Abstract

Cryptolestes (Coleoptera: Laemophloeidae) are economically important pests, which damage stored products in granaries and mills. Because it is difficult to distinguish Cryptolestes species morphologically, we develop here a high sensitivity real-time PCR method for distinguishing five species of Cryptolestes (C. ferrugineus, C. pusillus, C. turcicus, C. pusilloides and C. capensis). The lower limit of DNA concentration required for species identification is 0.1 ng/μl for C. ferruginues and C. pusilloides, and 0.01 ng/μl for C. pusillus, C. turcicus, and C. capensis. This method successfully determined species assignments for previously unidentified Cryptolestes spp. Collected from the field and will facilitate Cryptolestes identification in the field of plant quarantine and stored product protection.

Abstract

The development of a high resolution high-field 1 GHz NMR spectrometer has been undertaken by Battelle Pacific Northwest Laboratories (BPNL). In preparation for the design of the superconducting magnet for that instrument, BPNL sponsored a program to measure the properties at high field (up to 23.5 T) and low temperature (1.8 K) of a number of commercially available superconductors. Lengths of these conductors were obtained and tested as short samples and in small coils. The critical current densities, index values and strain dependence of these properties were characterized. The potential applications of these conductors in construction of a 1 GHz NMR magnet are discussed.

Abstract

The new National High Magnetic Field Laboratory (NHMFL) at Tallahassee, Florida is committed to putting into operation in 1995 a 45-T Hybrid Magnet System to support research in steady, high magnetic fields. This facility will be accessible by qualified users world-wide on the basis of proposal and review. The more prominent components of this system will be a 24-MW resistive insert and a 120-MJ superconducting outsert. But successful achievement of the performance goals for the 45-T Hybrid System will depend on a number of unique, state-of-the-art subsystems and components. This paper describes the requirements and specifications on the individual subsystems and components in the context of the overall performance goals and reviews the plan for putting the whole together.

Abstract

Primary pests such as Rhyzoperta dominica may increase the contents of dockage, dust, and frass in grain mass. Although it has been suggested that frass can affect the population growth of stored product pests and ecological interactions among primary and secondary pests in stored grain, this has not been validated experimentally. Therefore, this work experimentally tested the hypothesis that R. dominica wheat frass may support population increases in secondary pests such as Tribolium confusum, T. castaneum, and Oryzaephilus surinamensis for the first time. The effect of frass on secondary pest performance was compared with the effects of various physical qualities of wheat grain (i.e., intact grain kernels, grain fragments, flour, grain + frass) and an artificially enriched control diet (milled wheat kernels, oat flakes, and yeast). The results showed that the clean intact grain kernels did not support the population growth of any tested species, and the nutrient-rich control diet provided the best support. Frass was a significantly better food medium for O. surinamensis and T. castaneum than flour or cracked grain, while T. confusum performed equally well on flour and frass. Our results showed that in terms of food quality and suitability for the tested species, frass occupied an intermediate position between the optimized breeding diet and simple uniform cereal diets such as cracked grain or flour. The results suggest that (i) the wheat frass of primary pest R. dominica is a riskier food source for the development of the tested secondary pests than intact or cracked wheat grain or flour; (ii) frass has the potential to positively influence interspecific interactions between R. dominica and the tested secondary pests; and (iii) wheat grain should be cleaned if increases in R. dominica populations and/or accumulated frass are detected. Abstract

Legume proteins were shown to have insecticidal activity against stored-product pests. Grain enriched by bean (Phaseolus vulgaris) flour inhibits the growth of stored-product mites. In this study, we tested the toxicity of bean flour to storage mites under optimal conditions for their population growth (i.e. rearing diet, temperature 25°C and humidity optimum 85% RH). Bean flour was added to the diet in one of eight concentrations: 0, 0.01, 0.1, 0.5, 1, 2.5, 5, 10%). The population growth of Tyrophagus putrescentiae, Acarus siro and Aleuroglyphus ovatus initiating from a density of 50 mites per 0.2 g of diet was recorded for 21 days. The enrichment of grain with bean flour suppressed the population growth of the tested species. These differed in their sensitivity to bean flour. Population growth was decreased to 50% in comparison to the control (rC50) by the bean flour concentration of 0.02% in T. putrescentiae, 0.04% in A. siro, and by 4.87% in A. ovatus. The concentration of 5% bean flour in diets kept populations of A. siro and T. putrescentiae at the initial level. The results are discussed in the context of applying bean flour in the integrated control of stored-product mites.

Abstract

The detection of synathropic rodents may be difficult since they are animals with nocturnal activity. Methods of their detection and monitoring rely mostly on indirect signs of their activity such as the presence of faeces, urine, consumed foods and damaged materials. Our experimental hypothesis was that the production of fluorescent faeces - following consumption of fluorescent bait - may be used for rodent monitoring. For this purpose we studied the production of fluorescent faeces, temporal dynamics and detectability in wild black rat (Rattus rattus). Wild black rats were individually housed in experimental cages with the wire-mesh grid floor and faeces were collected in short-time intervals. The peak of fluorescent activity in faeces was detected 10-20 hours after bait ingestion. We found that there is only relatively short delay between bait consumption and defecation and fluorescent faeces are easily detectable at distance using an ultraviolet hand lamp. Thus, this method can contribute to effective monitoring of rodent pests.

Abstract

We measured the individual variation of the area-estimate of simple geometrical patterns (circle, rectangle) in ten people. We found that they tended to underestimate (max. 5 x) the correct area of the tested geometrical patterns. Consequently, we explored how the insecticides Ficam 80WP, K-Othrine 25, and Actellic EC50 are robust or sensitive to the measured extent of over-dosage (2x) or underdosage (5x). We also tested the effects of incorrect dosages of insecticides applied to porous filter paper nonporous glass and bioassayed with adult Sitophilus oryzae and Tribolium castaneum. We found that the tested insecticides were surprisingly robust to under-dosage on the glass surface but sensitive to under-dosage on the porous paper surface.

Abstract

Until now, there has been no record of Liotrophon punctulatus (Hymenoptera: Ichneumonidae) presence in the anthropogenic environment of mills, bakeries or pasta factories. This is the first report of the species parasitizing Ephestia kuehniella (Lepidoptera: Pyralidae) larvae. The host/parasitoid interaction was validated under laboratory conditions where fourth or fifth instar larvae of Ephestia kuehniella were provided ad libitum to L. punctulatus females. After two filial generations emergence, the validation process was considered to confirm the interaction.
