Abstract

The PANDA experiment at the FAIR facility adresses open questions in hadron physics with antiproton beams in the momentum range of 1.5-15 GeV/c. The antiprotons are stored and cooled in a High Energy Storage RING (HESR) with a momentum spread down to Dp/p = 4*10^-5. A high luminosity of up to 2*10^32 cm-2 s-1 can be achieved. An excellent hadronic particle identification (PID) will be provided by two Cherenkov detectors using the priciple of Detection of Internally Reflected Cherenkov light (DIRC). In the forward direction from polar angles of 5 degree to 22 degree, the Endcap Disc DIRC (EDD) separates pions from kaons up to momenta of 4 GeV/c. Between 22 degree and 140 degree the Barrel DIRC cleanly separates pions from kaons for momenta up to 3.5 GeV/c. This article describes the design of the Barrel DIRC and of the Endcap Disc DIRC and the validation of their designs in particle beams at the CERN PS., Comment: 10 pages, 8 figures

Abstract

The innovative Barrel DIRC (Detection of Internally Reflected Cherenkov light) counter will provide hadronic particle identification (PID) in the central region of the PANDA experiment at the new Facility for Antiproton and Ion Research (FAIR), Darmstadt, Germany. This detector is designed to separate charged pions and kaons with at least 3 standard deviations for momenta up to 3.5 GeV/c, covering the polar angle range of 22$^{\circ}$-140$^{\circ}$. An array of microchannel plate photomultiplier tubes is used to detect the location and arrival time of the Cherenkov photons with a position resolution of 2 mm and time precision of about 100 ps. The time imaging reconstruction has been developed to make optimum use of the observables and to determine the performance of the detector. This reconstruction algorithm performs particle identification by directly calculating the maximum likelihoods using probability density functions based on detected photon propagation time in each pixel, determined directly from the data, or analytically, or from detailed simulations., Comment: International Workshop on Fast Cherenkov Detectors (DIRC2019)

Abstract

The PANDA experiment will use cooled antiproton beams with high intensity stored1 in the High Energy Storage Ring at FAIR. Reactions on a fixed target producing charmed hadrons will shed light on the strong QCD. Three ring imaging Cherenkov counters are used for charged particle identification. The status of the Barrel DIRC (Detection of Internally Reflected Cherenkov light) is described. Its design is robust and its performance validated in experiments with test beams. Abstract

PANDA (anti-Proton ANnihiliation at DArmstadt) is planned to be one of the four main experiments at the future international accelerator complex FAIR (Facility for Antiproton and Ion Research) in Darmstadt, Germany. It is going to address fundamental questions of hadron physics and quantum chromodynamics using cooled antiproton beams with a high intensity and and momenta between 1.5 and 15 GeV/c. PANDA is designed to reach a maximum luminosity of 2x10^32 cm^2 s. Most of the physics programs require an excellent particle identification (PID). The PID of hadronic states at the forward endcap of the target spectrometer will be done by a fast and compact Cherenkov detector that uses the detection of internally reflected Cherenkov light (DIRC) principle. It is designed to cover the polar angle range from 5{\deg} to 22{\deg} and to provide a separation power for the separation of charged pions and kaons up to 3 standard deviations (s.d.) for particle momenta up to 4 GeV/c in order to cover the important particle phase space. This document describes the technical design and the expected performance of the novel PANDA Disc DIRC detector that has not been used in any other high energy physics experiment (HEP) before. The performance has been studied with Monte-Carlo simulations and various beam tests at DESY and CERN. Abstract

The DIRC technology (Detection of Internally Reflected Cherenkov light) offers an excellent possibility to minimize the form factor of Cherenkov detectors in hermetic high energy detectors. The PANDA experiment at FAIR in Germany will combine a barrel-shaped DIRC with a disc-shaped DIRC to cover an angular range of 5 to 140 degrees. Particle identification for pions and kaons with a separation power of 3 standard deviations or more will be provided for momenta between 0.5 GeV/c and 3.5 GeV/c in the barrel region and up to 4 GeV/c in the forward region. Even though the concept is simple, the design and construction of a DIRC is challenging. High precision optics and mechanics are required to maintain the angular information of the Cherenkov photons during multiple internal reflections and to focus the individual photons onto position sensitive photon detectors. These sensors must combine high efficiencies for single photons with low dark count rates and good timing resolution at high rates. The choice of radiation hard fused silica for the optical material and of MCP-PMT photon sensors is essential for DIRC detectors to survive in an environment of radiation and strong magnetic field. The two DIRC detectors differ in the focusing optics, in the treatment of chromatic dispersion and in the electronic readout systems. Abstract

The key component of the future PANDA experiment at FAIR is a fixed-target detector for collisions of antiprotons with a proton target up to a beam momentum of 15 GeV/c and is designed to address a large number of open questions in the hadron physics sector. In order to guarantee an excellent PID for charged hadrons in the polar angle range between $5^\circ$ and $22^\circ$, a new type of Cherenkov detector called Endcap Disc DIRC (EDD) has been developed for the forward endcap of the PANDA target spectrometer. Abstract

The PANDA experiment is one of the four large experiments being built at FAIR in Darmstadt. It will use a cooled antiproton beam on a fixed target within the momentum range of 1.5 to 15 GeV/c to address questions of strong QCD, where the coupling constant $\alpha_s \gtrsim 0.3$. The luminosity of up to $2 \cdot 10^{32} cm^{-2}s^{-1}$ and the momentum resolution of the antiproton beam down to \mbox{$\Delta$p/p = 4$\cdot10^{-5}$} allows for high precision spectroscopy, especially for rare reaction processes. Above the production threshold for open charm mesons the production of kaons plays an important role for identifying the reaction. The DIRC principle allows for a compact particle identification for charged particles in a hermetic detector, limited in size by the electromagnetic lead tungstate calorimeter. The Barrel DIRC in the target spectrometer covers polar angles between $22^\circ$ and $140^\circ$ and will achieve a pion-kaon separation of 3 standard deviations up to 3.5 GeV/$c$. Abstract

Plants can provide a cost-effective and scalable technology for production of therapeutic monoclonal antibodies, with the potential for precise engineering of glycosylation. Glycan structures in the antibody Fc region influence binding properties to Fc receptors, which opens opportunities for modulation of antibody effector functions. To test the impact of glycosylation in detail, on binding to human Fc receptors, different glycovariants of VRC01, a broadly neutralizing HIV monoclonal antibody, were generated in Nicotiana benthamiana and characterized. These include glycovariants lacking plant characteristic α1,3-fucose and β1,2-xylose residues and glycans extended with terminal β1,4-galactose. Surface plasmon resonance-based assays were established for kinetic/affinity evaluation of antibody-FcγR interactions, and revealed that antibodies with typical plant glycosylation have a limited capacity to engage FcγRI, FcγRIIa, FcγRIIb and FcγRIIIa; however, the binding characteristics can be restored and even improved with targeted glycoengineering. All plant-made glycovariants had a slightly reduced affinity to the neonatal Fc receptor (FcRn) compared with HEK cell-derived antibody. However, this was independent of plant glycosylation, but related to the oxidation status of two methionine residues in the Fc region. This points towards a need for process optimization to control oxidation levels and improve the quality of plant-produced antibodies.

Abstract

The key component of the future PANDA experiment at FAIR is a fixed-target detector for collisions of antiprotons with a proton target up to a beam momentum of 15 GeV/c and is designed to address a large number of open questions in the hadron physics sector. In order to guarantee an excellent PID for charged hadrons in the polar angle range between $5^\circ$ and $22^\circ$, a new type of Cherenkov detector called Endcap Disc DIRC (EDD) has been developed for the forward endcap of the PANDA target spectrometer. Abstract

Nowadays, robots are used in the retail market mostly for warehousing, while they could be of great help in different in-store logistics processes as discussed in previous chapters. The present chapter deals with the shelf replenishment task; its execution by a robot requires overcoming of technological and methodological barriers in the handling of single products rather than the boxes containing them. The challenges a robot has to face to replenish a supermarket shelf are all related to manipulation in narrow spaces of products with a large variety of size, shape, weight, and fragility. The solution proposed by REFILLS is based on a robotic system where perception is used at all hierarchical levels of the control architecture, from high-level task planning algorithms and motion planning to reactive control layers based on physics models, where tactile and visual perception are combined to achieve highly reliable manipulation of items. Experiments in an emulated supermarket shelf are carried out to demonstrate the effectiveness of the approach.

Abstract

Background: Improving cancer immunotherapy long-term clinical benefit is a major priority. It has become apparent that multiple axes of immune suppression restrain the capacity of T cells to provide anti-tumour activity including signalling through PD1/PD-L1 and LAG3/MHC-II., Methods: CB213 has been developed as a fully human PD1/LAG3 co-targeting multi-specific Humabody composed of linked V H domains that avidly bind and block PD1 and LAG3 on dual-positive T cells. We present the preclinical primary pharmacology of CB213: biochemistry, cell-based function vs. immune-suppressive targets, induction of T cell proliferation ex vivo using blood obtained from NSCLC patients, and syngeneic mouse model anti-tumour activity. CB213 pharmacokinetics was assessed in cynomolgus macaques., Results: CB213 shows picomolar avidity when simultaneously engaging PD1 and LAG3. Assessing LAG3/MHC-II or PD1/PD-L1 suppression individually, CB213 preferentially counters the LAG3 axis. CB213 showed superior activity vs. αPD1 antibody to induce ex vivo NSCLC patient T cell proliferation and to suppress tumour growth in a syngeneic mouse tumour model, for which both experimental systems possess PD1 and LAG3 suppressive components. Non-human primate PK of CB213 suggests weekly clinical administration., Conclusions: CB213 is poised to enter clinical development and, through intercepting both PD1 and LAG3 resistance mechanisms, may benefit patients with tumours escaping front-line immunological control., (© 2021. Abstract

Background: Simmondsia chinensis (jojoba) is the only plant known to store wax esters instead of triacylglycerols in its seeds. Wax esters are composed of very-long-chain monounsaturated fatty acids and fatty alcohols and constitute up to 60% of the jojoba seed weight. During jojoba germination, the first step of wax ester mobilization is catalyzed by lipases. To date, none of the jojoba lipase-encoding genes have been cloned and characterized. In this study, we monitored mobilization of storage reserves during germination of jojoba seeds and performed detailed characterization of the jojoba lipases using microsomal fractions isolated from germinating seeds., Results: During 26 days of germination, we observed a 60-70% decrease in wax ester content in the seeds, which was accompanied by the reduction of oleosin amounts and increase in glucose content. The activity of jojoba lipases in the seed microsomal fractions increased in the first 50 days of germination. The enzymes showed higher activity towards triacylglycerols than towards wax esters. The maximum lipase activity was observed at 60 °C and pH around 7 for triacylglycerols and 6.5-8 for wax esters. The enzyme efficiently hydrolyzed various wax esters containing saturated and unsaturated acyl and alcohol moieties. We also demonstrated that jojoba lipases possess wax ester-synthesizing activity when free fatty alcohols and different acyl donors, including triacylglycerols and free fatty acids, are used as substrates. For esterification reactions, the enzyme utilized both saturated and unsaturated fatty alcohols, with the preference towards long chain and very long chain compounds., Conclusions: In in vitro assays, jojoba lipases catalyzed hydrolysis of triacylglycerols and different wax esters in a broad range of temperatures. In addition, the enzymes had the ability to synthesize wax esters in the backward reaction. Our data suggest that jojoba lipases may be more similar to other plant lipases than previously assumed.

Abstract

Plants can provide a cost-effective and scalable technology for production of therapeutic monoclonal antibodies, with the potential for precise engineering of glycosylation. Glycan structures in the antibody Fc region influence binding properties to Fc receptors, which opens opportunities for modulation of antibody effector functions. To test the impact of glycosylation in detail, on binding to human Fc receptors, different glycovariants of VRC01, a broadly neutralizing HIV monoclonal antibody, were generated in Nicotiana benthamiana and characterized. These include glycovariants lacking plant characteristic α1,3-fucose and β1,2-xylose residues and glycans extended with terminal β1,4-galactose. Surface plasmon resonance-based assays were established for kinetic/affinity evaluation of antibody-FcγR interactions, and revealed that antibodies with typical plant glycosylation have a limited capacity to engage FcγRI, FcγRIIa, FcγRIIb and FcγRIIIa; however, the binding characteristics can be restored and even improved with targeted glycoengineering. All plant-made glycovariants had a slightly reduced affinity to the neonatal Fc receptor (FcRn) compared with HEK cell-derived antibody. However, this was independent of plant glycosylation, but related to the oxidation status of two methionine residues in the Fc region. This points towards a need for process optimization to control oxidation levels and improve the quality of plant-produced antibodies., (© 2019 The Authors. Abstract

In this study, a strategy based on polymeric immunoglobulin G scaffolds (PIGS) was used to produce a vaccine candidate for Mycobacterium tuberculosis. A genetic fusion construct comprising genes encoding the mycobacterial Ag85B antigen, an immunoglobulin γ-chain fragment and the tailpiece from immunoglobulin μ chain was engineered. Expression was attempted in Chinese Hamster Ovary (CHO) cells and in Nicotiana benthamiana. The recombinant protein assembled into polymeric structures (TB-PIGS) in N. benthamiana, similar in size to polymeric IgM. These complexes were subsequently shown to bind to the complement protein C1q and FcγRs with increased affinity. Modification of the N-glycans linked to TB-PIGS by removal of xylose and fucose residues that are normally found in plant glycosylated proteins also resulted in increased affinity for low-affinity FcγRs. Immunization studies in mice indicated that TB-PIGS are highly immunogenic with and without adjuvant. However, they did not improve protective efficacy in mice against challenge with M. tuberculosis compared to conventional vaccination with BCG, suggesting that additional or alternative antigens may be needed to protect against this disease. Nevertheless, these results establish a novel platform for producing polymeric antigen-IgG γ-chain molecules with inherent functional characteristics that are desirable in vaccines., (© 2018 The Authors. Abstract

Microalgae are unicellular eukaryotic organisms which represent an emerging alternative to other cell biofactories commonly used to produce monoclonal antibodies. Microalgae display several biotechnological advantages such as their rapid growth rate and their phototrophic lifestyle allowing low production costs as protein expression is solar-fueled. Recently, a fully assembled recombinant IgG antibody directed against Hepatitis B surface antigen is produced and secreted in the culture medium of the diatom Phaeodactylum tricornutum. A biochemical characterization of this recombinant antibody demonstrated that the Asn-297 is N-glycosylated by oligomannosides. In the immune system, antibodies interact with effector molecules and cells through their Fc part and the recognition of Fcγ receptors (FcγR) which are important for inducing phagocytosis of opsonized microbes. Interactions between IgG and FcγR are influenced by the N-glycan structures present on the Asn-297. In this study, the authors characterized the binding capacity of the anti-hepatitis B recombinant IgG produced in P. tricornutum to two human Fcγ receptors (FcγRI and IIIa) using a cellular binding assay and surface plasmon resonance (SPR). This allowed us to demonstrate that the alga-made antibody is able to bind FcγRI with a reduced affinity and engages FcyRIIIa with 3-times higher affinity compared to a control human IgG1., (© 2017 WILEY-VCH Verlag GmbH & Co. Abstract

Anti-CD20 recombinant antibodies are among the most promising therapeutics for the treatment of B-cell malignancies such as non-Hodgkin lymphomas. We recently demonstrated that an immunocytokine (2B8-Fc-hIL2), obtained by fusing an anti-CD20 scFv-Fc antibody derived from C2B8 mAb (rituximab) to the human interleukin 2 (hIL-2), can be efficiently produced in Nicotiana benthamiana plants. The purified immunocytokine (IC) bearing a typical plant protein N-glycosylation profile showed a CD20 binding activity comparable to that of rituximab and was efficient in eliciting antibody-dependent cell-mediated cytotoxicity (ADCC) of human PBMC against Daudi cells, indicating its fuctional integrity. In this work, the immunocytokine devoid of the typical xylose/fucose N-glycosylation plant signature (IC-ΔXF) and the corresponding scFv-Fc-ΔXF antibody not fused to the cytokine, were obtained in a glyco-engineered ΔXylT/FucT N. benthamiana line. Purification yields from agroinfiltrated plants amounted to 20-35 mg/kg of leaf fresh weight. When assayed for interaction with FcγRI and FcγRIIIa, IC-ΔXF exhibited significantly enhanced binding affinities if compared to the counterpart bearing the typical plant protein N-glycosylation profile (IC) and to rituximab. The glyco-engineered recombinant molecules also exhibited a strongly improved ADCC and complement-dependent cytotoxicity (CDC). Notably, our results demonstrate a reduced C1q binding of xylose/fucose carrying IC and scFv-Fc compared to versions that lack these sugar moieties. These results demonstrate that specific N-glycosylation alterations in recombinant products can dramatically affect the effector functions of the immunocytokine, resulting in an overall improvement of the biological functions and consequently of the therapeutic potential., (© 2017 Wiley Periodicals, Inc.)

Abstract

IL-4 is a pleiotropic cytokine that is highly Th2 polarizing. The ratio of IL-4 and its splice variant IL-4Δ2 observed in human health and disease suggests a role for both isoforms. In the present study, the biological function of murine IL-4Δ2 and the potential mechanism of action were studied. We report for the first time the generation of a functional, recombinant murine IL-4Δ2 form which is suggestive of its possible biological role in this species. Recombinant murine IL-4Δ2 inhibited IL-4 mediated cellular processes in macrophages and lymphocytes. Specifically, (i) it reversed IL-4 mediated inhibition of IFN-γ induced nitric oxide release by macrophages, (ii) inhibited IL-4 mediated induction of T cell proliferation, and (iii) prevented IL-4 stimulation of IgE synthesis by B cells. However, IL-4Δ2 did not compete with IL-4 for IL-4Rα binding and did not interfere with the downstream STAT-6 phosphorylation in T cells, suggesting an alternative mechanism for its antagonism of specific IL4-driven effects. Abstract

The principal involvement of cyclic nucleotides in regulating sperm functions is well established, but the factors controlling their generation and actions have not yet been entirely resolved. In particular, specific roles for cyclic (c)GMP in mammalian sperm are poorly understood. In this study, we have characterized comparatively the cAMP and cGMP signalling systems in ejaculated human sperm. Mean concentrations of cGMP (0.1 micromol/l) were found to be 100-fold lower than those of cAMP in non-stimulated cells, and adenylyl cyclase (AC) activities predominate by far guanylyl cyclase (GC) activities in both particulate and soluble protein fractions. By different experimental approaches (photoaffinity labelling, cyclase assays, immunoblotting), we provide evidence for the presence (guanylyl cyclase-A, soluble guanylyl cyclase, regulatory and catalytic subunits of cAMP-dependent protein kinase) or absence (guanylyl cyclase-B, natriuretic peptide clearance receptor, neuronal nitric oxide synthase, cGMP-dependent protein kinase I) of different factors involved in either cAMP or cGMP pathways. Functional studies showed that cGMP, at high concentrations, can enhance sperm protein tyrosine phosphorylation but not serine phosphorylation of glycogen synthase kinase. This study reveals that human sperm are characterized by an exceptional predominance of cAMP signalling and indicates potential roles for cGMP.
