For the production of sugars and biobased platform chemicals from lignocellulosic biomass, the hydrolysis of cellulose and hemicelluloses to water-soluble sugars is a crucial step. As the complex structure of lignocellulosic biomass hinders an efficient hydrolysis via acid hydrolysis, a suitable pretreatment strategy is of special importance. The pretreatment steam explosion was intended to increase the accessibility of the cellulose fibers so that the subsequent acid hydrolysis of the cellulose to glucose would take place in a shorter time. Steam explosion pretreatment was performed with beech wood chips at varying severities with different reaction times (25&ndash, 34 min) and maximum temperatures (186&ndash, 223 &deg, C). However, the subsequent acid hydrolysis step of steam-exploded residue was performed at constant settings at 180 &deg, C with diluted sulfuric acid. The concentration profiles of the main water-soluble hydrolysis products were recorded. We showed in this study that the defibration of the macrofibrils in the lignocellulose structure during steam explosion does not lead to an increased rate of cellulose hydrolysis. So, steam explosion is not a suitable pretreatment for acid hydrolysis of hardwood lignocellulosic biomass.

Hydroxymethylfurfural (HMF) wird als eine der wichtigsten bio-basierten Plattformchemikalien angesehen. Ziel der Arbeit ist ein Verfahren zur Gewinnung von HMF auf Basis von Lignocellulosen in Wasser zu entwickeln. Lignocellulosen, das heißt Holz oder Halmgüter, bestehen aus einem komplexen Verbund von Lignin, Hemicellulosen und Cellulose. Dieser komplexe Verbund muss zunächst aufgeschlossen werden, denn nur Hexosen, die in Cellulose und Hemicellulosen vorhanden sind, können als Ausgangsmaterial für HMF dienen (vgl. Abbildung 0.1). Die zweite Herausforderung besteht darin, dass Cellulose ein Polymer der Glucose, und nicht der Fructose, ist. Ein Blick in die Literatur zeigt, dass Glucose nur sehr langsam, möglicherweise nur über die Isomerisierung zu Fructose, zu HMF reagiert. Glucose sollte folglich zunächst in Fructose umgewandelt werden. Zunächst wurde der säurekatalysierte Aufschluss von Hölzern in einer halbkontinuierlichen Laboranlage untersucht. Die Cellulose wird bei 180 °C mit 0,05 mol/l H2SO4 nur langsam zu Glucose hydrolysiert. Bei 200 °C ist die Cellulose nach knapp 2 h vollständig abreagiert und bei 220 °C verkürzt sich dies auf 40 min. Die Hemicellulosen sind ohne Zugabe eines Katalysators unter hydrothermalen Bedingungen bei 180 °C noch größtenteils stabil, werden bei 220 °C aber hydrolysiert. Die Ausbeute von Glucose beim Aufschluss mit 0,05 mol/l H2SO4 erreicht nach 40 min Reaktionszeit bei einer mittleren Hydrolysetemperatur von 200 °C den höchsten Wert. Die Ausbeute ist bei 180 °C niedriger, während bei 220 °C Glucose in Folgereaktionen abgebaut wird. So steigen beispielsweise die Ausbeuten an Lävulinsäure und Ameisensäure an. Durch die Vorbehandlung von Holz mittels Steam-Explosion sollte die Zugänglichkeit der Cellulose-Fasern erhöht werden, damit die Hydrolyse der Cellulose zu Glucose in kürzerer Reaktionszeit abläuft. Trotz einer „Aufsprengung“ der Macrofibrillen in der Lignocellulosen-Struktur durch die Steam-Explosion kam es nicht zu einer Erhöhung der Hydrolysegeschwindigkeit der Cellulose. Bei der Isomerisierung von Glucose wurden die besten Fructoseausbeuten mit dem heterogenen Katalysator Hydrotalkit erzielt. In Parameterstudien mit reinen Glucose-Lösungen unter pH-neutralen Bedingungen wurden 25 Ma.-% Fructoseausbeute bei 38 Ma.-% Glucoseumsatz erreicht (Massenverhältnis von Hydrotalkit zu Glucose von 0,2 bei 90 °C für 60 min). Durch vorherige Neutralisation der sauren Produktflüssigkeit aus dem Aufschluss ist es gelungen, säurekatalysierte Nebenreaktionen der Glucose, welche während der Isomerisierung ablaufen, drastisch zu reduzieren. Bei der Neutralisation fallen nur geringe Mengen an organischen Verbindungen aus. Allerdings ist als Neutralisationsmittel NaOH gegenüber Ba(OH)2 zu favorisieren, denn es werden nach NaOH-Neutralisation höhere Fructoseausbeuten bei der Isomerisierung erhalten (16 Ma.-% Fructoseausbeute, 32 Ma.-% Glucoseumsatz). Für eine technische Realisierung der HMF-Synthese aus Hexose-Zuckern sind moderate Reaktions-bedingungen zu wählen, bei denen die Summe der Nebenprodukte möglichst gering ist. Dies können beispielsweise 200 °C und 14 min bei Katalyse mit 0,005 mol/l Schwefelsäure sein. Durch eine numerische Optimierung werden die Geschwindigkeitskonstanten eines Reaktionsmodells von Zuckern zu HMF bestimmt. Dieses Modell liefert einen Einblick in die Bildung der nicht charakterisierten Nebenprodukte. Diese Nebenprodukte entstehen laut der Kinetik vor allem ausgehend von HMF und weniger direkt von den Monosacchariden. Es wurden zwei Machbarkeitsstudien für die Teilschritte (1) Lignocellulosen zu Zuckern sowie (2) Zucker zu HMF einer hydrothermalen Bioraffinerie durchgeführt. Für die erste Machbarkeitsstudie zum Aufschluss von Lignocellulosen zu Zuckern bilden die Rohstoffkosten für Holz den größten Kostenposten. Hexosen sind nicht wirtschaftlich aus Lignocellulosen herstellbar, wenn sie das einzige Wertprodukt des Prozesses sind, insbesondere wenn der Vergleich mit dem Import von Zuckern wie Saccharose gezogen wird. Eine Vermarktung der Ligninfraktion ist dabei für eine wirtschaftliche Bioraffinerie unerlässlich. In einer zweiten Machbarkeitsstudie wurde die Herstellung von HMF aus Saccharose untersucht, wobei ein Kreisprozess inklusive Abtrennung von HMF projektiert wurde. Bei den hier bestimmten HMF-Herstellungskosten von 4,30 €/kgHMF sind die beiden größten Posten die Eduktkosten sowie die Energiekosten. Erst danach folgen die Abschreibungskosten für die Anlage.

Straws are agricultural residues that can be used to produce biomethane by anaerobic digestion. The methane yield of rice straw is lower than other straws. Steam explosion was investigated as a pretreatment to increase methane production. Pretreatment conditions with varying reaction times (12&ndash, 30 min) and maximum temperatures (162&ndash, 240 &deg, C) were applied. The pretreated material was characterized for its composition and thermal and morphological properties. When the steam explosion was performed with a moderate severity parameter of S0 = 4.1 min, the methane yield was increased by 32% compared to untreated rice straw. This study shows that a harsher pretreatment at S0 &gt, 4.3 min causes a drastic reduction of methane yield because inert condensation products are formed from hemicelluloses.

Hydroxymethylfurfural (HMF) has an outstanding position among bio-based platform chemicals, because high-value polymer precursors and fuel additives can be derived from HMF. Unfortunately, the large-scale industrial production of HMF is not yet realized. An open research question is the choice of hexose feedstock material. In this study, we used the highly available disaccharide sucrose for HMF synthesis. The conversion of sucrose was catalyzed by sulfuric acid in water media. Experiments were conducted at temperatures of 180, 200, and 220 °C with reaction times of 2–24 min. A carbon balance showed that the yield of unwanted side products rose strongly with temperature. We also developed a kinetic model for the conversion of sucrose, involving nine first-order reactions, to uncover the kinetics of the main reaction pathways. Within this model, HMF is produced exclusively via the dehydration of fructose. Glucose isomerizes slowly to fructose. Side products arise simultaneously from glucose, fructose, and HMF. A pathway from hexoses to xylose via reverse aldol reaction was also included in the model. We believe that sucrose is the ideal feedstock for large-scale production of HMF because it is more abundant than fructose, and easier to process than sugars obtained from lignocellulosic biomass. Phase noise from radio frequency (RF) oscillators is one of the major limiting factors affecting communication system performance. Phase noise directly effects short-term frequency stability, Bit-Error-Rate (BER), and phase-locked loop adjacent-channel interference. RF oscillator circuits contain at least one active device, usually a transistor. The active device has noise properties which generally dominate the noise characteristic limits of an oscillator. Since all noise sources, except thermal noise, are generally proportional to average current flow through the active device, it is logical that reducing the current flow through the device will lead to lower noise levels. A theory based on the time-varying properties of oscillators proposes that narrowing the current pulse width in the active device will decrease the time that noise is present in the circuit and therefore, decrease phase noise even further. The time-domain waveforms and phase noise of an active-biased 700MHz oscillator are analyzed, showing heavy saturation and high harmonic content. Redesigns of the example oscillator in active-bias and four-resistor-bias configurations show improved phase noise and lower harmonic levels at the output. Five oscillator designs of each bias configuration, each having a different pulse width, are simulated. As predicted by the theory, the narrowest current pulse corresponds to the lowest phase noise of the simulated oscillators. Master of Science

This study reports the use of hypotonic osmotic shock as a treatment step to enhance the recoveries of biofuel-convertible lipids and proteins from lipid-rich saltwater Nannochloropsis gaditana (N. gaditana) slurries (biomass content = ~140 mg biomass / g slurry, total lipid content = ~600 mg lipid /g biomass). The osmotic shock was induced through repeated washing of microalgal slurries with multiple batches of fresh water. Subjecting the slurries to 2 stages of freshwater washing resulted in a measurable damage to cell membranes (the uptake of membrane permeability marker increased by 6 folds), a partial loss of cell viability (only 64% of available cells were recoverable), and a minor release of free protein (~2 wt% of available protein) from the biomass into the interstitial space of the slurries. Hypotonic osmotic shock was revealed to be ineffective in rupturing N. gaditana slurries (only 13 ± 9% of available cells were ruptured after 2-stage washing) and, as such, had a limited prospect as a stand-alone cell disruption technology for the saltwater strain. The washing treatment, however, was found to be able to weaken the structural integrity of N. gaditana slurries and enhance the performance of subsequent mechanical or chemical cell disruption technologies when installed as a preparatory step. Applying the washing treatment prior to high-pressure homogenisation (HPH) and low-solvent-to-biomass ratio hexane extraction (hexane: slurry = 1:1 w/w) for the recovery of biofuel-convertible lipids increased the extent of cell rupture from 28 ± 8 to 46 ± 19% of available cells and more than doubled neutral lipid yield from 25.1 ± 2.0 to 64.6 ± 4.9 wt% of available neutral lipid. Initial analysis revealed that the washing treatment had a minimal energy cost (~6% of the total energy expenditure of downstream processing) and that its integration into HPH + hexane lipid recovery led to a 2.5 fold increase in the energy output of the biomass. Partnering the washing treatment with NaOH hydrolysis increased protein yield from 6.7 ± 2.4 to 31.9 ± 10.7 wt% of available protein.

Hydrolysis of lignocellulosic biomass is a crucial step for the production of sugars and biobased platform chemicals. Pretreatment experiments in a semi-continuous plant with diluted sulphuric acid as catalyst were carried out to measure the time-dependent formation of sugars (glucose, xylose, mannose), furfurals, and organic acids (acetic, formic, and levulinic acid) at different hydrolysis temperatures (180, 200, 220° C) of one representative of each basic type of lignocellulose: hardwood, softwood, and grass. The addition of the acid catalyst is followed by a sharp increase in the sugar concentration. Xylose and mannose were mainly formed in the initial stages of the process, while glucose was released slowly. Increasing the reaction temperature had a positive effect on the formation of furfurals and organic acids, especially on hydroxymehtylfurfural (HMF) and levulinic acid, regardless of biomass type. In addition, large amounts of formic acid were released during the hydrolysis of miscanthus grass. Structural changes in the solid residue show a complete hydrolysis of hemicellulose at 180° C and of cellulose at 200° C after around 120 min reaction time. The results obtained in this study can be used for the optimisation of the hydrolysis conditions and reactor design to maximise the yields of desired products, which might be sugars or furfurals.
