Abstract

Our objective was to develop alternatives to hormones for estrus synchronization in gilts. Gilts exhibit a pre-puberty period with high urinary estrone concentration during which boar exposure could induce the first ovulation. We searched for salivary biomarkers of this period. Urine and saliva were collected on six 140-day-old gilts until puberty for estrone assay, metabolome and steroidome analysis. We identified 23 metabolites and 28 steroids in saliva. The concentration of 8 of them showed significant variations at the pre-puberty period, they were candidate biomarkers. Saliva was collected from 30 gilts exposed to a boar and subjected to estrus detection from 150 to 175 days of age. Metabolome and steroidome analyses allowed the identification of 33 metabolites and 29 steroids in saliva. Their concentrations were not significantly different between receptive and non-receptive gilts. Thus, we could not identify salivary biomarkers of the period of receptivity to the boar effect.

Abstract

International audience; The objective of this study was to examine the effect of nutrition during the first 18 weeks of life on the physiological and transcriptional functionality of the hypothalamic (arcuate nucleus region), anterior pituitary and testes in Holstein-Friesian bull calves. Holstein-Friesian bull calves with a mean (+/- S.D.) age and bodyweight of 19 (+/- 8.2) days and 47.5 (+/- 5.3) kg, respectively, were assigned to either a HIGH (n=10) or LOW (n=10) plane of nutrition, to achieve an overall target growth rate of 1.2 or 0.5 kg/day, respectively. At 126 +/- 1.1 days of age, all calves were euthanised. Animal performance (weekly) and systemic concentrations of metabolic (monthly) and reproductive hormones (fortnightly) were assessed. Testicular histology, targeted gene and protein expression of the arcuate nucleus region, anterior pituitary and testes were also assessed using qPCR and immunohistochemistry, respectively. The expression of candidate genes in testicular tissue from post pubertal 19-month-old Holstein-Friesian bulls (n=10) was compared to that of the 18-week-old calves. Abstract

International audience; In order to follow the composition of parental broilers (Cobb500) during growth from hatching to adulthood (32 weeks of age), we evaluated kinetics of fattening, growth rate, reproduction parameters and body composition of the animals by using non-invasive tools such as medical imaging and blood sample analysing. The use of CT scanner allowed us to monitor the development of body composition on the same animals who are sleeping (fatness, bone, muscle, ovary and testis growth). These analyses were associated with biochemical blood analysis. Parental broilers fed the recommended diet by breeders. The analyses were performed with a CT scanner of bio-medical grade (Siemens As Definition ; 100K Volta and 120 mA / s). Five hundred images were performed every 0.6 mm, 0.45 Pitch and reconstruction filter Safire I26 was used to characterize fat tissues, skeletal and muscle. Image processing by thresholding was used to quantify and monitor the tissues from 4 weeks to 32 weeks of age. The acquired data were used to compare males and females. A significant difference in volume of adipose tissue was observed between male and female. From 20 weeks of age, shortly before the onset of lay, the females had 1.6 fold more adipose tissue than males (P < 0,001). In the female group, the increase in adipose tissue is associated to elevated plasma triglycerides levels (average 8 fold more in comparison to male at the beginning of the laying period. In addition, females, from 16 weeks of age, present a slow bone development in comparison to male (P < 0,001). The ratio bone/bodyweight is 30% lower in female compared to male chicken at 32 weeks of age. A section of the pectoral muscle volume was also analysed but no difference has been noted between both sexes. The correlation between the number of yellow follicles of hens and volume of ovarian adipose tissue determined in vivo was 0.80. Abstract

Estrus synchronization is important for optimal management of gilt reproduction in farms. Synthetic progestogens are used for this purpose, but there is growing demand for non-hormonal alternatives. Before puberty, gilts exhibit a “waiting period”, related to ovarian development and gonadotrophin secretions, during which external stimulation, such as boar exposure, could induce and synchronize first ovulation. Practical non-invasive tools for identification of this period in farms are lacking. During this period, urinary estrone levels are high, but urine sampling is difficult in group-housed females. Our aim was to search for steroidal biomarkers of this “waiting period” from immature to pubertal gilts through saliva monitoring. Six 144-to 147-day-old Large White gilts were subjected to ultrasound puberty diagnosis 3 times a week until first ovulation. Urine and saliva samples were collected at the same frequency for estrone assay and steroidome analysis respectively. Data were analyzed using the R software (nonparametric permutation test). Urinary estrone concentration significantly increased 2 weeks before puberty (detected at 182–192 days). Steroidome analysis quantified 28 steroids in saliva. Significant variations were detected within 2 weeks before puberty for dehydroepiandrosterone (decrease) and estradiol-17b (increase). These steroids could be biomarkers of the “waiting period”. These results confirm that non-invasive salivary sampling could allow the identification of the physiological status of the gilts and presumably the optimal time for application of the boar effect.

Abstract

Energy metabolism (in connection with growth, adipose tissue, etc.) is an important modulator of fertility. In order to improve performance in fertility, we have evaluated the feasibility to analyze the adipose tissue composition in 3 poultry species: chicken, turkey and guinea fowl by using a non-invasive method such as medical imaging (computed tomography, CTScan). In these 3 species, the adipose tissue composition varies between 10 and 15% depending the age. In particular, we carried out kinetics of fattening in the hen to associate it with the feeding, growth, and egg quality. In the experiment, meat-type chickens (Cobbgenetics) were bred with 2 diets conditions (ad libitum and restricted diet) from 3 weeks to 35 weeks of age. During this period, analyzes were performed with a biomedical scanner (Siemens, 100K Volta and 120mA / s). 500 images were taken every 0.6 mm. Image processing by thresholding was used to quantify adipose tissue. We calculated a correlation r² of 0.94 between the weight of the abdominal adipose tissue measured and the weight of the adipose tissue determined throughout the organism in vivo by computed tomography. The correlation between the chicken body weight and the volume of adipose tissue determined in vivo is r²= 0.82. The location and development of adipose tissue revealed significant differences as early as 6 weeks between ad libitum and restricted groups. In the ad libitum group, the increase in volume of adipose tissue is also associated with a high plasma triglyceride concentration and early onset of the laying period. Preliminary measurements of muscle, bone and liver volume have been measured, and allow opening new perspectives on the acquisition of new data that could be extrapolated with the feed and fertility parameters. In conclusion, this work has shown the importance of the CTscan to follow the evolution of the body composition in a non-invasive method, and to associate it with the growth parameters. Similar experiments are underway in turkey and guinea fowl., Le métabolisme énergétique (en lien avec la croissance, la masse du tissu adipeux,...) est un modulateur important de la fertilité. Afin d'améliorer les performances de fertilité, nous avons évalué la faisabilité d’analyser la composition en tissu adipeux de 3 espèces avicoles d’importance autour de la période de stimulation lumineuse : la poule, la dinde et la pintade en utilisant des outils non invasifs tels que l'imagerie médicale (tomodensitométrie, CTScan) qui permettent de localiser le tissu. Chez ces 3 espèces, lacomposition en tissu adipeux varie entre 10 et 15 % en fonction de l’âge. Plus particulièrement, nous avons réalisé une cinétique d'engraissement chez la poule afin de la mettre en lien avec l’alimentation, la croissance, et la qualité des oeufs. Pour cela, des poules de type chair (génétique Cobb) ont été nourries selon 2 régimes (ad libitum; régime restreint) de 3 semaines à 35 semaines d'âge. Sur cette période, des analyses ont été effectuées avec un scanner biomédical (Siemens Comme Définition; 100K Volta et 120 mA / s). 500 images ont été prises tous les 0,6 mm. Un traitement de l'image par seuillage a été utilisé pour quantifier le tissuadipeux. Nous avons calculé une corrélation r² de 0,94 entre le poids du tissu adipeux abdominal mesuré et le poids du tissu adipeux déterminé dans tout l’organisme in vivo par tomodensitométrie. La corrélation entre le poids corporel des poules et le volume de tissu adipeux déterminé in vivo est de r²=0,82. La localisation et le développement du tissu adipeux a révélé des différences significatives dès l’âge de 6 semaines entre les lots ad libitum et restreint. Dans le groupe ad libitum, l'augmentation de volume du tissu adipeux est également associée à une concentration élevée en triglycérides plasmatiques et un début de la période de ponte plus précoce. Des mesures préliminaires de masse musculaire, de volume osseux et du foie ont été réalisées, etpermettent d’ouvrir sur l’acquisition de nouvelles données qui pourraient être extrapolées avec les paramètres de fertilité ou de l’alimentation utilisée. En conclusion, ce travail montre l’importance du CTscan pour suivre l'évolution de la composition corporelle de façon non invasive, et de l’associer aux paramètres de croissance. Des expériences similaires sont en cours chez la dinde et la pintade.

Abstract

Effect of food restriction and fatty acid supplementation on metabolic parameters and plasma levels ofseveral adipokines in reproductive hens In livestock, reproductive hens are subjected to restricted diet to limit the decline of fertility associated with body mass changes. Thus, we have established an in vivo protocol of assessing the effect of different levels of restricted diet with or without polyunsaturated fatty acid supplementation (FAS) on metabolic parameters and plasma levels of adipokines (chemerin, visfatin, adiponectin) in reproductive hens. Abstract

Synchronizing oestrus in gilts is an important tool for pig producers to optimize the management of reproduction. Synthetic progestogens are used routinely in pig farms for this purpose, but there is a need for alternative non-hormonal breeding tools. Before puberty, gilts exhibit a “waiting period”, related to ovarian development and gonadotrophin secretions, during which an external stimulation, such as boar exposure, could induce and synchronize the first ovulation. Our aim was to characterize this “waiting period” and search for biomarkers of this period using non-invasive tools. During this period, an increase of urinary oestrone concentration has been observed, but urinary sampling is difficult in group-housed females. Salivary samples are non-invasive and easier to perform. Thus, the aim of this study was to evaluate steroid concentrations in the saliva of immature to pubertal gilts in order to search for biomarkers of the “waiting period”. Trans-abdominal ultrasonographies were performed over a period of 5 weeks on six 140-day-old Large White gilts until puberty detection. Urinary samples were collected for oestrone assay to detect the “waiting period”. Salivary samples were collected for steroidome analysis using gas chromatography coupled with tandem mass spectrometry to detect potential biomarkers of the “waiting period”. Urinary oestrone concentration increased 2 weeks before puberty. Steroidome analysis allowed the quantification of 28 steroids, 13 of them showing significant differences between weeks. Dehydroepiandrosterone concentrations decreased and estradiol-17 concentrations increased significantly just before the “waiting period”. These steroids could be biomarkers of this period. Present results confirm that non-invasive salivary samples could allow the identification of the physiological status of the gilts and the optimal time for application of the boar effect.

Abstract

Remerciements :Jean-Luc Touzé and Eric Venturi for help in performing ultrasonography; International audience; Current vaccines to Escherichia coli mastitis have shown some albeit limited efficacy. Their mode of action has not been documented, and immune responses protecting the mammary gland against E. coli are not completely understood. To improve our knowledge of mammary gland immune protection, cows immunized either intramuscularly or intramammarily with the E. coli P4 were submitted to a homologous mastitis challenge. A third group of mock-immunized cows serve as challenge controls. Local immunization modified favorably the course of infection, by improving bacterial clearance while limiting inflammation. Systemic clinical signs and reduction in milk secretion were also contained. This occurred with a modification of the cytokine profile, such as an increase in IFN-γ and a reduction in TNF-α concentrations in milk. Concentrations of IL-17A and IL-22 increased in milk at the onset of the inflammatory response and remained high up to the elimination of bacteria, but concentrations did not differ between groups. Accelerated bacteriological cure was not linked to an increase in the initial efficiency of phagocytosis in milk. Results support the idea that antibodies did not play a major role in the improvement, and that cell-mediated immunity is the key to understanding E. coli vaccine-induced protection of the mammary gland.

Abstract

International audience; Estimation of body composition in live cattle is difficult and usually performed using subjective approaches (visual and handling evaluation). Today, there is no reference methodological framework for the standardization of the determination of body fatness in cattle by ultrasonography. The objective of this study was to compare ultrasonographic measurements of fat and muscle subcutaneous tissue thickness at the buttock, lumbar, back and rib level with the conventional body condition scoring method. We performed ultrasonography on 160 cows from 5 different breeds (Normande, Holstein, Montbeliarde, Charolaise, Salers) by selecting populations of animals with the widest range of body condition scores possible (from 0.5 to 4.5 on a 0-5 scale). The study was carried out with four experimenters working with two different ultrasound devices and repeating the measurements two times. Low variation between assessors was observed at the lumbar and the rib sites. Measurements performed on dairy cows allow good correlation between body scores and ultrasound indicators at the buttock (fat, r=0.709), lumbar (fat and muscle, r=0.829) and rib (fat and muscle, r=0.707)) sites. Abstract

National audience; Depuis plus de 20 ans maintenant, la sélection de caractères positifs par les organismes de sélection porcine a permis l’amélioration des lignées hyperprolifiques. Cependant, il demeure une variabilité importante de la qualité des truies reproductrices (variabilité de l’âge à la puberté, variabilité des performances reproductives des truies (syndrome deuxième portée), forte occurrence d’anomalies héréditaires, …) et une grande hétérogénéité de la qualité des porcelets au sein d’une même portée dès la naissance (mortalité périnatale, croissance, …). Ce projet avait plusieurs objectifs tant au niveau technique que scientifique : - Au niveau de l’élevage porcin, le financement de ce projet devait permettre la mise en place de solutions techniques permettant la saisie informatisée des données zootechniques sur le terrain. - Au niveau biologique, le phénotypage fin du troupeau de truies Large White de race pure devait permettre d’identifier les paramètres mesurables les plus pertinents pour étudier l’aptitude des truies à la reproduction et aussi permettre d'identifier des familles d’animaux phéno-déviants ou touchés par des anomalies héréditaires.

Before puberty, gilts exhibit a “waiting period”, related to ovarian development and gonadotrophin secretions, during which urinary oestrone concentration increase. During this “waiting period”, an external stimulation, such as boar exposure, could induce the first ovulation. As non-invasive tools are required to increase knowledge about the “waiting period”, the aim of this work is to search for specific biomarkers of this period in urine. Trans-abdominal ultrasonographies were carried out for 5 weeks in six 140 days old Large White gilts until puberty detection (week -5 to week -1 before puberty). Urinary samples were collected for oestrone assay and metabolome analysis using 1H Nuclear Magnetic Resonance. Gilts were then slaughtered 7 days after puberty detection for puberty confirmation. Urinary oestrone concentration increases from week -2 to the day of puberty detection. Metabolome analysis allows the identification of 78 spectral bins, 8 of them showing significant differences between weeks. Metabolites whose concentration significantly increase or decrease during the “waiting period” (such as Trigonelline, N-acetyl-X…) could be interesting biomarkers of this period. These results confirm that non invasive urinary samples could make it possible to detect optimal time for application of boar effect. Potential urinary biomarkers of this period have been identified. This could contribute to decreasing the number of females mated while they are pre-pubertal.

Abstract

Effective methods for synchronizing estrus in gilts are crucial for strict implementation of batch management and optimum reproductive performances. Altrenogest treatments are used on a routine basis in pig farms, but there is growing demand for alternative non‐hormonal breeding tools. Before puberty, gilts exhibit a “waiting” period, related to ovarian development and gonadotrophin secretions, during which external stimulation, such as boar exposure, could induce the first ovulation. As noninvasive tools are required to increase knowledge about the waiting period, the aim of this work is to search for specific biomarkers of this period in saliva. Trans‐abdominal ultrasonography was carried out for 5 weeks in six 140‐day old Large White gilts until puberty detection (week ‐5 to week ‐1 before puberty). We collected saliva samples as well as urinary samples for estrone assay to detect the waiting period. Gilts were then slaughtered 7 days after puberty detection for puberty confirmation. The salivary metabolome was analyzed using 1H Nuclear Magnetic Resonance. Urinary estrone assays are in progress. The concentrations of some saliva metabolites significantly increased (propionate and butyrate for example) or decreased (pyruvate and fumarate for example) between week ‐5 and week ‐1 before puberty. The identification of other metabolites whose salivary concentration significantly varied is in progress. These results suggest that some salivary metabolites could be biomarkers of the pre‐pubertal stage in gilts. Salivary monitoring may help to investigate optimal time for application of boar effect and could contribute to decreasing the number of females mated while they are pre‐pubertal.

Abstract

Toxicology of Reproductive and Endocrine System; This introductory chapter offers a basic overview of male reproduction, specifically the anatomy and physiology of the male reproductive system. Comprehension of normal anatomy and physiology is necessary to (1) fully understand the severity of toxicant-induced damage to structures and/or functions of the male reproductive system, (2) design more powerful experiments that analyze potential male reproductive toxicants, and (3) identify numerous potential targets of toxicants in the male reproductive system. This chapter begins with an overview of the hypothalamic-pituitary-testicular axis and its interaction with additional components of the male reproductive system. This is then followed by detailed presentation of the composition of the testis and the interrelationships of the testicular cells, spermatogenesis, the excurrent ducts that carry spermatozoa out of the testis, the accessory sex glands that supply the seminal plasma of semen, the cellular communication within the testis, the reproductive tract's development, and normal male sexual behavior. The chapter's conclusion emphasizes the foundational importance of the male reproductive system to desgining, conducting, and interpreting reproductive toxicology research.

Abstract

National audience; Clinical mastitis is a germ-related teat and udder infection prevalent in dairy cows. The severity of the disease depends on the involved bacteria and the host reaction. Milk production but also teat and udder morphology can be irreversibly impacted. Our first goal was to study the incidence of clinical mastitis on milk production parameters in our dairy herd, with a special emphasis on the measure of electrical conductivity of individual udder quartiers. Our second goal was to use ultrasonography to illustrate changes caused by clinical mastitis on udder parenchyma and teats’ cistern, aiming at validating new indicators. Our results show a significant reduction of milk yield during and after a clinical mastitis episode, the loss of milk production reaching up to 80% in the most severe cases. Moreover, changes in milk composition during mastitis induce a significant increase in milk mean conductivity in the 4 days surrounding the first clinical signs and dramatic fluctuations of this parameter over at least a 28-day period surrounding detection of the pathology. Ultrasonography of udders and teats affected by mastitis reveals distention of the udder parenchyma and reduction of teat cistern volume during inflammation. This work shows that sonographic examination of bovine teats is a useful non-invasive method to be used in mastitis control during and after clinical episodes. Those morphometric indicators could help dairy cattle breeders to decide whether they should cull a cow after clinical mastitis.; La mammite clinique est une inflammation sévère du trayon et de la glande mammaire fréquente dans l’espèce bovine. En fonction de l’agent bactérien impliqué et de la réaction de l’hôte, les symptômes sont variables et peuvent, dans certains cas, provoquer une chute importante de la production laitière et altérer de façon irréversible la glande mammaire et le trayon des animaux atteints par cette pathologie. Notre premier objectif était de vérifier l’impact des mammites cliniques sur les paramètres de production laitière et les mesures de conductivité électrique du lait par quartier sur les vaches Holstein de l’élevage INRA de Nouzilly. Notre deuxième objectif était de mettre en évidence par échographie, les changements qualitatifs et quantitatifs observés au niveau de la glande mammaire et des trayons atteints de mammite clinique, dans le but de valider de nouveaux indicateurs morphométriques. Nos résultats montrent clairement une baisse de la production laitière pendant et après les épisodes de mammite clinique observés pendant l’étude, cette diminution pouvant atteindre 80% de la production antérieure à la mammite pour certaines vaches. En parallèle, l’analyse des données de conductivité par quartier révèle une augmentation significative de ce paramètre dans les 4 jours qui entourent l’apparition des signes cliniques et des fluctuations importantes de ce paramètre sur les quartiers touchés par une mammite pendant les 28 jours qui entourent les signes cliniques. Grâce aux examens échographiques, il est possible de mettre en évidence une inflammation au niveau de la glande mammaire et une réduction du volume de la citerne des trayons dans les quartiers atteints pendant une mammite. L’examen échographique réalisé 15 jours après la fin du traitement antibiobique permet d’évaluer la qualité de la récupération ou les séquelles d’une mammite sur la glande mammaire et le trayon atteints. Ces premiers résultats pourraient aboutir à la validation de nouveaux indicateurs morphométriques et servir d’aide à la réforme pour les éleveurs.

Temperature is projected toincrease from 1.8 to 4.0°C from 1980-1999 to 2090-2099. Hence, heat stress-related costs in pigproduction will be amplified in the future, both in temperate areas (summer heat waves) and tropicalareas (hot and humid environment). Meanwhile, world pig production is moving rapidly to tropicaland subtropical regions reaching now more than 50% of the total production. The world developmentof pig production has been achieved through improvement of animal genetics and management intemperate countries. However, selection performed in optimally controlled conditions has increasedthe sensitivity of animals to high ambient temperature. Heat stressed pigs reduce their feed intakewhich impair their growth or reproduction performances. Management solutions are available toattenuate the effect of heat stress on pigs, such as environmental solutions (water or feedingmanagement). However, these solutions are technically and economically difficult to implement. Thegenetic selection for improving environmental adaptation in pig production is the most promising longterm option. The PigHeaT project aims 1) at identifying QTLs for heat adaptation, by examining directresponses to find genes involved in metabolic ways, indirect responses to find genes affecting growthor robustness to environmental variations, 2) at better understanding the physiological mechanismsunderlying heat adaptation. It will provide tools for improving breeding strategies to face theupcoming global warming, and knowledge to better comprehend the physiological reactions ofanimals submitted to short and long term heat stress. The PigHeaT project is based on originalbiological resources and original experimental facilities. The studied population will be a backcrossbetween Large White pigs, productive but poorly thermotolerant breed, and Creole pigs, lowproductive but highly thermotolerant breed. The progeny issued from this backcross will express allpossible levels of thermal tolerance and production performances when submitted to heat stress,depending on the alleles received from their parents. High throughput phenotyping, metabolomics onall the progeny, and transcriptomics on a subset of extreme pigs selected on thermal toleranceresponse, will be applied. It will allow to refine the phenotypes and to achieve a high level of accuracyin QTL detection in the frame of the PigHeaT project. Additionally, the design will benefit from theunique combination of experimental facilities available at INRA: the first part of the project will relyon the backcross population raised in the experimental facilities located in the West Indies(Guadeloupe, tropical environment). The concomitant production of the same population in theexperimental facilities available in temperate France (Charente Maritime) will allow the detection ofgenetic by environment (GxE) effects for the QTL detected in Guadeloupe. Moreover, a heat wavephenomenon will be systematically simulated in the temperate environment at the end of the growingperiod. As a result, chromosomal regions robust or susceptible to GxE interactions will be identified,GxE being either tropical vs temperate, or tropical vs heat wave. Finally, an integrated analysis of the(fine) phenotypes and QTL will be proposed to better understand the metabolic pathways involved inheat stress responses. The respective use of the QTL and biological knowledge in further breedingstrategies will finally be considered.

Abstract

Hypo- and hyperthyroidism alter testicular functions in the young. Among T3 receptors, TRalpha1 is ubiquitous, and its previously described knockout leads to an increase in testis weight and sperm production. We tested, for the first time, the hypothesis that TRalpha1-dependent regulation of Sertoli cell (SC) proliferation was directly regulated by TRalpha1 present in these cells. Thus, after crossing with the AMH-Cre line, we generated and analyzed a new line that expressed a dominant-negative TRalpha1 isoform (TRalpha(AMI)) in SCs only. So-called TRalpha(AMI)-SC (TRalpha(AMI/+) Cre(+)) mice exhibited similar phenotypic features to the knockout line: heavier testicular weight and higher sperm reserve, in comparison with their adequate controls (TRalpha(AMI/+) Cre(-)). SC density increased significantly as a result of a higher proliferative index at ages Postnatal Day (P) 0 and P3. When explants of control testes were cultured (at age P3), a significant decrease in the proliferation of SCs was observed in response to an excess of T3. This response was not observed in the TRalpha(AMI)-SC and knockout lines. Finally, when TRalpha(AMI) is present in SCs, the phenotype observed is similar to that of the knockout line. This study demonstrates that T3 limits postnatal SC proliferation by activation of TRalpha1 present in these cells. Moreover, quantitative RT-PCR provided evidence that regulation of the Cdk4/JunD/c-myc pathway was involved in this negative control.
