Mycobacterium tuberculosis (Mtb) endures a combination of metal scarcity and toxicity throughout the human infection cycle, contributing to complex clinical manifestations. Pathogens counteract this paradoxical dysmetallostasis by producing specialized metal trafficking systems. Capture of extracellular metal by siderophores is a widely accepted mode of iron acquisition, and Mtb iron-chelating siderophores, mycobactin, have been known since 1965. Currently, it is not known whether Mtb produces zinc scavenging molecules. Here, we characterize low-molecular-weight zinc-binding com- pounds secreted and imported by Mtb for zinc acquisition. These molecules, termed kupyaphores, are produced by a 10.8 kbp bio- synthetic cluster and consists of a dipeptide core of ornithine and phenylalaninol, where amino groups are acylated with isonitrile- containing fatty acyl chains. Kupyaphores are stringently regulated and support Mtb survival under both nutritional deprivation and intoxication conditions. A kupyaphore-deficient Mtb strain is unable to mobilize sufficient zinc and shows reduced fitness upon infection. We observed early induction of kupyaphores in Mtb-infected mice lungs after infection, and these metabolites dis- appeared after 2 wk. Furthermore, we identify an Mtb-encoded isonitrile hydratase, which can possibly mediate intracellular zinc release through covalent modification of the isonitrile group of kupyaphores. Mtb clinical strains also produce kupyaphores during early passages. Our study thus uncovers a previously unknown zinc acquisition strategy of Mtb that could modulate host–pathogen interactions and disease outcome. [ABSTRACT FROM AUTHOR]

Arsenic present in water bodies causes devastating effects on aquatic organisms and indirectly poses a hazardous threat to human existence. There is an urgent need to develop potential and convincing technologies to troubleshoot this problem. In the present study, an adsorbent has been prepared using the waste red mud from hazardous aluminium industry and doping it with calcium-alginate beads (ARMCB) for the effective removal of As(III) from wastewater. The concentration of As(III) was reduced from 0.101 mg/L to 0.008 mg/L after adsorption which effectively met the economic and environmental conditions imposed by WHO (>0.01 mg/L). Further, the statistical Response Surface Methodology (RSM) is adopted to analyze the combined effects of four operational parameters namely: pH, sorbent dosage, contact time and initial concentration on the adsorption of As(III) from the synthetic contaminated water samples. A high correlation coefficient (R2) value of 0.9672 projected by ANOVA confirmed the satisfactory regression of the developed model. The maximum adsorption capacity is found to be 1.807 mg/g at optimum operating conditions. The surface characterization of the adsorbent before and after adsorption by SEM, EDX, XRD, and FTIR confirms the potentiality of the adsorbent towards As(III) ions. Thermodynamic, adsorption isotherms and kinetic analysis respectively projected the endothermic Langmuir model adsorption of As(III) and the pseudo-second-order rate kinetics of the sorption mechanism. The present work proposes the synthesis of robust biochar from Gracilaria Rhodophyta red weeds for sequential removal of Al(III) and fluoride from wastewater. The sorption experiments have been modeled by preliminary optimization of operational parameters using 2 4 factorial statistical modeling. The model has estimated an optimum sequential synergetic removal of 44.5 mg/g of Al(III) and 2.1 mg/g of fluoride onto the biochar. FESEM, BET, XRD, EDX, and FTIR established the potentiality of biochar toward synergetic sorption of Al(III) and fluoride. The thermodynamic analysis projected that the adsorption is physisorption in nature. The adsorption of Al(III) and fluoride follows the Langmuir and Freundlich isotherm models, respectively, and the kinetic analysis established the pseudo-second-order deposition of Al(III) and fluoride ions. The synthesized adsorbent is regenerative enough and could achieve synergetic removal of Al(III) and fluoride ions from industrial- and groundwater-contaminated water bodies. PRACTITIONER POINTS: Biochar from seaweeds is explored in the sequential removal of Al(III) and F - ions. Statistical model is developed for % adsorption and tested for reliability by ANOVA. GRBC sorbed 44.5 and 2.1 mg/g of Al(III) and F - ions, respectively, at optimum levels. FESEM, EDX, XRD, and FTIR characterization confirm the potentiality of the GRBC.
