This paper addresses the critical issue of missing data in power demand time series by emphasizing the relevance of imputation-based approaches in data-driven technologies. A comparative analysis of imputation methods is performed, where the reference from the state of the art is selected as K-Nearest Neighbors (KNN) applied in the time domain. Two innovative methods are proposed. The former method is defined as Historical Data Informed Regression Technique (H-DIRT) and is based on incorporating historical data for setting up a multivariate linear regression and then imputing through the estimated relation between the missing power demand measurement and the historical data. When the available historical data are insufficient, the algorithm proceeds by averaging or by a linear interpolation between the first available measurement before and after the missing value. The latter proposed method is defined as Seasonal KNN (SKNN) and it is based on enriching the data set with features related to yearly, seasonal, weekly and daily trends and then proceeding by baseline KNN. Experiments are set up with random and continuous data clipping, even with rather extreme pruning (up 70% of the data). The results in general demonstrate a significant improvement in imputation accuracy compared to the state of the art. The average error metrics (like Mean Absolute Error and Root Mean Square Error) for the SKNN method are in the order of respectively one third and one half those of the baseline KNN, in the cases of random and continuous data clipping. In general, the SKNN method provides more accurate results and better captures the statistical features of the data set to impute. Anyway, if the share of data to impute is not too large, the H-DIRT method provides comparable accuracy at a much lower computational cost. Hence, this study presents an easily implementable and computationally affordable approach for improving, in various contexts, the state of the art in power demand data imputation. It establishes a foundation for future exploration into trends, seasonal factors, and external variables influencing power load parameters.

Summary: In perinatal HIV infection, early antiretroviral therapy (ART) initiation is recommended but questions remain regarding infant immune responses to HIV and its impact on immune development. Using single cell transcriptional and phenotypic analysis we evaluated the T cell compartment at pre-ART initiation of infants with perinatally acquired HIV from Maputo, Mozambique (Towards AIDS Remission Approaches cohort). CD8+ T cell maturation subsets exhibited altered distribution in HIV exposed infected (HEI) infants relative to HIV exposed uninfected infants with reduced naive, increased effectors, higher frequencies of activated T cells, and lower frequencies of cells with markers of self-renewal. Additionally, a cluster of CD8+ T cells identified in HEI displayed gene profiles consistent with cytotoxic T lymphocytes and showed evidence for hyper expansion. Longitudinal phenotypic analysis revealed accelerated maturation of CD8+ T cells was maintained in HEI despite viral control. The results point to an HIV-directed immune response that is likely to influence reservoir establishment.

Abstract Measurements of the gravitational constant G are notoriously difficult. Individual state-of-the-art experiments have managed to determine the value of G with high precision: although, when considered collectively, the range in the measured values of G far exceeds individual uncertainties, suggesting the presence of unaccounted for systematic effects. Here, we propose a Bayesian framework to account for the presence of systematic errors in the various measurement of G while proposing a consensus value, following two paths: a parametric approach, based on the maximum entropy principle, and a non-parametric one, the latter being a very flexible approach not committed to any specific functional form. With both our methods, we find that the uncertainty on this fundamental constant, once systematics are included, is significantly larger than what quoted in CODATA 2018. Moreover, the morphology of the non-parametric distribution hints towards the presence of several sources of unaccounted for systematics. In light of this, we recommend a consensus value for the gravitational constant $$G = 6.6740^{+0.0015}_{-0.0015} \times 10^{-11}\ \textrm{m}^3\ \textrm{kg}^{-1}\ \textrm{s}^{-2}$$ G = 6 . 6740 - 0.0015 + 0.0015 × 10 - 11 m 3 kg - 1 s - 2 .

Background: The persistence of HIV-1-infected cells during antiretroviral therapy is well documented but may be modulated by early initiation of antiretroviral therapy in infants. Methods: Here, we longitudinally analyzed the proviral landscape in nine infants with vertical HIV-1 infection from Mozambique over a median period of 24 months, using single-genome, near full-length, next-generation proviral sequencing. Results: We observed a rapid decline in the frequency of intact proviruses, leading to a disproportional under-representation of intact HIV-1 sequences within the total number of HIV-1 DNA sequences after 12-24 months of therapy. In addition, proviral integration site profiling in one infant demonstrated clonal expansion of infected cells harboring intact proviruses and indicated that viral rebound was associated with an integration site profile dominated by intact proviruses integrated into genic and accessible chromatin locations. Conclusion: Together, these results permit rare insight into the evolution of the HIV-1 reservoir in infants infected with HIV-1 and suggest that the rapid decline of intact proviruses, relative to defective proviruses, may be attributed to a higher vulnerability of genome-intact proviruses to antiviral immunity. Technologies to analyze combinations of intact proviral sequences and corresponding integration sites permit a high-resolution analysis of HIV-1 reservoir cells after early antiretroviral treatment initiation in infants.

Summary: Background: Despite antiretroviral treatment (ART), immune dysfunction persists in children with perinatal HIV infection (HEI). Here we investigated the impact of HIV status on maternal antibody (Ab) passage, long-term vaccine induced immunity and B-cell maturation. Methods: 46 HIV Exposed Uninfected (HEU), 43 HEI, and 15 HIV unexposed uninfected (HUU) infants were vaccinated with 3 doses of DTaP-HepB-Hib-PCV10-OP at 2, 3, and 4 months at Matola Provincial Hospital, Maputo, Mozambique. Tetanus toxoid specific (TT) IgG, HIV Ab and B-cell phenotype characteristics were evaluated at entry, pre-ART, 5, 10, and 18 months in this longitudinal cohort study. Findings: Baseline (maternal) plasma TT Ab levels were significantly lower in HEI compared to both HEU and HUU and a faster decay of TT Ab was observed in HEI compared to HEU with significantly lower TT Ab levels at 10 and 18 months of age. TT unprotected (UP) (≤0.1 IU/mL) HEI showed higher HIV-RNA at entry and higher longitudinal HIV viremia (Area Under the Curve) compared to TT protected (P) HEI. A distinct HIV-Ab profile was found at entry in HEI compared to HEU. B-cell phenotype showed a B-cell perturbation in HEI vs HEU infants at entry (mean age 40.8 days) with lower transitional CD10+CD19+ B-cells and IgD+CD27− naive B-cells and an overall higher frequency of IgD−CD27− double negative B-cell subsets in HEI. Interpretation: B-cell perturbation, presenting with higher double negative IgD−CD27− B-cells was observed in neonatal age and may play a major role in the B-cell exhaustion in HEI. The ability to maintain TT protective Ab titers over time is impaired in HEI with uncontrolled viral replication and the current vaccination schedule is insufficient to provide long-term protection against tetanus. Construction is known as one of the most dangerous industries in terms of worker safety. Collisions due the excessive proximity of workers to moving construction vehicles are one of the leading causes of fatal and non-fatal accidents on construction sites internationally. Proximity warning systems (PWS) have been proposed in the literature as a solution to detect the risk for collision and to alert workers and equipment operators in time to prevent collisions. Although the role of sensing technologies for situational awareness has been recognised in previous studies, several factors still need to be considered. This paper describes the design of a prototype sensor-based PWS, aimed mainly at small and medium-sized construction companies, to collect real-time data directly from construction sites and to warn workers of a potential risk of collision accidents. It considers, in an integrated manner, factors such as cost of deployment, the actual nature of a construction site as an operating environment and data protection. A low-cost, ultra-wideband (UWB)-based proximity detection system has been developed that can operate with or without fixed anchors. In addition, the PWS is compliant with the General Data Protection Regulation (GDPR) of the European Union. A privacy-by-design approach has been adopted and privacy mechanisms have been used for data protection. Future work could evaluate the PWS in real operational conditions and incorporate additional factors for its further development, such as studies on the timely interpretation of data.

BackgroundIdentifying subphenotypes within heterogeneous diseases may have an impact in terms of therapeutic options. In this study, we aim to assess different subphenotypes in children living with human immunodeficiency virus (HIV-1), according to the clinical, virological, and immunological characteristics.MethodsWe collected clinical and sociodemographic data, baseline viral load (VL), CD4 and CD8 count and percentage, age at initiation of ART, HIV DNA reservoir size in peripheral blood mononuclear cells (PBMCs), cell-associated RNA (CA-RNA), ultrasensitive VL, CD4 subsets (T effector CD25+, activated memory cells, Treg cells), humoral-specific HIV response (T-bet B cells), innate response (CD56dim natural killer (NK) cells, NKp46+, perforin), exhaustion markers (PD-1, PD-L1, DNAM), CD8 senescence, and biomarkers for T-lymphocyte thymic output (TREC) and endothelial activation (VCAM). The most informative variables were selected using an unsupervised lasso-type penalty selection for sparse clustering. Hierarchical clustering was performed using Pearson correlation as the distance metric and WARD.D2 as the clustering method. Internal validation was applied to select the best number of clusters. To compare the characteristics among clusters, boxplot and Kruskal Wallis test were assessed.ResultsThree subphenotypes were discovered (cluster1: n=18, 45%; cluster2: n=11, 27.5%; cluster3: n=11, 27.5%). Patients in cluster1 were treated earlier, had higher baseline %CD4, low HIV reservoir size, low western blot score, higher TREC values, and lower VCAM values than the patients in the other clusters. In contrast, cluster3 was the less favorable. Patients were treated later and presented poorer outcomes with lower %CD4, and higher reservoir size, along with a higher percentage of CD8 immunosenescent cells, lower TREC, higher VCAM cytokine, and a higher %CD4 PD-1. Cluster2 was intermediate. Patients were like those of cluster1, but had lower levels of t-bet expression and higher HIV DNA reservoir size.ConclusionsThree HIV pediatric subphenotypes with different virological and immunological features were identified. The most favorable cluster was characterized by a higher rate of immune reconstitution and a slower disease progression, and the less favorable with more senescence and high reservoir size. In the near future therapeutic interventions for a path of a cure might be guided or supported by the different subphenotypes.

The Internet of Things (IoT) approach relies on the use of the Internet Protocol (IP) as a pervasive network protocol. IP acts as a “glue” for interconnecting end devices (on the field side) and end users, leveraging on very diverse lower-level and upper-level protocols. The need for scalability would suggest the adoption of IPv6, but the large overhead and payloads do not match with the constraints dictated by common wireless solutions. For this reason, compression strategies have been proposed to avoid redundant information in the IPv6 header and to provide fragmentation and reassembly of long messages. For example, the Static Context Header Compression (SCHC) protocol has been recently referenced by the LoRa Alliance as a standard IPv6 compression scheme for LoRaWAN-based applications. In this way, IoT end points can seamlessly share an end-to-end IP link. However, implementation details are out of the specifications’ scope. For this reason, formal test procedures for comparing solutions from different providers are important. In this paper, a test method for assessing architectural delays of real-world deployments of SCHC-over-LoRaWAN implementations is presented. The original proposal includes a mapping phase, for identifying information flows, and a subsequent evaluation phase, in which flows are timestamped and time-related metrics are computed. The proposed strategy has been tested in different use cases involving LoRaWAN backends deployed all around the world. The feasibility of the proposed approach has been tested by measuring the end-to-end latency of IPv6 data in sample use cases, obtaining a delay of less than 1 s. However, the main result is the demonstration that the suggested methodology permits a comparison of the behavior of IPv6 with SCHC-over-LoRaWAN, allowing the optimization of choices and parameters during deployment and commissioning of both infrastructure components and software.

BackgroundDespite a successful antiretroviral therapy (ART), adolescents living with perinatally acquired HIV (PHIV) experience signs of B-cell hyperactivation with expansion of ‘namely’ atypical B-cell phenotypes, including double negative (CD27-IgD-) and termed age associated (ABCs) B-cells (T-bet+CD11c+), which may result in reduced cell functionality, including loss of vaccine-induced immunological memory and higher risk of developing B-cells associated tumors. In this context, perinatally HIV infected children (PHIV) deserve particular attention, given their life-long exposure to chronic immune activation.MethodsWe studied 40 PHIV who started treatment by the 2nd year of life and maintained virological suppression for 13.5 years, with 5/40 patients experiencing transient elevation of the HIV-1 load in the plasma (Spike). We applied a multi-disciplinary approach including immunological B and T cell phenotype, plasma proteomics analysis, and serum level of anti-measles antibodies as functional correlates of vaccine-induced immunity.ResultsPhenotypic signs of B cell hyperactivation were elevated in subjects starting ART later (%DN T-bet+CD11c+ p=0.03; %AM T-bet+CD11c+ p=0.02) and were associated with detectable cell-associated HIV-1 RNA (%AM T-bet+CD11c+ p=0.0003) and transient elevation of the plasma viral load (spike). Furthermore, B-cell hyperactivation appeared to be present in individuals with higher frequency of exhausted T-cells, in particular: %CD4 TIGIT+ were associated with %DN (p=0.008), %DN T-bet+CD11c+ (p=0.0002) and %AM T-bet+CD11c+ (p=0.002) and %CD4 PD-1 were associated with %DN (p=0.048), %DN T-bet+CD11c+ (p=0.039) and %AM T-bet+CD11c+ (p=0.006). The proteomic analysis revealed that subjects with expansion of these atypical B-cells and exhausted T-cells had enrichment of proteins involved in immune inflammation and complement activation pathways. Furthermore, we observed that higher levels of ABCs were associated a reduced capacity to maintain vaccine-induced antibody immunity against measles (%B-cells CD19+CD10- T-bet+, p=0.035).ConclusionWe identified that the levels of hyperactivated B cell subsets were strongly affected by time of ART start and associated with clinical, viral, cellular and plasma soluble markers. Furthermore, the expansion of ABCs also had a direct impact on the capacity to develop antibodies response following routine vaccination.

Early initiation of antiretroviral therapy and adherence to achieve viral load suppression (VLS) are crucial for reducing morbidity and mortality of perinatally HIV-infected infants. In this descriptive cohort study of 39 HIV perinatally infected infants, who started treatment at one month of life in Mozambique, we aimed to describe the viral response over 2 years of follow up. VLS ≤ 400 copies/mL, sustained VLS and viral rebound were described using a Kaplan–Meier estimator. Antiretroviral drug transmitted resistance was assessed for a sub-group of non-VLS infants. In total, 61% of infants reached VLS, and 50% had a rebound. Cumulative probability of VLS was 36%, 51%, and 69% at 6, 12 and 24 months of treatment, respectively. The median duration of VLS was 7.4 months (IQR 12.6) and the cumulative probability of rebound at 6 months was 30%. Two infants had resistance biomarkers to drugs included in their treatment regimen. Our findings point to a low rate of VLS and high rate of viral rebound. More frequent viral response monitoring is advisable to identify infants with rebound and offer timely adherence support. It is urgent to tailor the psychosocial support model of care to this specific age group and offer differentiated service delivery to mother–baby pairs.

HIV eradication is hindered by the existence of latent HIV reservoirs in CD4+ T cells. Therapeutic strategies targeting latent cells are required to achieve a functional cure, however the study of latently infected cells from HIV infected persons is extremely challenging due to the lack of biomarkers that uniquely characterize them. In this study, the dual reporter virus HIVGKO was used to investigate latency establishment and maintenance in lymphoid-derived CD4+ T cells. Single cell technologies to evaluate protein expression, host gene expression, and HIV transcript expression were integrated to identify and analyze latently infected cells. FDA-approved, JAK1/2 inhibitors were tested in this system as a potential therapeutic strategy to target the latent reservoir. Latent and productively infected tonsillar CD4+ T cells displayed similar activation profiles as measured by expression of CD69, CD25, and HLADR, however latent cells showed higher CXCR5 expression 3 days post-infection. Single cell analysis revealed a small set of genes, including HIST1-related genes and the inflammatory cytokine, IL32, that were upregulated in latent compared to uninfected and productively infected cells suggesting a role for these molecular pathways in persistent HIV infection. In vitro treatment of HIV-infected CD4+ T cells with physiological concentrations of JAK1/2 inhibitors, ruxolitinib and baricitinib, used in clinical settings to target inflammation, reduced latent and productive infection events when added 24 hr after infection and blocked HIV reactivation from latent cells. Our methods using an established model of HIV latency and lymphoid-derived cells shed light on the biology of latency in a crucial anatomical site for HIV persistence and provides key insights about repurposing baricitinib or ruxolitinib to target the HIV reservoir.

The size of the latent HIV reservoir is associated with the timing of therapeutic interventions and overall health of the immune system. Here, we demonstrate that T cell phenotypic signatures associate with viral reservoir size in a cohort of HIV vertically infected children and young adults under durable viral control, and who initiated anti-retroviral therapy (ART)

Antigen-primed cluster of differentiation (CD) 4+ T follicular helper (Tfh) cells interact with B cells in the germinal centers (GCs) of lymph nodes to generate vaccine-induced antibody (Ab) responses. In the circulation, peripheral Tfh (pTfh) cells, a subset of memory CD4 T cells, serve as surrogates for GC Tfh because of several functional and phenotypic similarities between them. We investigated features of H1N1 influenza antigen-specific pTfh (Ag.pTfh) in virologically controlled HIV+ volunteers on antiretroviral therapy (ART) and healthy control (HC) participants selected from a seasonal influenza vaccine responsiveness study. Selection of the participants was made based on age, defined as young (18-40 y) and old (>60 y) and on their classification as a vaccine responder (VR) or vaccine nonresponder (VNR). VRs demonstrated expansion of CD40L+ and CD69+ Ag.pTfh, with induction of intracellular interleukin 21 (IL-21) and inducible costimulator (ICOS) post vaccination; these responses were strongest in young HC VRs and were less prominent in HIV+ individuals of all ages. Ag.pTfh in VNRs exhibited dramatically different characteristics from VRs, displaying an altered phenotype and a cytokine profile dominated by cytokines IL-2, tumor necrosis factor alpha (TNF-α), or IL-17 but lacking in IL-21. In coculture experiments, sorted pTfh did not support the B cell IgG production in VNRs and were predominantly an inflammatory T helper 1 (Th1)/T helper 17 (Th17) phenotype with lower ICOS and higher programmed cell death protein 1 (PD1) expression. Induction of IL-21 and ICOS on Ag.pTfh cells are negatively affected by both aging and HIV infection. Our findings demonstrate that dysfunctional Ag.pTfh cells with an altered IL-21/IL-2 axis contribute to inadequate vaccine responses. Approaches for targeting inflammation or expanding functional Tfh may improve vaccine responses in healthy aging and those aging with HIV infection.

The management of the distribution network is becoming increasingly important as the penetration of distributed energy resources is increasing. Reliable knowledge of the real-time status of the network is essential if algorithms are to be used to help distribution system operators define network configurations. State Estimation (SE) algorithms are capable of producing such an accurate snapshot of the network state but, in turn, require a wide range of information, e.g., network topology, real-time measurement and power profiles from customers/productions. Those profiles which may, in principle, be provided by smart meters are not always available due to technical limitations of existing Advanced Metering Infrastructure (AMI) in terms of communication, storage and computing power. That means that power profiles are only available for a subset of customers. The paper proposes an approach that can overcome these limitations: the remaining profiles, required by SE algorithms, are generated on the basis of customer-related information, identifying clusters of customers with similar features, such as the same contract and pattern of energy consumption. For each cluster, a power profile estimator is generated using long-term power profiles of a limited sub-set of customers, randomly selected from the cluster itself. The synthesized full power profile, representing each customer of the distribution network, is then obtained by scaling the power profile estimator of the cluster to which the customer belongs, by the monthly energy exchanged by that customer, data that are easily available. The feasibility of the proposed approach was validated considering the distribution grid of Unareti SpA, an Italian Distribution System Operator (DSO), operating in northern Italy and serving approximately one million customers. The application of the proposed approach to the actual infrastructure shows some limitations in terms of the accuracy of the estimation of the power profile of the customer. In particular, the proposed methodology is not fully able to properly represent clusters composed of customers with a large variability in terms of power exchange with the distribution network. In any case, the root mean square error of the synthesized full power profile with the respect to validation power profiles belonging to the same cluster is, in the worst case, on the order of 6.3%, while in the rest of cases is well below 5%. Thus, the proposed approach represents a good compromise between accuracy in representing the behavior of customers on the network and resources (in terms of computational power, data storage and communication resources) to achieve that results.

The current perspective about urban development expects 70% of energy consumption will be concentrated in the cities in 2050. In addition, a growing density of people in the urban context leads to the need for increased security and safety for citizens, which imply a better lighting infrastructure. Smart solutions are required to optimize the corresponding energy effort. In developing countries, the cities’ lighting is limited and the lighting world map is strongly significant about the urban density of the different areas. Nevertheless, in territories where the illumination level is particularly high, such as urban contexts, the conditions are not homogenous at the microscale level and the perceived security is affected by artificial urban lighting. As an example, 27.2% of the families living in the city of Milan, ombardy Region, Italy, consider critical the conditions of lighting in the city during the night, although the region has diffused infrastructure. The paper aims to provide a local illuminance geographic information system (GIS) mapping at the neighborhood level that can be extended to the urban context. Such an approach could unveil the need to increase lighting to enhance the perceived safety and security for the citizens and promote a higher quality of life in the smart city. Lighting mapping can be matched with car accident mapping of cities and could be extended to perceived security among pedestrians in urban roads and green areas, also related to degradation signs of the built environment. In addition, such an approach could open new scenarios to the adaptive street lighting control used to reduce the energy consumption in a smart city: the perceived security of an area could be used as an additional index to be considered during the modulation of the level of the luminosity of street lighting. An example of a measurement set-up is described and tested at the district level to define how to implement an extensive monitoring campaign based on an extended research schema.

The adoption of the distributed generation paradigm is introducing several changes in the design and operation of modern distribution networks. Modern grid codes are becoming more and more complex, and the adoption of smart protection systems is becoming mandatory. However, the adoption of newer and smarter units is only half of the story. Proper communication networks must be provided as well, and the overall costs may become critical. In this work, the adoption of the Long-Range Wide Area Network (LoRaWAN) technology is suggested as a viable approach to implement the coordination of Interface Protection Systems. A proper communication architecture based on the LoRaWAN Class B technology was proposed and evaluated in order to assess its feasibility for the considered application. A scalability analysis was carried out, by computing the number of devices that can be handled by a single LoRaWAN Gateway (GW) and the maximum expected time of response between a triggering event and the arrival of the related coordination command. The results of the study showed that up to 312 devices can be managed by a single GW, by assuring a maximum response time of 22.95 s. A faster maximum response time of 6.2 s is also possible by reducing the number of managed devices to 12.

T follicular helper (Tfh) cells are a subset of CD4 T cells that provide critical signals to antigen-primed B cells in germinal centers to undergo proliferation, isotype switching, and somatic hypermutation to generate long-lived plasma cells and memory B cells during an immune response. The quantity and quality of Tfh cells therefore must be tightly controlled to prevent immune dysfunction in the form of autoimmunity and, on the other hand, immune deficiency. Both Tfh and B cell perturbations appear during HIV infection resulting in impaired antibody responses to vaccines such as seasonal trivalent influenza vaccine, also seen in biologic aging. Although many of the HIV-associated defects improve with antiretroviral therapy (ART), excess immune activation and antigen-specific B and T cell responses including Tfh function are still impaired in virologically controlled HIV-infected persons on ART. Interestingly, HIV infected individuals experience increased risk of age-associated pathologies. This review will discuss Tfh and B cell dysfunction in HIV infection and highlight the impact of chronic HIV infection and aging on Tfh–B cell interactions.

The present study was aimed at investigating the role of coping strategies in predicting emotional distress following breast cancer, over and above the illness severity, operationalized in terms of the type of surgery performed. In order to achieve this goal, two groups of newly diagnosed breast cancer women were selected and compared on the basis of the type of surgical treatment received. A subsample of 30 women with quadrantectomy and sentinel lymph-node biopsy (SLNB) and a subsample of 31 patients with mastectomy and axillary dissection (MAD) filled in the Brief Cope scale and Hospital Anxiety and Depression Scale. Summarizing, results showed that emotional support, venting, and humor explained a statistically significant increment of variance in psychological distress indices. Implication for clinical practice and future research were discussed.

Despite effective antiretroviral therapy (ART), HIV-infected individuals with apparently similar clinical and immunological characteristics can vary in responsiveness to vaccinations. However, molecular mechanisms responsible for such impairment, as well as biomarkers able to predict vaccine responsiveness in HIV-infected children, remain unknown. Following the hypothesis that a B cell qualitative impairment persists in HIV-infected children (HIV) despite effective ART and phenotypic B cell immune reconstitution, the aim of the current study was to investigate B cell gene expression of HIV compared to age-matched healthy controls (HCs) and to determine whether distinct gene expression patterns could predict the ability to respond to influenza vaccine. To do so, we analyzed prevaccination transcriptional levels of a 96-gene panel in equal numbers of sort-purified B cell subsets (SPBS) isolated from peripheral blood mononuclear cells using multiplexed RT-PCR. Immune responses to H1N1 antigen were determined by hemaglutination inhibition and memory B cell ELISpot assays following trivalent-inactivated influenza vaccination (TIV) for all study participants. Although there were no differences in terms of cell frequencies of SPBS between HIV and HC, the groups were distinguishable based upon gene expression analyses. Indeed, a 28-gene signature, characterized by higher expression of genes involved in the inflammatory response and immune activation was observed in activated memory B cells (CD27+CD21−) from HIV when compared to HC despite long-term viral control (>24 months). Further analysis, taking into account H1N1 responses after TIV in HIV participants, revealed that a 25-gene signature in resting memory (RM) B cells (CD27+CD21+) was able to distinguish vaccine responders from non-responders (NR). In fact, prevaccination RM B cells of responders showed a higher expression of gene sets involved in B cell adaptive immune responses (APRIL, BTK, BLIMP1) and BCR signaling (MTOR, FYN, CD86) when compared to NR. Overall, these data suggest that a perturbation at a transcriptional level in the B cell compartment persists despite stable virus control achieved through ART in HIV-infected children. Additionally, the present study demonstrates the potential utility of transcriptional evaluation of RM B cells before vaccination for identifying predictive correlates of vaccine responses in this population.

In this paper a general model for the estimation of the uncoordinated charging costs of Electric Vehicles (EVs) in the presence of distributed and intermittent generation, and variable electricity tariffs is presented. The proposed method aims at estimating the monthly average cost of uncoordinated charging of a single EV depending on the hour at which the EV is plugged into the EV Supply Equipment (EVSE). The feasibility and relevance of the proposed model is verified by applying the considered cost estimation method to a suitable use case. A single EV charging service offered at a public building equipped with a Photovoltaic (PV) system has been considered as reference case. The proposed model has been applied to the PV production and loads consumption data collected during one year, and the results of the study compared with the Time-Of-Use (TOU) electricity tariff. The application of the proposed model identified noticeable deviations among the computed EV charging costs and the reference TOU profile, with differences up to 40%, depending on the considered month and on the time of charging during the day. It can be concluded that such model could be used to properly detect opportunities of energy savings, and to define dedicated EV price signals that could help to promote the optimal use of distributed energy resources.

The rising concerns about global warming and environmental pollution are increasingly pushing towards the replacement of road vehicles powered by Internal Combustion Engines (ICEs). Electric Vehicles (EVs) are generally considered the best candidates for this transition, however, existing power grids and EV management systems are not yet ready for a large penetration of EVs, and the current opinion of the scientific community is that further research must be done in this field. The so-called Vehicle-to-Grid (V2G) concept plays a relevant role in this scenario by providing the communication capabilities required by advanced control and Demand-Side Management (DSM) strategies. Following this research trend, in this paper the communication requirements for the DSM of EVs in urban environments are discussed, by focusing on the mobile communication among EVs and smart grids. A specific system architecture for the DSM of EVs moving inside urban areas is proposed and discussed in terms of the required data throughput. In addition, the use of a Low-Power Wide-Area Network (LPWAN) solution—the Long-Range Wide Area Network (LoRaWAN) technology—is proposed as a possible alternative to cellular-like solutions, by testing an experimental communication infrastructure in a real environment. The results show that the proposed LPWAN technology is capable to handle an adequate amount of information for the considered application, and that one LoRa base station is able to serve up to 438 EVs per cell, and 1408 EV charging points.

In this paper, we present a conceptual study on a Virtual Power Plant (VPP) architecture for the optimal management of Distributed Energy Resources (DERs) owned by prosumers participating in Demand-Side Management (DSM) programs. Compared to classical VPP architectures, which aim to aggregate several DERs dispersed throughout the electrical grid, in the proposed VPP architecture the supervised physical domain is limited to single users, i.e., to single Points of Delivery (PODs) of the distribution network. The VPP architecture is based on a service-oriented approach, where multiple agents cooperate to implement the optimal management of the prosumer’s assets, by also considering different forms of Demand Response (DR) requests. The considered DR schemes range from Price-Based DRs to Event-Based DRs, covering both the normal operating functions and the emergency control requests applied in modern distribution networks. With respect to centralized approaches, in this study the control perspective is moved from the system level to the single prosumer’s level, who is allowed to independently provide flexible power profiles through the aggregation of multiple DERs. A generalized optimization model, formulated as a Mixed-Integer Linear Programming (MILP) problem, is also introduced. Such a model is able to compute the optimal scheduling of a prosumer’s assets by considering both DR requests and end-users’ requirements in terms of comfort levels while minimizing the costs.

Se realizó una encuesta en algunos Departamentos de Radiología de Santiago, en la cual se consultó a funcionarios no administrativos de éstos (médicos, tecnólogos y auxiliares paramédicos) sobre los atributos que consideran más importantes en un jefe de Departamento de Diagnóstico por Imagen, mediante la elección de 5 de 12 aspectos enunciados. Se analizaron los resultados, relacionándolos con el tipo de cargo desempeñado, edad y lugar de trabajo. En este estudio se concluye que los atributos más valorados en un líder, por los diferentes estamentos dentro de un Servicio de Imaginología, difieren dependiendo de los cargos de los funcionarios, por lo cual, para lograr un trabajo en equipo armónico y fructífero, el líder debe conocer y poner en práctica estos aspectos de manera personalizada, en función de los intereses de sus subalternos.A survey was performed in a number of Units of Radiology in Santiago, Chile, aimed at obtaining from each medical staff (physicians, technologists and ancillary personnel) estimations on what the Head of a Department of Radiology ideal profile should be. Five out of twelve desirable characteristics proposed had to be selected for statistical purposes. Results were analyzed in correlation with positions, age and workplace of those polled. From our findings we conclude that most valuable leading traits vary from different work units, mainly depending on positions held within each medical Imaging Department. Thus, a proactive leadership demands awareness of such considerations in order to improve organizational practices and results.
