Abstract

Diese Übersichtsarbeit verfolgt das Ziel, über bakterielle kontraktile Phagenderivate (englisch CPTPs) und ihr Potenzial als effiziente und Schadorganismus-spezifische Alternativen zu konventionellen chemischen Pflanzenschutzmitteln in der Land-/Forstwirtschaft und im Gartenbau zu informieren. CPTPs werden von verschiedenen Bakterien in diversen Habitaten für den interbakteriellen Konkurrenzkampf sowie zur Beeinflussung eukaryotischer Wirte, wie Pilze und Insekten, verwendet. Diese Arbeit präsentiert interessante und bemerkenswerte Beispiele für den vielfältigen Einsatz von CPTPs als leistungsfähige biologische Bekämpfungsmittel. Wir stellen die verschiedenen Typen von CPTPs vor und legen einen besonderen Fokus auf diejenigen, die eine Wirkung gegen Schadinsekten besitzen. Zusätzlich stellen wir zwei kürzlich etablierte Webservices vor, die das permanent wachsende Wissen über CPTPs mit einem Auswahlverfahren für die besten Bakterienkandidaten kombinieren, um eine zielgerichtete Anwendung der CPTPs in der nachhaltigen Pflanzenproduk­tion zu ermöglichen.

2. Visual chunking as a strategy for spatial thinking in STEM

Abstract

Abstract Working memory capacity is known to predict the performance of novices and experts on a variety of tasks found in STEM (Science, Technology, Engineering, and Mathematics). A common feature of STEM tasks is that they require the problem solver to encode and transform complex spatial information depicted in disciplinary representations that seemingly exceed the known capacity limits of visuospatial working memory. Understanding these limits and how visuospatial information is encoded and transformed differently by STEM learners presents new avenues for addressing the challenges students face while navigating STEM classes and degree programs. Here, we describe two studies that explore student accuracy at detecting color changes in visual stimuli from the discipline of chemistry. We demonstrate that both naive and novice chemistry students’ encoding of visuospatial information is affected by how information is visually structured in “chunks” prevalent across chemistry representations. In both studies we show that students are more accurate at detecting color changes within chemistry-relevant chunks compared to changes that occur outside of them, but performance was not affected by the dimensionality of the structure (2D vs 3D) or the presence of redundancies in the visual representation. These studies support the hypothesis that strategies for chunking the spatial structure of information may be critical tools for transcending otherwise severely limited visuospatial capacity in the absence of expertise.

Abstract

Ariel, the Atmospheric Remote-sensing Infrared Exoplanet Large-survey, was adopted as the fourth medium-class mission in ESA's Cosmic Vision programme to be launched in 2029. During its 4-year mission, Ariel will study what exoplanets are made of, how they formed and how they evolve, by surveying a diverse sample of about 1000 extrasolar planets, simultaneously in visible and infrared wavelengths. It is the first mission dedicated to measuring the chemical composition and thermal structures of hundreds of transiting exoplanets, enabling planetary science far beyond the boundaries of the Solar System. The payload consists of an off-axis Cassegrain telescope (primary mirror 1100 mm x 730 mm ellipse) and two separate instruments (FGS and AIRS) covering simultaneously 0.5-7.8 micron spectral range. The satellite is best placed into an L2 orbit to maximise the thermal stability and the field of regard. The payload module is passively cooled via a series of V-Groove radiators; the detectors for the AIRS are the only items that require active cooling via an active Ne JT cooler. 6. Spectral study of Fe-rich vermiculite bearing rocks and implications for mineralogy of Oxia Planum, ExoMars 2022 Rover landing site

Abstract

In 2022 rover, ESA-Roscosmos will launch rover to land at Oxia Planum on Mars. Oxia Planum is a wide, Noachian-age, phyllosilicate-bearing plain located between Mawrth and Ares Valles. The bedrock phyllosilicate deposits at Oxia Planum is Fe,Mg-rich and one of the largest exposures of this type on Mars, with a thickness of more than 10 m [1]. When characterized in the near infrared by CRISM and OMEGA instruments, the bedrock deposits exhibit absorptions that suggest the presence of Fe,Mg-rich phyllosilicates with significant amounts of Fe2+ in octahedral sites (i.e., trioctahedral) [2]. The typical spectrum is best matched by saponite or vermiculite, but an accurate spectral match is lacking so far. Because precise mineral composition of the bedrock is not fully understood, limited conclusions can be drawn regarding the aqueous evolution and habitability potential of Oxia – the main scientific aim of the ExoMars 2022 mission. To fill this gap, and to better prepare for in-situ analyses by the ExoMars2022 rover, we performed a survey of potential terrestrial analogue rocks. We have identified terrestrial deposits of Fe-rich, trioctahedral vermiculite and perform spectral comparisons with the Martian substrate. Analogues from two terrestrial sites were obtained: (1) vermiculitized chlorite-schists from Blue Spur, Otago, New Zealand and [3] (2) basaltic tuffs and pyroclastics from Granby, Massachusetts, USA, with Fe-rich clays filling amygdales [4]. The samples were characterized by X-ray diffraction to understand chemistry and structure of clay minerals and by NIR spectroscopy in order to provide further structural details and to assess the match to Oxia Planum bedrock clays. XRD was performed on bulk rock samples as well as for separated clay fraction minerals. Near Infrared (NIR) spectra for each sample were collected using a reflectance spectrometer in the near-infrared (0.8–4.2 μm) mode. Analysis was performed on powdered samples, under ambient temperature and pressure conditions. Both analogues have been added to a newly built Planetary Terrestrial Analogue Library (PTAL) rock collection and spectral library. Deposits in Blue Spur formed by erosion and deposition of greenschist-facies quartzo-feldspathic schists rich in Fe-rich chlorite. Due to rapid erosion and non-oxidizing or mild oxidative conditions [3] chlorite remained unoxidized when buried. Later interactions with groundwater caused formation of trioctahedral vermiculite and progressive alteration of deposit led to illitization and kaolinitization For our purposes we have collected samples of pristine schist clasts, greenish (unoxidized) vermiculitized material and pale, illitized clasts [3]. The XRD patterns of oriented clay fraction separates suggest the presence of chlorite in pristine material, trioctahedral vermiculite in greenish samples that underwent alteration in anoxic conditions, and interstratified illite-vermiculite in more oxidized samples.NIR spectroscopy analysis of Otago samples tends to indicate that the nature and overall Fe content is broadly similar to the phyllosilicates at Oxia. Furthermore, illitization of the vermiculite seen spectrally in Otago is likely limited for Oxia. This means that vermiculite at Oxia is a rather pure, one-phase mineral that underwent only minor (if any) post-depositional alterations toward illite. The Granby Tuff consists of basaltic flows and volcanic ash units. In places, it contains zones of vesicular basalts that have amygdales filled with calcite and dark-brown clays. The clays filling the amygdales are of apparent hydrothermal origin, that is precipitated in gas vesicles from solutions generated by hydrothermal alteration of the basaltic tuff. The clays were reported to be either saponite or vermiculite [4]. The XRD patterns of the oriented clay fraction separates from these tuffs reveal the presence of well-crystallized, trioctahedral vermiculite and various amounts of saponite in different samples. In NIR spectroscopy, Granby samples show quite a good spectral match to Oxia and indicate that saponite, present in Granby, is absent from Oxia spectra. Oxia deposits are likely pure vermiculite rather than a mixture of saponite and vermiculite. Additionally, other spectral features suggest that the Granby clays have different Fe/Mg ratios than minerals of Oxia Planum, or a different (plausibly lower) oxidation state of Fe in the clay structure. Our spectral analogue study shows that rocks containing Fe2+-rich vermiculite, locally with octahedral Al and (oxidized) Fe3+, are likely the main component of bedrock phyllosilicates at Oxia Planum. In terms of clay structure and its di- versus trioctahedral nature, Oxia deposits are matched best by a vermiculite-saponite from hydrothermally-derived settings (Granby Tuffs), although the contribution of saponite must be minor at Oxia. Comparison of Otago vermiculite to Oxia shows high Fe content in Oxia and a similar oxidation state. Spectral inconsistencies between Otago and Oxia related to presence of Al in the clay structure allow us to conclude that Oxia bedrock deposits were likely not oxidized nor illitized after formation. Abstract

Published by BMJ., Objectives: To determine factors associated with COVID-19-related death in people with rheumatic diseases. Methods: Physician-reported registry of adults with rheumatic disease and confirmed or presumptive COVID-19 (from 24 March to 1 July 2020). The primary outcome was COVID-19-related death. Age, sex, smoking status, comorbidities, rheumatic disease diagnosis, disease activity and medications were included as covariates in multivariable logistic regression models. Analyses were further stratified according to rheumatic disease category. Results: Of 3729 patients (mean age 57 years, 68% female), 390 (10.5%) died. Independent factors associated with COVID-19-related death were age (66-75 years: OR 3.00, 95% CI 2.13 to 4.22; >75 years: 6.18, 4.47 to 8.53; both vs ≤65 years), male sex (1.46, 1.11 to 1.91), hypertension combined with cardiovascular disease (1.89, 1.31 to 2.73), chronic lung disease (1.68, 1.26 to 2.25) and prednisolone-equivalent dosage >10 mg/day (1.69, 1.18 to 2.41; vs no glucocorticoid intake). Moderate/high disease activity (vs remission/low disease activity) was associated with higher odds of death (1.87, 1.27 to 2.77). Rituximab (4.04, 2.32 to 7.03), sulfasalazine (3.60, 1.66 to 7.78), immunosuppressants (azathioprine, cyclophosphamide, ciclosporin, mycophenolate or tacrolimus: 2.22, 1.43 to 3.46) and not receiving any disease-modifying anti-rheumatic drug (DMARD) (2.11, 1.48 to 3.01) were associated with higher odds of death, compared with methotrexate monotherapy. Other synthetic/biological DMARDs were not associated with COVID-19-related death. Conclusion: Among people with rheumatic disease, COVID-19-related death was associated with known general factors (older age, male sex and specific comorbidities) and disease-specific factors (disease activity and specific medications). The association with moderate/high disease activity highlights the importance of adequate disease control with DMARDs, preferably without increasing glucocorticoid dosages. Caution may be required with rituximab, sulfasalazine and some immunosuppressants.

Abstract

INTRODUCTION:The objective of the study was to compare compliance and acceptability of a new auto-injector (AI) versus syringe for administration of methotrexate (MTX) in patients with rheumatoid arthritis (RA).METHODS:We conducted a randomized, open-label, parallel group study comparing AI to pre-filled syringe (PFS). Adult patients with RA (ACR/EULAR 2010) receiving MTX (orally or by injection) for at least 3 months were allocated to AI or PFS for 6 months and then were allocated to AI for 6 further months. Two co-primary endpoints were defined at M6: percentage of patients with compliance at least 80%; change in functional capacity assessed by Health Assessment Questionnaire (HAQ). Secondary endpoints included quality of life (RaQoL), RA activity (DAS28), and acceptability. Local safety at injection site was assessed at each visit.RESULTS:Two-hundred and sixty-five patients were randomized. The main analysis was conducted on per protocol set (99 AI and 98 PFS). Compliance was 96.2% in AI and 98.9% in PFS. Good complier rates were 89.9% and 94.9%, thus a difference of - 5.0% (- 18.9%; 8.9%). HAQ remained stable in both groups. No difference was found on RaQoL, change in RA activity, and safety profile. Autonomy, acceptability, and patient satisfaction were better with AI, and patients having had the experience of both AI and PFS preferred AI (p, ClinicalTrials.gov identifier, NCT02553018.

Abstract

Industrialized tomato production faces a decrease in flavors and nutritional value due to conventional breeding. Moreover, tomato production heavily relies on nitrogen and phosphate fertilization. Phosphate uptake and improvement of fruit quality by arbuscular mycorrhizal (AM) fungi are well-studied. We addressed the question of whether commercially used tomato cultivars grown in a hydroponic system can be mycorrhizal, leading to improved fruit quality. Tomato plants inoculated with Rhizophagus irregularis were grown under different phosphate concentrations and in substrates used in industrial tomato production. Changes in fruit gene expression and metabolite levels were checked by RNAseq and metabolite determination, respectively. The tests revealed that reduction of phosphate to 80% and use of mixed substrate allow AM establishment without affecting yield. By comparing green fruits from non-mycorrhizal and mycorrhizal plants, differentially expressed genes (DEGs) were found to possibly be involved in processes regulating fruit maturation and nutrition. Red fruits from mycorrhizal plants showed a trend of higher BRIX values and increased levels of carotenoids in comparison to those from non-mycorrhizal plants. Free amino acids exhibited up to four times higher levels in red fruits due to AM, showing the potential of mycorrhization to increase the nutritional value of tomatoes in industrialized production.

Abstract

We reviewed the impact of fungal volatile organic compounds (VOCs) on soil-inhabiting organisms and their physiological and molecular consequences for their targets. Because fungi can only move by growth to distinct directions, a main mechanism to protect themselves from enemies or to manipulate their surroundings is the secretion of exudates or VOCs. The importance of VOCs in this regard has been significantly underestimated. VOCs not only can be means of communication, but also signals that are able to specifically manipulate the recipient. VOCs can reprogram root architecture of symbiotic partner plants or increase plant growth leading to enlarged colonization surfaces. VOCs are also able to enhance plant resistance against pathogens by activating phytohormone-dependent signaling pathways. In some cases, they were phytotoxic. Because the response was specific to distinct species, fungal VOCs may contribute to regulate the competition of plant communities. Additionally, VOCs are used by the producing fungus to attack rivaling fungi or bacteria, thereby protecting the emitter or its nutrient sources. In addition, animals, like springtails, nematodes, and earthworms, which are important components of the soil food web, respond to fungal VOCs. Some VOCs are effective repellents for nematodes and, therefore, have applications as biocontrol agents. In conclusion, this review shows that fungal VOCs have a huge impact on soil fauna and flora, but the underlying mechanisms, how VOCs are perceived by the recipients, how they manipulate their targets and the resulting ecological consequences of VOCs in inter-kingdom signaling is only partly understood. These knowledge gaps are left to be filled by future studies.

Abstract

Finally, we would like to recognize the help and support of ESA, Roscosmos, the European states and agencies participating in the ExoMars program, and NASA. We really are doing this together for the benefit of all., The second ExoMars mission will be launched in 2020 to target an ancient location interpreted to have strong potential for past habitability and for preserving physical and chemical biosignatures (as well as abiotic/prebiotic organics). The mission will deliver a lander with instruments for atmospheric and geophysical investigations and a rover tasked with searching for signs of extinct life. The ExoMars rover will be equipped with a drill to collect material from outcrops and at depth down to 2 m. This subsurface sampling capability will provide the best chance yet to gain access to chemical biosignatures. Abstract

Introduction Anterior cervical discectomy and fusion has been associated with the development of adjacent segment degeneration (ASD), with clinical incidence of approximately 3% per year. Cervical total disc arthroplasty (TDA) has been proposed as an alternative to prevent ASD. Hypotheses TDA in optimal placement using an elastic‐core cervical disc (RHINE, K2M Inc., Leesburg, Virginia) will replicate natural kinematics and will improve with optimal vs anterior placement. Methods Seven C3‐T1 cervical cadaver spines were tested intact first, then after one‐level TDA at C5‐C6 anterior placement, after TDA at C5‐C6 optimal placement, after two‐level TDA at C5‐C6 and C6‐C7 optimal placement, and finally after two‐level TDA at C5‐C6 lateral placement and C6‐C7 optimal placement. The specimens were subjected to: Flexion‐Extension moments (+1.5 Nm) with compressive preloads of 0 N and 150 N, lateral bending (LB) and axial rotation (AR) (+1.5 Nm) without preload. Results C5‐C6 TDA in optimal placement resulted in a non‐significant increase in flexion‐extension ROM compared to intact under 0 N and 150 N preload (P > 0.05). Abstract

International audience; Extracellular matrices (ECMs) are central to the advent of multicellular life, and their mechanical properties are modulated by and impinge on intracellular signaling pathways that regulate vital cellular functions. High spatial-resolution mapping of mechanical properties in live cells is, however, extremely challenging. Thus, our understanding of how signaling pathways process physiological signals to generate appropriate mechanical responses is limited. We introduce fluorescence emission-Brillouin scattering imaging (FBi), a method for the parallel and all-optical measurements of mechanical properties and fluorescence at the submicrometer scale in living organisms. Using FBi, we showed that changes in cellular hydrostatic pressure and cytoplasm viscoelasticity modulate the mechanical signatures of plant ECMs. We further established that the measured "stiffness" of plant ECMs is symmetrically patterned in hypocotyl cells undergoing directional growth. Finally, application of this method to Arabidopsis thaliana with photoreceptor mutants revealed that red and far-red light signals are essential modulators of ECM viscoelasticity. By mapping the viscoelastic signatures of a complex ECM, we provide proof of principle for the organism-wide applicability of FBi for measuring the mechanical outputs of intracellular signaling pathways. As such, our work has implications for investigations of mechanosignaling pathways and developmental biology.

Abstract

The bioactive lipid ceramide is produced by the enzyme ceramide synthase, which exists in several isoforms in most eukaryotic organisms. Here, we investigated functional differences between the three ceramide synthase isoforms in Arabidopsis thaliana. The biochemical properties of the three ceramide synthases were investigated by comparing lipid profiles of yeast strains expressing LOH1, LOH2 or LOH3 with those of wild-type and loh1, loh2 and loh3 knockout plants. Expression profiles of the ceramide synthases and of the pathogenesis-related gene PR-1 were investigated by real-time PCR. Each ceramide synthase isoform showed a characteristic preference regarding acyl-CoA chain length as well as sphingoid base hydroxylation, which matches the pattern of ceramide and glucosylceramide species found in leaves. After extended culture under short-day conditions, loh1 plants showed spontaneous cell death accompanied by enhanced expression of PR-1. The levels of free trihydroxy sphingoid bases as well as ceramide and glucosylceramide species with C(16) fatty acid were significantly elevated while species with C(20) -C(28) fatty acids were reduced. These data suggest that spontaneous cell death in the loh1 line is triggered either by the accumulation of free trihydroxy sphingoid bases or ceramide species with C(16) fatty acid.

Abstract

The functions of the minor phospholipid phosphatidylinositol-4,5-bisphosphate [PtdIns(4,5)P2] during vegetative plant growth remain obscure. Here, we targeted two related phosphatidylinositol 4-phosphate 5-kinases (PI4P 5-kinases) PIP5K1 and PIP5K2, which are expressed ubiquitously in Arabidopsis thaliana. A pip5k1 pip5k2 double mutant with reduced PtdIns(4,5)P2 levels showed dwarf stature and phenotypes suggesting defects in auxin distribution. The roots of the pip5k1 pip5k2 double mutant had normal auxin levels but reduced auxin transport and altered distribution. Fluorescence-tagged auxin efflux carriers PIN-FORMED (PIN1)–green fluorescent protein (GFP) and PIN2-GFP displayed abnormal, partially apolar distribution. Furthermore, fewer brefeldin A–induced endosomal bodies decorated by PIN1-GFP or PIN2-GFP formed in pip5k1 pip5k2 mutants. Inducible overexpressor lines for PIP5K1 or PIP5K2 also exhibited phenotypes indicating misregulation of auxin-dependent processes, and immunolocalization showed reduced membrane association of PIN1 and PIN2. PIN cycling and polarization require clathrin-mediated endocytosis and labeled clathrin light chain also displayed altered localization patterns in the pip5k1 pip5k2 double mutant, consistent with a role for PtdIns(4,5)P2 in the regulation of clathrin-mediated endocytosis. Further biochemical tests on subcellular fractions enriched for clathrin-coated vesicles (CCVs) indicated that pip5k1 and pip5k2 mutants have reduced CCV-associated PI4P 5-kinase activity. Together, the data indicate an important role for PtdIns(4,5)P2 in the control of clathrin dynamics and in auxin distribution in Arabidopsis.

Cyanobacterial toxins have been shown to have a far-reaching impact-from aquatic organisms to human health. Aquatic organisms are typically exposed in their natural environment to toxic cyanobacteria, and exposure can occur via ingestion of cyanobacterial cells or by bioaccumulation of water-borne toxin. The aquaculture and fisheries of crustaceans are among the most important seafood industries. Concomitant with the growth of this industry, the importance of the health of crustaceans increased. The black tiger prawn is the major cultivated prawn in Australia. The aquaculture of these prawns takes place in shallow ponds, where blooms, often of cyanobacteria, develop. Cyanobacterial toxins were hypothesized to contribute to the mortality of prawns. Many aquatic organisms have the possibility of detoxifying cyanobacterial toxins via conjugation to glutathione. The presence of several classes of the cytosolic glutathione S-transferase system in black tiger prawns-mu, pi, theta, alpha, and tau-was shown using different substrates for measurement. Injection experiments with microcystin-LR and feeding experiments with nodularin revealed elevation of GST activity in different types of prawn tissue in parallel with reduction in the GST classes. Correlation analyses of toxin content of the prawns with GST activity showed that low toxin content was correlated with high elevation of enzymes and high toxin content with low elevation of enzymes.
