The study investigated the antibacterial activity of crude extracts of C. edwardsii, Combretum krausii, and Maytenus nemorosa as well as their interactions with selected antibiotics against drug resistant bacterial strains. Using the rapid p-iodonitrotetrazolium chloride colorimetric assay, minimum inhibitory concentration values of plant extracts and antibiotics were determined. The interactions of plant extracts and antibiotics were studied using a checkerboard method. The MICs of the plant extracts and antibiotics were in the range of 0.037-6.25 and 0.001-2.5 mg/ml, respectively. The plant fractions tested in the present study displayed varying levels of antibacterial activity depending on the bacterial strains. Generally, Staphylococcus aureus was the most susceptible of the three strains of bacteria while the other two beta-lactamase producing Gram-negative bacteria were the most resistant. The hexane leaf extract of M. nemorosa was the most active (MIC = 37 µg/ml) against S. aureus. Ethyl acetate leaf extract of C. krausii was the most active against Klebsiella pneumoniae and ethyl acetate leaf extract of C. edwardsii was the most active against Escherichia coli. Synergistic interactions were detected in 13% of the combinations against E. coli, 27% of the combinations against K. pneumoniae and 80% of the combinations against S. aureus. The few synergistic interactions observed in the present study suggest that the crude extracts of the leaves of M. nemorosa, C. edwardsii, and C. krausii could be potential sources of broad spectrum antibiotic resistance modifying compounds. Background Due to the development of resistance to synthetic insecticides, adverse effects to human health, non-target organisms and the environment, there is an urgent need to develop new insecticides, which are effective, safe, biodegrable and target-specific. This study was undertaken to evaluate the adulticidal activity of 10 plants used traditionally as mosquito repellents in South Africa. Methods The dried plant materials were extracted with dichloromethane (DCM) and ethanol (EtOH). The extracts were evaluated for adulticidal activity against Anopheles arabiensis mosquitoes, a potent malaria vector in South Africa. Adult mortality was observed after 24 h of exposure. Results All the extracts showed adulticidal activity. The highest activity was observed in both DCM and EtOH extracts of Aloe ferox leaves with 98 and 86% mosquito mortality, respectively. The DCM extract of A. ferox leaves was then subjected to dose-dependent bioassay to determine the EC50 value. The extract exhibited the EC50 value of 4.92 mg/ml. Conclusion The results of the present study showed that the DCM extract of A. ferox leaves may have the potential to be used as insecticide against An. arabiensis. Kelpak® - a liquid seaweed concentrate made from the kelp Ecklonia maxima (Osbeck) Papenfuss - is used as a natural biostimulantto promote rooting and improve yield in crops. Plant-soil environmental conditions and the chemistry of water used for irrigation may affect the efficiency of Kelpak. The effect of pH (pH 4.5, 6.5 and 8.5) and water hardness (200 mg/L and 400 mg/L Ca2+) on the growth-promoting ability of Kelpak was assessed using the mungbean rooting bioassay and in a pot trial with Swiss chard. Kelpak promoted rooting in all the treatments in the mungbean bioassay with maximum rooting generally achieved with 20% Kelpak. With 20% Kelpak, the addition of 200 mg/L and 400 mg/L Ca2+ decreased rooting at pH 4.5, increased rooting at pH 6.5 and did not affect rooting at pH 8.5. A similar trend was observed in the pot trial with Swiss chard: leaf and root (fresh weight) and pigment content (chl a, chl b and carotenoids) improved with the addition of 200 mg/L Ca2+ + 5% Kelpak at pH 6.5 or pH 8.5, while Kelpak was able to partially mask the negative effect of 200 mg/L Ca2+ at pH 4.5. Smoke from burning vegetation is widely recognised as a germination cue for seed germination and recent reports suggest that smoke treatments can improve seedling vigour also. We investigated the effect of smoke-water on seedling vigour and changes of the global transcriptome in the early post-germination phase in maize. Application of smoke-water improved the germination characteristics and seedling vigour. The transcriptional response of embryos and emerging radicles 24 and 48 h after the onset of smoke treatment was investigated. The microarray study revealed a number of smoke-responsive genes amongst which stress- and abscisic acid (ABA)-related genes were over-represented. The global promoter analysis of the smoke-responsive genes revealed a tight correlation with the results obtained from Gene Ontology annotations. This concerted over-expression shows that smoke treatment induces stress and ABA-related responses in the early post-germination phase which leads to better adaptation to environmental stress factors occurring during germination, eventually resulting in greater seedling vigour. Respiratory ailments are major human killers, especially in developing countries. Tuberculosis (TB) is an infectious disease causing a threat to human healthcare. Many South African plants are used in the traditional treatment of TB and related symptoms, but there has not been a sufficient focus on evaluating their antimicrobial properties. The aim of this study was to evaluate the antimicrobial properties of plants used traditionally to treat TB and related symptoms against microorganisms (Klebsiella pneumoniae, Staphylococcus aureus, and Mycobacterium aurum A+) associated with respiratory infections using the microdilution assay. Ten plants were selected based on a survey of available literature of medicinal plants used in South Africa for the treatment of TB and related symptoms. The petroleum ether, dichloromethane, 80% ethanol, and water extracts of the selected plants were evaluated for antibacterial activity. Out of 68 extracts tested from different parts of the 10 plant species, 17 showed good antimicrobial activities against at least one or more of the microbial strains tested, with minimum inhibitory concentration ranging from 0.195 to 12.5 mg/mL. The good antimicrobial properties of Abrus precatorius, Terminalia phanerophlebia, Indigofera arrecta, and Pentanisia prunelloides authenticate their traditional use in the treatment of respiratory diseases. Thus, further pharmacological and phytochemical analysis is required.
