The effects of climate change are becoming increasingly hazardous for our ecosystem. Climate resilient landscaping, which promotes the use of native plants, has the potential to simultaneously decrease the rate of climate change, enhance climate resilience, and combat biodiversity losses. Native plants and their associated microbiome form a holo-organism; interaction between plants and microbes is responsible for plants' growth and proper functioning. In this study, we were interested in exploring the soil and root microbiome composition associated with Shepherdia utahensis, a drought hardy plant proposed for low water use landscaping, which is the hybrid between two native hardy shrubs of Utah, S. rotudifolia and S. argentea. The bulk soil, rhizosphere, root, and nodule samples of the hybrid Shepherdia plants were collected from three locations in Utah: the Logan Campus, the Greenville farm, and the Kaysville farm. The microbial diversity analysis was conducted, and plant growth-promoting bacteria were isolated and characterized from the rhizosphere. The results suggest no difference in alpha diversity between the locations; however, the beta diversity analysis suggests the bacterial community composition of bulk soil and nodule samples are different between the locations. The taxonomic classification suggests Proteobacteria and Actinobacteriota are the dominant species in bulk soil and rhizosphere, and Actinobacteriota is solely found in root and nodule samples. However, the composition of the bacterial community was different among the locations. There was a great diversity in the genus composition in bulk soil and rhizosphere samples among the locations; however, Frankia was the dominant genus in root and nodule samples. Fifty-nine different bacteria were isolated from the rhizosphere and tested for seven plant growth-promoting (PGP) traits, such as the ability to fix nitrogen, phosphates solubilization, protease activity, siderophore, Indole Acetic Acid (IAA) and catalase production, and ability to use ACC as nitrogen source. All the isolates produced some amount of IAA. Thirty-one showed at least four PGP traits and belonged to Stenotrophomonas, Chryseobacterium, Massilia, Variovorax, and Pseudomonas. The family Araceae, comprising ornamentals including Anthurium, Dieffenbachia, Philodendron, Colocasia, and Zantedeschia, is susceptible to Xanthomonas pathogens. Previous analyses have established heterogeneity in aroid strains, yet unresolved taxonomic positions and dynamics between Xanthomonas and frequently associated Stenotrophomonas in aroids necessitate in-depth genetic investigation to resolve these complex relationships. This study utilized multilocus sequence analysis of housekeeping genes atpD, dnaA, dnaK, gltA, and gyrB to investigate 59 aroid strains, selected based on hosts, time, and geographical origins. After adding sequences from additional strains from NCBI GenBank, analysis of 161 concatenated sequences indicated that all aroid strains fell within Xanthomonas and Stenotrophomonas. Thirty-six strains isolated from Anthurium grouped under X. phaseoli, with outliers including one strain each in X. arboricola and X. sacchari and two in Stenotrophomonas. Six strains from Caladium, Dieffenbachia, and Philodendron formed host-specific subgroups within X. euvesicatoria. One strain from Dieffenbachia aligned with X. campestris, whereas strains from Colocasia, Aglaonema, and Spathiphyllum clustered with X. sacchari. Apart from the zantedeschia strain described as X. arboricola pv. zantedeschiae, two colocasia, one epipremnum, and one anthurium strain joined the X. arboricola group. Overall, this study revealed significant heterogeneity among aroid strains, with anthurium strains clustering closely despite distant geographical origins. The analysis underscores the complexity of host-pathogen specificity within Xanthomonas and emphasizes the need for further taxonomic clarification through whole-genome analysis of representative strains. The findings of this research will facilitate strain selection for inclusivity and exclusivity panels in developing diagnostic assays for X. phaseoli and xanthomonads affecting aroids. Background: Endometrial hyperplasia (EH) is a precursor to endometrial cancer, and the role of the microbiome in its development is unclear. Results: The present study investigated the uterine microbiome in patients with benign uterine conditions and endometrial hyperplasia. A significant structural shift in the uterine microbiome of patients with endometrial hyperplasia compared to those with benign conditions was found. Delftia, Serratia and Stenotrophomonas were significantly enriched in endometrial hyperplasia samples and associated with the presence of endometrial hyperplasia. Conclusions: The novel finding suggested that increased abundance of Delftia, Serratia and Stenotrophomonas is associated with the presence of endometrial hyperplasia. Introduction: Stenotrophomonas is a prominent genus owing to its dual nature. Species of this genus have many applications in industry and agriculture as plant growth-promoting rhizobacteria and microbial biological control agents, whereas species such as Stenotrophomonas maltophilia are considered one of the leading gram-negative multi-drug-resistant bacterial pathogens because of their high contribution to the increase in crude mortality and significant clinical challenge. Pathogenic Stenotrophomonas species and most clinical isolates belong to the Stenotrophomonas maltophilia complex (SMc). However, a strain highly homologous to S. terrae was isolated from a patient with pulmonary tuberculosis (TB), which aroused our interest, as S. terrae belongs to a relatively distant clade from SMc and there have been no human association reports. Methods: The pathogenicity, immunological and biochemical characteristics of 610A2T were systematically evaluated. Results: 610A2T is a new species of genus Stenotrophomonas, which is named as Stenotrophomonas pigmentata sp. nov. for its obvious brown water-soluble pigment. 610A2T is pathogenic and caused significant weight loss, pulmonary congestion, and blood transmission in mice because it has multiple virulence factors, haemolysis, and strong biofilm formation abilities. In addition, the cytokine response induced by this strain was similar to that observed in patients with TB, and the strain was resistant to half of the anti-TB drugs. Conclusions: The pathogenicity of 610A2T may not be weaker than that of S. maltophilia. Its isolation extended the opportunistic pathogenic species to all 3 major clades of the genus Stenotrophomonas, indicating that the clinical importance of species of Stenotrophomonas other than S. maltophilia and potential risks to biological safety associated with the use of Stenotrophomonas require more attention. Stenotrophomonas strains, which are often described as plant growth promoting (PGP) bacteria, are ubiquitous in many environments. A total of 213 genomes of strains of Stenotrophomonas were analyzed using comparative genomics to better understand the ecological roles of these bacteria in the environment. The pan-genome of the 213 strains of Stenotrophomonas consists of 27,186 gene families, including 710 core gene families, 11,039 unique genes and 15,437 accessory genes. Nearly all strains of Stenotrophomonas harbor the genes for GH3-family cellulose degradation and GH2- and GH31-family hemicellulose hydrolase, as well as intact glycolysis and tricarboxylic acid cycle pathways. These abilities suggest that the strains of this genus can easily obtain carbon and energy from the environment. The Stenotrophomonas strains can respond to oxidative stress by synthesizing catalase, superoxide dismutase, methionine sulfoxide reductase, and disulfide isomerase, as well as managing their osmotic balance by accumulating potassium and synthesizing compatible solutes, such as betaine, trehalose, glutamate, and proline. Each Stenotrophomonas strain also contains many genes for resistance to antibiotics and heavy metals. These genes that mediate stress tolerance increase the ability of Stenotrophomonas strains to survive in extreme environments. In addition, many functional genes related to attachment and plant colonization, growth promotion and biocontrol were identified. In detail, the genes associated with flagellar assembly, motility, chemotaxis and biofilm formation enable the strains of Stenotrophomonas to effectively colonize host plants. The presence of genes for phosphatesolubilization and siderophore production and the polyamine, indole-3-acetic acid, and cytokinin biosynthetic pathways confer the ability to promote plant growth. These strains can produce antimicrobial compounds, chitinases, lipases and proteases. Each Stenotrophomonas genome contained 1-9 prophages and 17-60 genomic islands, and the genes related to antibiotic and heavy metal resistance and the biosynthesis of polyamines, indole-3-acetic acid, and cytokinin may be acquired by horizontal gene transfer. Aim Tarantulas are one of the largest predatory arthropods in tropical regions. Tarantulas though not lethal to humans, their venomous bite kills small animals and insect upon which they prey. To understand the abiotic and biotic components involved in Neotropical tarantula bites, we conducted a venom-microbiomics study in eight species from Costa Rica. Methods and results We determined that the toxin profiles of tarantula venom are highly diverse using shotgun proteomics; the most frequently encountered toxins were ω-Ap2 toxin, neprilysin-1, and several teraphotoxins. Through culture-independent and culture-dependent methods, we determined the microbiota present in the venom and excreta to evaluate the presence of pathogens that could contribute to primary infections in animals, including humans. The presence of opportunistic pathogens with hemolytic activity was observed, with a prominence of Stenotrophomonas in the venoms. Other bacteria found in venoms and excreta with hemolytic activity included members of the genera Serratia, Bacillus, Acinetobacter, Microbacterium , and Morganella. Conclusions Our data shed light on the venom- and gut-microbiome associated with Neotropical tarantulas. This information may be useful for treating bites from these arthropods in both humans and farm animals, while also providing insight into the toxins and biodiversity of this little-explored microenvironment. Healthy seeds are the foundation of healthy plants. Planting healthy seeds contributes to securing crop productivity and seed germplasm conservation. In this study, we have identified microbes associated with seeds of three cherry tomato genotypes and demonstrated their negative effect on general seed health. Through a combined morpho-cultural and molecular characterisation (using multi-loci analysis of the ITS, β-tubulin, tef1α, and gapdh gene regions for fungi and 16s rDNA for bacteria), we have identified three fungi (Nigrospora sphaerica, N. lacticolonia, and Curvularia aeria), and two bacteria (Citrobacter freundii, and Stenotrophomonas maltophilia) from healthy-looking tomato seeds. These fungi and bacteria, through seed-soaked-inoculation, caused seed discoloration, lesions, and low germination. To our knowledge, these are the first reports of Nigrospora sphaerica, N. lacticolonia, Curvularia aeria, Citrobacter freundii, and Stenotrophomonas maltophilia on tomato seeds and demonstrated their negative impact on seed health. Endotoxins are pyrogenic lipopolysaccharides from Gram-negative bacteria that are known to induce fever, septic shock, and multiple organ failure, posing a substantial risk to human health. Drinking water systems are especially prone to home microbiomes containing a large variety of Gram-negative bacteria. Consumption of water from these systems in developed countries is generally regarded as non-hazardous to humans due to the low number of non-pathogenic bacterial cells per milliliter and oral admission. To assess potential risks posed by endotoxins in drinking water systems, we conducted a conventional microbiological investigation on a local community water system in the north of Germany and mined the resulting data to investigate the endotoxin contents of some of the most abundant microbiota found during these analyses. Using a Limulus amoebocyte lysate (LAL) -based endotoxin detection method, average normalized endotoxin content was determined. Although the average culturable amounts of microbiota in the drinking water system were insufficient to exert endotoxin levels critical to human health, peaks and acute contaminations may pose substantial health risks. Introduction: Cadmium (Cd) is a highly toxic heavy metal which contaminates agricultural soils and is easily absorbed by plants. Brassica rapa L. is one of the most popular vegetables in China and is known to accumulate Cd in its roots and aerial tissues. Methods: A highly Cd-resistant bacterium (‘CD2’) was isolated and identified. Its ability to immobilize Cd(II) in medium was studied. Strain CD2 were added into Cd-polluted soil to ameliorate Cd accumulation in B. rapa. The underlying mechanisms of ‘CD2’ to reduce Cd accumulation in B. rapa. were analyzed by transcriptomics. Results and discussion: Strain CD2 was classified as belonging to the genus Stenotrophomonas. Strain CD2 was found to be able to remove 0.1 mmol/L Cd(II) after 36 h by intracellular sequestration and by producing biofilm, exopolysaccharide, and H2S. When applied to Cd-contaminated soil, ‘CD2’ significantly increased the content of nonbioavailable Cd by 212.70%. Furthermore, ‘CD2’-inoculated B. rapa exhibited a 51.16% decrease in the Cd content of roots and a 55.56% decrease in the Cd content of aerial tissues. Transcriptome analysis identified 424 differentially expressed genes (DEGs) in the roots and 501 DEGs in the aerial tissues of uninoculated Cd-exposed plants. By comparison, 1107 DEGs were identified in the roots and 1721 DEGs were identified in the aerial tissues of ‘CD2’-inoculated Cd-exposed plants. In both treatment groups, genes related to vacuolar sequestration were upregulated, resulting in inhibited Cd transport. In addition, both catalase and glutathione transferase were induced in uninoculated plants, while the oxidative stressrelated genes CPK and RBOH belonged to ‘plant-pathogen interactions’ were upregulated in ‘CD2’-inoculated plants. Moreover, inoculation with ‘CD2’ resulted in the enrichment of phenylpropane metabolism; cutin, suberine, and wax biosynthesis; and the AP2, Dof, WOX, Trihelix, B3, EIL, and M-type_MADS transcription factors; as well as the downregulation of zinc transporters and blue copper proteins. All of these changes likely contributed to the reduced Cd accumulation in ‘CD2’-inoculated B. rapa. The results of this study suggest that Stenotrophomonas sp. Background A culture of the green algae Chlamydomonas reinhardtii was accidentally contaminated with three different bacteria in our laboratory facilities. This contaminated alga culture showed increased algal biohydrogen production. These three bacteria were independently isolated. Methods The chromosomic DNA of one of the isolated bacteria was extracted and sequenced using PacBio technology. Tentative genome annotation (RAST server) and phylogenetic trees analysis (TYGS server) were conducted. Diverse growth tests were assayed for the bacterium and for the alga-bacterium consortium. Results Phylogenetic analysis indicates that the bacterium is a novel member of the Stenotrophomonas genus that has been termed in this work as S. goyi sp. nov. A fully sequenced genome (4,487,389 base pairs) and its tentative annotation (4,147 genes) are provided. The genome information suggests that S. goyi sp. nov. is unable to use sulfate and nitrate as sulfur and nitrogen sources, respectively. Growth tests have confirmed the dependence on the sulfur-containing amino acids methionine and cysteine. S. goyi sp. nov. and Chlamydomonas reinhardtii can establish a mutualistic relationship when cocultured together. Conclusions S. goyi sp. nov. could be of interest for the design of biotechnological approaches based on the use of artificial microalgae-bacteria multispecies consortia that take advantage of the complementary metabolic capacities of their different microorganisms.

The skin microbiome of amphibians can influence host susceptibility towards the fungal pathogen Batrachochytrium dendrobatidis (Bd), while simultaneously having the potential to be altered by Bd. Severe Bd infections are known to alter the amphibian skin microbiome; however, little is known about microbiome interactions in amphibians with low infection intensity. In addition to disease dynamics, environmental factors may influence the microbiome. To test for patterns in bacterial diversity based on pathogen infection and environmental factors, 399 Columbia spotted frogs (Rana luteiventris) were sampled throughout northern Idaho and northeastern Washington across two years. Bd prevalence and intensity were measured in 376 frogs, revealing a prevalence of 69%, but generally low infection intensity (Mean = 127 Bd zoospore equivalents among infected frogs). Skin bacterial communities were characterized in 92 frogs using 16S rRNA gene amplicon sequencing. Our results indicated correlations of decreasing Shannon diversity and evenness as infection intensity increased. Latitude was correlated with bacterial richness and Faith's Phylogenetic Diversity measures, indicating increased diversity in northern locations. Beta diversity (UniFrac) analyses revealed that skin microbiomes were distinct between infected and uninfected frogs, and infection intensity had a significant effect on microbiome composition. Site explained the majority of microbiome variation (weighted UniFrac: 57.5%), suggesting a combination of local habitat conditions explain variation, as only small proportions of variation could be explained by year, month, temperature, elevation, and latitude individually. Bacterial genera with potential for Bd-inhibitory properties were found with differential relative abundance in infected and uninfected frogs, with higher Stenotrophomonas and lower Pseudomonas relative abundance observed in infected frogs. Further study may indicate if Bd inhibition by members of the skin microbiome is an influence behind the low infection intensities observed and whether low Bd infection intensities are capable of altering skin microbiome composition.

ABSTRACT Acinetobacter baumannii-calcoaceticus species complex (ACB) and Stenotrophomonas maltophilia (SM) are opportunistic, non-fermentative organisms that can cause serious hospital-acquired infections in immunocompromised patients. These pathogens are inherently resistant to several common drug classes and often acquire other resistance mechanisms, making them difficult to treat. In this study, we analyzed the susceptibility of 1,029 contemporary ACB and 1,522 SM isolates to minocycline (MIN) and levofloxacin (LEV) as well as meropenem (MER) for ACB and trimethoprim-sulfamethoxazole (TMP/SMX) for SM using the CLSI broth microdilution method. Isolates were collected as a part of the SENTRY Antimicrobial Surveillance Program from 2014 to 2021. Pneumonia in hospitalized patients was the most common infection from which ACB (57.0%) and SM (73.9%) were isolated. MIN had the highest in vitro activity for ACB (86.2%) and SM (99.5%). The activity of ACB and SM to all three agents varied over the period studied. MIN activity to ACB decreased in 2020 (80.6%) but rebounded in 2021 (86.2%). LEV and MER showed an overall trend of increasing susceptibility for ACB, with slightly lower activity in 2020–2021. MIN and TMP/SMX (>98.3% and >93.7%, respectively) activities were stable against SM isolates. LEV activity decreased from 84.3% (2015) to 69.2% (2018). The activity of MIN remained stable and higher than other agents tested for both ACB and SM, pathogens that have limited therapeutic alternatives. These in vitro data suggest that MIN is a useful treatment option for infections caused by ACB or SM. IMPORTANCE Acinetobacter baumannii-calcoaceticus species complex and Stenotrophomonas maltophilia are opportunistic, non-fermentative Gram-negative organisms that can cause serious hospital-acquired infections in immunocompromised patients. These pathogens are inherently resistant to several common drug classes and often acquire other resistance mechanisms, making them difficult to treat. In this study, we analyzed the trends of susceptibility of over 2,500 U.S. bacterial isolates collected from hospitalized patients over an 8-year period to minocycline, which is used to treat infections caused by these pathogens. These in vitro data suggest that minocycline is a useful treatment option for infections caused by Acinetobacter baumannii-calcoaceticus species complex or Stenotrophomonas maltophilia.

Acclimatisation of orchid seedlings propagated in vitro is a slow process that can be accelerated with the use of plant growth-promoting microorganisms and humic substances. The aim of this study was to evaluate the plant's response to inoculation with plant growth-promoting bacteria and humic acids, as well as to their combined use, during the period of acclimatisation of Cymbidium sp. orchids. The treatments were control; application of humic acids; inoculation with Stenotrophomonas maltophilia; inoculation with Burkholderia cepacia; inoculation with S. maltophilia and B. cepacia; inoculation with S. maltophilia combined with application of humic acids; inoculation with B. cepacia combined with application of humic acids; inoculation with S. maltophilia and B. cepacia combined with application of humic acids. A total of 150 days after acclimatisation, the plants were biometrically and nutritionally evaluated. Inoculation with the bacterial strains separately promoted seedlings with greater N content; however, only inoculation with S. maltophilia, resulted in plants with more total dry matter in relation to the control. There was no response from application of humic acids in an isolated manner or in combination with the bacterial strains. The results indicate the biotechnological potential of the bacteria S. maltophilia in promoting the growth of the Cymbidium sp. orchid. [ABSTRACT FROM AUTHOR]

White rot, caused by Stromatinia cepivora (Anamorph: Sclerotium cepivorum Berk), is a serious soil-borne disease of the onion that restricts its cultivation and production worldwide. Herein, we isolated and characterized a plant growth-promoting rhizobacterium Stenotrophomonas maltophilia from healthy onion roots and an endophytic bacterium Serratia liquefaciens from healthy bean leaves. Both isolates showed strong fungistatic activity against S. cepivora using the dual culture and culture filtrate methods. This effect might be due to the presence of several volatile compounds, especially menthol in both culture filtrates as shown with a GC-MS analysis. Additionally, the root drench application of cell-free culture filtrates of S. maltophilia and S. liquefaciens significantly reduced the incidence and severity of white rot disease on treated onion plants, which was associated with the activation of both enzymatic (POX and PPO) and non-enzymatic (phenolics and flavonoids) antioxidant defense machineries of S. cepivora-infected onion plants. Moreover, the culture filtrates of both bacterial bioagents remarkably enhanced the growth (as expressed by root length, plant height, and number of leaves) and yield parameters (as indicated by bulb circumference, fresh weight of the bulb, and bulb yield per plot) of treated onion plants under field conditions during two successive seasons (2020/2021 and 2021/2022). This might be because of a reduced disease severity and/or the accumulation of the main auxin, indole-3-acetic acid (IAA), and its precursor, the amino acid tryptophan. Our findings suggest that both bioagents might be utilized as eco-friendly alternative control measures to reduce the utilization of chemical fungicides entirely or partially for the safer production of onion in S. cepivora-infested soils. Stenotrophomonas maltophilia is an environmental bacterium as well as an emerging opportunistic multidrug-resistant pathogen. They use the endogenous diffusible signal factor (DSF) quorum sensing (QS) system to coordinate population behavior and regulate virulence processes but can also respond to exogenous N-acyl-homoserine lactone (AHL) signals produced by neighboring bacteria. The effect of these QS signals on the global gene expression of this species remains, however, unknown. Whole-transcriptome sequencing analyses were performed for exponential cultures of S. maltophilia K279a treated with exogenous DSF or AHLs. Addition of DSF and AHLs signals resulted in changes in expression of at least 2-fold for 28 and 82 genes, respectively. Interestingly, 22 of these genes were found upregulated by both QS signals, 14 of which were shown to also be induced during the stationary phase. Gene functions regulated by all conditions included lipid and amino acid metabolism, stress response and signal transduction, nitrogen and iron metabolism, and adaptation to microoxic conditions. Among the common top upregulated QS core genes, a putative TetR-like regulator (locus tag SMLT2053) was selected for functional characterization. This regulator controls its own b-oxidation operon (Smlt2053-Smlt2051), and it is found to sense long-chain fatty acids (FAs), including the QS signal DSF. Gene knockout experiments reveal that operon Smlt2053-Smlt2051 is involved in biofilm formation. Overall, our findings provide clues on the effect that QS signals have in S. maltophilia QS-related phenotypes and the transition from the exponential to the stationary phase and bacterial fitness under high-density growth. IMPORTANCE The quorum sensing system in Stenotrophomonas maltophilia, in addition to coordinating the bacterial population, controls virulence-associated phenotypes, such as biofilm formation, motility, protease production, and antibiotic resistance mechanisms. Biofilm formation is frequently associated with the persistence and chronic nature of nosocomial infections. In addition, biofilms exhibit high resistance to antibiotics, making treatment of these infections extremely difficult. The importance of studying the metabolic and regulatory systems controlled by quorum sensing autoinducers will make it possible to discover new targets to control pathogenicity mechanisms in S. maltophilia. Stenotrophomonas maltophilia is one of the most frequently isolated multidrug-resistant nosocomial opportunistic pathogens. It contributes to disease progression in cystic fibrosis (CF) patients and is frequently isolated from wounds, infected tissues, and catheter surfaces. On these diverse surfaces S. maltophilia lives in single-species or multispecies biofilms. Since very little is known about common processes in biofilms of different S. maltophilia isolates, we analyzed the biofilm profiles of 300 clinical and environmental isolates from Europe of the recently identified main lineages Sgn3, Sgn4, and Sm2 to Sm18. The analysis of the biofilm architecture of 40 clinical isolates revealed the presence of multicellular structures and high phenotypic variability at a strain-specific level. Further, transcriptome analyses of biofilm cells of seven clinical isolates identified a set of 106 shared strongly expressed genes and 33 strain-specifically expressed genes. Surprisingly, the transcriptome profiles of biofilm versus planktonic cells revealed that just 9.43% ± 1.36% of all genes were differentially regulated. This implies that just a small set of shared and commonly regulated genes is involved in the biofilm lifestyle. Strikingly, iron uptake appears to be a key factor involved in this metabolic shift. Further, metabolic analyses implied that S. maltophilia employs a mostly fermentative growth mode under biofilm conditions. The transcriptome data of this study together with the phenotypic and metabolic analyses represent so far the largest data set on S. maltophilia biofilm versus planktonic cells. This study will lay the foundation for the identification of strategies for fighting S. maltophilia biofilms in clinical and industrial settings.IMPORTANCE Microorganisms living in a biofilm are much more tolerant to antibiotics and antimicrobial substances than planktonic cells are. Thus, the treatment of infections caused by microorganisms living in biofilms is extremely difficult. Nosocomial infections (among others) caused by S. maltophilia, particularly lung infection among CF patients, have increased in prevalence in recent years. The intrinsic multidrug resistance of S. maltophilia and the increased tolerance to antimicrobial agents of its biofilm cells make the treatment of S. maltophilia infection difficult. The significance of our research is based on understanding the common mechanisms involved in biofilm formation of different S. maltophilia isolates, understanding the diversity of biofilm architectures among strains of this species, and identifying the differently regulated processes in biofilm versus planktonic cells. Crop nutrition depends on zinc for enzymatic, oxidative, and metabolic processes. In the current study 20 different bacteria were isolated from five soil samples collected from different fields in Egypt. Bacterial isolates were screened for their ability to solubilize insoluble zinc oxide and zinc carbonate. The ability of selected isolates to tolerate soluble zinc was determined using different concentrations of (ZnSO4). Three bacterial isolates were selected with efficiency in solubilizing zinc oxide and zinc carbonate while tolerating high levels of soluble zinc. Molecular identification by 16S rRNA sequencing of the chosen isolates identified them as B3 (Acinetobacter calcoaceticus), B5 (Bacillus proteolyticus) and C6 (Stenotrophomonas pavanii). Sodium alginate beads formulated with the isolated bacteria were tested for stability under different storage conditions for 3 months. A pot experiment was conducted to study and compare the effect of using chosen isolates as an in vivo Zn solubilizer with amended ZnCO3 either alone or embedded in beads as carrier in the soil and its effect on growth parameters of Zea mays after 2 months. There was an increase in Zn uptake in all treatments compared to the control. However, plants grown in a pot treated with ZnCO3 and Acinetobacter calcoaceticus showed the highest zinc content and plant dry weight as compared to the control. Finally, selected isolates in both free and encapsulated forms showed improved plant growth parameters and higher zinc content and can be applied as biofertilizers to enhance soil fertility. Introduction: stenotrophomonas maltophilia is a clinically relevant opportunistic and nosocomial pathogen with increasing concerns regarding antibiotic resistance. Accurate diagnosis and identification are crucial for effective treatment, and misidentification can occur, thereby emphasizing the need for appropriate laboratory testing and surveillance. This review aimed to evaluate the epidemiology, pathogenesis, diagnosis, treatment, and antimicrobial resistance of Stenotrophomonas spp. Materials and Methods. A systematic literature review was conducted using the PubMed Central Database. Inclusion criteria included studies published in open-access scientific journals within the last five years, reporting information on Stenotrophomonas spp. epidemiology, pathogenesis, diagnosis, treatment, and/or antimicrobial resistance. The synthesis of the results involved a narrative synthesis of the findings from the included studies. Results. A total of 25 articles met the inclusion criteria and provided valuable insights into Stenotrophomonas spp. infections. The distribution of reported cases by country, sample type, and antimicrobial resistance patterns was summarized. The prevalence of resistance to various antibiotics was also assessed, highlighting the need for continuous surveillance. Conclusion. This analysis revealed the presence of antimicrobial resistance in Stenotrophomonas spp., particularly in S. maltophilia. The high prevalence of antibiotic resistance underscores the importance of ongoing surveillance and control measures to combat antibiotic resistance. The diverse distribution of S. maltophilia across different sample types emphasizes the need for accurate diagnosis and identification. Addressing antimicrobial resistance in Stenotrophomonas spp. is essential for global public health.

To visualize the personalized distributions of pathogens and chemical environments, including microbial metabolites, pharmaceuticals, and their metabolic products, within and between human lungs afflicted with cystic fibrosis (CF), we generated three-dimensional (3D) microbiome and metabolome maps of six explanted lungs from three cystic fibrosis patients. These 3D spatial maps revealed that the chemical environments differ between patients and within the lungs of each patient. Although the microbial ecosystems of the patients were defined by the dominant pathogen, their chemical diversity was not. Additionally, the chemical diversity between locales in the lungs of the same individual sometimes exceeded interindividual variation. Thus, the chemistry and microbiome of the explanted lungs appear to be not only personalized but also regiospecific. Previously undescribed analogs of microbial quinolones and antibiotic metabolites were also detected. Furthermore, mapping the chemical and microbial distributions allowed visualization of microbial community interactions, such as increased production of quorum sensing quinolones in locations where Pseudomonas was in contact with Staphylococcus and Granulicatella, consistent with in vitro observations of bacteria isolated from these patients. Visualization of microbe-metabolite associations within a host organ in early-stage CF disease in animal models will help elucidate the complex interplay between the presence of a given microbial structure, antibiotics, metabolism of antibiotics, microbial virulence factors, and host responses.IMPORTANCE Microbial infections are now recognized to be polymicrobial and personalized in nature. Comprehensive analysis and understanding of the factors underlying the polymicrobial and personalized nature of infections remain limited, especially in the context of the host. By visualizing microbiomes and metabolomes of diseased human lungs, we reveal how different the chemical environments are between hosts that are dominated by the same pathogen and how community interactions shape the chemical environment or vice versa. We highlight that three-dimensional organ mapping methods represent hypothesis-building tools that allow us to design mechanistic studies aimed at addressing microbial responses to other microbes, the host, and pharmaceutical drugs.

The airway fluids of cystic fibrosis (CF) patients contain local pH gradients and are more acidic than those of healthy individuals. pH is a critical factor that is often overlooked in studies seeking to recapitulate the infection microenvironment. We sought to determine the impact of pH on the physiology of a ubiqituous yet understudied microbe, Stenotrophomonas maltophilia Phylogenomics was first used to reconstruct evolutionary relationships between 74 strains of S. maltophilia (59 from CF patients). Neither the core genome (2,158 genes) nor the accessory genome (11,978 genes) distinguish the CF and non-CF isolates; however, strains from similar isolation sources grouped into the same subclades. We grew two human and six CF S. maltophilia isolates from different subclades at a range of pH values and observed impaired growth and altered antibiotic tolerances at pH 5. Transcriptomes revealed increased expression of both antibiotic resistance and DNA repair genes in acidic conditions. Although the gene expression profiles of S. maltophilia in lab cultures and CF sputum were distinct, we found that the same genes associated with low pH were also expressed during infection, and the higher pH cultures were more similar to sputum metatranscriptomes. Our findings suggest that S. maltophilia is not well adapted to acidity and may cope with low pH by expressing stress response genes and colonizing less acidic microenvironments. As a whole, our study underlines the impact of microenvironments on bacterial colonization and adaptation in CF infections.IMPORTANCE Understanding bacterial responses to physiological conditions is an important priority for combating opportunistic infections. The majority of CF patients succumb to inflammation and necrosis in the airways, arising from chronic infection due to ineffective mucociliary clearance. Steep pH gradients characterize the CF airways but are not often incorporated in standard microbiology culture conditions. Stenotrophomonas maltophilia is a prevalent CF opportunistic pathogen also found in many disparate environments, yet this bacterium's contribution to CF lung damage and its response to changing environmental factors remain largely understudied. Here, we show that pH impacts the physiology and antibiotic susceptibility of S. maltophilia, with implications for the development of relevant in vitro models and assessment of antibiotic sensitivity.

Non-fermenting Gram-negative Achromobacter xylosoxidans, Burkholderia cepacia complex, and Stenotrophomonas maltophilia species cause healthcare-associated infections, often showing resistance to first-line drugs such as trimethoprim-sulfamethoxazole (TMP-SXT). The aim of this study was to determine the effect of curcumin-chitosan nanocomplexes on biofilm-producing clinical isolates of non-fermenting Gram-negative bacilli. A. xylosoxidans, B. cepacia complex, and S. maltophilia clinical isolates were identified by MALDI-TOF mass spectrometry. Antimicrobial susceptibility was determined by broth microdilution. Curcumin (Cur), chitosan (Chi), and sodium tripolyphosphate (TPP) were encapsulated by ionotropic gelation in magnetic nanoparticles (MNP) and were assessed by scanning electron microscopy (SEM) and Fourier-transform infrared (FTIR). Biofilm inhibition and eradication by Cur-Chi-TPP-MNP with TMP-SXT was assessed. Cur-Chi-TPP-MNP in combination with TMP-SXT showed biofilm inhibition activity in A. xylosoxidans (37.5 µg/mL), B. cepacia (18.75 µg/mL), and S. maltophilia (4.69–18.75 µg/mL) and low biofilm eradication activity in all three strains (150 − 300 µg/mL). Cur-Chi-TPP-MNP in combination with TMP-SXT was able to inhibit biofilm and in lower effect to eradicate established biofilms of clinical isolates of A. xylosoxidans, B. cepacia complex, and S. maltophilia species. Simple Summary: The implementation of new strategies to define beneficial bacteria for improving tolerance to stressful conditions in plants is becoming a very relevant topic. The use of microbiota associated with seeds may generate a new solution to improving the efficiency of bioinoculants. Legume production suffers from increasingly accentuated climatic conditions such as heat and drought periods. This group of plants can be especially protected against stress by applying strains isolated from seeds of wild legume varieties. This study shows how strains isolated from seeds of alfalfa and pitch clover are capable of improving the development of lentil plants under drought conditions. Agricultural production is being affected by increasingly harsh conditions caused by climate change. The vast majority of crops suffer growth and yield declines due to a lack of water or intense heat. Hence, commercial legume crops suffer intense losses of production (20–80%). This situation is even more noticeable in plants used as fodder for animals, such as alfalfa and pitch trefoil, since their productivity is linked not only to the number of seeds produced, but also to the vegetative growth of the plant itself. Thus, we decided to study the microbiota associated with their seeds in different locations on the Iberian Peninsula, with the aim of identifying culturable bacteria strains that have adapted to harsh environments and that can be used as biotreatments to improve plant growth and resistance to stress. As potentially inherited microbiota, they may also represent a treatment with medium- and long-term adaptative effects. Hence, isolated strains showed no clear relationship with their geographical sampling location, but had about 50% internal similarity with their model plants. Moreover, out of the 51 strains isolated, about 80% were capable of producing biofilms; around 50% produced mid/high concentrations of auxins and grew notably in ACC medium; only 15% were characterized as xerotolerant, while more than 75% were able to sporulate; and finally, 65% produced siderophores and more than 40% produced compounds to solubilize phosphates. Thus, Paenibacillus amylolyticus BB B2-A, Paenibacillus xylanexedens MS M1-C, Paenibacillus pabuli BB Oeiras A, Stenotrophomonas maltophilia MS M1-B and Enterobacter hormaechei BB B2-C strains were tested as plant bioinoculants in lentil plants (Lens culinaris Medik.), showing promising results as future treatments to improve plant growth under stressful conditions. Abstract Background The potential pathogenic role of Stenotrophomonas maltophilia in lung disease and in particular in cystic fibrosis is unclear. To develop further understanding of the biology of this taxa, the taxonomic position, antibiotic resistance and virulence factors of S. maltophilia isolates from patients with chronic lung disease were studied. Results A total of 111 isolates recovered between 2003 and 2016 from respiratory samples from patients in five different countries were included. Based on a cut-off of 95%, analysis of average nucleotide identity by BLAST (ANIb) showed that the 111 isolates identified as S. maltophilia by Matrix-assisted laser desorption/ionization time of flight mass spectrometry (MALDI-TOF/MS) belonged to S. maltophilia (n = 65), S. pavanii (n = 6) and 13 putative novel species (n = 40), which each included 1–5 isolates; these groupings coincided with the results of the 16S rDNA analysis, and the L1 and L2 ß-lactamase Neighbor-Joining phylogeny. Chromosomally encoded aminoglycoside resistance was identified in all S. maltophilia and S. pavani isolates, while acquired antibiotic resistance genes were present in only a few isolates. Nevertheless, phenotypic resistance levels against commonly used antibiotics, determined by standard broth microbroth dilution, were high. Although putative virulence genes were present in all isolates, the percentage of positive isolates varied. The Xps II secretion system responsible for the secretion of the StmPr1–3 proteases was mainly limited to isolates identified as S. maltophilia based on ANIb, but no correlation with phenotypic expression of protease activity was found. The RPF two-component quorum sensing system involved in virulence and antibiotic resistance expression has two main variants with one variant lacking 190 amino acids in the sensing region. Conclusions The putative novel Stenotrophomonas species recovered from patient samples and identified by MALDI-TOF/MS as S. maltophilia, differed from S. maltophilia in resistance and virulence genes, and therefore possibly in pathogenicity. Revision of the Stenotrophomonas taxonomy is needed in order to reliably identify strains within the genus and elucidate the role of the different species in disease.

Immobilization is a commonly used method in response to the need to increase the resistance of microorganisms to the toxic effects of xenobiotics. In this study, a plant sponge from Luffa cylindrica was used as a carrier for the immobilization of the Stenotrophomonas maltophilia KB2 strain since such a carrier meets the criteria for high-quality carriers, i.e., low price and biodegradability. The optimal immobilization conditions were established as a temperature of 30 °C, pH 7.2, incubation time of 72 h, and an optical density of the culture of 1.4. The strain immobilized in such conditions was used for the biodegradation of naproxen, and an average rate of degradation of 3.8 µg/hour was obtained under cometabolic conditions with glucose. The obtained results indicate that a microbiological preparation based on immobilized cells on a luffa sponge can be used in bioremediation processes where it is necessary to remove the introduced carrier. Background: Tracheostomy bypasses physical barriers that decrease microbial access to the lower airway, which can lead to changes to the lung microbiota. Patients often become chronically colonized with potential pathogens. This study described the incidence and prevalence of specific organisms in a 5-y cohort of children with tracheostomy., Methods: This retrospective cohort of children aged 0-18 y with tracheostomy identified associations between microbial species and subject characteristics including reason for tracheostomy, gastrostomy tube (G-tube), fundoplication, and ventilator use using chi-square test or Fisher exact test., Results: Of 113 eligible patients, 79 (57% male) met study inclusion criteria. Reasons for tracheostomy included airway obstruction secondary to craniofacial anomalies in 16 children (20%), upper-airway obstruction in 14 subjects (17.3%), neuromuscular disorder in 19 subjects (24%), bronchopulmonary dysplasia with or without pulmonary hypertension in 17 subjects (21%), and congenital heart disease in 13 subjects (16%). Most (69%) used a ventilator for at least 6 h/d; 63% had a G-tube; 41% also had a Nissen fundoplication. Of the 20% with upper-airway obstruction, one third were ventilator dependent, unlike other diagnoses where 57% used a ventilator. Staphylococcus aureus (52/113), Pseudomonas aeruginosa (43/113), and Stenotrophomonas species (39/113) were the most frequently identified bacterial species. Most microbes identified were not associated with subjects underlying diagnoses, ventilator use, or feeding type. However, there was a significant association between upper-airway obstruction and group B Streptococcus species and G-tube with P. aeruginosa ., Conclusions: This retrospective single-site descriptive cohort analysis of pediatric subjects with long-term tracheostomy identified trends in microbial prevalence. The presence of specific bacterial strains was more likely to follow individual subject trajectories than sequential appearance of species. P. aeruginosa was associated with G-tube and Streptococcus species with upper-airway obstruction. Treatment options for carbapenem-resistant gram-negative bacilli (CR-GNB), especially metallo-β-lactamase (MBL)-producing CR-GNB, are limited. Aztreonam (ATM) in combination with avibactam (AVI) has shown potential for treating MBL-producing carbapenem-resistant Enterobacterales (CREs) and Stenotrophomonas maltophilia . However, data on ATM in combination with other β-lactamase inhibitors (BLIs) are limited. We performed a multicenter study to evaluate the in vitro activities of ATM in combination with AVI, vaborbactam (VAB), relebactam (REL), tazobactam (TAZ) as well as with their commercially available formulations against CREs and S. maltophilia using broth microdilution. AVI restored ATM activity for MBL-producing CREs (ATM: 9.8% vs ATM-AVI: 78.0%) and S. maltophilia (ATM: 0% vs ATM-AVI: 93.3%). REL also moderately restored activity of ATM in MBL-producing CREs (ATM: 9.8% vs ATM-REL: 42.7%) and S. maltophilia (ATM: 0% vs ATM-REL: 68.9%). VAB and TAZ demonstrated very limited effect on the activity of ATM against CR-GNB evaluated. The combination of ATM with ceftazidime-AVI (CAZ-AVI) demonstrated maximum activity against CREs. Abstract The objective of this work was to isolate endophytic bacteria from peach palm (Bactris gasipaes var. gasipaes) plants and to evaluate the effects of their inoculation on the plant seedlings. Bacteria were isolated from the leaves and roots of the seedlings and from the meristems of peach palm plants in vitro. The isolates were characterized phenotypically and, then, 15 of them, representing different phenotypic groups, were selected and identified by partial sequencing of the 16S rRNA gene. Afterward, these isolates and two commercial strains of Azospirillum brasilense (Ab-V5 and Ab-V6) were inoculated in the peach palm seedlings. After 76 days, the seedlings were evaluated for plant development. The following six genera were identified based on the sequencing: Pseudomonas, Enterobacter, Rhizobium, Stenotrophomonas, Klebsiella, and Erwinia. Out of the 15 inoculated isolates, 9 had a positive effect on the root dry mass of palm peach, with CNPF 77 (Enterobacter sp.), CNPF 100 (Rhizobium sp.), and CNP 179 and CNPF 277 (Stenotrophomonas sp.) standing out. Peach palm seedlings harbor endophytic bacteria which are able to increase root dry matter.

White rot, caused by Stromatinia cepivora (Anamorph: Sclerotium cepivorum Berk), is a serious soil-borne disease of the onion that restricts its cultivation and production worldwide. Herein, we isolated and characterized a plant growth-promoting rhizobacterium Stenotrophomonas maltophilia from healthy onion roots and an endophytic bacterium Serratia liquefaciens from healthy bean leaves. Both isolates showed strong fungistatic activity against S. cepivora using the dual culture and culture filtrate methods. Additionally, the root drench application of cell-free culture filtrates of S. maltophilia and S. liquefaciens significantly reduced the incidence and severity of white rot disease on treated onion plants, which was associated with the activation of both enzymatic (POX and PPO) and non-enzymatic (phenolics and flavonoids) antioxidant defense machineries of S. cepivora-infected onion plants. Moreover, the culture filtrates of both bacterial bioagents remarkably enhanced the growth (as expressed by root length, plant height, and number of leaves) and yield parameters (as indicated by bulb circumference, fresh weight of the bulb, and bulb yield per plot) of treated onion plants under field conditions during two successive seasons (2020/2021 and 2021/2022). This might be because of a reduced disease severity and/or the accumulation of the main auxin, indole-3-acetic acid (IAA), and its precursor, the amino acid tryptophan. Our findings suggest that both bioagents might be utilized as eco-friendly alternative control measures to reduce the utilization of chemical fungicides entirely or partially for the safer production of onion in S. cepivora-infested soils.

The increasing antibiotic resistance due to inappropriate administration of antibiotics is a huge concern for the scientific society, recurring the demand for novel antibiotics against antibiotic‐resistant bacteria. In this study, ABN18 bacterial strain was isolated from sewage wastewater with antibacterial substance production ability and was identified as Stenotrophomonas sp. by partial 16S rRNA sequencing. The isolated bacterial strain was Gram‐negative, long oval‐shaped, motile, nonendospore forming with an average length of 1.7025 μm determined by environmental scanning electron microscope (ESEM). Stenotrophomonas sp. ABN18 was found to have a flexible carbohydrate preference, only unable to grow in sorbitol. The strain showed moderate antibiotic resistance with significant sensitivity against antibiotics such as ceftazidime, doxycycline hydrochloride, and rifampicin. Antibacterial activity of ABN18 was found to be more active against Gram‐negative strains as compared with Gram‐positive bacterial strains. The well‐diffusion study confirmed the extracellular production of the antibacterial substance and its presence in the fermentation broth. Moreover, the temperature, pH, and incubation time were optimized at 33.17°C, 7.45, and 88.8 h, respectively, to maximize antibacterial substance production, and the highest zone of inhibition diameter of 22 mm against E. coli was achieved. The gastrointestinal (GI) microbiome in fish plays significant roles in health and disease resistance. This investigation was accomplished to enumerate, characterize and identify the potential probiotic bacteria from three Indian Major Carps (IMCs), viz., rohu (Labeo rohita), catla (Catla catla) and mrigal (Cirrhinus mrigala) using culture dependent methods. Altogether, 105 pathogen-inhibitory bacteria (out of 1216 isolates) were detected from three IMCs by double layer assay. 16S rRNA partial gene sequence analyses and BLAST search in the NCBI GenBank unveiled that 94.29% of the pathogen inhibitory bacteria were bacilli (99 strains) and Bacillus licheniformis by far the most common (28%). The primarily selected 27 pathogen-inhibitory strains (cumulative inhibition score ≥13) produced extracellular enzymes, while 15 of them produced all the six exo-enzymes studied (amylase, protease, lipase, cellulase, phytase and xylanase). Gut stability of the strains became apparent by their ability to grow in fish mucus and tolerance to diluted bile-juice. Finally, 14 strains were noticed as γ-hemolytic and susceptible to the commonly used antibiotics. Further, intra-peritoneal injection with γ-hemolytic strains did not induce any pathological signs or mortalities in fish, and thus were considered as safe. These 14 γ-hemolytic isolates were represented by the genus Bacillus (13) and Stenotrophomonas (1), which might form probiotic consortia for prospective use in carp culture.

Abstract Background Azithromycin is commonly prescribed drug for individuals with cystic fibrosis (CF), with demonstrated benefits in reducing lung function decline, exacerbation occurrence and improving nutrition. As azithromycin has antimicrobial activity against components of the uncultured microbiome and increasingly the CF microbiome is implicated in disease pathogenesis – we postulated azithromycin may act through its manipulation. Herein we sought to determine if the CF microbiome changed following azithromycin use and if clinical benefit observed during azithromycin use associated with baseline community structure. Results Drawing from a prospectively collected biobank we identified patients with sputum samples prior to, during and after initiating azithromycin and determined the composition of the CF microbial community by sequencing the V3-V4 region of the 16S rRNA gene. We categorized patients as responders if their rate of lung function decline improved after azithromycin initiation. Thirty-eight adults comprised our cohort, nine who had not utilized azithromycin in at least 3 years, and 29 who were completely naïve. We did not observe a major impact in the microbial community structure of CF sputum in the 2 years following azithromycin usage in either alpha or beta-diversity metrics. Seventeen patients (45%) were classified as Responders – demonstrating reduced lung function decline after azithromycin. Responders who were naïve to azithromycin had a modest clustering effect distinguishing them from those who were non-Responders, and had communities enriched with several organisms including Stenotrophomonas, but not Pseudomonas. Conclusions Azithromycin treatment did not associate with subsequent large changes in the CF microbiome structure. However, we found that baseline community structure associated with subsequent azithromycin response in CF adults.

Background: The potential pathogenic role of Stenotrophomonas maltophilia in lung disease and in particular in cystic fibrosis is unclear. To develop further understanding of the biology of this taxa, the taxonomic position, antibiotic resistance and virulence factors of S. maltophilia isolates from patients with chronic lung disease were studied. Results: A total of 111 isolates recovered between 2003 and 2016 from respiratory samples from patients in five different countries were included. Based on a cut-off of 95%, analysis of average nucleotide identity by BLAST (ANIb) showed that the 111 isolates identified as S. maltophilia by Matrix-assisted laser desorption/ionization time of flight mass spectrometry (MALDI-TOF/MS) belonged to S. maltophilia (n = 65), S. pavanii (n = 6) and 13 putative novel species (n = 40), which each included 1–5 isolates; these groupings coincided with the results of the 16S rDNA analysis, and the L1 and L2 ß-lactamase Neighbor-Joining phylogeny. Chromosomally encoded aminoglycoside resistance was identified in all S. maltophilia and S. pavani isolates, while acquired antibiotic resistance genes were present in only a few isolates. Nevertheless, phenotypic resistance levels against commonly used antibiotics, determined by standard broth microbroth dilution, were high. Although putative virulence genes were present in all isolates, the percentage of positive isolates varied. The Xps II secretion system responsible for the secretion of the StmPr1–3 proteases was mainly limited to isolates identified as S. maltophilia based on ANIb, but no correlation with phenotypic expression of protease activity was found. The RPF two-component quorum sensing system involved in virulence and antibiotic resistance expression has two main variants with one variant lacking 190 amino acids in the sensing region. Conclusions: The putative novel Stenotrophomonas species recovered from patient samples and identified by MALDI-TOF/MS as S. maltophilia, differed from S. maltophilia in resistance and virulence genes, and therefore possibly in pathogenicity. Revision of the Stenotrophomonas taxonomy is needed in order to reliably identify strains within the genus and elucidate the role of the different species in disease. [ABSTRACT FROM AUTHOR]

Aims: Plant Growth-Promoting Rhizobacteria (PGPR) support plant growth by alleviating plant stresses, among which those triggered by saline soils. We isolated Stenotrophomonas sp. SRS1 from salt-resistant Carex distans (distant sedge) roots to understand how this growth promotion was enabled and whether an active contribution of the bacteria and/or plant was required. Methods: Various growth assays were used to analyze the effect of bacterial inoculation on Arabidopsis thaliana and Solanum lycopersicum (cherry tomato MicroTom) growth. Furthermore, droplet microfluidics, bacterial genome mining, and bacterial and plant gene expression analysis combined with plant mutant analysis were used for in-depth analysis. Results: SRS1 application enhanced plant growth in both saline and nonsaline environments. The fresh weight of SRS1-inoculated plants was higher than that of noninoculated plants, whereas the fresh weight ratio between SRS1-inoculated and noninoculated plants differed whether the plants were grown on agar plates, white sand or in soil. We demonstrated that the strain grew well in high salt-containing media and that, besides plant-growth-promotion-related genes, the bacterium contained various active stress genes. Interestingly, inoculation with the strain increased the induction of plant genes related to abscisic acid and auxin signaling pathways under saline conditions. Conclusions: SRS1 inoculation promoted the growth of Arabidopsis and MicroTom tomato under saline and nonsaline conditions, also when the plants were grown in white sand and potting soil. Overall, genetic traits related to stress alleviation, derived from both the bacteria and the plants, play a crucial role in the impact of this novel PGPR strain on plant performance. Ochratoxin A is a secondary metabolite that acts as a mycotoxin and is produced by Aspergillus, Penicillium , and other fungal species. It is a threat to animal and human health due to nephrotoxic, carcinogenic, and genotoxic properties and its widespread incidence in agricultural products. To reduce this threat, biological remediation processes are of growing interest. The aerobic gram-negative bacterium Stenotrophomonas sp. 043-1a, isolated from soil, was previously shown to degrade ochratoxin A into the non-toxic ochratoxin α and l -phenylalanine (Schatzmayr et al. 2002). However, the enzyme or enzymes catalyzing this reaction in this strain remained elusive. Here, we report the targeted purification of Stenotrophomonas sp. 043-1a lysate via ammonium sulfate precipitation, size-exclusion chromatography, and hydrophobic interaction chromatography to identify the ochratoxin A degrading enzymes by subsequent peptide fragment fingerprinting. The metallo-dependent hydrolase Chr1_3858681_3267 and a member of the peptidase S9 family, Chr1_3858681_771, were shown to degrade ochratoxin A. Stenotrophomonas maltophilia is an opportunistic pathogen and a dreaded cause of bacteraemia with 30-day mortality rates ranging from 14 to 69%. The purpose of this cohort study was to evaluate outcomes of S. maltophilia bacteraemia, at Rigshospitalet, a tertiary hospital in Copenhagen, Denmark. We included all patients with a blood culture positive for S. maltophilia, from January 1, 2015 to April 1, 2020. We extracted data on antimicrobial susceptibility, treatment, central venous catheter intervention and severe haematological disease. Sixty-one cases of S. maltophilia bacteraemia were identified. The overall 90-day mortality was 18%. Sixty percent of patients had a central venous catheter intervention performed. Seventy-nine percent of patients were treated with trimethoprim/sulfamethoxazole (TMP/SMX). Patients with central venous catheter intervention had significantly better survival than those without (HR: 0.16 [95% CI: 0.03–0.73]). Severe haematological disease and patients, who received intensive care unit (ICU) care, were at higher risk of death than other patients (HR: 5.93 [95% CI: 1.18 − 29.94] and HR: 8.37 [95% CI: 1.79 − 39.20], respectively). We found no evidence that any antibiotic regime was superior with regard to 90-day mortality. We did not find evidence to support a change in the current standard-of-care regimen of TMP/SMX and CVC removal. Larger clinical trials are needed to guide such recommendations. Various technological and biomedical applications rely on the ability of materials to emit light (photoluminescence [PL]), and, among them, metal nanoparticles (NPs) and semi-conductor Quantum Dots (QDs) represent ideal candidates as sensing probes and imaging tools, portraying better PL features than conventional organic dyes. However, the knowledge of PL behavior of semiconductor NPs – i.e., selenium; SeNPs – is still in its infancy, especially for those synthesized by microorganisms. Considering the essential role played by biogenic SeNPs as antimicrobial, anticancer, and antioxidant agents, or food supplements, their PL properties must be explored to take full advantage of them as eco-friendly and versatile tools. Here, PL features of SeNPs produced by the Se-tolerant Stenotrophomonas maltophilia SeITE02 strain, compared with chemogenic ones, are investigated, highlighting the PL dependency on the NP size. Indeed, PL emission shifted from indigo-blue (emission wavelength λem 400–450 nm) to green-yellow (λem 480–570 nm) and orange-red (λem 580–700 nm) for small (ca. 50 nm) and big (ca. 100 nm) SeNPs respectively, revealing the versatility of an environmental bacterial isolate to synthesize diverse PL probes. Besides, biogenic SeNPs show PL lifetime comparable to those of the most used fluorophores, supporting their potential application as markers for (bio)imaging.
