Abstract The scale of wildfire impacts to the built environment is growing and will likely continue under rising average global temperatures. We investigate whether and at what destruction threshold wildfires have influenced human mobility patterns by examining the migration effects of the most destructive wildfires in the contiguous U.S. between 1999 and 2020. We find that only the most extreme wildfires (258+ structures destroyed) influenced migration patterns. In contrast, the majority of wildfires examined were less destructive and did not cause significant changes to out- or in-migration. These findings suggest that, for the past two decades, the influence of wildfire on population mobility was rare and operated primarily through destruction of the built environment.

Summary:. Onycholysis secondary to nail bed scarring can be a challenging problem for hand surgeons, given the relatively few techniques available for nail bed lengthening. Here, we present the case of a patient with upper extremity onycholysis, secondary to porphyria cutanea tarda. We used a technique first described by Lemperle et al in 2003 to successfully lengthen the nail bed and improve the appearance of the nail plate. This involved full-thickness excision of a crescent-shaped wedge of the distal nail bed in all five right sided digits in a single operation. We found this technique to be simple and effective, meeting our patient’s goals after just one excision.

5. Evaluation of resazurin phenoxazine dye as a highly sensitive cell viability potency assay for natural killer cell‐derived extracellular vesicle‐based cancer biotherapeutics

Abstract Natural killer cell‐derived extracellular vesicles (NK‐EVs) are candidate biotherapeutics against various cancers. However, standardised potency assays are necessary for a reliable assessment of NK‐EVs' cytotoxicity. This study aims to thoroughly evaluate a highly sensitive resazurin phenoxazine‐based cell viability potency assay (measurement of the cellular redox metabolism) for quantifying the cytotoxicity of NK‐EVs against leukaemia K562 cells (suspension model) and breast cancer MDA‐MB‐231 cells (adherent model) in vitro. The assay was evaluated based on common analytical parameters setforth by regulatory guidelines, including specificity, selectivity,accuracy, precision, linearity, range and stability. Our results revealed that this resazurin‐based cell viability potency assay reliably and reproducibly measured a dose‐response of NK‐EVs’ cytotoxic activity against both cancer models. The assay showed precision with 5% and 20% variation for intra‐run and inter‐run variability. The assay signal showed specificity and selectivity of NK‐EVs against cancer target cells, as evidenced by the diminished viability of cancer cells following a 5‐hour treatment with NK‐EVs, without any detectable interference or background. The linearity analysis of target cancer cells revealed strong linearity for densities of 5000 K562 and 1000 MDA‐MB‐231 cells per test with a consistent range. Importantly, NK‐EVs’ dose‐response for cytotoxicity showed a strong correlation (|ρ| ∼ 0.8) with the levels of known cytotoxic factors associated with the NK‐EVs’ corona (FasL, GNLY, GzmB, PFN and IFN‐γ), thereby validating the accuracy of the assay. The assay also distinguished cytotoxicity changes in degraded NK‐EVs, indicating the ability of the assay to detect the potential loss of sample integrity. Compared to other commonly reported bioassays (i.e., flow cytometry, cell counting, lactate dehydrogenase release assay, DNA‐binding reporter assay and confluence assay), our results support this highly sensitive resazurin‐based viability potency assay as a high‐throughput and quantitative method for assessing NK‐EVs’ cytotoxicity against both suspension and adherent cancer models for evaluating NK‐EVs’ biotherapeutics.

9. Human Milk Supports Robust Intestinal Organoid Growth, Differentiation, and Homeostatic Cytokine Production

Background and Aims: Necrotizing enterocolitis is a severe gastrointestinal complication of prematurity. Using small intestinal organoids derived from fetal tissue of a gestational age similar to an extremely preterm infant, this study aims to assess the effect of diet on intestinal epithelial growth and differentiation to elucidate the role nutrition type plays in intestinal development and modifies the risk for necrotizing enterocolitis. Methods: Organoids were cultured for 5 days in growth media and 5 days in differentiation media supplemented 1:40 with 4 different diets: parental milk, donor human milk, standard formula, or extensively hydrolyzed formula. Images were captured daily and organoids were quantified. Organoids were preserved for RNA sequencing and immunofluorescence staining with Ki67, cleaved caspase 3, and chromogranin-A. Media was saved for cytokine/chemokine and growth factor analysis. Results: Human milk supplementation improved growth and differentiation of intestinal organoids generating larger organoids during the growth phase and organoids with longer and wider buds during differentiation compared to formula. Ki67 staining confirmed the proliferative nature of milk-supplemented organoids and chromogranin A staining proved that MM-supplemented organoids induced highest enteroendocrine differentiation. Human milk supplementation also upregulated genes involved in Wnt signaling and fatty acid metabolism pathways and promoted a homeostatic immune landscape, including via increased secretion of tumor necrosis factor-related apoptosis-inducing ligand among other cytokines. Conversely, organoids supplemented with formula had a downregulation of cell-cycle-promoting genes and a more inflammatory immune signature, including a reduced level of leukemia inhibitory factor. Conclusion: Our results demonstrate that parental milk, and to a lesser extent donor human milk, support robust intestinal epithelial proliferation, differentiation, and homeostatic cytokine production, suggesting a critical role for factors enriched in human milk in intestinal epithelial health.

Abstract Protein-based virus-like particles (P-VLPs) are commonly used to spatially organize antigens and enhance humoral immunity through multivalent antigen display. However, P-VLPs are thymus-dependent antigens that are themselves immunogenic and can induce B cell responses that may neutralize the platform. Here, we investigate thymus-independent DNA origami as an alternative material for multivalent antigen display using the receptor binding domain (RBD) of the SARS-CoV-2 spike protein, the primary target of neutralizing antibody responses. Sequential immunization of mice with DNA-based VLPs (DNA-VLPs) elicits protective neutralizing antibodies to SARS-CoV-2 in a manner that depends on the valency of the antigen displayed and on T cell help. Importantly, the immune sera do not contain boosted, class-switched antibodies against the DNA scaffold, in contrast to P-VLPs that elicit strong B cell memory against both the target antigen and the scaffold. Thus, DNA-VLPs enhance target antigen immunogenicity without generating scaffold-directed immunity and thereby offer an important alternative material for particulate vaccine design.

13. Coastal Sea Ice Concentration Derived from Marine Radar Images: A Case Study from Utqiaġvik, Alaska

We apply the Canny edge algorithm to imagery from the Utqiaġvik coastal sea ice radar system (CSIRS) to identify regions of open water and sea ice and quantify ice concentration. The radar-derived sea ice concentration (SIC) is compared against the (closest to the radar field of view) 25 km resolution NSIDC Climate Data Record (CDR) and the 1 km merged MODIS-AMSR2 sea ice concentrations within the ∼11 km field of view for the year 2022–2023, when improved image contrast was first implemented. The algorithm was first optimized using sea ice concentration from 14 different images and 10 ice analysts (140 analyses in total) covering a range of ice conditions with landfast ice, drifting ice, and open water. The algorithm is also validated quantitatively against high-resolution MODIS-Terra in the visible range. Results show a correlation coefficient and mean bias error between the optimized algorithm, the CDR and MODIS-AMSR2 daily SIC of 0.18 and 0.54, and ∼−1.0 and 0.7%, respectively, with an averaged inter-analyst error of ±3%. In general, the CDR captures the melt period correctly and overestimates the SIC during the winter and freeze-up period, while the merged MODIS-AMSR2 better captures the punctual break-out events in winter, including those during the freeze-up events (reduction in SIC). Remnant issues with the detection algorithm include the false detection of sea ice in the presence of fog or precipitation (up to 20%), quantified from the summer reconstruction with known open water conditions. The proposed technique allows for the derivation of the SIC from CSIRS data at spatial and temporal scales that coincide with those at which coastal communities members interact with sea ice. Moreover, by measuring the SIC in nearshore waters adjacent to the shoreline, we can quantify the effect of land contamination that detracts from the usefulness of satellite-derived SIC for coastal communities.

Marek’s disease (MD), caused by the Marek’s disease virus, is a lymphoproliferative disease in chickens that can be controlled by vaccination. However, the current vaccines can limit tumor growth and death but not virus replication and transmission. The present study aimed to evaluate host responses following intramuscular injection of an mRNA vaccine encoding gB and pp38 proteins of the MDV within the first 36 h. The vaccine was injected in low and high doses using prime and prime-boost strategies. The expression of type I and II interferons (IFNs), a panel of interferon-stimulated genes, and two key antiviral cytokines, IL-1β and IL-2, were measured in spleen and lungs after vaccination. The transcriptional analysis of the above genes showed significant increases in the expression of MDA5, Myd88, IFN-α, IFN-β, IFN-γ, IRF7, OAS, Mx1, and IL-2 in both the spleen and lungs within the first 36 h of immunization. Secondary immunization increased expression of all the above genes in the lungs. In contrast, only IFN-γ, MDA5, MyD88, Mx1, and OAS showed significant upregulation in the spleen after the secondary immunization. This study shows that two doses of the MDV mRNA vaccine encoding gB and pp38 antigens activate innate and adaptive responses and induce an antiviral state in chickens.

Summary: Extracellular vesicles (EVs) are entering the clinical arena as novel biologics for infectious diseases, potentially serving as the immunogenic components of next generation vaccines. However, relevant human assays to evaluate the immunogenicity of EVs carrying viral antigens are lacking, contributing to challenges in translating rodent studies to human clinical trials. Here, we engineered EVs to carry SARS-CoV-2 Spike to evaluate the immunogenicity of antigen-carrying EVs using human peripheral blood mononuclear cells (PBMCs). Delivery of Spike EVs to PBMCs resulted in specific immune cell activation as assessed through T cell activation marker expression. Further, Spike EVs were taken up largely by antigen-presenting cells (monocytes, dendritic cells and B cells). Taken together, this human PBMC-based system models physiologically relevant pathways of antigen delivery, uptake and presentation. In summary, the current study highlights the suitability of using human PBMCs for evaluating the immunogenicity of EVs engineered to carry antigens for infectious disease therapeutics.

Abstract Natural killer cell‐derived extracellular vesicles (NK‐EVs) have shown promising potential as biotherapeutics for cancer due to their unique attributes as cytotoxic nanovesicles against cancer cells and immune‐modulatory activity towards immune cells. However, a biomanufacturing workflow is needed to produce clinical‐grade NK‐EVs for pre‐clinical and clinical applications. This study established a novel biomanufacturing workflow using a closed‐loop hollow‐fibre bioreactor to continuously produce NK‐EVs from the clinically relevant NK92‐MI cell line under serum‐free, Xeno‐free and feeder‐free conditions following GMP‐compliant conditions. The NK92 cells grown in the bioreactor for three continuous production lots resulted in large quantities of both NK cell and NK‐EV biotherapeutics at the end of each production lot (over 109 viable cells and 1013 EVs), while retaining their cytotoxic payload (granzyme B and perforin), pro‐inflammatory cytokine (interferon‐gamma) content and cytotoxicity against the human leukemic cell line K562 with limited off‐target toxicity against healthy human fibroblast cells. This scalable biomanufacturing workflow has the potential to facilitate the clinical translation of adoptive NK cell‐based and NK‐EV‐based immunotherapies for cancer with GMP considerations.

The objective of this manuscript is to provide selective examples of the work of the Pan American Health Organization/World Health Organization (PAHO/WHO) Collaborating Centre for Research and Training in Parasite Epidemiology and Control which contribute to the WHO goal of eliminating neglected tropical diseases by 2030. This PAHO/WHO CC specifically aligns its activities with the Sustainable Development Goals and with the goals outlined in the WHO Road Map for Neglected Tropical Diseases 2021-2030. Its role is to contribute to advancing global action on NTDs, primarily through policy development and knowledge translation. Three important projects have recently been completed: 1. Finalizing the Monitoring and Evaluation Framework for the NTD Road Map (published May 2021; this PAHO/WHO CC was a member of the working group); 2. Developing new guidelines for the preventive chemotherapy of Taenia solium taeniasis (published September 2021; this PAHO/WHO CC was co-Chair; and 3. Formulating a policy brief on deworming for adolescent girls and women of reproductive age (published January 2022; this PAHO/WHO CC is co-lead). These projects are the result of the integration of expertise and experience from multiple partners, including from PAHO and WHO (where both organizations provided key leadership), this PAHO/WHO CC, government ministries, civil society organizations and universities, among others. In conclusion, this PAHO/WHO CC contributes timely guidance to country-led evidence-informed public health policy, to cost-effective program implementation and to the identification of priority research topics – all focused, ultimately, on eliminating NTD-attributable morbidity by 2030.

Canada, like other nations with colonizing histories and ongoing colonial practices marginalizing Indigenous peoples, is searching for pathways leading to reconciliation. The Truth and Reconciliation Commission called on the social work profession to engage in the decolonization of social work structures and processes, including how it educates entrants in the profession. This article reports on a project that used focus groups with social work faculty in two Canadian universities to explore perceptions about the ways change might be accomplished. From faculty viewpoints, four main themes are identified: coming to know about colonization, assimilation, and genocide; struggling with whose story it is to tell; questioning individual and collective responsibilities in decolonizing social work; and accepting truth first and trauma stories for reconciliation.

Abstract Currently available mRNA vaccines are extremely safe and effective to prevent severe SARS-CoV-2 infections. However, the emergence of variants of concerns (VOCs) has highlighted the importance of high population-based vaccine rates to effectively suppress viral transmission and breakthrough infections. While initially left out from vaccine efforts, children have become one of the most affected age groups and are key targets to stop community and household spread. Antibodies are central for vaccine-induced protection and emerging data points to the importance of additional Fc effector functions like opsononophagocytosis or cytotoxicity, particularly in the context of VOCs that escape neutralizing antibodies. Here, we observed delayed induction and reduced magnitude of vaccine-induced antibody titers in children 5-11 years receiving two doses of the age-recommended 10 μg dose of the Pfizer SARS-CoV-2 BNT162b2 vaccine compared to adolescents (12–15 years) or adults receiving the 30 μg dose. Conversely, children mounted equivalent or more robust neutralization and opsonophagocytic functions at peak immunogenicity, pointing to a qualitatively more robust humoral functional response in children. Moreover, broad cross-VOC responses were observed across children, with enhanced IgM and parallel IgG cross-reactivity to VOCs in children compared to adults. Collectively, these data argue that despite the lower magnitude of the BNT162b2-induced antibody response in children, vaccine-induced immunity in children target VOCs broadly and exhibit enhanced functionality that may contribute to the attenuation of disease.

Polo-like kinase 1 (PLK1) is a serine/threonine kinase required for mitosis and cytokinesis. As cancer cells are often hypersensitive to partial PLK1 inactivation, chemical inhibitors of PLK1 have been developed and tested in clinical trials. However, these small molecule inhibitors alone are not completely effective. PLK1 promotes numerous molecular and cellular events in the cell division cycle and it is unclear which of these events most crucially depend on PLK1 activity. We used a CRISPR-based genome-wide screening strategy to identify genes whose inactivation enhances cell proliferation defects upon partial chemical inhibition of PLK1. Genes identified encode proteins that are functionally linked to PLK1 in multiple ways, most notably factors that promote centromere and kinetochore function. Loss of the kinesin KIF18A or the outer kinetochore protein SKA1 in PLK1-compromised cells resulted in mitotic defects, activation of the spindle assembly checkpoint and nuclear reassembly defects. We also show that PLK1-dependent CENP-A loading at centromeres is extremely sensitive to partial PLK1 inhibition. Our results suggest that partial inhibition of PLK1 compromises the integrity and function of the centromere/kinetochore complex, rendering cells hypersensitive to different kinetochore perturbations. We propose that KIF18A is a promising target for combinatorial therapies with PLK1 inhibitors.

Wildfires are becoming more frequent in parts of the globe, but predicting where and when wildfires occur remains difficult. To predict wildfire extremes across the contiguous United States, we integrate a 30-yr wildfire record with meteorological and housing data in spatiotemporal Bayesian statistical models with spatially varying nonlinear effects. We compared different distributions for the number and sizes of large fires to generate a posterior predictive distribution based on finite sample maxima for extreme events (the largest fires over bounded spatiotemporal domains). A zero-inflated negative binomial model for fire counts and a lognormal model for burned areas provided the best performance. This model attains 99% interval coverage for the number of fires and 93% coverage for fire sizes over a six year withheld data set. Dryness and air temperature strongly predict extreme wildfire probabilities. Housing density has a hump-shaped relationship with fire occurrence, with more fires occurring at intermediate housing densities. Statistically, these drivers affect the chance of an extreme wildfire in two ways: by altering fire size distributions, and by altering fire frequency, which influences sampling from the tails of fire size distributions. We conclude that recent extremes should not be surprising, and that the contiguous United States may be on the verge of even larger wildfire extremes.

Abstract Background Systemic racism in Canadian healthcare may be observed through racially inequitable outcomes, particularly for Indigenous people. Nursing approaches intending to respond to racism often focus on culture without critically addressing the roots of racist inequity directly. In contrast, the critical race theory approach used in this study identifies whiteness as the underlying problem; a system of racial hierarchy that accords value to white people while it devalues everyone else. Methods This qualitative study seeks to add depth to the understanding of how whiteness gets performed by nursing faculty and poses antiracism education as a necessary tool in addressing the systemic racism within Canadian healthcare. The methodology of poststructural discourse analysis is used to explore the research question: how do white nursing faculty draw on common discourses to produce themselves following introductory antiracism education? Results Analysis of data reveals common patterns of innocent and superior white identity constructions including benevolence, neutrality, Knowing, and exceptionalism. While these patterns are established in other academic fields, the approaches and results of this study are not yet common in nursing literature. Conclusions The findings highlight the need for antiracism education at personal and policy levels beginning in nursing programs.

We apply the Canny edge algorithm to imagery from the Utqiaġvik coastal sea ice radar system (CSIRS) to identify regions of open water and sea ice and quantify ice concentration. The radar-derived sea ice concentration (SIC) is compared against the (closest to the radar field of view) 25 km resolution NSIDC Climate Data Record (CDR) and the 1 km merged MODIS-AMSR2 sea ice concentrations within the ∼11 km field of view for the year 2022–2023, when improved image contrast was first implemented. The algorithm was first optimized using sea ice concentration from 14 different images and 10 ice analysts (140 analyses in total) covering a range of ice conditions with landfast ice, drifting ice, and open water. The algorithm is also validated quantitatively against high-resolution MODIS-Terra in the visible range. Results show a correlation coefficient and mean bias error between the optimized algorithm, the CDR and MODIS-AMSR2 daily SIC of 0.18 and 0.54, and ∼−1.0 and 0.7%, respectively, with an averaged inter-analyst error of ±3%. In general, the CDR captures the melt period correctly and overestimates the SIC during the winter and freeze-up period, while the merged MODIS-AMSR2 better captures the punctual break-out events in winter, including those during the freeze-up events (reduction in SIC). Remnant issues with the detection algorithm include the false detection of sea ice in the presence of fog or precipitation (up to 20%), quantified from the summer reconstruction with known open water conditions. The proposed technique allows for the derivation of the SIC from CSIRS data at spatial and temporal scales that coincide with those at which coastal communities members interact with sea ice. Moreover, by measuring the SIC in nearshore waters adjacent to the shoreline, we can quantify the effect of land contamination that detracts from the usefulness of satellite-derived SIC for coastal communities. This paper argues that employee retention remains a key priority for many employers, even as they become less likely to offer stable employment. In the face of increased job insecurity, researchers have shown that workers have adopted personalized career progression strategies that emphasize inter-firm mobility. Such strategies heighten employer concerns about employee turnover. The paper argues that the industrial relations literature would gain from closer study of organizational practices that restrict employee mobility, i.e., 'worker capture strategies.' Such practices stand out from other mobility-reducing ones because they are unilateral and aim to reduce turnover without necessarily fostering a sense of commitment or loyalty among employees. The literature on precarious work has focused on the higher frequency of layoffs and downsizing and the shift away from the standard employment relationship over the past few decades. This paper argues that employee retention remains nonetheless a key priority for many employers, even as they become less likely to offer stable employment. In the face of increased job insecurity, researchers have shown that workers have adopted individualized career progression strategies in which inter-firm mobility plays a great role. Such strategies heighten employer concerns about employee turnover. This paper reviews the state of the literature on organizational practices that address employer concerns about employee retention. It then argues that this literature would gain from closer study of organizational practices that restrict employee mobility, which it labels 'worker capture strategies.' They include non-compete and non-solicitation clauses in employment contracts, no-poaching agreements between firms, and TRAP clauses. Such practices stand out from others because of their one-sided nature and their use as a turnover reduction strategy that does not necessarily drive a sense of commitment or loyalty among employees. Global climate change and associated environmental extremes present a pressing need to understand and predict social–environmental impacts while identifying opportunities for mitigation and adaptation. In support of informing a more resilient future, emerging data analytics technologies can leverage the growing availability of Earth observations from diverse data sources ranging from satellites to sensors to social media. Yet, there remains a need to transition from research for knowledge gain to sustained operational deployment. In this paper, we present a research-to-commercialization (R2C) model and conduct a case study using it to address the wicked wildfire problem through an industry–university partnership. We systematically evaluated 39 different user stories across eight user personas and identified information gaps in public perception and dynamic risk. We discuss utility and challenges in deploying such a model as well as the relevance of the findings from this use case. We find that research-to-commercialization is non-trivial and that academic–industry partnerships can facilitate this process provided there is a clear delineation of (i) intellectual property rights; (ii) technical deliverables that help overcome cultural differences in working styles and reward systems; and (iii) a method to both satisfy open science and protect proprietary information and strategy. The R2C model presented provides a basis for directing solutions-oriented science in support of value-added analytics that can inform a more resilient future.

Researchers in Earth and environmental science can extract incredible value from high- resolution (sub-meter, sub-hourly or hyper-spectral) remote sensing data, but these data can be difficult to use. Correct, appropriate and competent use of such data requires skills from remote sensing and the data sciences that are rarely taught together. In practice, many researchers teach themselves how to use high-resolution remote sensing data with ad hoc trial and error processes, often resulting in wasted effort and resources. In order to implement a consistent strategy, we outline ten rules with examples from Earth and environmental science to help academic researchers and professionals in industry work more effectively and competently with high-resolution data.

Inherited GPI deficiencies (IGDs) are a subset of congenital disorders of glycosylation that are increasingly recognized as a result of advances in whole-exome sequencing (WES) and whole-genome sequencing (WGS). IGDs cause a series of overlapping phenotypes consisting of seizures, dysmorphic features, multiple congenital malformations, and severe intellectual disability. We present a study of six individuals from three unrelated families in which WES or WGS identified bi-allelic phosphatidylinositol glycan class S (PIGS) biosynthesis mutations. Phenotypes included severe global developmental delay, seizures (partly responding to pyridoxine), hypotonia, weakness, ataxia, and dysmorphic facial features. Two of them had compound-heterozygous variants c.108G>A (p.Trp36∗) and c.101T>C (p.Leu34Pro), and two siblings of another family were homozygous for a deletion and insertion leading to p.Thr439_Lys451delinsArgLeuLeu. The third family had two fetuses with multiple joint contractures consistent with fetal akinesia. They were compound heterozygous for c.923A>G (p.Glu308Gly) and c.468+1G>C, a splicing mutation. Flow-cytometry analyses demonstrated that the individuals with PIGS mutations show a GPI-AP deficiency profile. Expression of the p.Trp36∗ variant in PIGS-deficient HEK293 cells revealed only partial restoration of cell-surface GPI-APs. In terms of both biochemistry and phenotype, loss of function of PIGS shares features with PIGT deficiency and other IGDs. This study contributes to the understanding of the GPI-AP biosynthesis pathway by describing the consequences of PIGS disruption in humans and extending the family of IGDs.
