening of a poised fragment library of 768 compounds for binding to these RNAs, employing three different 1H‐based 1D NMR binding assays. The screening identified common as well as RNA‐element specific hits. The results allow selection of the most promising of the 15 RNA elements as putative drug targets. Based on the identified hits, we derive key functional units and groups in ligands for effective targeting of the RNA of SARS‐CoV‐2., The RNA genome of SARS‐CoV‐2 contains 15 independently folded regulatory RNA elements. By NMR‐based fragment screening, the RNA target space for small molecule binding, functional mapping and inhibition is defined.
