Abstract Unsustainable wild meat hunting poses a significant threat to wildlife and tropical forest ecosystems. While high levels of extraction linked to commercial trade have received significant attention, the sustainability of subsistence hunting by Indigenous Peoples in Africa has been less studied. Understanding how changing lifestyles, particularly the sedentarisation of former hunter-gatherers, have affected the use of forest resources is crucial for wildlife conservation and livelihoods. The spatial management of hunting through the establishment of no-take zones, which act as sources for adjacent hunting areas, offers promise for the sustainability of Indigenous livelihoods. We conducted an extensive camera trap study in hunting areas subject to source-sink dynamics used by 10 sedentarised Baka communities. We compared species richness, occupancy, abundance, and community composition to a relatively non-hunted reference area in the adjacent Dja Faunal Reserve. Subsistence hunting by the Baka had a limited impact on species richness but significantly altered community composition and the abundance of carnivores, seed dispersers and granivores. These changes highlight that even the spatial management of hunting may have consequences for the sustainability of hunting systems and the functional ecology of tropical forests.

Abstract Caves are primary sites for studying human and animal subsistence patterns and genetic ancestry throughout the Palaeolithic. Iberia served as a critical human and animal refugium in Europe during the Last Glacial Maximum (LGM), 26.5 to 19 thousand years before the present (cal kya). Therefore, it is a key location for understanding human and animal population dynamics during this event. We recover and analyse sedimentary ancient DNA (sedaDNA) data from the lower archaeological stratigraphic sequence of El Mirón Cave (Cantabria, Spain), encompassing the (1) Late Mousterian period, associated with Neanderthals, and (2) the Gravettian (c. 31.5 cal kya), Solutrean (c. 24.5–22 cal kya), and Initial Magdalenian (d. 21–20.5 cal kya) periods, associated with anatomically modern humans. We identify 28 animal taxa including humans. Fifteen of these taxa had not been identified from the archaeozoological (i.e., faunal) record, including the presence of hyenas in the Magdalenian. Additionally, we provide phylogenetic analyses on 70 sedaDNA mtDNA genomes of fauna including the densest Iberian Pleistocene sampling of C. lupus. Finally, we recover three human mtDNA sequences from the Solutrean levels. These sequences, along with published data, suggest mtDNA haplogroup continuity in Iberia throughout the Solutrean/Last Glacial Maximum period.

The slowing and reduction of auditory responses in the brain are recognized side effects of increased pure tone thresholds, impaired speech recognition, and aging. However, it remains controversial whether central slowing is primarily linked to brain processes as atrophy, or is also associated with the slowing of temporal neural processing from the periphery. Here we analyzed electroencephalogram (EEG) responses that most likely reflect medial geniculate body (MGB) responses to passive listening of phonemes in 80 subjects ranging in age from 18 to 76 years, in whom the peripheral auditory responses had been analyzed in detail (Schirmer et al., 2024). A soil spectrum generated by any spectrometer requires a calibration model to estimate soil properties from it. To achieve best results, the assumption is that locally calibrated models offer more accurate predictions. However, achieving this higher accuracy comes with associated costs, complexity, and resource requirements, thus limiting widespread adoption. Furthermore, there is a lack of comprehensive frameworks for developing and utilizing soil spectral libraries (SSLs) to make predictions for specific samples. While calibration samples are necessary, there is the need to optimize SSL development through strategically determining the quantity, location, and timing of these samples based on the quality of the information in the library. This research aimed to develop a spatially optimized SSL and propose a use-framework tailored for predicting soil properties for a specific farmland context. Consequently, the Moroccan SSL (MSSL) was established utilizing a stratified spatially balanced sampling design, using six environmental covariates and FAO soil units. Subsequently, various criteria for calibration sample selection were explored, including a spatial autocorrelation of spectra principal component (PC) scores (spatial calibration sample selection), spectra similarity memory-based learner (MBL), and selection based on environmental covariate clustering. Twelve soil properties were used to evaluate these calibration sample selections to predict soil properties using the near infrared (NIR) and mid infrared (MIR) ranges. Among the methods assessed, we observed distinct precision improvements resulting from spatial sample selection and MBL compared to the use of the entire MSSL. Notably, the Lin’s Concordance Correlation Coefficient (CCC) values using the spatial calibration sample selection was improved for Olsen extractable phosphorus (OlsenP) by 41.3% and Mehlich III extractable phosphorus (P_M3) by 8.5% for the MIR spectra and for CEC by 25.6%, pH by 13.0% and total nitrogen (Tot_N) by 10.6% for the NIR spectra in reference to use of the entire MSSL. Utilizing the spatial autocorrelation of the spectra PC scores proved beneficial in identifying appropriate calibration samples for a new sample location, thereby enhancing prediction performance comparable to, or surpassing that of the use of the entire MSSL. This study signifies notable advancement in crafting targeted models tailored for specific samples within a vast and diverse SSL.

Abstract In the last few decades, the field of ancient DNA has taken a new direction towards using sedimentary ancient DNA (sedaDNA) for studying human and mammalian population dynamics as well as past ecosystems. However, the screening of numerous sediment samples from archaeological sites remains a time-consuming and costly endeavor, particularly when targeting hominin DNA. Here, we present a novel high-throughput method that facilitates the fast and efficient analysis of sediment samples by applying a pooled testing approach. This method combines multiple extracts, enabling early parallelization of laboratory procedures and effective aDNA screening. Pooled samples with detectable aDNA signals undergo detailed analysis, while empty pools are discarded. We have successfully applied our method to multiple sediment samples from Middle and Upper Paleolithic sites in Europe, Asia, and Africa. Notably, our results reveal that an aDNA signal remains discernible even when pooled with four negative samples. We also demonstrate that the DNA yield of double-stranded libraries increases significantly when reducing the extract input, potentially mitigating the effects of inhibition. By embracing this innovative approach, researchers can analyze large numbers of sediment samples for aDNA preservation, achieving significant cost reductions of up to 70% and reducing hands-on laboratory time to one-fifth.

Abstract Analyzing irregularities in walking patterns helps detect human locomotion abnormalities that can signal health changes. Traditional observation-based assessments have limitations due to subjective biases and capture only a single time point. Ambient and wearable sensor technologies allow continuous and objective locomotion monitoring but face challenges due to the need for specialized expertise and user compliance. This work proposes a seismograph-based algorithm for quantifying human gait, incorporating a step extraction algorithm derived from mathematical morphologies, with the goal of achieving the accuracy of clinical reference systems. To evaluate our method, we compared the gait parameters of 50 healthy participants, as recorded by seismographs, and those obtained from reference systems (a pressure-sensitive walkway and a camera system). Participants performed four walking tests, including traversing a walkway and completing the timed up-and-go (TUG) test. In our findings, we observed linear relationships with strong positive correlations (R 2 > 0.9) and tight 95% confidence intervals for all gait parameters (step time, cycle time, ambulation time, and cadence). We demonstrated that clinical gait parameters and TUG mobility test timings can be accurately derived from seismographic signals, with our method exhibiting no significant differences from established clinical reference systems.

Background: In genomics, highly sensitive point mutation detection is particularly relevant for cancer diagnosis and early relapse detection. Next-generation sequencing combined with unique molecular identifiers (UMIs) is known to improve the mutation detection sensitivity. Methods: We present an open-source bioinformatics framework named Interface for Point Mutation Identification (IMPI) with a graphical user interface (GUI) for processing especially small-scale NGS data to identify variants. IMPI ensures detailed UMI analysis and clustering, as well as initial raw read processing, and consensus sequence building. Furthermore, the effects of custom algorithm and parameter settings for NGS data pre-processing and UMI collapsing (e.g., UMI clustered versus unclustered (raw) reads) can be investigated. Additionally, IMPI implements optimization and quality control methods; an evolution strategy is used for parameter optimization. Results: IMPI was designed, implemented, and tested using BCR::ABL1 fusion gene kinase domain sequencing data. In summary, IMPI enables a detailed analysis of the impact of UMI clustering and parameter setting changes on the measured allele frequencies. Conclusions: Regarding the BCR::ABL1 data, IMPI’s results underlined the need for caution while designing specialized single amplicon NGS approaches due to methodical limitations (e.g., high PCR-mediated recombination rate). This cannot be corrected using UMIs.

IntroductionThe Colorado potato beetle (Leptinotarsa decemlineata) (CPB) is a major threat to potato crops in Europe, Asia, and America. In Europe, it is expected that climate change will cause the species to expand its range northwards and allow for an additional generation per year in regions where it is already established. The effectiveness of insecticides, a common method of control, is decreasing due to the emergence of resistance to several substances. Therefore, research into alternative measures is becoming increasingly necessary. One preventive measure that has already been shown to have great effects in past studies is the use of organic mulch. For organic mulching, conserved or freshly cut plant material is spread on the field usually when the potato emerges. Previous research has mainly used cereal straw for this purpose in plots that are relatively small compared to actual agricultural fields. Materials and methodsTo address the lack of large-scale field trials in the literature, a series of on-station and on-farm field trials were conducted in this study. Organic, legume-rich mulches were used primarily, which so far have not been extensively investigated. Over a period of four years and in a total of six field trials, CPB and larval infestations were assessed. Results and discussionOur results show effects on the initial infestation of young plants by adult CPB and on the emergence time of the first two larval stages. By the time of flowering, when the feeding effect of CPB on yield is known to be greatest, L3 and L4 larvae were reduced by about 30% in the on-station trials and by about 40% in the on-farm trials. In addition, when CPB larval stages were used to calculate a weighted daily leaf consumption per stage, total calculated leaf consumption was on average about 40% lower in the mulch treatments across all sampling dates and all trials. The recorded yields were equal or higher in the mulched treatments. In conclusion, organic mulching can effectively reduce CPB damage even on a large scale and can have a positive impact on yield. Further agronomic benefits of mulching, as well as drawbacks and putative mechanisms of action, are discussed.

Abstract Epstein-Barr virus (EBV) latent membrane protein 1 (LMP1) drives viral B cell transformation and oncogenesis. LMP1’s transforming activity depends on its C-terminal activation region 2 (CTAR2), which induces NF-κB and JNK by engaging TNF receptor-associated factor 6 (TRAF6). The mechanism of TRAF6 recruitment to LMP1 and its role in LMP1 signalling remains elusive. Here we demonstrate that TRAF6 interacts directly with a viral TRAF6 binding motif within CTAR2. Functional and NMR studies supported by molecular modeling provide insight into the architecture of the LMP1-TRAF6 complex, which differs from that of CD40-TRAF6. The direct recruitment of TRAF6 to LMP1 is essential for NF-κB activation by CTAR2 and the survival of LMP1-driven lymphoma. Disruption of the LMP1-TRAF6 complex by inhibitory peptides interferes with the survival of EBV-transformed B cells. In this work, we identify LMP1-TRAF6 as a critical virus-host interface and validate this interaction as a potential therapeutic target in EBV-associated cancer.

Aims: Despite limited clinical scientific backing, an additional trochanteric stabilizing plate (TSP) has been advocated when treating unstable trochanteric fractures with a sliding hip screw (SHS). We aimed to explore whether the TSP would result in less post operative fracture motion, compared to SHS alone. Methods: Overall, 31 patients with AO/OTA 31-A2 trochanteric fractures were randomized to either a SHS alone or a SHS with an additional TSP. To compare postoperative fracture motion, radiostereometric analysis (RSA) was performed before and after weightbearing, and then at four, eight, 12, 26, and 52 weeks. With the “after weightbearing” images as baseline, we calculated translations and rotations, including shortening and medialization of the femoral shaft. Results: Similar migration profiles were observed in all directions during the course of healing. At one year, eight patients in the SHS group and 12 patients in the TSP group were available for analysis, finding a clinically non-relevant, and statistically non-significant, difference in total translation of 1 mm (95% confidence interval -4.7 to 2.9) in favour of the TSP group. In line with the migration data, no significant differences in clinical outcomes were found. Conclusion: The TSP did not influence the course of healing or postoperative fracture motion compared to SHS alone. Based on our results, routine use of the TSP in AO/OTA 31-A2 trochanteric fractures cannot be recommended. The TSP has been shown, in biomechanical studies, to increase stability in sliding hip screw constructs in both unstable and intermediate stable trochanteric fractures, but the clinical evidence is limited. This study showed no advantage of the TSP in unstable (AO 31-A2) fractures in elderly patients when fracture movement was evaluated with radiostereometric analysis. Abstract Histopathological examination of tissue samples is essential for identifying tumor malignancy and the diagnosis of different types of tumor. In the case of lymphoma classification, nuclear size of the neoplastic lymphocytes is one of the key features to differentiate the different subtypes. Based on the combination of artificial intelligence and advanced image processing, we provide a workflow for the classification of lymphoma with regards to their nuclear size (small, intermediate, and large). As the baseline for our workflow testing, we use a Unet++ model trained on histological images of canine lymphoma with individually labeled nuclei. As an alternative to the Unet++, we also used a publicly available pre-trained and unmodified instance segmentation model called Stardist to demonstrate that our modular classification workflow can be combined with different types of segmentation models if they can provide proper nuclei segmentation. Subsequent to nuclear segmentation, we optimize algorithmic parameters for accurate classification of nuclear size using a newly derived reference size and final image classification based on a pathologists-derived ground truth. Our image classification module achieves a classification accuracy of up to 92% on canine lymphoma data. Compared to the accuracy ranging from 66.67 to 84% achieved using measurements provided by three individual pathologists, our algorithm provides a higher accuracy level and reproducible results. Our workflow also demonstrates a high transferability to feline lymphoma, as shown by its accuracy of up to 84.21%, even though our workflow was not optimized for feline lymphoma images. By determining the nuclear size distribution in tumor areas, our workflow can assist pathologists in subtyping lymphoma based on the nuclei size and potentially improve reproducibility. Our proposed approach is modular and comprehensible, thus allowing adaptation for specific tasks and increasing the users’ trust in computer-assisted image classification.

Abstract The intermediate-conductance calcium-activated potassium channel KCa3.1 has been proposed to be a new potential target for glioblastoma treatment. This study analyzed the effect of combined irradiation and KCa3.1-targeting with TRAM-34 in the syngeneic, immune-competent orthotopic SMA-560/VM/Dk glioma mouse model. Whereas neither irradiation nor TRAM-34 treatment alone meaningfully prolonged the survival of the animals, the combination significantly prolonged the survival of the mice. We found an irradiation-induced hyperinvasion of glioma cells into the brain, which was inhibited by concomitant TRAM-34 treatment. Interestingly, TRAM-34 did neither radiosensitize nor impair SMA-560’s intrinsic migratory capacities in vitro. Exploratory findings hint at increased TGF-β1 signaling after irradiation. On top, we found a marginal upregulation of MMP9 mRNA, which was inhibited by TRAM-34. Last, infiltration of CD3+, CD8+ or FoxP3+ T cells was not impacted by either irradiation or KCa3.1 targeting and we found no evidence of adverse events of the combined treatment. We conclude that concomitant irradiation and TRAM-34 treatment is efficacious in this preclinical glioma model.

Abstract In multiple myeloma spatial differences in the subclonal architecture, molecular signatures and composition of the microenvironment remain poorly characterized. To address this shortcoming, we perform multi-region sequencing on paired random bone marrow and focal lesion samples from 17 newly diagnosed patients. Using single-cell RNA- and ATAC-seq we find a median of 6 tumor subclones per patient and unique subclones in focal lesions. Genetically identical subclones display different levels of spatial transcriptional plasticity, including nearly identical profiles and pronounced heterogeneity at different sites, which can include differential expression of immunotherapy targets, such as CD20 and CD38. Macrophages are significantly depleted in the microenvironment of focal lesions. We observe proportional changes in the T-cell repertoire but no site-specific expansion of T-cell clones in intramedullary lesions. In conclusion, our results demonstrate the relevance of considering spatial heterogeneity in multiple myeloma with potential implications for models of cell-cell interactions and disease progression.

In this study, we investigate an innovative option for the ecological management of agricultural land. The focus is on the use of phase change materials (PCMs) for passive temperature regulation in greenhouses and fruit crop fields in order to reduce yield losses due to unforeseen late frost events. The use of PCMs represents a novel approach to enhancing crop growth and extending growing seasons without relying on conventional energy-intensive methods, providing a stable microclimate that can protect plants from cold stress. This passive regulation of temperature helps to reduce the need for fossil fuel-based heating systems, thereby lowering greenhouse gas emissions and operational costs. The application of PCMs in agricultural settings is particularly innovative as it leverages naturally occurring temperature variations to create a self-sustaining, low-maintenance solution that aligns with the principles of sustainable farming. This approach not only improves energy efficiency but also contributes to the resilience of agricultural practices in the face of climate variability. This study focuses on the possible use of PCMs in passive heating modules for the protection of potted plants in greenhouses. Various PCMs such as paraffin, beeswax, and shea butter were tested. Experiments were then conducted, using one kind of paraffin-based PCM, in a specially designed module. In addition, an FEM simulation model (CFD) was built and tested. The model was used to perform detailed analyses of the heat transfer efficiency, fluid dynamics, and overall performance of the modules. The model can also be used for optimization purposes (e.g., efficiency improvements).

Conflicts between human populations and Nile crocodiles are widespread with crocodiles posing significant threats to fisherfolk and riverine communities across r-Saharan Africa. Hundreds of deadly attacks take place annually, and mortality rates may range from 50% to 100%. Attitudes and perceptions towards crocodiles were studied using structured questionnaires among fisherfolk along the River Nile and the Sudd wetlands in South Sudan. Local communities used crocodiles for their meat and skin/leather trades. The meat is regarded to enhance longevity, sexual potency, and protection against witchcraft. Crocodiles are perceived as a main threat to lives and livelihoods as they restrict people’s freedom of movement along water bodies, attack livestock and humans, and devastate fishing equipment. To assess whether responses were influenced by the intensity of crocodile threats, published data on fatal crocodile attacks on humans and livestock were analysed using Generalised Linear Models (GLMs). This analysis indicated a direct link between the number of crocodile attacks and human attitudes. Crocodiles were generally feared and hated, and there was the agreement of the need to destroy breeding habitats. However, some attitudes were complex and nuanced as highlighted by the agreement of local communities on the need to destroy Nile Crocodile breeding habitats on the one hand and the need to establish crocodile sanctuaries as the the preferred strategy to mitigate risks and conflict on the other hand. There is a need for the creation of a crocodile sanctuary in the Sudd wetlands to minimise the risks of illegal hunting and to buffer the increasing pressure on crocodiles due to human population growth and economic upturn after the civil war.

The integration of deep learning-based tools into diagnostic workflows is increasingly prevalent due to their efficiency and reproducibility in various settings. We investigated the utility of automated nuclear morphometry for assessing nuclear pleomorphism (NP), a criterion of malignancy in the current grading system in canine pulmonary carcinoma (cPC), and its prognostic implications. We developed a deep learning-based algorithm for evaluating NP (variation in size, i.e., anisokaryosis and/or shape) using a segmentation model. Its performance was evaluated on 46 cPC cases with comprehensive follow-up data regarding its accuracy in nuclear segmentation and its prognostic ability. Its assessment of NP was compared to manual morphometry and established prognostic tests (pathologists’ NP estimates (n = 11), mitotic count, histological grading, and TNM-stage). The standard deviation (SD) of the nuclear area, indicative of anisokaryosis, exhibited good discriminatory ability for tumor-specific survival, with an area under the curve (AUC) of 0.80 and a hazard ratio (HR) of 3.38. The algorithm achieved values comparable to manual morphometry. In contrast, the pathologists’ estimates of anisokaryosis resulted in HR values ranging from 0.86 to 34.8, with slight inter-observer reproducibility (k = 0.204). Other conventional tests had no significant prognostic value in our study cohort. Fully automated morphometry promises a time-efficient and reproducible assessment of NP with a high prognostic value. Further refinement of the algorithm, particularly to address undersegmentation, and application to a larger study population are required.

The sedentarization of Pygmies in the Congo Basin has triggered a profound transformation in their traditional lifestyles, particularly affecting dietary habits and food consumption. We employed 24-hour dietary recalls in 10 sedentarized Baka Pygmy villages in southeastern Cameroon, gathering data on diet composition, diversity (Household Dietary Diversity Score, HDDS), and nutrient intake per adult male equivalent (AME) from 67 homes (28% of all households). Our findings revealed that 62% of consumed foods were agricultural produce, 29% were locally produced or purchased products, and the remaining 9% comprised items sourced or hunted from the wild. The average HDDS per village was low (4.1±1.56) and mean total energy intake was 1734.9±1,031.8 kcal/AME, with significant contributions from cultivated foods. There was a negative correlation between the consumption of cultivated and wild foods. Moreover, a considerable proportion of households (78.7%, ranging from 22.4% to 97%) exhibited nutrient consumption below the lower 95% uncertainty interval found in a Cameroonian nutrient supply study. Additionally, 78.3% of respondents fell below WHO/FAO recommendations for 21 nutrients, even after adjusting for the Baka’s shorter stature. This high prevalence of insufficient nutrient intake underscores the urgent need for targeted interventions to address nutritional deficiencies within this population. We show Baka households rely more on cultivated foods and are less dependent on wild sources. Understanding the profound transformation in dietary patterns and its repercussions on the health and overall well-being of the studied marginalized Indigenous communities is pivotal in devising strategies to enhance their survival. This shift in dietary profiles often stems from complex factors, including socioeconomic challenges, environmental changes, and cultural shifts. To address these issues effectively, a comprehensive approach that integrates cultural sensitivity, community engagement, and sustainable practices is imperative.

Summary: Background: Minimally invasive fusion of the sacroiliac joint as treatment for low back pain may reduce pain and improve function compared to non-operative treatment, although clear evidence is lacking. The aim of this trial was to evaluate the effect of minimally invasive sacroiliac joint fusion compared to sham surgery on sacroiliac joint pain reduction. Methods: In this double-blind randomized sham surgery-controlled trial patients with clinical diagnosis of sacroiliac joint pain confirmed with sacroiliac joint injection were included at two university hospitals in Sweden and Norway. Patients were randomized by the operating surgeon at each site to minimally invasive sacroiliac joint fusion or sham surgery. The primary endpoint was group difference in sacroiliac joint pain on the operated side at six months postoperatively, measured by the Numeric Rating Scale (0–10). Un-blinding and primary analysis were performed when all patients had completed six months follow-up. The trial is closed for new participants and was registered at clinicaltrials.gov: NCT03507049. Findings: Between September 1st, 2018 and October 22nd, 2021, 63 patients were randomized, 32 to the surgical group, 31 to the sham group. Mean age was 45 years (range 26–63) and 59 of 63 (94%) patients were female. The mean reduction in the operated sacroiliac joint from baseline to six months postoperative was 2.6 Numeric Rating Scale points in the surgical group and 1.7 points in the sham group (mean between groups difference −1.0 points; 95% CI, −2.2 to 0.3; p = 0.13). Interpretation: This double-blind randomized controlled trial could not prove that minimally invasive fusion of the sacroiliac joint was superior to sham surgery at six months postoperative. The computational detection of R-peaks in an electrocardiogram (ECG) is particularly important for assessing vital signs in ambulatory settings. Algorithmic approaches are mainly open source and available for research purposes, primarily in the Python environment on stationary computers. This motivates the need for more support of mobile environments and their particular demands, such as context awareness and their limited computational resources.PeakSwift is a comprehensive Swift package designed for detecting R-peaks in single-lead ECG, tailored for the iOS environment. Its core functional features include on-device R-peak detection algorithms, quality assessment, and context-aware algorithm selection. PeakSwift is lightweight, extensible and modular. To evaluate its performance, a dedicated iOS application was developed, and a set of experiments was conducted using over 10,000 single-lead ECGs.The results demonstrated that PeakSwift achieves high reproducibility in detecting R-peaks compared to NeuroKit. In addition, its computational runtime was competitive.

The ongoing controversies about the neural basis of tinnitus, whether linked with central neural gain or not, may hamper efforts to develop therapies. We asked to what extent measurable audiometric characteristics of tinnitus without (T) or with co-occurrence of hyperacusis (TH) are distinguishable on the level of cortical responses. To accomplish this, electroencephalography (EEG) and concurrent functional near-infrared spectroscopy (fNIRS) were measured while patients performed an attentionally demanding auditory discrimination task using stimuli within the individual tinnitus frequency (fTin) and a reference frequency (fRef). Resting-state-fMRI-based functional connectivity (rs-fMRI-bfc) in ascending auditory nuclei (AAN), the primary auditory cortex (AC-I), and four other regions relevant for directing attention or regulating distress in temporal, parietal, and prefrontal cortex was compiled and compared to EEG and concurrent fNIRS activity in the same brain areas. We observed no group differences in pure-tone audiometry (PTA) between 10 and 16 kHz. However, the PTA threshold around the tinnitus pitch was positively correlated with the self-rated tinnitus loudness and also correlated with distress in T-groups, while TH experienced their tinnitus loudness at minimal loudness levels already with maximal suffering scores. The T-group exhibited prolonged auditory brain stem (ABR) wave I latency and reduced ABR wave V amplitudes (indicating reduced neural synchrony in the brainstem), which were associated with lower rs-fMRI-bfc between AAN and the AC-I, as observed in previous studies. In T-subjects, these features were linked with elevated spontaneous and reduced evoked gamma oscillations and with reduced deoxygenated hemoglobin (deoxy-Hb) concentrations in response to stimulation with lower frequencies in temporal cortex (Brodmann area (BA) 41, 42, 22), implying less synchronous auditory responses during active auditory discrimination of reference frequencies. In contrast, in the TH-group gamma oscillations and hemodynamic responses in temporoparietal regions were reversed during active discrimination of tinnitus frequencies. Our findings suggest that T and TH differ in auditory discrimination and memory-dependent directed attention during active discrimination at either tinnitus or reference frequencies, offering a test paradigm that may allow for more precise sub-classification of tinnitus and future improved treatment approaches.

We present two-dimensional near-infrared temperature maps of the canonical hot Jupiter WASP-43b using a phase-curve observation with JWST NIRSpec/G395H. From the white-light planetary transit, we improve constraints on the planet’s orbital parameters and measure a planet-to-star radius ratio of ${0.15883}_{-0.00053}^{+0.00056}$ . Using the white-light phase curve, we measure a longitude of maximum brightness of ${6.9}_{-0\buildrel{\circ}\over{.} 5}^{+0\buildrel{\circ}\over{.} 5}$ east of the substellar point and a phase-curve offset of ${10.0}_{-0\buildrel{\circ}\over{.} 8}^{+0\buildrel{\circ}\over{.} 8}$ . We also find a ≈4 σ detection of a latitudinal hotspot offset of $-{13.4}_{-1\buildrel{\circ}\over{.} 7}^{+3\buildrel{\circ}\over{.} 2}$ , the first significant detection of a nonequatorial hotspot in an exoplanet atmosphere. We show that this detection is robust to variations within planetary parameter uncertainties, but only if the transit is used to improve constraints, showing the importance of transit observations to eclipse mapping. Maps retrieved from the NRS1 and NRS2 detectors are similar, with hotspot locations consistent between the two detectors at the 1 σ level. Our JWST data show brighter (hotter) nightsides and a dimmer (colder) dayside at the shorter wavelengths relative to fits to Spitzer 3.6 and 4.5 μ m phase curves. Through comparison between our phase curves and a set of general circulation models, we find evidence for clouds on the nightside and atmospheric drag or high metallicity reducing the eastward hotspot offset.

L 98-59 d is a Super-Earth planet orbiting an M-type star. We performed retrievals on the transmission spectrum of L 98-59 d obtained using NIRSpec G395H during a single transit, from JWST Cycle 1 GTO 1224. The wavelength range of this spectrum allows us to detect the presence of several atmospheric species. We found that the spectrum is consistent with a high mean molecular weight atmosphere. The atmospheric spectrum indicates the possible presence of the sulfur-bearing species H _2 S and SO _2 , which could hint at active volcanism on this planet if verified by future observations. We also tested for signs of stellar contamination in the spectrum and found signs of unocculted faculae on the star. The tentative signs of an atmosphere on L 98-59 d presented in this work from just one transit bodes well for possible molecular detections in the future, particularly as it is one of the best targets among small exoplanets for atmospheric characterization using JWST.

Detecting atmospheres around planets with a radius below 1.6 R _⊕ , commonly referred to as rocky planets, has proven to be challenging. However, rocky planets orbiting M dwarfs are ideal candidates due to their favorable planet-to-star radius ratio. Here, we present one transit observation of the Super-Earth L98-59 d (1.58 R _⊕ and 2.31 M _⊕ ), at the limit of rocky/gas-rich, using the JWST NIRSpec G395H mode covering the 2.8–5.1 μ m wavelength range. The extracted transit spectrum from a single transit observation deviates from a flat line by 2.6 σ –5.6 σ , depending on the data reduction and retrieval setup. The hints of an atmospheric detection are driven by a large absorption feature between 3.3 and 4.8 μ m. A stellar contamination retrieval analysis rejected the source of this feature as being due to stellar inhomogeneities, making the best fit an atmospheric model including sulfur-bearing species, suggesting that the atmosphere of L98-59 d may not be at equilibrium. This result will need to be confirmed by the analysis of the second NIRSpec G395H visit in addition to the NIRISS SOSS transit observation.

Diffuse reflectance spectroscopy has been extensively employed to deliver timely and cost-effective predictions of a number of soil properties. However, although several soil spectral laboratories have been established worldwide, the distinct characteristics of instruments and operations still hamper further integration and interoperability across mid-infrared (MIR) soil spectral libraries. In this study, we conducted a large-scale ring trial experiment to understand the lab-to-lab variability of multiple MIR instruments. By developing a systematic evaluation of different mathematical treatments with modeling algorithms, including regular preprocessing and spectral standardization, we quantified and evaluated instruments' dissimilarity and how this impacts internal and shared model performance. We found that all instruments delivered good predictions when calibrated internally using the same instruments' characteristics and standard operating procedures by solely relying on regular spectral preprocessing that accounts for light scattering and multiplicative/additive effects, e.g., using standard normal variate (SNV). When performing model transfer from a large public library (the USDA NSSC-KSSL MIR library) to secondary instruments, good performance was also achieved by regular preprocessing (e.g., SNV) if both instruments shared the same manufacturer. However, significant differences between the KSSL MIR library and contrasting ring trial instruments responses were evident and confirmed by a semi-unsupervised spectral clustering. For heavily contrasting setups, spectral standardization was necessary before transferring prediction models. Non-linear model types like Cubist and memory-based learning delivered more precise estimates because they seemed to be less sensitive to spectral variations than global partial least square regression. In summary, the results from this study can assist new laboratories in building spectroscopy capacity utilizing existing MIR spectral libraries and support the recent global efforts to make soil spectroscopy universally accessible with centralized or shared operating procedures.

Federated learning (FL) enables the optimization of machine learning models on distributed clients without sharing local data. The integration of FL into a mobile environment is becoming more feasible due to increasing on-device processing capabilities. However, there is limited open-source support for the iOS platform.The article introduces a Swift-based client implementation of the user-friendly FL framework Flower. The objective is facilitating FL client processes based on a modular and easy-to-integrate software development kit.A benchmark test demonstrates consistent stability and performance using the software, further motivating its use for research.

BackgroundSensory overload and sensory deprivation have both been associated with negative health outcomes in critically ill patients. While there is a lack of any clear treatment or prevention strategies, immersive virtual reality is a promising tool for addressing such problems, but which has not been repetitively tested in random samples. Therefore, this study aimed to determine how critically ill patients react to repeated sessions of immersive virtual reality.MethodsThis exploratory study was conducted in the mixed medical–surgical intermediate care unit of the University Hospital of Bern (Inselspital). Participants (N = 45; 20 women, 25 men; age = 57.73 ± 15.92 years) received two immersive virtual reality sessions via a head-mounted display and noise-canceling headphones within 24 h during their stay in the unit. Each session lasted 30-min and showed a 360-degree nature landscape. Abstract Stress is a part of everyday life which can be counteracted by evoking the relaxation response via nature scenes presented using immersive virtual reality (VR). The aim of this study was to determine which sensory aspect of immersive VR intervention is responsible for the greatest relaxation response. We compared four conditions: auditory and visual combined (audiovisual), auditory only, visual only, and no artificial sensory input. Physiological changes in heart rate, respiration rate, and blood pressure were recorded, while participants reported their preferred condition and awareness of people, noise, and light in the real-world. Over the duration of the stimulation, participants had the lowest heart rate during the audiovisual and visual only conditions. They had the steadiest decrease in respiration rate and the lowest blood pressure during the audiovisual condition, compared to the other conditions, indicating the greatest relaxation. Background Critical illness may lead to activation of the sympathetic system. The sympathetic stimulation may be further increased by exogenous catecholamines, such as vasopressors and inotropes. Excessive adrenergic stress has been associated with organ dysfunction and higher mortality. β-Blockers may reduce the adrenergic burden, but they may also compromise perfusion to vital organs thus worsening organ dysfunction. To assess the effect of treatment with β-blockers in critically ill adults, we conducted a systematic review and meta-analysis of randomized controlled trials.Materials and methods We conducted a search from three major databases: Ovid Medline, the Cochrane Central Register for Controlled Trials and Scopus database. Two independent reviewers screened, selected, and assessed the included articles according to prespecified eligibility criteria. We assessed risk of bias of eligible articles according to the Cochrane guidelines. AimZinc deficiencies are widespread in many soils, limiting crop growth and contributing to Zn deficiencies in human diets. This study aimed at understanding soil factors influencing grain Zn concentrations and uptake of crops grown in different landscape positions in West Amhara, Ethiopia.MethodsOn-farm experiments were conducted in three landscape positions, with five farmers’ fields as replicates in each landscape position, and at three sites. Available Zn from the soil (Mehlich 3, M3, Zn) and applied fertilizer (NET_FERT Zn, estimated based on adsorption/desorption characteristics and applied Zn) were related to the actual grain Zn concentration and uptake of teff, wheat, and maize. Zinc fertilizer treatments tested were Zn applied at planting (basal), basal plus side dressing and a control with no Zn applied.ResultsZn treatments had a significant effect on grain Zn concentration (increase by up to 10%) but the effect on grain yield was variable. Differences in crop Zn concentrations along the landscape positions were observed but not at all sites and crops. Trial results showed that soils with higher soil pH and Soil Organic Carbon (SOC) (typical of footslope landscape positions) tended to adsorb more applied Zn (reduce NET_FERT Zn) than soils with lower soil pH and SOC (typical of upslope landscape positions). Zn availability indicators (M3, NET_FERT Zn, clay%) explained 14-52% of the observed variation in grain Zn concentrations, whereas macronutrient indicators (Total N, exchangeable K) together with M3 Zn were better in predicting grain Zn uptake (16 to 32% explained variability). Maize had the lowest grain Zn concentrations but the highest grain Zn uptake due to high yields.ConclusionWe found that the sum of indigenous and fertilizer Zn significantly affects grain Zn loadings of cereals and that the associated soil parameters differ between and within landscape positions. Therefore, knowledge of soil properties and crop characteristics helps to understand where agronomic biofortification can be effective.

Background Left ventricular (LV) global longitudinal strain (GLS) provides incremental prognostic information over LV ejection fraction in patients with heart failure (HF) and secondary mitral regurgitation. We examined the prognostic impact of LV GLS improvement in this population. Methods and Results The COAPT (Cardiovascular Outcomes Assessment of the MitraClip Percutaneous Therapy for Heart Failure Patients With Functional Mitral Regurgitation) trial randomized symptomatic patients with HF with severe (3+/4+) mitral regurgitation to transcatheter edge‐to‐edge repair with the MitraClip device plus maximally tolerated guideline‐directed medical therapy (GDMT) versus GDMT alone. LV GLS was measured at baseline and 6‐month follow‐up. The relationship between the improvement in LV GLS from baseline to 6 months and the composite of all‐cause death or HF hospitalization between 6‐ and 24‐month follow‐up were assessed. Among 383 patients, 174 (45.4%) had improved LV GLS at 6‐month follow‐up (83/195 [42.6%] with transcatheter edge‐to‐edge repair+GDMT and 91/188 [48.4%] with GDMT alone; P=0.25). Abstract Autonomic and vascular failures are common phenotypes of sepsis, typically characterized by tachycardia despite corrected hypotension/hypovolemia, vasopressor resistance, increased arterial stiffness and decreased peripheral vascular resistance. In a 5-day swine experiment of polymicrobial sepsis we aimed at characterizing arterial properties and autonomic mechanisms responsible for cardiovascular homeostasis regulation, with the final goal to verify whether the resuscitation therapy in agreement with standard guidelines was successful in restoring a physiological condition of hemodynamic profile, cardiovascular interactions and autonomic control. Twenty pigs were randomized to polymicrobial sepsis and protocol-based resuscitation or to prolonged mechanical ventilation and sedation without sepsis. The animals were studied at baseline, after sepsis development, and every 24 h during the 3-days resuscitation period. Beat-to-beat carotid blood pressure (BP), carotid blood flow, and central venous pressure were continuously recorded. The two-element Windkessel model was adopted to study carotid arterial compliance, systemic vascular resistance and characteristic time constant τ. Effective arterial elastance was calculated as a simple estimate of total arterial load. Cardiac baroreflex sensitivity (BRS) and low frequency (LF) spectral power of diastolic BP were computed to assess autonomic activity. Sepsis induced significant vascular and autonomic alterations, manifested as increased arterial stiffness, decreased vascular resistance and τ constant, reduced BRS and LF power, higher arterial afterload and elevated heart rate in septic pigs compared to sham animals. This compromised condition was persistent until the end of the experiment, despite achievement of recommended resuscitation goals by administered vasopressors and fluids. Vascular and autonomic alterations persist 3 days after goal-directed resuscitation in a clinically relevant sepsis model. We hypothesize that the addition of these variables to standard clinical markers may better profile patients’ response to treatment and this could drive a more tailored therapy which could have a potential impact on long-term outcomes.

Abstract Subarachnoid hemorrhage (SAH) is a serious condition, and a myocardial injury or dysfunction could contribute to the outcome. We assessed the prevalence and prognostic impact of cardiac involvement in a cohort with SAH. This is a prospective observational multicenter study. We included 192 patients treated for non-traumatic subarachnoid hemorrhage. We performed ECG recordings, echocardiographic examinations, and blood sampling within 24 h of admission and on days 3 and 7 and at 90 days. The primary endpoint was the evidence of cardiac involvement at 90 days, and the secondary endpoint was to examine the prevalence of a myocardial injury or dysfunction. The median age was 54.5 (interquartile range [IQR] 48.0–64.0) years, 44.3% were male and the median World Federation of Neurological Surgeons (WFNS) score was 2 (IQR 1–4). At day 90, 22/125 patients (17.6%) had left ventricular ejection fractions ≤ 50%, and 2/121 patients (1.7%) had evidence of a diastolic dysfunction as defined by mitral peak E-wave velocity by peak eʹ velocity (E/eʹ) > 14. There was no prognostic impact from echocardiographic evidence of cardiac complications on neurological outcomes. The overall prevalence of cardiac dysfunction was modest. We found no demographic or SAH-related factors associated with 90 days cardiac dysfunction.

Abstract In Covid-19, anticoagulation with heparin is often administered to prevent or treat thromboembolic events. Heparin-induced thrombocytopenia (HIT) is a severe complication of heparin treatment, caused by heparin-dependent, platelet activating anti-platelet factor 4 (PF4)/heparin antibodies. Diagnosis of HIT is based on the combination of clinical parameters, allowing to determine the pretest probability, and laboratory testing for anti-PF4/heparin antibodies and confirmatory functional assays, such as the heparin-induced platelet activation (HIPA) test. We report the case of a patient with severe Covid-19 pneumonia requiring ECMO treatment, who developed recurrent clotting of the ECMO filter and a drop in platelet count under heparin treatment. He was therefore suspected to have HIT and the anticoagulation was switched to argatroban. Despite high clinical probability and high titres of anti-PF4/heparin antibodies, the functional HIPA test was negative. Nevertheless, argatroban was continued rather than to reinstate anticoagulation with heparin. Reevaluation 7 days later then demonstrated a strongly positive functional HIPA test and confirmed the diagnosis of HIT. Under anticoagulation with argatroban the patient gradually improved and was finally weaned off the ECMO. In conclusion, this case highlights the critical importance of clinical judgement, exploiting the 4 T score, given that Covid-19 patients may present a different pattern of routine laboratory test results in HIT diagnostics. The possibility of a false negative HIPA test has to be considered, particularly in early phases of presentation. In cases of a discrepancy with high clinical probability of HIT and/or high titre anti-PF4/heparin antibodies despite a negative HIPA test, a reevaluation within 3 to 5 days after the initial test should be considered in order to avoid precipitant reestablishment of unfractionated heparin, with potentially fatal consequences.

BackgroundExposure to elevated sound pressure levels within the intensive care unit is known to negatively affect patient and staff health. In the past, interventions to address this problem have been unsuccessful as there is no conclusive evidence on the severity of each sound source and their role on the overall sound pressure levels. Therefore, the goal of the study was to perform a continuous 1 week recording to characterize the sound pressure levels and identify negative sound sources in this setting.MethodsIn this prospective, systematic, and quantitative observational study, the sound pressure levels and sound sources were continuously recorded in a mixed medical–surgical intensive care unit over 1 week. Abstract Using connected sensing devices to remotely monitor health is a promising way to help transition healthcare from a rather reactive to a more precision medicine oriented proactive approach, which could be particularly relevant in the face of rapid population ageing and the challenges it poses to healthcare systems. Sensor derived digital measures of health, such as digital biomarkers or digital clinical outcome assessments, may be used to monitor health status or the risk of adverse events like falls. Current research around such digital measures has largely focused on exploring the use of few individual measures obtained through mobile devices. However, especially for long-term applications in older adults, this choice of technology may not be ideal and could further add to the digital divide. Moreover, large-scale systems biology approaches, like genomics, have already proven beneficial in precision medicine, making it plausible that the same could also hold for remote-health monitoring. In this context, we introduce and describe a zero-interaction digital exhaust: a set of 1268 digital measures that cover large parts of a person’s activity, behavior and physiology. Making this approach more inclusive of older adults, we base this set entirely on contactless, zero-interaction sensing technologies. Applying the resulting digital exhaust to real-world data, we then demonstrate the possibility to create multiple ageing relevant digital clinical outcome assessments. Paired with modern machine learning, we find these assessments to be surprisingly powerful and often on-par with mobile approaches. Lastly, we highlight the possibility to discover novel digital biomarkers based on this large-scale approach.

While working on an identification guide to the sawflies of Fennoscandia, we encountered numerous taxonomic problems, for some of which we present solutions. Dicrostema Benson, 1952 is a new synonym of Phymatoceropsis Rohwer, 1916, and not congeneric with Paracharactus MacGillivray, 1908. Two species occurring in Europe are transferred to Phymatoceropsis. Dolerus aericepsellus Heidemaa and Mutanen sp. nov. and Heptamelus viitasaarii Liston, Mutanen and Prous sp. nov. are described from Finland. Abia brevicornis Leach, 1817 nom. rev. is the valid name of Abia nitens auct. nec Linnaeus, and Abia nitens (Linnaeus, 1758) is the valid name for what has recently been called Abia sericea (Linnaeus, 1767). Tenthredo haemorrhoidalis Fabricius, 1781 is treated as an unplaced species of Hymenoptera, possibly Ichneumonoidea. Calameuta variabilis (Mocsáry, 1886) is the valid name of the species recently generally called C. haemorrhoidalis. Abstract Background It remains elusive how the characteristics, the course of disease, the clinical management and the outcomes of critically ill COVID-19 patients admitted to intensive care units (ICU) worldwide have changed over the course of the pandemic. Methods Prospective, observational registry constituted by 90 ICUs across 22 countries worldwide including patients with a laboratory-confirmed, critical presentation of COVID-19 requiring advanced organ support. Abstract Bee pollen is recommended as dietary supplement due to immunostimulating functions including antioxidant, anti-inflammatory and anti-carcinogenic properties. Nevertheless, the effectiveness of such properties is still not well understood. As diet can be associated with animal performance, microbiota modulation and potentially factor for cancer, this study aimed to analyze if bee pollen could influence growth, gut microbial and skin cutaneous melanoma development in zebrafish. Control diets based on commercial flakes and Artemia were compared with the same diet supplemented with bee pollen. Fish weight gain, increased length, intestinal bacteria metagenomics analysis, serum amyloid A gene expression and cutaneous melanoma transplantation assays were performed. Bee pollen affected microbiota composition and melanoma development. Differential abundance revealed higher abundance in the control group for Aeromonadaceae family, Aeromonas and Pseudomonas genus, A. sobria, A. schubertii, A. jandaei and P. alcaligenes species compared with pollen diet group. Pollen group presented higher abundance for Chromobacterium genus and for Gemmobacter aquaticus, Flavobacterium succinicans and Bifidobacterium breve compared with control group. Unexpectedly, fish fed with bee pollen showed higher tumor growth rate and larger tumor size than control group. This is the first study to report intestinal microbial changes and no protective cancer properties after bee pollen administration.

Abstract Background The COVID‐19 pandemic has greatly increased the incidence and clinical importance of critical illness myopathy (CIM), because it is one of the most common complications of modern intensive care medicine. Current diagnostic criteria only allow diagnosis of CIM at an advanced stage, so that patients are at risk of being overlooked, especially in early stages. To determine the frequency of CIM and to assess a recently proposed tool for early diagnosis, we have followed a cohort of COVID‐19 patients with acute respiratory distress syndrome and compared the time course of muscle excitability measurements with the definite diagnosis of CIM. Methods Adult COVID‐19 patients admitted to the Intensive Care Unit of the University Hospital Bern, Switzerland requiring mechanical ventilation were recruited and examined on Days 1, 2, 5, and 10 post‐intubation. Clinical examination, muscle excitability measurements, medication record, and laboratory analyses were performed on all study visits, and additionally nerve conduction studies, electromyography and muscle biopsy on Day 10. Muscle excitability data were compared with a cohort of 31 age‐matched healthy subjects. Diagnosis of definite CIM was made according to the current guidelines and was based on patient history, results of clinical and electrophysiological examinations as well as muscle biopsy. Results Complete data were available in 31 out of 44 recruited patients (mean [SD] age, 62.4 [9.8] years). Of these, 17 (55%) developed CIM. Muscle excitability measurements on Day 10 discriminated between patients who developed CIM and those who did not, with a diagnostic precision of 90% (AUC 0.908; 95% CI 0.799–1.000; sensitivity 1.000; specificity 0.714). On Days 1 and 2, muscle excitability parameters also discriminated between the two groups with 73% (AUC 0.734; 95% CI 0.550–0.919; sensitivity 0.562; specificity 0.857) and 82% (AUC 0.820; CI 0.652–0.903; sensitivity 0.750; specificity 0.923) diagnostic precision, respectively. All critically ill COVID‐19 patients showed signs of muscle membrane depolarization compared with healthy subjects, but in patients who developed CIM muscle membrane depolarization on Days 1, 2 and 10 was more pronounced than in patients who did not develop CIM. Conclusions This study reports a 55% prevalence of definite CIM in critically ill COVID‐19 patients. Furthermore, the results confirm that muscle excitability measurements may serve as an alternative method for CIM diagnosis and support its use as a tool for early diagnosis and monitoring the development of CIM.

Gait abnormalities in older adults are linked to increased risks of falls, institutionalization, and mortality, necessitating accurate and frequent gait assessments beyond traditional clinical settings. Current methods, such as pressure-sensitive walkways, often lack the continuous natural environment monitoring needed to understand an individual’s gait fully during their daily activities. To address this gap, we present a Lidar-based method capable of unobtrusively and continuously tracking human leg movements in diverse home-like environments, aiming to match the accuracy of a clinical reference measurement system. We developed a calibration-free step extraction algorithm based on mathematical morphology to realize Lidar-based gait analysis. Clinical gait parameters of 45 healthy individuals were measured using Lidar and reference systems (a pressure-sensitive walkway and a video recording system). Each participant participated in three predefined ambulation experiments by walking over the walkway. We observed linear relationships with strong positive correlations (R2>0.9) between the values of the gait parameters (step and stride length, step and stride time, cadence, and velocity) measured with the Lidar sensors and the pressure-sensitive walkway reference system. Moreover, the lower and upper 95% confidence intervals of all gait parameters were tight. The proposed algorithm can accurately derive gait parameters from Lidar data captured in home-like environments, with a performance not significantly less accurate than clinical reference systems.

A series of syringaldehyde imines with para-substituted anilines have been synthesized in a good yield, and their crystal structures have been analyzed. The orientation of the syringaldehyde hydroxyl group plays in important role in the intermolecular hydrogen-bonding pattern of the molecules. The O–H…N hydrogen bonding interactions primarily determine the three-dimensional packing of the molecules, even though they make up a relatively small percentage of intermolecular interactions in the molecules. The three structures with the p-hydroxy group cis to the imine group give hydrogen-bonded zigzag chains in the monoclinic crystals, while the structure with a trans hydroxy group crystallize in a hexagonal space group (R3¯) and form hydrogen-bonded hexamers. The hexagonal structure also displays Br…Br interactions, forming additional hexameric clusters. The analysis of published p-hydroxyphenyl imine crystal structures from the Cambridge Crystallographic Database revealed patterns in the length of the hydrogen bonding interactions based on steric congestion around the hydroxyl group.

Abstract Background Protein truncating variants in ATM, BRCA1, BRCA2, CHEK2, and PALB2 are associated with increased breast cancer risk, but risks associated with missense variants in these genes are uncertain. Methods We analyzed data on 59,639 breast cancer cases and 53,165 controls from studies participating in the Breast Cancer Association Consortium BRIDGES project. We sampled training (80%) and validation (20%) sets to analyze rare missense variants in ATM (1146 training variants), BRCA1 (644), BRCA2 (1425), CHEK2 (325), and PALB2 (472). We evaluated breast cancer risks according to five in silico prediction-of-deleteriousness algorithms, functional protein domain, and frequency, using logistic regression models and also mixture models in which a subset of variants was assumed to be risk-associated. Results The most predictive in silico algorithms were Helix (BRCA1, BRCA2 and CHEK2) and CADD (ATM). Increased risks appeared restricted to functional protein domains for ATM (FAT and PIK domains) and BRCA1 (RING and BRCT domains). For ATM, BRCA1, and BRCA2, data were compatible with small subsets (approximately 7%, 2%, and 0.6%, respectively) of rare missense variants giving similar risk to those of protein truncating variants in the same gene. For CHEK2, data were more consistent with a large fraction (approximately 60%) of rare missense variants giving a lower risk (OR 1.75, 95% CI (1.47–2.08)) than CHEK2 protein truncating variants. There was little evidence for an association with risk for missense variants in PALB2. The best fitting models were well calibrated in the validation set. Conclusions These results will inform risk prediction models and the selection of candidate variants for functional assays and could contribute to the clinical reporting of gene panel testing for breast cancer susceptibility.

Abstract Realistic targets for soil organic carbon (SOC) concentrations are needed, accounting for differences between soils and land uses. We assess the use of SOC/clay ratio for this purpose by comparing changes over time in (a) the National Soil Inventory of England and Wales, first sampled in 1978–1983 and resampled in 1994–2003, and (b) two long-term experiments under ley-arable rotations on contrasting soils in the East of England. The results showed that normalising for clay concentration provides a more meaningful separation between land uses than changes in SOC alone. Almost half of arable soils in the NSI had degraded SOC/clay ratios ( 1/8, respectively. Given the wide range of soils and land uses across England and Wales in the datasets used to test these targets, they should apply across similar temperate regions globally, and at national to sub-regional scales.

Abstract Background Reliable and objective assessment of psychomotor skills in physiotherapy students’ education is essential for direct feedback and skill improvement. The aim of this study is to determine the interrater reliability in the assessment process of physiotherapy students and to analyse the assessment behaviour of the examiners. Methods Physiotherapy teachers from two different schools assessed students from two different schools performing proprioceptive neuromuscular facilitation (PNF) patterns. An evaluation sheet with a 6-point rating scale and 20 evaluation criteria including an overall rating was used for assessment. The interrater reliability was determined calculating an intraclass-correlation coefficient (ICC) and Krippendorff’s alpha. The assessment behaviour of the examiners was further analysed calculating the location parameters and showing the item response distribution over item in form of a Likert plot. Results The ICC estimates were mostly below 0.4, indicating poor interrater reliability. This was confirmed by Krippendorff’s alpha. The examiners showed a certain central tendency and intergroup bias. Discussion and conclusion The interrater reliability in this assessment format was rather low. No difference between the two physiotherapy schools concerning the interrater reliability could be identified. Despite certain limitations of this study, there is a definite need for improvement of the assessment process in physiotherapy education to provide the students with reliable and objective feedback and ensure a certain level of professional competence in the students. Trial registration The study was approved by the ethics committee of the Medical Faculty RWTH Aachen University (EK 340/16).

When houseflies find optimal conditions to develop, they rapidly increase their population size negatively impacting both humans and animals through nuisance and the transmission of pathogens. Musca domestica Linnaeus, 1758 (Diptera: Muscidae) shows a preference for animal faeces and for this reason it is a serious pest in animal breeding facilities. To prevent proliferation of houseflies and to evaluate the efficiency of the control methods, it is important to routinely monitor the activity of this dipteran in such facilities. There are several types of traps for sampling houseflies in breeding sites, here we used colored pan-traps to evaluate the efficiency of the trap and its color in sampling M. domestica in a cage poultry facility located in the city of Nepomuceno, Minas Gerais, Brazil. To do so, we set up yellow, white, blue and red pan-traps next to the cages of poultries and collected flies once a week for a period of a year. Although the density of M. domestica was high during the entire period of sampling, more flies were collected in white (monthly average of 470.83 flies) than in yellow (327.55), blue (267.14) and red traps (199.63). Hence, we argue that this poultry farm needs to improve its housefly monitoring program and we suggest a continuous use of white pan-traps to monitor the efficiency of the pest control methods implemented in the facility.

Rotational rice systems, involving pastures, other crops and/or livestock, are common in temperate South America, exemplified by the rice-pasture-livestock system of Uruguay which combines very high rice yields with tight nitrogen (N) balances. The generally good nutrient use efficiency in these systems provides a template for nutrient management in other mixed farming systems, if the underlying processes can be sufficiently well quantified and understood. Here, we studied N balances in rice–non-rice rotations in a long-term experiment in Uruguay, with the aim of parameterizing and testing the DNDC model of N dynamics for such systems for use in future work. The experiment includes three rotations: continuous rice (RI-CONT), rice-soybean (RI-SOY) and rice-pasture (RI-PAST). We considered 9 years of data on N balances (NBAL), defined as all N inputs minus all N outputs; N surplus (NSURP), defined as all N inputs minus only N outputs in food products; and N use efficiency (NUE), defined as the fraction of N inputs removed in food products. We parameterized DNDC against measured yield and input and output data, with missing data on N losses inferred from the N balance and compared with literature values. The model performance was assessed using standard indices of mean error, agreement and efficiency. The model simulated crop yields and rice cumulative N uptake very well, and soil N reasonably well. The values of NBAL were +45 and−20 kg N ha−1 yr−1 in RI-CONT and RI-SOY, respectively, and close to zero in RI-PAST (−6 kg N ha−1 yr−1). Values of NSURP decreased in the order RI-CONT >> RI-SOY > RI-PAST (+115, +25 and +13 kg N ha−1 yr−1, respectively). Values of NUE (84, 54, and 48% for RI-SOY, RI-PAST, and RI-CONT, respectively) decreased as NBAL increased. The sensitivity of DNDC's predictions to the agronomic characteristics of the different crops, rotations and water regimes agreed with expectations. We conclude that the DNDC model as parameterized here is suitable for exploring how to optimize N management in these systems.
